1.Isolation and Culture of Alveolar Type Ⅱ Epithelial Cells and Injurious Effects of Phospholipase A_2 on the Cultured Cells

A method for isolation and primary culture of guinea pig alveolar type Ⅱ epithelial cells is reported. The injurious effects of phospholipase A2 (PLA2) on the cultured cells were studied. Trypsinizing solution was instilled into the bronchoalveolar space. The lungs were digested, sectioned, vortexed and filtered. The yield of original cell mixture was 172.3?106 cells/guinea pig, of which 33.6% were type Ⅱcells. At 48 h of culture following short term selective attachment 7.9?106 cells were obtained, of which 66.1% were type Ⅱ ceils. The purity could further be raised to 76.2% (63.9% -92%) if pretreated with ethyienediarnine tetraacetic acid (EDTA) before trypsinization to remove attached cells. The characteristics of cultured cells were observed under phase contrast microscopy, light microscopy and electron microscopy. After exposure of the cells to 300U/ml PLA2 for 4h at 37℃, detachment and lactic dehydrogenase (LDH) release of the cultured cells increased significantly. It is suggested that PLA, may damage and promote detachment of cultured type Ⅱ cells and it may play an important part in the development of some diseases.
3.Analysis on gene polymorphism of MTHFR C677Tin Han population of south China area

Objective To investigate the distribution situation of methylenetetrahydrofolate reductase (MTHFR) C677T gene polymorphism among healthy Han population ,patients with primary hypertension and patients with hypertension complicating coro‐nary heart disease(CHD) in south China area .Methods MTHFR C677T gene polymorphism in 359 cases was detected by adopting the microarray method .The differences of genotypes frequencies were statistically compared between males and females .Results The wild type CC genotypes were common ,accounting for 54 .9% ,the mutation type CT accounted for 33 .1% and TT for 12 .0% . The distribution difference of wild type and mutation type had statistical difference between the male and female populations (P<0 .05);the TT frequency distribution had statistical difference among the patients with hypertension complicating CHD ,healthy population and patients with primary hypertension (P<0 .05) .Conclusion MTHFR C677T gene polymorphism distribution has difference between male and female populations ,and is related to the CHD occurrence in the patients with primary hypertension .
4.Deep Sternal Wound Infection after Cardiac Surgery

OBJECTIVE Deep sternal wound infection (DSWI) is one of the most serious complications after the open heart cardiac surgery. To explore the risk factors and appropriate treatment methods,we performed this study retrospectively. METHODS Between Jan 2001 and Jan 2006,1123 patients underwent various cardiac surgical procedures via a median sternotomy in the Cardiovascular Center of Beijing Tongren Hospital. These patients were divided into two groups by whether suffered from DSWI. Univariate analyses were performed for possible risk factors,and Logistic regression was used in multivariate analysis. RESULTS Eight among 1123 (0.71%) patients suffered from DSWI. No one died from wound infection. Results from both single variate analysis and multivariate analysis showed that only age significantly associated with DSWI. No other independent risk factors significantly associated with DSWI. Blood culture in acute DSWI group tended to be positive,while in chronic group it tended to be negative. All patients with DSWI were effectively cured. CONCLUSIONS Age is an independent risk factor for DSWI. Early debridement with closed chest catheter irrigation and antibiotics using are strongly recommended as an easy and effective way to treat DWSI. Large-scale multicenter studies are still needed to determine reliable risk factors for DSWI.
5.Platelet-activating Factor Mediates Hydrogen Peroxide Induced Endothelial-leukocyte Adhesion

The effects of hydrogen peroxide (H2O2) on endothelial-polymorphonuclear cells (EC-PMN) adhesion and their mechanisms wsre studied in cultured bovine pulmonary artery endothelial monolayers in vitro. H2O2 at various concentrations (10-1, 10-2, 10-3mol/L respectively) stimulated EC dependent PMN adhesion, of which l02mol/L H2O2 was the most potent one, increasing adhesion to 2.3 times that of the control. Pretreatment of PMNs with SRI 63-441, a platelet-activating factor (PAF) receptor antagonist, had no effect on H2O2 induced EC-PMN adhesion. Pretreatment of ECs with SRI 63-441 before H2O2 exposure significantly decreased PMN adherence to ECs. Pretreatment of ECs with phospholipase A2 inhibitor p-bromophenacyl-bromide or cahnodulin antagonist chlorpromazine and aildum ion chelate EGTA obviously decreased H2O2 induced increment of EC-PMN adhesion. These results suggest that H2O2 may activate ECs, causing the inflow of extracellular calcium or the release of calcium from intracellular deposits. Increased intracellular Ca2+ may bind with calmodulin to activate phospholipase A2 thus initiating PAF synthesis and promoting EC-PMN adhesion.

To study the effects of high salt intake together with cold stress on rat cardiac renin angiotensin aldosterone system (RAAS) in rats. Male Wistar rats were randomly divided into four groups: control, high salt (8%NaCl), cold stress (5 2℃) and compound group (8%NaCl+cold stress). Blood pressure and body weight were measured once a week. The experiment ran for 8 weeks. 7.Locking compression plate fixation for distal radius comminuted fractures

Objective To evaluate the effectiveness of locking compression plate (LCP) in treatment of comminuted fractures of distal end of radius. Methods A retrospective analysis of the clinical data was done for the 24 patients with comminuted fracture of distal end of radius who had been treated by locking compression plate fixation from August 2002 to August 2003. Results The follow up duration averaged 7.8 months. The satisfactory rate of functional outcome with LCP fixation was 91.6 percent. Conclusion Although LCP can provide the outstanding stability theoretically, it has not resulted in more exciting outcomes in treatment of comminuted fractures of distal end of radius than traditional methods.
8.Application of hydroxyapatite-calcium phosphate in nasal augmentation

ObjectiveTo investigate the practicability and maneuverability of applying partical hydroxylaptite artificial bone to rehabilitate saddle nose as well as removing materials from the malposition of nasal implant.MethodsWe used the incision beside nasal columella and detached atunnel in deep of nasal aponeurosis.Then we put hydroxyapatite into the detached tunnel,at the same time press position of operation from skin to obtain a nice form.At last we sewed up the incision.Results In 15000 cases of nasal implant operations with follow-up within last 18 years,the successful cases were 94 %,while the failing ones were 6 %.ConclusionsApplying partical hydroxylaptite artificial bone to saddle nose and nasal implant can obtain a satisfactory,long-tcrm and stable result.It can he an optional surgical method in nasal augmentation.
9.Curative effect of surgical treatment for severe hypertensive intracerebral hemorrhages in basal ganglia and broken into ventricles

Objective To investigate the clinical treatment of severe hypertensive intracerebral hemorrhage in basal ganglia and broken into ventricles .Methods 16 patients with severe hypertensive intracerebral hemorrhage in basal ganglia and broken into ventricles received microsurgical treatment using posterior central gyrus posterior -lateral fissure upper approach .The hematoma cavity and ventricle were opened ,and brain surgical department drainage was placed in the hematoma cavity in the operation .The curative effect was analyzed retrospectively 6 months after operation.Results According to Glasgow Outcome Scale ( GOS):6 cases had good recovery;4 cases had moderate disability;2 cases had severe disability;1 case was in persistent vegetative state and 3 cases were dead .Conclusion Craniotomy microsurgical operation using posterior central gyrus posterior -lateral fissure upper approach is an effec-tive treatment of severe hypertensive intracerebral hemorrhage in basal ganglia and broken into ventricles .The fatality rate was reduced .
10.Platelet-activating Factor Mediates Endotoxin-induced Lung Injury in Dogs and Its Mechanism

Intravenous administration of E. coli endotoxin (2mg/kg) caused a persistent increase in platelet activating factor (PAF) contents in arterial blood and bronchoalveolar fluid (BALF). Meanwhile phospholipase A2(PLA2) activity in serum and BALF was elevated and mean arterial blood pressure declined. The total cell number, expecially polymorphonuclear leukocytes and lymphocytes, and protein concentration in BALF were markedly increased 6 h after endotoxin injection. Lung index and extravascular lung water content of endotoxin-treated animals were significantly highter than those of controls. Pretreatment with PAF receptor antagonist SRI 63-441 blocked the increase in PLA, activity and attenuated endotoxin-induced hypotension and acute lung injury. The results suggest that PAF mediates endotoxin-induced lung injury, and leukocyte activation by PAF and the subsequent release of oxygen metabolites and lysoenzymes are important intermediate mechanisms leading to high permeability pulmonary edema.
