1.Expressions of transcription factor activator protein-2α, estrogen receptor-β and matrixmetalloproteinase-9 in lichen planus lesions

Objective To determine the expressions of transcription factor activator protein-2α (AP-2α), estrogen receptor-β (ER-β) and matrixmetalloproteinase-9 (MMP-9) in lichen planus (LP) lesions and their significance. Methods Tissue samples were resected from the lesions of 30 patients with LP and normal skin of 30 human controls. An immunohistochemical method using streptavidin-peroxidase (SP) was performed to detect the expression of ER-β and MMP-9, reverse transcription-PCR and Western blot to measure the mRNA and protein expressions of AP-2α, ER-β and MMP-9 in these specimens. Results There was a significant decrease in the mRNA expressions of AP-2α (0.488 ± 0.039 vs. 1.008 ± 0.023, P < 0.01 ) and ER-β (0.365 ± 0.032 vs. 0.998 ± 0.036, P < 0.01), together with an increase in the expression of MMP-9 (1.237 ± 0.027 vs. 0.567 ± 0.015, P< 0.01) in the LP lesions compared with the control specimens; similar results were observed for the protein expressions of AP-2α, ER-β and MMP-9. In LP lesions, the expression of AP-2α was positively correlated with that of ER-β (R = 0.89, P < 0.01), but negatively correlated with that of MMP-9 (r = -0.91, P < 0.01). Conclusions The down-regulation of ER-β expression and up-regulation of MMP-9 expression may be ascribed to the abnormal regulation of upstream target genes mediated by the decreased expression of AP-2α, which may be involved in the pathogenesis of LP.

Copy number variation (CNV) is one kind of structural variations,which exists extensively in human genome.In addition,it is the crucial source of inter-individual gene differences.Recently,with the successful development of gene array and gene sequencing,molecular methods for CNV testing are more and more standardizing and automating.Therefore,ongoing attentions have been attached to the effects and mechanisms of CNV in complicated diseases like autoimmune diseases.Molecular methods for CNV testing can be divided into two categories:target screening and the whole genome scanning platforms.Currently,the studies about CNV and autoimmune diseases mainly focused on systemic lupus erythematosus and rheumatoid arthritis.Considering the limitations of measurement methods and testing costs,it is still in the infant.
3.Research progress in radiotherapy for liver tumors reported in 2013 ASTRO Annual Meeting

With the development of radiotherapy techniques,the efficacy of radiotherapy for liver tumors has been improved,and radiothera-py can be considered as an alternative treatment for patients who are not suitable for surgery.The progress in liver tumor radiotherapy repor-ted in the 55th annual meeting of the American Society for Therapeutic Radiology and Oncology (ASTRO)in 2013,particularly the efficacy and safety of stereotactic body radiation therapy for liver tumors,is reviewed.In the future,radiotherapy may be increasingly used as an ef-fective method in the treatment of liver tumors.The main studies reported at the meeting are sorted as follows in this article.

Objective The effect of acidity and severity of gastric atrophy on Helicobactor pylori (H.pylori) colonization was studied. Methods 95 patients with chronic atrophic gastritis (CAG) and positive H.pylori test were categorized into groups.Group 1,48 cases having moderate atrophic antritis with various degrees of fundic atrophy.Group 2,47 cases having mild fundic atrophy with various degrees of antritis. Assessment was made of basic and maximum acid output (BAO, MAO ) as well as of gastrin. H.pylori was tested with Warthin-Starry staining. 5.Dysfunctional Attitudes and Cognitive Bias in Major Depressive Disorder

Objective:To examine whether dysfunctional attitudes and cognitive bias change with fluctuations in depressive symptoms. Methods:122 patients with MDD (major depressive disorder) were assessed with DAS (dysfunctional attitude scale),CBQ (cognitive bias questionnaire) ,and HRSD (Hamilton rating scale for depression) before and after the 8-week treatment. 51 normal subjects were assessed with the DAS, CBQ and SDS as controls.Results:(1) Compared with normal controls, at the baseline MDD patients had significantly higher total DAS scores (158.2?34.6 vs. 119.3?23.4,t=8.559, P=0.000) and the score of CBQ (5.19?3.77 vs. 1.69?1.83, t=8.208, P=0.000). (2) After 8 weeks, the HRSD scores of patients reduced to Median 7.0, and the DAS and CBQ scores significantly lower than those in baseline (DAS total score from 158.2?34.6 to 143.3?29.3, t=3.611, P=0.000; the median of CBQ decreased from 5.0 to 3.0;z=4.670, P=0.000). (3) The DAS score (131.1?25.7) and CBQ score (median 3.0) of full recovery patients with full recovery remained higher than those of controls (t=2.397, P=0.018 ;z=3.990, P=0.000).Conclusion:Dysfunctional attitudes and cognitive bias were influenced by fluctuations in stata or demonstrateing stability over time. They were all state-trait features.

7.The correlation between sarcopenia and energy intake in people of advanced age

Objective:To analyze influencing factors for sarcopenia in people of advanced age, in order to provide insight and evidence for the prevention and treatment of sarcopenia in people belonging to this age group.Methods:Data from 167 people of advanced age seeking care at our department from December 2014 to July 2017 were retrospectively analyzed.According to the diagnostic criteria for sarcopenia of the Asian Working Group for Sarcopenia, subjects were divided into the sarcopenia group( n=46, 27.5%)and the non-sarcopenia group( n=121). Differences in body composition, energy intake, quantities and proportions of three major nutrients were analyzed between males and females.Related influencing factors for sarcopenia were analyzed by using multiple linear regression. Results:Compared with the non-sarcopenia group, the sarcopenia group had lower body mass index, waist-hip ratio, fat mass, total energy intake and protein( P<0.05)but higher age and fat intake( P<0.05). Values for grip strength, muscle mass, index of skeletal muscle, adjusted muscle mass by body mass index, total energy intake, carbohydrates, fat and protein were lower and the percentage of body fat was higher in females than in males( P<0.05). Multiple Logistic regression analysis showed that insufficient intake of protein( β=-0.290, OR=0.748, 95% CI: 0.569-0.984, P<0.05), reduction of body fat mass( β=-2.673, OR=0.069, 95% CI: 0.010-0.488, P<0.05)and excessive visceral fat accumulation( β=0.739, OR=2.094, 95% CI: 1.219-3.597, P<0.01)were correlated with sarcopenia in people of advanced age. Conclusions:The occurrence of sarcopenia is higher in people of advanced age and is related to insufficient intake of protein, reduction of body fat mass and excessive visceral fat accumulation.Individualized nutrition evaluation and support should be carried out as early as possible for people in this age group.
8.Reversed-phase ion pair high performance liquid chromatography in determination of berberine and palmatine in Phellodendron chinense Schneid.and its granules

Objective:To determine berberine and palmatine in Phellodendron chinense Schneid.and its granules.Methods: The reversed-phase ion pair high performance liquid chromatography(RP-HPLC) was used and the validation of the method was tested.The chromatography condition was with Lichrospher C18 column(4.6 mm?250 mm,5 ?m), mobile phase was ACN∶25 mmol/L NaH 2PO 4∶25 mmol/L SDS(2∶1∶1),flow speed was 1.0 ml/min,detection wavelength was 345 nm,and temperature of column was 25℃,Phellodendron chinense Schneid.and its granules were extracted with methanol solution of hydrochloric acid.Results: The theoretical plate number of berberine and palmatine were 14 906 and 14 847,the resolution were 2.33 and 2.86,the tailing factor were 1.09 and 1.06,respectively; all the parameters were suitable for determination.The calibration curves were linear in the range of 40-500 ng,Y=698 278X-3 846,r=1.000(berberine) and 20-250 ng, Y= 536 632X- 7 738, r=0.999 9,r=0.999 4(palmatine).The intra-day and inter-day precision(RSD) at low,middle and high injection amount was all less than 2.5%(berberine) and 1.5%(palmatine).The stability(RSD) was 0.66%(berberine) and 0.70% (palmatine) in 48 h.The recurrence(RSD,n=5) was 0.11%(berberine) and 0.12%(palmatine).The limits of detection was 2.0 ng(berberine) and 1.0 ng(palmatine).The recoveries were 100.4% (RSD=0.12%,n=3) for berberine and 99.80%(RSD=0.22%,n=3) for palmatine.The contents of berberine and palmatine in 3 batch of Phellodendron chinense Schneid.and 5 batch of its granules were determined.Conclusion: Our method can be used for determination of berberine and palmatine in Phellodendron chinense Schneid.and its granules, which is simple and reliable.
9.EXPERIMENTAL STUDY ON OSSEOINTEGRATION AND BONE FORMATION OF DENTAL IMPLANTS IN RATS WITH OSTEOPOROSIS

The aim of the study was to observe osseointegration of dental implants in osteoporotic rats. Rats were randomly divided into groups A(SHAM) and B(OVX), and pure titanium screws were implanted in the distal half of the right femurs. 1 5 and 3 months after implantation, half of the rats in each group were sacrificed and right femurs were observed by X ray photography, scan electron microscopy as well as light microscopy. The results showed that 1.5 and 3 months after implantation, the osseointegration index (OI) of group A was higher than that of group B. But the OI of each group was almost the same at different intervals. 1 5 months after implantation, the amount of new bone formation in group A was more abundant than that in group B, and it increased in group A but decreased in group B 3 months after implantation.The results confirmed that with higher BMD osseointegration was better. Osteoporotic changes could decrease OI and the amount of new bone formation at implant bone interface in rats. Osseointegration would not improve without outside factors.
10.Expression of VEGF -C and p63 in early esophageal carcinoma and intraepithelial neoplasia

Objective To explore expression and clinical value of VEGF-C and p63 in early esophageal carcinoma and intraepithelial neoplasia. Methods 146 cases were randomized into normal esophageal mucosa, low level intraepithelial tumor, high level intraepithelial tumor and early esophageal carcinoma. The expression of VEGF-C and p63 were detected by using the immunohistochemistry dyeing.Results The expression of VEGF-C immunohistochemistry dyeing had statistical differences among different levels(X~2= 47.455, P <0.001). Normal esophageal mucosa v.s. high level intraepithelial tumor (X~2=36.721, P <0.001), Normal esophageal mucosa v.s. early esophageal carcinoma (X~2=26.483, P <0.001), low level intraepithelial tumor v.s. high level intraepithelial tumor(X~2= 10.025, P<0.0083), low level intraepithelial tumor v.s. early esophageal carcinoma(X~2=16.734, P<0.001). There was a significant correlation between pathological classification and the expression amount of VEGF-C (r = 0.462, P <0.001). The expression of p63 had statistical differences among different levels(X~2=28.962, P <0.05). There was a significant difference on normal esophageal mucosa comparing with low level, high level intraepithelial tumor or early esophageal carcinoma (X~2=12.735, P =0.005, X~2=20.421, P<0.001, X~2=20.854, P<0.001). There was a significant correlation between pathological classification and the expression of p63 (r= 0.272, P<0.05). Conclusion There is a significant correlation in the express of either VEGF-C or p63 comparing with either intraepithelial tumor or early esophageal carcinoma. It may be an early warning indicator.
