Objective:Exploring the significance of micrometastases of lymph nodes in cases of pT 3N 0M 0 gastric cancer.Methods:We examined 2230 lymph nodes that had been removed from 95 patients with stage Ⅱ gastric cancer(pT 3N 0M 0) with immunostaining of cytokeratin.Results:In 38 of 95(40%) patients with gastric cancer,evidence of micrometastases was found in 290 of 2230(13%) lymph nodes.The incidence of micrometastases in undifferentiated carcinoma was higher than that in differentiated carcinoma.An analysis of survival demonstrated the 5-year survival of patients with micrometastases in their resected lymph nodes was significantly lower than that of patients without micrometastases( P
2.Research progress of the function of high mobility group A2 protein in malignant tumors

The high mobility group A2 (HMGA2) is a non-histone chromatin protein. It has no transcriptional activity of itself but could alter chromatin structure to regulate the transcription of other genes. HMGA2 gene is weakly expressed or does not express in normal tissues. The expression of HMGA2 is up-regulated during embryogenesis and malignant neoplasias. In several malignant tumors, HMGA2 is used as a diagnostic molecular marker or an independent prognostic indicator. Furthermore, HMGA2 not only elicits epithelial-mesenchymal transition but also maintains the differentiation potential and self-regeneration capability of stem cells. It plays an important role in initiation, proliferation, and metastasis of malignant neoplasias.
3.Sclerostin monoclonal antibody in the treatment of osteoporosis

BACKGROUND:Sclerostin can negatively regulate the bone metabolism, and the sclerostin monoclonal
antibody can antagonize the negative regulation effect, inhibit bone resorption and promote bone formation. OBJECTIVE:To explore the mechanism and application progress of sclerostin monoclonal antibody in the treatment of osteoporosis.
METHODS:An online search of PubMed database, CNKI database, VIP database and Wanfang database
between May 2005 and May 2013 was performed by the first author to search the related articles with the key words of“osteoporosis, antibody, sclerostin, Wnt, SOST”in both English and Chinese. Articles related to
sclerostin monoclonal antibody were included. For the articles in the same field, those published earlier or in the authorized journals were preferred.
RESULTS AND CONCLUSION:A total of 170 articles were obtained after initial search, and final y 54 articles related to sclerostin monoclonal antibody were included for review according to the inclusion criteria. The
sclerostin can block Wnt pathway through combining with low-density lipoprotein receptor-related protein 5/6, thus inhibiting the differentiation and mineralization of osteoblasts. By specifical y binding to sclerostin, the
sclerosin monoclonal antibody can indirectly promote bone formation and restrain bone absorption which has great significance in the treatment of osteoporosis. Meanwhile, compared with the other treatment method, the specific targeting of sclerostin and the binding specificity of sclerostin monoclonal antibody provide application advantages for the treatment of osteoporosis.
4.Expression and significance of Livin in gastric cancer tissues

Objective To detect the expression of Livin in gastric cancer tissues and cells, and investigate its relation to tumorigenesis and progression of gastric cancer cells. Methods (1) The expression of Livin in gastric cancer tissues was located by immunocytochemisty. (2) Livin mRNA and protein expression in adjacent tissues of carcinoma, gastric ulcers and gastric cancer tissues were detected by RT-PCR and Western blot. Results The expression of Livin in the nucleus and cytoplasm of gastric cancer cell line SGC-7901 was observed. The positive rate of Livin expression in the gastric cancer tissues was 67% (20/30). There were no expression of Livin-α and Livin-β in the adjacent tissues of carcinoma and the gastric ulcers. The positive rate of Livin-α expression in the gastric cancer tissues was positively correlated with TNM stage of gastric cancer (x2=11.719, P<0.05). The expression rate of Livin in the differentiation and location of gastric cancer, age and gender of patients had no statistical difference (x2=1.607, 0.000, 0.156, 0.419, P>0.05). Conclusion Correlation of high Livin-α expression and pathologic staging of gastric cancer indicates that Livin-α may play an important role in the carcinogenesis and progression of gastric cancer.
5.Construction of lentiviral vector pLeno-DCE-PDGF-B and its effect on invasion ability of SGC7 9 0 1 gastric carcinoma cells

Objective To construct a lentiviral vector that stably express platelet-derived growth factor B (PDGF-B)and to in-vestigate effect of PDGF-B overexpression on invasion of SGC7901 gastric carcinoma cell.Methods The lentiviral vector pLeno-DCE-PDGF-B was constructed and transfected into 293T cells.The supernatant containing the lentivirus particles was harvested to determine the virus titer and high titer lentivirus particles was gathered.Then PDGF-B lentiviral vector was transfected into SGC7901 cells for construction of stable PDGF-B overexpression SGC7901 cells.Western-blot and immunofluorescence were used for evaluation of the construction of stable PDGF-B overexpression SGC7901 cells.Transwell test was used for detection the inva-sion of SGC7901 cells.Results The lentiviral vector was correctly constructed and verified by sequencing.High titer PDGF-B lenti-viral particles were acquired successfully.After transfection,SGC7901 cells were green color by detection of fluorescence micro-scope;and the expression of PDGF-B protein in transfected SGC7901 cells were much higher than that in normal SGC7901 cells.Al-so,PDGF-B overexpression increased the invasion ability of SGC7901 cells.Conclusion The PDGF-B lentiviral particles are ob-tained after successful construction and package.The stable PDGF-B overexpression SGC7901 gastric carcinoma cell line is estab-lished,which proves that the PDGF-B overexpression might increase the invasion ability of gastric carcinoma cells.
6.Change of survivin gene expression in gastric cancer cell line BGC-823 induced by hydroxycamptothecine

Background and purpose:The antitumor drugs produce a marked effect through the induction of apoptosis. The resistance to chemotherapy has been found to be associated with abnormal expression of genes related to apoptosis. We designed this experiment to investigate the role of survivin gene in the resistance to chemotherapy. Methods:After being treated with Hydroxycamptothecine (OPT,21.96mmol/L), the apoptosis in gastric cancer cell line BGC-823 cells were evaluated by either flow cytometry(FCM) or the terminal deoxynucleotidy transfer mediated dUTP-biotin nick end labeling(TUNEL). The expression of Survivin was detected by reverse transcription-Polymerase chain reaction(RT-PCR) and immunohistochemistry.Results:Compared to control group, the OPT can induced apoptosis in BGC-823[(12.68?0.17)% vs (3.35?0.13)% by FCM,(16.00?1.23)% vs (2.78? 0.84)% by TUNEL[, OPT could upregulate survivin expression in BGC-823 cells (1.489?0.041)% vs (0.756?0.037)% in terms of level of mRNA,(0.928?0.046) vs (0.303?0.032) in terms of protein expression).Conclusions:The survivin gene may be associated to the resistance to chemotherapy through the induction of gene expression.
7.The research progress of DNA methylation in bladder cancer

Bladder cancer is a malignant tumor of the urinary system with the highest and still increasing incidence rate in China in recent years. While most patients with a diagnosis of non-muscle invasive bladder cancer (NMIBC) have a good prognosis, NMIBC is prone to relapse and progress to muscular invasive cancer (MIBC) after treatment, leading to a poor prognosis. At present, diagnosis and postoperative follow-up of bladder cancer still rely on invasive cystoscopy, and there is a lack of effective treatment for advanced tumors. The results show that methylation, as a chemical modification of DNA, is related to the occurrence and development of bladder cancers. This article reviews the research progress of bladder cancer DNA methylation in diagnosis, monitoring and treatment in recent years.

9.Application of the systematic education platform for patient with permanent colostomy in improving self-care ability

Objective To evaluate the effect of the systematic education platform for patient with colostomy in improving self‐care ability .Methods A total of 198 colostomy patients from a Third‐Grade Class A Hospitals were randomly assigned to ei‐ther learn colostomy nursing knowledge by the systematic education platform or routine health education condition .The patients were investigated by a questionnaire about Exercise of Self‐Care Agency Scale ,ESCA before discharge ,1 month and 3 months after discharge .Results After the systematic education platform for 3 month ,the total score(120 .87 ± 10 .11) and each factor scores of ESCA scale[self‐protection skills(30 .51 ± 2 .57) ,self nursing responsibility(23 .95 ± 2 .64) ,self concept(26 .23 ± 1 .69) ,health knowledge level (39 .78 ± 3 .12 )] were all significantly improved after the systematic education platform ( P < 0 .05 ) . Conclusion The systematic education platform can improve ostomy patients′self‐care ability and quality of life .
10.Risk factors of elderly patients with abdominoperineal resection for rectal carcinoma

Objective To analyze the relative risk factors of elderly patients with abdominoperineal resection for rectal carcinoma.Methods The risk factors for hospital death and postoperative complications in 137 elderly patients who had undergone abdominoperineal resection for rectal carcinoma from July 1995 to July 2005 were analyzed retrospectively.Results The risk factors included coexistent diseases,hemoglobin level less than 70 g/L,body mass index(BMI)less than 18.5 kg/m2,intraoperative blood loss more than 1 000 ml,operating time longer than 6 h.The relative risk ratio were 4.76,5.85,6.49,5.47,3.90 respectively for hospital death,and 2.02,2.27,1.83,2.01,1.70 for postoperative complications.Conclusion The risk factors of the elderly patients with abdominoperineal resection for rectal carcinoma should be considered carefully during the perioperative period.
