1.Dental pulp stem cells in regenerative medicine:application and development

BACKGROUND:Dental pulp stem cel s are characterized by multi-lineage differentiation and proliferation abilities and are easy to obtain, so they are becoming an issue of concern in regenerative medicine. OBJECTIVE:To provide clues and direction for further study by analyzing progress of domestic and overseas research on dental pulp stem cel s, and summarizing their application in regenerative medicine. METHODS:The“dental pulp stem cel , regenerative medicine, tissue engineering”in Chinese and English served as the search terms to search articles related to dental pulp stem cel s and regenerative medicine, published from 2000 to 2015 in Medline, PubMed, CNKI, Wanfang and Cqvip databases. Total y 46 articles were selected for overview. RESULTS AND CONCLUSION:Dental pulp stem cel s, which hold the capacity of self-renewal and multi-lineage differentiation, are relatively easy to obtain, and exhibit a great potential in regenerative medicine. The research of dental pulp stem cel s in repairing bone defects has entered the clinical trial phase, but the research of cel differentiation into other tissues is stil in basic trial phase and needs further development.
2.Clinical Analysis on Brachial Plexus Injury with 41 Cases

Objective To observe the long-term result of brachial plexus injury.Methods41 cases of brachial plexus injury were divided into the non-operation group (n=11), neurolysis group (n=17), never repair group (n=13). Retrospective analysis of the charts and follow-up survey of the functional recovery of shoulder joint and elbow joint were carried out. Gilbert score and Mallett score were adopted at the follow-up.ResultsIn the non-operation group, 9 cases were graded as excellent, 1 case as good, 1 case as bad. In the neurolysis group, 2 cases were graded as excellent and good. In the never repair group, 2 cases of shoulder joint obtained excellent, 4 cases of elbow joint were grade as good.ConclusionThe simple surgical result of brachial plexus injury is not satisfactory, it should be combined with postoperative rehabilitation.
3.Optimization of parameters of passive cutaneous anaphylaxis in rats

OBJECTIVE To optimize the parameters of passive cutaneous anaphylaxis(PCA)in rats immunized by ovalbumin(OVA). METHODS 1-2 month-old Sprague-Dawley rats were immu?nized by ip injection of OVA(0.2,1.0 and 5.0 mg per rat)mixed with complete Freund′s adjuvant once every other day 3 times. Serum was collected on the 12th-16th days after final immunization. Then the rats were intracutaneously injected with sensitized serum and then stimulated by iv injection of the same dose of OVA mixed with Evans blue after a latent period of 0.5,1.5,3,6,12,24,36,48 and 60 h. Finally,the diameters of blue spots in the skin were measured at stimulation. RESULTS Serum total-IgE(T-IgE)and OVA-specific IgE(sIgE)levels increased significantly and reached the peak on the 3rd-7th days and 12th-16th days after final immunization,respectively. There was no correlation between the serum T-IgE level and OVA-sIgE level when the rats were immunized with OVA at OVA 0.2-5.0 mg per rat. The rats experienced PCA after injection of OVA 1.0 and 5.0 mg per rat. Diameters of blue spots in the skin reached the maximum value after rats were sensitized for 0.5-3 h. Moreover,the shape,color and size of blue spots were better 30-60 min after stimulation. CONCLUSION Optimized PCA is as follows:1-2 month-old rats are immunized on the 1st,3rd and 5th days by ip injection of OVA 1.0-5.0 mg. The immunizing serum is collected at 12-16 d after final immunization. The rats are stimulated by OVA and Evans blue after a latent period of 0.5-3 h. Diameters of blue spots in rats′ skin are then measured 30-60 min after stimulation.
4.Effect of Passive Exercise on Neural Functional Recovery in Rabbits after Peripheral Nerve Crushed Injury

Objective To investigate the effect of passive exercise on neural functional recovery after peripheral nerve crushed injury.MethodsThe effect of passive exercise on early peripheral nerve regeneration and recovery of motor function were observed with electrophysiological and histological indexes compared with that of the splinting group.ResultsThe nerve conduction of training group was faster than that in the splinting group,and the latency of compound muscle action potentials(CMAP)was shorter(P<0.05).The thickness of myelin sheath,average numbers of myelinated nerve fibera per area and diameter of regenerating axon in training group were larger than those in the splinting group(P<0.05).The wet weight of sural triceps of training group were bigger than that of the splinting group(P<0.01).ConclusionThe passive exercise can improve the early recovery of motor function and neural regeneration after peripheral nerve crushed injury.
5.Effects of remote education based on short video on pain symptoms and rehabilitation compliance in patients with degenerative osteoarthritis

Objective:To explore effects of remote education based on short video on pain symptoms and rehabilitation compliance in patients with degenerative osteoarthritis.Methods:A total of 132 patients with degenerative osteoarthritis who were treated in Department of Orthopedics of Beijing Jishuitan Hospital were selected as the research objects from March 2018 to June 2020. According to the order of admission, they were divided into the control group and the observation group, with 66 cases in each group. The control group received routine health education, while the observation group received short video-based remote education. The differences of pain symptoms and rehabilitation compliance were compared between the two groups after intervention.Results:After intervention, the pain scores of the observation group during exercise [ (2.4±1.0) vs. (3.5±1.1) ]and tenderness point [ (2.7±1.0) vs. (3.9±1.2) ]were lower than those of the control group. The scores of medication [ (5.2±1.0) vs. (4.5±1.2) ], diet [ (2.2±0.6) vs. (1.6±0.8) ]and exercise [ (2.4±0.6) vs. (1.7±0.5) ]compliance were higher than those of control group. The above differences were statistically significant ( P<0.05) . Conclusions:Application of remote education based on short video in patients with degenerative osteoarthritis can reduce pain symptoms and improve rehabilitation compliance.
6.Application of nursing intervention based on well-being PERMA model in patients with spinal fracture accompanying spinal cord injury

Objective:To explore the application effect of nursing interventions based on well-being PERMA model in patients with spinal fracture accompanying spinal cord injury.Methods:Using the convenient sampling method, a total of 116 patients with spinal fracture accompanying spinal cord injury who received treatment in the Comprehensive Orthopedics Department of Beijing Jishuitan Hospital from January 2018 to December 2019 were selected as the research objects. The patients were divided into the control group and the observation group using the random number table method, with 58 cases in each group. The control group received routine nursing intervention, while the observation group received nursing intervention based on well-being PERMA model. The scores of subjective well-being, activity of daily living, motor function and quality of life were compared between the two groups before intervention and after intervention (3 months after discharge) .Results:Before intervention, there was no statistically significant difference between the two groups ( P>0.05). After the intervention, the scores of subjective well-being, motor function and quality of life in the observation group were higher than those in the control group, and the score of activity of daily living were lower than those in the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Application of nursing intervention based on well-being PERMA model in patients with spinal fracture accompanying spinal cord injury can improve subjective well-being and quality of life of patients, and improve their activity of daily living and motor function.
7. Clinical effects of free superficial femoral artery femoral triangle perforator flap in the repair of skin and soft tissue defects in extremities

Objective:
To investigate the clinical effects of free superficial femoral artery femoral triangle perforator flap in the repair of skin and soft tissue defects in extremities.
Methods:
From January 2016 to November 2017, 14 patients (9 males and 5 females, aged 19 to 54 years) with skin and soft tissue defects in extremities accompanied with tendon and bone exposure were admitted to our unit. The size of skin and soft tissue defects after debridement ranged from 7 cm×3 cm to 10 cm×7 cm. The defects were repaired with free superficial femoral artery femoral triangle perforator flaps, with size ranging from 13.0 cm×2.0 cm to 20.0 cm×4.5 cm. The medial femoral cutaneous nerve was applied to the flap. The perforator flap was grafted onto the medial femoral cutaneous nerve in 6 patients. The donor sites were sutured directly. The survival of flaps and the follow-up of patients were observed.
Results:
All flaps of 14 patients survived successfully. The recipient sites and donor sites were healed completely in 13 patients, and 1 patient with partial skin necrosis at the edge of flap was healed after treatment. All patients were followed up for 6 months to 1 year after the operation. The flaps were in good shape, with nearly normal color and soft texture and no cicatrix contracture deformity. The flaps recovered protective sense in 6 patients who had medial femoral cutaneous nerve grafting, and the sensory recovery of the flap was slightly worse in the remaining 8 patients. There was no significant complications on the appearance and walking of the donor thigh in 14 patients, only a linear scar was left on the inner thigh, and no numbness was felt in the donor sites of patients.
Conclusions
The free superficial femoral artery femoral triangle perforator flap is an ideal therapy for repairing skin and soft tissue defects in extremities.
8.Inter-hemispheric Functional Connections Are More Vulnerable to Attack Than Structural Connection in Patients With Irritable Bowel Syndrome

Irritable bowel syndrome (IBS) is a prevalent functional gastrointestinal disease characterized by recurrent abdominal pain and bowel dysfunction. However, the majority of previous neuroimaging studies focus on brain structure and connections but seldom on the inter-hemispheric connectivity or structural asymmetry. This study uses multi-modal imaging to investigate the abnormal changes across the 2 cerebral hemispheres in patients with IBS.
Methods:
Structural MRI, resting-state functional MRI, and diffusion tensor imaging were acquired from 34 patients with IBS and 33 healthy controls. The voxel-mirrored homotopic connectivity, fractional anisotropy, fiber length, fiber number, and asymmetry index were calculated and assessed for group differences. In addition, we assessed their relevance for the severity of IBS.
Results:
Compared with healthy controls, the inter-hemispheric functional connectivity of patients with IBS showed higher levels in bilateral superior occipital gyrus, middle occipital gyrus, precuneus, posterior cingulate gyrus, and angular gyrus, but lower in supplementary motor area. The statistical results showed no significant difference in inter-hemispheric anatomical connections and structural asymmetry, however negative correlations between inter-hemispheric connectivity and the severity of IBS were found in some regions with significant difference.
Conclusions
The functional connections between cerebral hemispheres were more susceptible to IBS than anatomical connections, and brain structure is relatively stable. Besides, the brain areas affected by IBS were concentrated in default mode network and sensorimotor network.

10.Comparative study on specific chromatograms and main active components of wild and cultivated rhizomes of Paris polyphylla var. yunnanensis.

The present study is to compare specific chromatograms and main acitive components between wild and cultivated rhizomes of Paris polyphylla var. yunnanensis by HPLC. HPLC analysis was performed on a Waters XSelect HSS T3 C₁₈ clumn (4.6 mm×250 mm, 5 μm), with a mobile phase consisting of acetonitrile (A)-water (B) at a flow rate of 1 mL•min⁻¹ (0-50 min,30%-50%A;50-80 min,50% A,80-85 min,50%-30%A;85-100 min,30% A). The detection wavelength was 203 nm and the column temperature was controlled at 30 ℃, and the injection volume was 10 μL. HPLC specific chromatograms of wild and cultivated rhizomes of P. polyphylla var. yunnanensis were established and nine steroidal saponins were simultaneously determined by the above method. The mean contents of paris saponin Ⅶ, paris saponin H and total average contents of four pennogenyl saponins in Rhizomes of wild samples were significantly higher than those of cultivated ones. However, this result is opposite from the average content of paris saponin Ⅰ and total average contents of five dioscins in the wild and cultivated samples. Because the significant differences occurred for the specific chromatograms and main active components between the wild and cultivated P. polyphylla var. yunnanensis, much more pharmacological and clinical researches are therefore necessary.
