1.Investigation on radon concentration and dose estimation at some areas in Zhejiang Province

Objective To investigate the concentrations of indoor radon (222Rn) and its daughter products as well as indoor thoron (220Rn) in selected houses in Yuhang district and Sanmen county,Zhejiang province,and estimate their annual effective doses to the population.Methods Solid state nuclear track detectors were used in selected dwellings in Yuhang district and Sanmen county,and the detectors were placed in bedrooms or living rooms.Without changing the ventilation habits of residents,These detectors were continuously placed from March to September in 2009.Results Indoor 222 Rn and 220Rn concentrations in low-rise buildings were the highest among all types of houses.The indoor concentration of 222 Rn had no relation with the building age (F = 0.53,P > 0.05),but that of 220 Rn was dependent on the building age (F = 3.56,P < 0.05).Moreover,the investigation demonstrated indoor 220 Rn concentrations in houses with no decoration were higher than in the houses decorated (t = 2.33,P <0.05).The average indoor concentrations of 222Rn and 220Rn in Yuhang district were 32.5 Bq/m3 and 314.3 Bq/m3,respectively,and the annual effective doses were 0.88 mSy and 0.42 mSv respectively.The average indoor concentrations of 222Rn and 220Rn in Sanmen county were 26.8 Bq/m3 and 399.5 Bq/m3,and the annual effective doses were 0.72 mSy and 0.53 mSv respectively.Conclusion The concentrations of indoor 222 Rn in some areas of Zhejiang province are at natural background level,and the concentrations of indoor 220Rn in rural areas are relatively higher.The total annual effective dose from 220Rn and its progeny was larger than that from 222Rn and its progeny by 50 percents.
2. Preliminary study on the period of CDT therapy for limb lymphedema

Objective:
To explore the appropriate treatment cycle of complex decongestive Therapy (CDT) for extremity lymphedema, which is the summary of the relationship between treatment time and curative effect.
Methods:
From January to December 2015, 203 patients with limb lymphedema were divided into four stages according to the degree of edema and the degree of fibrosis, that is, four groups. All patients were given CDT treatment for a course of treatment (4 weeks). During the treatment period, the multi frequency bioelectrical resistance body composition analyzer was used to monitor the change of tissue moisture and the tape diameter was used to measure the circumference of the affected limb. SPSS software is used to analyze the data.

3. Pathological study on mast cells and their released protease and transforming growth factor-β1 in lymphedema skin tissue

Objective:
To explore the relationship between protease-chymase secreted by mast cells, and activated transforming growth factor-β1(TGF-β1), in skin with secondary lymphedema(SLE)in lower extremity, so as toidentify the key factors in fibrosis of lymphedema.
Methods:
In this study, the affected limb skin of 7 SLE patients was includedas the experimental group, and normal skin tissue of the lower limb of 7 volunteers was used as controls. The skin samples were assayed by Masson staining, and the expressions of chymase and TGF-β1 were assayed by immunohistochemistry, immunofluorescent staining and enzyme-linked immunosorbent assay.
Results:
There was obvious fibrosis in the skin of lower extremity in patients with lymphedema. The number of MCs andthe expressions of chymase, latency-associated peptide TGF-β1 (LAP TGF-β1) and TGF-β1 were all significantly increased in fibrotic skin in lymphedema, compared with those in normal skin. At the same time, the chymase-containing mast cells accumulated in the lymphatic vessels, with higher expression of TGF-β1.
Conclusions
The expression of chymase and TGF-β1 was significantly increased in the fibrotic skin insecondary lower extremity lymphedema. The increased expression of chymase in the skin may activate more TGF-β1 expression, and the increased TGF-β1 may promote skin fibrosis in SLE.
4. The anti-photoaging effects of human umbilical cord mesenchymal stem cells derived extracellular vesicles on dermal fibroblasts

Objective:
To investigate the anti-photoaging effects of human umbilical cord mesenchymal stem cells derived extracellular vesicles (hucMSC-EVs) on dermal fibroblasts.
Methods:
Human fibroblasts were pretreated with hucMSC of different concentrations for 24 hours before ultraviolet B (UVB) irradiation. Immediately post irradiation, the intracellular reactive oxygen species (ROS) were analyzed using reactive oxygen detection kit. After 72 hours irradiation, cell proliferation was assessed using CCK-8 assay, and the percentage of senescent cells was evaluated by β-galactosidase (SA-β-gal) staining.
Results:
The UVB irradiation increased intracellular ROS level, decreased the cell proliferation from 100% to(77.33±2.89)%, and increased the percentage of SA-β-gal positive senescent cells from (6.70±0.46)% to (17.67±1.53)%. The hucMSC-EVs pretreatment decreased the intracellular ROS level, stimulated cell proliferation from (77.33±2.89)% to (90.67±2.52)% and( 96.00±5.57)% , and decreased the positive rate of SA-β-gal positive cells from (17.67±1.53)% to (8.38±0.56)% and (7.07±1.10)%.
Conclusions
The hucMSC-EVs preconditioning may protect dermal fibroblasts from UVB induced photoaging, by reducing intracellular ROS.
5. Diagnosis of primary lymphedema with indocyanine green lymphography

Objective:
To evaluate the diagnostic value of indocyanine green(ICG)lymphography in primary lymphedema of extremities.
Methods:
61 patients with limb lymphedema were enrolled in the study. The contralateral healthy limbs were used as controls. After intradermal injection of Indocyanine Green, dynamic observation of lymphatic vessels and lymph flow was performed using PDE near-infrared fluorescent instrument. The frequency of lymphatic contraction was calculated.
Results:
Lymphatic vessel images were clearly visualized in healthy limbs. Inguinal or axillary lymph nodes can be visualized when examined 30 minutes after injection. Lymphatic contraction frequency was 1(0.33-5.00)time per minute. 94.8% lymphedematous limb demonstrated delayed lymphatic vessel or lymph nodes. "Dermal backflow" and diffused shadow pattern can be observed in all affected limbs. Abnormal lymphatic vasculature and contraction can be visualized. The frequency of lymphatic contraction in the affected limb was higher compared to controls: 2.5(0.5-7.0) times per minute.
Conclusions
ICG lymphography possess high sensitivity for lymphedema diagnosis. ICG lymphography furthers our understanding of pathophysiological alterations of lymphatic disorders.
6.Investigation on levels of gross radioactivity in drinking water and 90Sr in food before and after operation of Sanmen Nuclear Power Plant

Objective:To investigate the levels of gross radioactivity in drinking water and of 90Sr in food around Sanmen nuclear power plant (NPP) before and after operation. Methods:From 2012 to 2019, 7 drinking water monitoring sites were selected around the Sanmen NPP to collect source water, factory water and peripheral water, respectively, in order to measure and analyze the total radioactivity level in drinking water. Based on the dietary habits of local residents, 4 kinds of food such as grains, vegetables, freshwater fish and marine fish grown locally were sampled to measure and analyze the activity concentration of 90Sr. Results:Before the operation of Sanmen NPP, the total α radioactivity and total β radioactivity in drinking water around the NPP were 0.001-0.063 Bq/L and 0.019-0.210 Bq/L, respectively. After operation, the total α radioactivity and total β radioactivity were 0.001-0.084 Bq/L and 0.025-0.079 Bq/L, respectively. Both were lower than the national standard value given in the GB 5749-2006 " Sanitary standards for drinking water" . There was no significant change in the radioactivity level of source water, factory water and peripheral water during the wet and dry periods. The activity concentration of 90Sr in these four types of food ranged from 0.037 to 1.216 Bq/kg before operation and from 0.049 to 0.692 Bq/kg after operation, all lower than the standard value of " Standard for the concentration in radioactive substances in food" (GB 14882-94). Conclusions:Before and after the operation of Sanmen NPP, the total α and total β radioactivity in drinking water and 90Sr activity concentration in food are stable, below the national standard limit.

Objective:To investigate and analyze the radioactivity levels of 90Sr and 137Cs in drinking water and 137Cs in food after the installation of the first AP1000 nuclear power unit in China. Methods:From 2012 to 2019, four drinking water monitoring points around AP1000 nuclear power unit located at Sanmen nuclear power plant site were collected during the wet season and dry season, 90Sr and 137Cs and radioactivity concentrations were determined in drinking water. Local rice, cabbage, crucian and mullet were collected to determine the radioactivity concentration of 137Cs. Results:From 2012 to 2019, the radioactivity concentrations of 90Sr and 137Cs in drinking water were 1.2-9.8 mBq/L and 0.2-8.1 mBq/L, respectively. The radioactivity concentration of 137Cs in food were 1.1×10 -2-2.8×10 -1 Bq/kg, lower than the limits specified in the Limited concentrations of radioactive materials in foods (GB 14882-94). Conclusions:After the installation of the first AP1000 nuclear power unit in China, the radioactivity levels of 90Sr and 137Cs in drinking water and 137Cs in foods are stable, without environmental impact identified.
8.Analysis of monitored results of death cause in residents around Sanmen nuclear power plant from 2015 to 2019

Objective:To investigate the data on death cause of residents around Sanmen nuclear power plant from 2015 to 2019.Methods:The data on death cause of residents in Sanmen county from 2015 to 2019 were collected, and the top 10 diseases in death causes ranking and radiation-related malignant tumors were analyzed.Results:The average mortality rate of residents in Sanmen county was 575.87 per 100 000 population from 2015 to 2019, lower than the national level and close to that in Zhejiang Province. The top 10 death causes remain unchanged, higher than 95.79% of total deaths. The mortality rate of malignant tumor increased from 159.23 per 100 000 population in 2015 to 191.51 per 100 000 population in 2019 (χ 2=15.889, P<0.05). There was no significant difference in the proportion of mortality from radiation-related tumor(leukemia and thyroid cancer) in Sanmen county in recent 5 years ( P>0.05). Conclusions:From 2015 to 2019, the death rates of residents in Sanmen county were relatively stable. The effects of the operation of the nuclear power plant on the health of the local residents need to be continuously monitored.
9. The application of Whole-Mount immuno fluorescence staining technique in the study of lymphatic morphology in mice

Objective:
To elaborate the characteristics and advantages of Whole-Mount immune fluorescence staining by observing the lymphatic vessels of mice.
Methods:
The ear skin tissue, the hindlimb lymphatic vessels and the mesenteric lymphatic vessels were harvested from normal C57 mice. The tissue samples were subjected to whole-tissue immunofluorescence staining.These tissue samples were fixed by paraformaldehyde, blocked by bovine serum and incubated in primary and secondary antibodies. Then, the lymphatic vessels were observed and analyzed in these samples with a confocal laser-scanning microscope.
Results:
The capillary lymphatic vessels and lymphatic endothelial cells can be clearly showed in the ear skin. The valves and smooth muscles can be clearly showed in the hindlimb and mesenteric lymphatic vessels by Whole-Mount immunofluorescence staining.
Conclusions
The whole-tissue immunofluorescence staining technique can observe the external morphology of lymphatic vessels clearly and stereoscopically, and can deeply observe the internal structure of lymphatic vessels. This technique can provide more accurate study on physiology and pathology of lymphatic vessels.
