The essence of myelodysplastic syndrome (MDS) is a malignant clonal bone marrow myeloid neoplasm. Both
basic researches and clinical studies should firmly grasp this essence of MDS and should not be interferenced by
confounding factors. Comprehensive diagnosis can improve the accuracy of MDS diagnosis by multiple indicators that reflect
the malignant nature, such as cell dysplasia, function abnormalities, cytogenetic changes and gene mutations. The therapy of
MDS should also eradicate the MDS clone, change the disease progression, stimulating normal hematopoiesis, prolong the
survival and improve the quality of life.
4.Isolation,incubation and identification of parenchymal neural stem cells in adult mouse spinal cord

Objective To isolate and identify the adult neural stem cells from the parenchyma of spinal cord in adult mouse.Methods The parenchymal spinal cord from adult mouse was dissected and dissociated by mechanical trituration.The tissue suspension was cultured in serum-free DMEM/F12 medium supplemented with EGF and B27.The cell colonies generated from a single cell were screened by limited dilution and incubated with BrdU.The cell colonies were transferred into medium with serum to induce differentiation.The cells were identified with antibodies to Nestin,BrdU,MAP2 and GFAP by immunofluorescence staining.Results The cells were cultured for seven days to generate proliferative neurospheres.The majority of cells in these neurospheres expressed Nestin and were differentiated into MAP2-positive cells and GFAP-positive cells in medium containing with fetal bovine serum.Conclusion A significant number of neural stem cells are present in the parenchymal adult mouse spinal cord and can proliferate and also give rise to neurons and glia in vitro.
5.Advances of studies on T follicular helper cells.

T follicular helper cells (TFH cells) provides a helper function to B cells. It is one of the most important subsets of effector T cells in lymphoid tissues. The features of TFH cells include chemokine receptor expression (CXCR5), location/migration (B cell follicles), and function (B cell help). TFH cells produce a "helper" cytokine-IL-21, which can stimulate B cells to differentiate into Ab-forming cells through IL-21R. Dysregulation of TFH cell function likely contributes to the pathogenesis of immune-related diseases. In this article, the discovery of CXCR5 and TFH cells, origin of TFH cells, cytokines related with TFH cell function, migration and location of TFH effector cells and memory cells as well as dysregulation of TFH cell function and related diseases were reviewed.

6.A new C21 steroidal saponins from Periplocae Cortex.

To study the chemical constituents of Periplocae Cortex, the separation and purification of 70% alcohol extract were carried out by column chromatographies on AB-8 macroporous resin, silica gel and preparative HPLC. The structure of the compounds were identified by NMR and TOF-MS. A new compound was isolated and identified as 21-O-methyl-Δ5-pregnene-3β, 14β, 17β, 21-tetraol-20-one-3-O-β-D-oleandropyranosyl(1-->4)-β-D-cymaropyranosyl-(1-->4)-β-D-cymaropyranosyl (1), named as periplocoside P.

8.Expression and subcellular localization of the ORF4 gene of barley yellow dwarf virus GAV strain in baculovirus-insect cell expression system.

According to published nucleotide sequences, ORF4 gene of barley yellow dwarf virus GAV (BYDV-GAV) was synthesized by reverse transcription-polymerase chain reaction (RT-PCR). The BYDV-GAV ORF4 gene was expressed in baculovirus -insect cell expression system efficiently, and western bolt analysis confirmed its expression product. Confocal laser scanning microscopy showed that GFP: ORF4 fusion protein was associated with the nuclear envelope of insect cells. By expressing the N- and C-terminal regions of ORF4-encoding product (P4) in insect cells combined with structure prediction, it was found that the N-terminal region of P4 containing four a-helices is required for targeting P4 to the nuclear envelope. These results provide a base for biological function of ORF4 gene during systemic infection of BYDV-GAV in host plants further.

9.Study on the optimal harvesting time of Rumex gmelini by analyzing the contents of principal components.

OBJECTIVETo establish a method used for optimization of harvesting time and determine the best time for harvesting Rumex gmelini.
METHODAn HPLC method was applied to determinate the contents of seven active constituents(resveratrol, polydatin, chrysophanol 1-glucoside, nepodin, emodin, chrysophanol and physcion)of R. gmelini at different development stage. The result was analyzed by principal component analysis.
RESULTThe accumulation of active constituents showed a regular pattern.
CONCLUSIONThe best harvesting time of R. gmelini is early July.

10.Study on correlation of soil nutrients and content of active constituents in root of Rumex gmelini.

OBJECTIVETo provide scientific basis for the selection of agrotype and property fertilization for Rumex gmelini cultivated in compliance with good agricultural practice (GAP).
METHODHPLC method was applied to determinate the content of seven active constituents (resveratrol, polydatin, chrysophanol 1-glucoside, nepodin, emodin, chrysophanol and physcion) of annual R. gmelini. And the correlation between soil nutrients and content of active constituents in the root of R. gmelini were analyzed by stepwise regression analysis.
RESULTSeven regression equation were established. The statistic significance was found in three of them.
CONCLUSIONThe soil with high total K level is not suitable for R. gmelini cultivation. But the higher available N, available P, available K level of soil is suitable.
