1.Appilication of Femoral Neck Anteversion in Developmental Dislocation of the Hip by Digital Anatomy

Objective To discuss the femoral neck anteversion(FNA) of developmental dislocation of the hip(DDH) and guide operation with visual and digitalized picture in the dimensional(3D) CT.Methods Ninety children with unilateral DDH were selected,and they were analyzed using 3D CT.Children whose FNA exceed 45 degree received the subtrochanter osteotomy with images from different direction,FNA of hip was measured respectively before and after operation and was measured in normal and abnormal hips respectively,FNA of hip received respectively statistical treatment.Ninety patients (90 hips) were followed up ranging from 3 months to 2 years with the mean of 13 months.Results In the group younger than 18 months,the FNA whose in normal hips was(19.40?3.512)degree,the FNA while(68.45?12.272)degree in dysplasia hips respectively,the FNA measured after operation was (20.45?2.940) degree;in the group elder than 6 years,there were significantly statistical differences,the FNA in normal hips was(19.44?3.561)degree,in dysplasia hips respectively was(73.49?12.678)degree,while the FNA measured after operation was(18.28?1.931)degree.Clinical assessment was performed according to Mckay′s classification.The results showed that the overall excellent or good rate was 95.6%.Conclusions 3D CT method is a new accurate and convenient and reduplicative method for measuring FNA.It will be more helpful for related operations when 3D images are considered.

5.Isolation,incubation and identification of parenchymal neural stem cells in adult mouse spinal cord

Objective To isolate and identify the adult neural stem cells from the parenchyma of spinal cord in adult mouse.Methods The parenchymal spinal cord from adult mouse was dissected and dissociated by mechanical trituration.The tissue suspension was cultured in serum-free DMEM/F12 medium supplemented with EGF and B27.The cell colonies generated from a single cell were screened by limited dilution and incubated with BrdU.The cell colonies were transferred into medium with serum to induce differentiation.The cells were identified with antibodies to Nestin,BrdU,MAP2 and GFAP by immunofluorescence staining.Results The cells were cultured for seven days to generate proliferative neurospheres.The majority of cells in these neurospheres expressed Nestin and were differentiated into MAP2-positive cells and GFAP-positive cells in medium containing with fetal bovine serum.Conclusion A significant number of neural stem cells are present in the parenchymal adult mouse spinal cord and can proliferate and also give rise to neurons and glia in vitro.
6.Epstein-Barr Virus Serology in Early Detection and Screening of Nasopharyngeal Carcinoma

Nasopharyngeal carcinoma (NPC) is a common canceramong Chinese, especially Southern Chinese; except for a few other ethnic groups with moderate incidence, it is otherwise a rare cancer in the world.NPC has a male dominance of about 3:1 and mainly afflicts people in midlife. There is now compelling evidences to suggest that Epstein-Barr virus (EBV), a category Ⅰ human tumor virus defined by UICC (International Agency for Research on Cancer) in 1997, is a causal agent of NPC and is most likely to be involved in the multi-step and multi-factorial development of NPC. In this article, the role of EBV in pathogenesis of NPC is reviewed briefly, and principle applications of EBV antibodies and EBV DNA as markers of NPC are outlined. Based on current knowledge of EBV antibody responses by NPC and taking available testing technologies into account,serologic screening strategy to facilitate efficient early detection of NPC is formulated.
7.Study of sequence variations of Epstein-Barr virus LMP1 gene in nasopharyngeal carcinoma.

OBJECTIVETo detect the sequence variations frequently found within the N- and C-terminal regions of Epstein-Barr virus (EBV) LMP1 gene in nasopharyngeal carcinoma (NPC) and to study the underlying mechanisms.
METHODSFresh tumor tissues were sampled from 63 patients with untreated NPC encountered in Affiliated Tumor Hospital of Sun Yat-sen University, Guangzhou. The N-terminal region of EBV LMP1 gene was amplified with nested polymerase chain reaction (PCR), followed by XhoI enzyme digestion. Nested PCR was also employed to detect the 30 base pairs deletion within the C-terminal region. Four-colored fluorescence terminator sequencing method was applied for bi-directional solid-phase sequencing of the 8 representative PCR products in 4 cases of NPC. The DNA sequence within the N- and C-terminal regions of LMP1 gene was then analyzed.
RESULTSThere were 4 patterns of sequence variations, namely, wt-XhoI/wt-LMP1 (4 cases, 6.3%), wt-XhoI and XhoI-loss/del-LMP1 (4 cases, 6.3%), wt-XhoI/del-LMP1 (5 cases, 7.9%) and XhoI-loss/del-LMP1 (50 cases, 79.5%), detected in the 63 studied cases. Sequence analysis showed that the EBV LMP1 gene had underwent non-synonymous and synonymous substitutions, as compared with the prototype of B95-8 cells. The ratio of non-synonymous to synonymous substitutions was 2.25.
CONCLUSIONSXhoI-loss/del-LMP1 is the predominant sequence variation pattern of EBV LMP1 gene in NPC from Guangzhou. The XhoI-loss variation seems to develop on top of del-LMP1. When compared with the EBV LMP1 gene in peripheral blood B-lymphocytes of virus carriers and in preinvasive epithelial lesions (reported previously), it is likely that the sequence variation patterns of LMP1 gene may represent 4 different phases of intrahost evolution of EBV during nasopharyngeal carcinogenesis.

8.The application of forward and reversal flow axial island flap based on the superficial temporal artery in the orbital skin defects.

OBJECTIVETo investigate the application of forward and reversal flow axial island flap in the orbital skin defects.
METHODSAccording to the distribution of superficial temporal artery as well as the anastomoses with the branches of supraorbital artery and supratrochlear artery, etc, the frontal, scalp, preauricular and postauricular island flaps were designed. The flaps were displaced through subcutaneous tunnel to cover all kinds of orbital skin defects.
RESULTS13 cases of forward flow flap and 9 of reversal flow flap out of 22 cases in all were observed. One postauricular reversal flow axial island flap showed the obstruction of venous refluence in early postoperative stage, however, after active treatment, mere the distal epidermal necrosis was revealed. There were six re-operations on the postoperative flap hypertrophy. The others were in good shape and functional state.
CONCLUSIONSThe superficial temporal artery is one of most vital blood supply in the upper face. It has wide anastomoses with the supraorbital artery and supratrochlear artery. As the colors and the nature of the frontal, preauricular and post auricular skin are close to orbital skin, the application of forward and reversal flow axial island flap based on the superficial temporal artery can be used in the reconstruction of all kinds of orbital skin defects.

9.Observation on therapeutic effect of tic disorders treated with local acupuncture.

OBJECTIVETo observe the clinical efficacy on tic disorders (TD) treated with local acupuncture.
METHODSThe entire data of 196 cases, of which, 114 cases in acupuncture group were treated mainly with local acupuncture and 82 cases in western medication group were taken Tiapride tablets orally. YGTSS method was adopted for scoring and assessing the clinical efficacy.
RESULTSThe apparent improvement was achieved in YGTSS motor tic score in two groups. The markedly effective rates were 90.4% and 84.2% in acupuncture group and western medication group, respectively (P < 0.05), without significant difference in statistics. The effective rates of transient tic disorder (TTD) in acupuncture group and western medication group were 100.0% and 83.3%, respectively (P < 0.05), which indicated that the efficacy of acupuncture on TTD was advantageous to medical therapy. The effective rates of three kinds of TD in acupuncture group were 100.0%, 88.2% and 84.2% separately, of which, the efficacy of acupuncture on TTD was advantageous as compared with that on chronic tic disorder (CTD) and Toureett's syndrome (TS) (P < 0.05).
CONCLUSIONThe overall efficacy on TD is similar between acupuncture and medical therapy, but the clinical efficacy of acupuncture on TTD is superior to medical therapy. Meanwhile, the efficacy of acupuncture on TTD is advantageous to that on CTD and TS. Hence, the intervention at early time can achieve satisfactory efficacy and probably prevent from CTD or TS.

10.Apoptosis of human gastric cancer cells induced by omega-3 polyunsaturated fatty acids.

OBJECTIVETo investigate the effect of omega-3 polyunsaturated fatty acids(omega-3PUFAs) on the apoptosis of human gastric cancer cell line SGC-7901 and to explore the potential mechanisms.
METHODSCells were treated with eicosapentaenoic acid (20:5 omega-3,EPA) or docosahexaenoic acid (22:6 omega-3, DHA) at concentrations of 10, 20 and 40 microg/ml. Cell growth and apoptosis were analyzed with MTT assay, cell morphology, DNA electrophoresis and flow cytometry. Mitochondrial membrane potential ( triangle right psi mt) was measured by fluorescent probe rhodamine 123. The distribution of cytochrome C in mitochondria and cytosol was determined by enzyme-linked immunoadsorbent assay. The composition of mitochondrial membrane phospholipid(MMP)was examined by gas chromatography.
RESULTSBoth EPA and DHA markedly inhibited the SGC-7901 cell growth and induced apoptosis in a time- and dose-dependent manner. After incubation of the cells with 40 microg/ml EPA or DHA for 24 hours, the level of Deltapsimt siginificantly decreased (P<0.001), and cytochrome C largely released into cytosol from mitochondria. The proportions of EPA and DHA in MMP rapidly elevated while that of arachidonic acid sharply decreased.
CONCLUSIONSomega-3PUFAs inhibit the growth of gastric cancer cells through promoting apoptosis. Compositional and functional alterations in mitochondrial membrane may be an important initiator of apoptosis induced by omega-3PUFAs.
