1.Association of non-HLA gene polymorphisms with CMV infection after hematopoietic stem cell transplantation

Objective To explore the non-HLA gene polymorphisms that influence CMV infection after hematopoietic stem cell transplantation (HSCT).Method Non-HLA gene (ACE,CD14,MPO,MBL) single nucleotide polymorphisms were determined by using sequence-specific primer polymerase chain reaction (PCR-SSP) and sequencing in 64 pairs of donors and recipients before HSCT and the differences of non-HLA gene were analysed in CMV positive and negative patients Results The distribution of ACE gene single nucleotide polymorphism was DD (14/128,10.9%),ID (72/128,56.3%),and Ⅱ (42/128,38.8%).The distribution of CD14-159 allele gene single nucleotide polymorphism was CC (18/128,14.1%),CT (81/128,63.3%),and TT (29/128,22.7%).The distribution of MPO-463 allele gene single nucleotide polyrnorphism was G (100)/128,78.1%),A (2/128,1.6%),and GA (26/128,20.3%).The distribution of MBL gene single nucleotide polymorphism was H (28/128,21.9%),HL (73/128,57.0%),L (27/128,21.1%),Y (87/128,68.0%),YX (38/128,29.7%),X (3/128,2.3%),A (94/128,73.4%),AB (32/ 128,25.0%),and B (2/128,1.6%).The allele frequency of ACE,CD14 and MPO shoed no significant differcence between CMV positive and negative patients The gene frequency of MBL-HL was increased in CMV positive group.Conclusion MBL gene single nucleotide polymorphisrns may influence CMV infection after HSCT.
2.Influence of home nursing intervention on quality of life for stroke nasal patients with indwelling gastric tube

Objective To explore the influence of home nursing intervention on the adverse events and the ability of caregivers in cerebrovascular disease nasal patients with indwelling gastric tube. Methods Totals of 63 cases with nasal feeding discharge from September 2011 to August 2012 from our department were selected as the control group and 68 cases with nasal feeding discharge from September 2012 to August 2013 from our department were selected as the intervention group. The general information of patients in two groups, the incidence of nasal mucosa mechanical injury and the complications of indwelling gastric tube were retrospectively analyzed. The relevant knowledge and ability of caregivers were compared. Results The incidences of gastric tube out, reflux and aspiration, blocking tube, the nasal mucosa mechanical injury, the complications of constipation, diarrhea, gastric retention, malnutrition in the intervention group were 7. 4%, 0. 0%, 0. 0%, 2. 9%, 2. 9%, 23. 5%, 41. 2%, 14. 7% and 14. 7%, respectively. Those incidences in the control group were 30. 1%, 36. 5%, 36. 5%, 25. 3%, 23. 8%, 58. 7%, 71. 4%, 41. 3% and 36. 5%, respectively. The incidences of adverse events of the intervention group were significantly lower than those of the control group (χ2 =11. 37, 30. 11, 30. 11, 13. 91, 12. 61, 16. 82, 9. 41, 11. 58, 8. 25, respectively;P <0. 05). The extent of gaining the relevant knowledge and the ability of caregivers in the intervention group were 91. 2%, 88. 2% and 85. 3%, respectively. Those incidences in the control group were 52. 4%, 41. 3% and 55. 6%, respectively. The incidences of the intervention group were significantly higher than those of the control group (χ2 =24. 70, 31. 99, 14. 04, respectively;P <0. 01). Conclusions The home nursing intervention can improve the treatment compliance of patients and the ability of family caregivers. It can also avoid and reduce the occurrence of adverse events, therefore it can enhance the quality of life of the patients.
3.Coagulation function changes after CD19-CAR-T cells immunotherapy for B-ALL and its related factors

Objective:To investigate the changes of various cytokines and coagulation function in B cell acute lymphoblastic leukemia(ALL) patients with different CRS scores during CD19-CAR-T cell immunotherapy.Methods:87 patients with B-ALL hospitalized in the First Affiliated Hospital of Soochow University and 30 normal controls were enrolled into this study from July 2018 to October 2020. The age of the patients was 32(20, 56) years old and 36(41.4%) were female. All these coagulation indicators, prothrombin time (PT), activated partial thromboplastin time (APTT), D-dimer, fibrinogen (Fg) were analyzed by automatic blood coagulation in B-ALL patients before and after treated with CAR-T cell. The ratio of CD19-CAR-T cells and the expression of IL-6, IL-10, IFN-γ, TFN-α, IL-2, IL-4, and IL-17A were analyzed using flow cytometry. The patients′ clinical parameters were detected, and the CRS classification of severity was made according to the standard of consensus.Results:Patients with CRS>3 had prolonged PT and APTT, increased D-dimer, and decreased fibrinogen ( P<0.05). The levels of cytokines of IFN-γ, IL-6, and IL-10 were significantly higher in patients with CRS>3 than that in controls ( P<0.05).The D-dimer level is positively correlated with IL-10. Conclusion:Patients with severe CRS grading have significant coagulation dysfunction in CD19-CAR-T cell immunotherapy. Cytokines IFN-γ, IL-6, and IL-10 may affect coagulation function and CRS grading during CD19-CAR-T cell immunotherapy.
4.Effect of vessel number on computational fluid dynamics in vascular networks

BACKGROUND:Mechanical factors can affect the angiogenic ability of vascular endothelial cells.How the vessel number affects the hydrodynamic properties of microvessels remains to be clarified. OBJECTIVE:To investigate the influence of vessel number on the hydrodynamics of vascular networks based on computational fluid dynamics. METHODS:Three three-dimensional models of vascular network with different vessel numbers were constructed using the Geometry module of ANSYS 19.0 software,and then the vascular network was meshed to tetrahedral elements in Mesh module.The vascular network was assumed to rigid wall without slip,and the blood was assumed to laminar,viscous,and incompressible Newtonian fluid.Blood density,velocity,and a series of blood viscosity coefficients were also established.The Navier-Stokes equation was used for calculation.Hydrodynamic properties of different parts of vascular network with different vessel numbers were analyzed and compared. RESULTS AND CONCLUSION:The streamline,velocity,and mass flow all had the same trend in the vascular network,that is,the outlet and inlet were higher and the middle junction of vascular network was lower.The more the number of vessels,the thinner the blood flow lines in each part of the vascular network.Also,the velocity,mass flow,and wall shear decreased with the increase of the number of blood vessels.Therefore,the changes in vessel number could influence the hydrodynamic environment in the vascular network.Computational fluid dynamics indicates that the changes in vessel numbers can influence the hydrodynamic properties of blood,and provides a new idea for treating bone hypoperfusion-induced diseases(fracture nonunion,bone defect,osteoporosis,etc.)through tonifying kidney and activating blood circulation based on the coupling between angiogenesis and osteogenesis.
