Objective To study the effects of wedelolactone on the expression of P-glycoprotein (P-gp) and to explore the multi-drug resistance reversing mechanism of wedelolactone in K562/A02 cells in vitro. Methods The half maximal inhibitory concentration of ADM in K562/A02 was determined by MTT method. The protein expression level of P-gp was determined by Western blot after wedelolactone pretreatment. Results Wedelolactone remarkably enhanced chemo-sensitivity to ADM of K562/A02 cells. After 0.2, 2, 20 μmol/L different concentration of wedelolactone treatment in 24 h, the relative reversal efficiency of K562/A02 to ADM was 23.5%, 47.1% and 67.7%, respectively.according to the results of Western blot, wedelolactone was shown to efficiently inhibit the expression of P-gp (P<0.05).The relative efficiency of K562/A02 to ADM was 25.4%,46% and 55.6%, respectively. Conclusion Wedelolactone could modulate P-gp expression, and P-gp expression down regulation may be one of the MDR reversal mechanisms in K562/A02 cells by wedelolactone.

Objective To investigate the effect of clotrimazole on apoptosis of hepatic cells after ischemia-reperfusion injury and its mechanism. Methods Hepatic ischemia-reperfusion rat model was established. Thirty-two male Sprague-Dawley rats were randomly allocated into sham-operated group, model control group, low dose clotrimazole group and high dose clotrimazole group. Apoptosis in hepatic tissue was assessed by TUNEL method. Protein expression levels of CYP3A1,Bcl-2,Bax and PARP were measured by Western blotting. Results As compared with model control group, the apoptosis rate, tissue injury, activity of plasma enzymes and the Bax/Bcl-2 expression ratio were reduced in low and high dose clotrimazole groups. The apoptotic index in both clotrimazole-treated groups was lower than that of model control group with statistically significant difference. CYP3A1 expression was significantly induced by clotrimazole compared to the sham-operated group. Conclusion Clotrimazole may inhibit apoptosis of hepatic cells by up-regulating Bcl-2 and down-regulating Bax, thus produce a protective effect on hepatic ischemia-reperfusion injury and it is also related to the inhibition of PARP shear.

Objective: To investigate the expression changes of Bcl-2-associated athanogene 1(BAG-1) and its regulatory effect on the glucocorticoid receptor(GR) activity in rat alveolar macrophages in conditions of cell inflammation and glucocorticoid therapy. Methods: The expression changes of BAG-1 were detected by Western blot after lipopolysaccharide(LPS) and Dexamethasone(Dex) treatment of rat alveolar macrophages(AMs),the interaction between BAG-1 and GR determined by immune coprecipitation experiment, and the transcriptional activation of GR measured by relative luciferase activity assay. Results:After LPS and Dex treatment, the expression of BAG-1L in total protein increased but that of BAG-1S remained changed,BAG-1L rather than BAG-1S was detected in nuclear protein and its expression increased gradually within 24 hours, the interaction between BAG-1L and GR was observed in nucleoli, and the transcriptional activation of GR decreased, with a negative correlation between BAG-1L expression and GR activity. Conclusion:LPS and Dex acting on rat alveolar macrophages, the expression of BAG-1L increases, which, coupled with GR,translocates into nucleoli and inhibits GR activity. This might be the important mechanism that underlies glucocorticoid resistance in inflammation.

Objective:To investigate the protective effect of Momordica charantia polysaccharide (MCP) on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice and the possible mechanism. Methods:MCP was extracted from Momordica charantia (MC). Fifteen C57BL/6J mice were randomly divided into three groups with five in each group: control group, DSS group and DSS+ MCP group. The body weight and disease activity index (DAI) of the mice were monitored every day. Mouse colon tissues and serum samples were collected. Pathological changes in intestinal tissues and the expression of inflammatory factors, CD4 + T cells, neutrophils and macrophages were analyzed by HE staining, ELISA, RT-qPCR and flow cytometry. Results:MCP alleviated the DSS-induced UC in mice by restoring body weight and stool consistency and reducing bleeding. Moreover, MCP could repair the mucosal barrier function of colon tissues, decreasing inflammatory cell infiltration and lessening the edema in mucosal layer and muscle layer, and therefore protect the damaged intestinal tract of mice. The expression of inflammatory cytokines (TNF-α and IL-1β) and the level of CD4 + T cells were decreased in the colonic tissues of MCP-treated mice. Conclusions:MCP ameliorated DSS-induced UC in mice through inhibiting weight loss, repairing colonic tissue damage, improving immune system disorder and decreasing the expression of inflammatory cytokines. This study provided reference for further study of MCP as a potential dietary intervention in the treatment of UC.

Objective:To analyze the current situation and requirements of education for health technicians in maternal and child health care institutions, and put forward feasible strategies and measures to improve the comprehensive quality and professional level of the talent team of maternal and child health care institutions. Methods:Questionnaire survey was carried out on education needs of health technical staff of 11 maternal and child health care hospitals in 4 provinces (regions), and provincial, municipal and district-level medical institutions. The survey results were recorded by Epidata 3.1. SPSS 22.0 software was used for statistical analysis. Results:A total of 1 678 questionnaires were included in the analysis. A total of 1 313 people received training, accounting for 78.2%. The main reason for not receiving training was that the unit didn't arrange (180 people), accounting for 49.3%(180/365). There were 779 people who had more than 3 days of training, accounting for 59.3%. There were 384 people who were trained in superior general hospitals, accounting for 29.2%, and 268 people were trained in superior maternal and child health institutions, accounting for 20.4%. There were 837 people who learned the content of new professional progress, accounting for 50.8%(837/1 648). According to the interview, there were still some requirements for thematic training, further education, online learning, continuing education and standardized training. Conclusion:Maternal and child health care institutions have accelerated the construction of professional personnel, intensified training, and thoroughly implemented health personnel training programs, established a long-term mechanism, increased funding, improved training content, ensured the quality of training, and made a good job in hierarchical training to meet the learning needs of personnel at all levels. This is of great significance for strengthening the technical personnel of maternal and child health care institutions and improving their service capacity.

【Objective】 To review the scientific research advances in the field of preschool children′s nutrition from 2012 to 2022, in order to provide reference for relevant policy formulation and further scientific exploration in this area. 【Methods】 High-quality literature published both domestically and internationally between 2012 and 2022 was retrieved through Web of Science (WOS) and China National Knowledge Infrastructure (CNKI) database. Excel and CiteSpace software were applied to visualize the research hotspots and frontier trends. 【Results】 A total of 1 816 English articles and 110 Chinese articles were included in the analysis. The cooperation network density of research institutions and authors was found to be low. Excluding the search terms, the top three English keywords were vitamin A (0.74), intervention (0.67), obesity prevention (0.55); the top three Chinese keywords were "anemia" (0.41), "obesity" (0.38), and "nutritional status" (0.18). In Chinese and English clusters, "anemia" and "physical activity" ranked first, respectively. The latest burst words identified were "dietary pattern" and "diet quality". 【Conclusions】 The number of English literature published in the field of preschool children′s nutrition is increasing, but it has shown a downward trend in the past three years in China. There are few studies in this field in China. The research hotspots primarily focus on the nutritional status of preschool children, with less attention paid to intervention research and methodological exploration. Further research in the field of preschool children′s nutrition in China may need to focus on the external determinants of preschool children′s nutrition, such as parental education, diet and food supply.
