1.Molecular features of CD71 expression in renal tissue in IgA nephropathy

Objective To study the molecular pathologic features of CD71 and the correlation between CD71 expression and IgA deposit in IgA nephropathy(IgAN),and to investigate the role of CD71 in pathogenesis of IgAN in order to direct the therapeutic strategies.Methods According to clinical manifestations and pathological diagnosis,170 cases of renal biopsy were divided into IgA deposition nephritis(primary group),non-IgAN with IgA deposition(secondary group)and no IgA deposited nephritis(control group).Immunofluorescence double-staining using TRITC-conjugated anti CD71 antibody and FITC-conjugated anti-human IgA antibody was performed to illustrate the expression of CD71 and IgA respectively by confocal microscopy.Results No CD71 expression was found in control group,while the CD71 was positively expressed in primary and secondary groups to various levels.The intensity of expression of CD71 in primary group was more intense than that in the secondary group.The expression of CD71 in the IgAN tissues correlated closely with the accumulation of IgA.CD71 and IgA were distributed in mesangial area and capillary lumina of glomeruli of IgAN(primary group).The study showed that CD71 expression and IgA deposition were co-localized under confocal fluorescence microscopy.Conclusion CD71 is highly expressed in the glomeruli of primary IgAN.The expression of CD71 in the IgAN tissues correlats closely with the accumulation of IgA.CD71 and IgA are co-localized under confocal fluorescence microscopy.The present study reveals that CD71 is one of IgA receptors and involves in the course of IgAN.The results suggests that CD71 may play an important role in immunopathogenesis of IgAN.
2.Orthokeratology for treating myopia:a report of clinical application

Objective To make a preliminary analysis of orthokeratology(OK) for treating myopia.Methods OK lenses were chosen to treat myopia with or without astigmatism according to the myopia characterizations, then changes of their visions, refractions, corneal curvature and eccedtricities after being treated for ld, 1、2、3、4 weeks were observed once every 4weeks and were followed up for 18 months.Results Significant changes were observed after being treated. Slight myopia: the diopters decreased-2.75D±0.50D,the horizontal meridians flattened 1.64±0.21, the vertical 0.92±0.27 and the eccedtricities declined 0.47±0.11; Middle myopia: the diopters decreased -5.50D±0.75D,the horizontal meridians flattened 2.16±0.45, the vertical 1.61±0.67, and the eccedtricities declined 0.38±0.07; High myopia:the diopters did -6.75D±0.50D,the horizontal did 3.86±0.56, the vertical 2.97±0.67 and the eccedtricities did 0.32±0.10. The predicabilities of slight and middle myopia were superior to high myopia's.Conclusion Orthokeratology is a non-traumatic therapy and the clinical application should be careful and scientific.
3.Effect of He-Ne laser verus low-level CO2 laser irradiation on accelerating fracture healing

BACKGROUND: The effects of low-level laser biostimulation have been proved by various experimental studies and clinical application, which are manifested as tissue repairing, analgesia, antiinflammation, etc.OBJECTIVE: To observe the effects of He-Ne laser and low-level CO2 laser irradiation on fracture healing.DESIGN: A randomized controlled animal experiment. SETTING: Research Room of Laser Medicine, Chengde Medical College.MATERIALS: Forty-eight healthy male New Zealand rabbits, weighing 2.0-2.5 kg, were used.METHODS: The experiments were carried out in the Research Room of Laser Medicine, Chengde Medical College from 1998 to 2003. ①The 48 rabbits were induced into models of experimental fracture of the left radius, and then they were divided into three groups with 16 rabbits in each group: control group, He-Ne laser irradiation group and CO2 laser irradiation group. The fracture areas of the animals in the He-Ne and CO2 laser irradiation groups were respectively irradiated with 28 mW/cm2 He-Ne laser and 150 mW/cm2 CO2 laser for 10 minutes, once a day. ② The animals were killed on the 15th and 35th days after taking roentgenogram respectively. At 15 days, the collagen and calcium contents in callus were detected. At 35 days, the biomechanic anti-torsion strength was examined.MAIN OUTCOME MEASURES: ①Results of roentgenogram in each group; ② Collagen and calcium contents in callus; ③ Results of biomechanic anti-torsion strength.RESULTS: Totally 45 rabbits were involved in the analysis of results. ① On the 15th and day after fracture, the roentgenogram, collagen and calcium contents in callus in the He-Ne and CO2 laser irradiation groups were better than those in the control group, and the collagen and calcium contents in callus were higher in the CO2 laser irradiation group than in the He-Ne laser irradiation group [(341.9±30.1) vs (302.1±28.7) mg/g; (197.1±19.7)vs (156.5±17.6) mg/g, P< 0.05]. ②On the 35th days after fracture, the biomechanic anti-torsion functions in the He-Ne and CO2 laser irradiation groups were superior to that in the control group, and there were no significant difference between the two laser irradiation groups. ③ The results of roentgenogram in the He-Ne and CO2 laser irradiation groups were better than that in the control group both on the 15th and 35th days after fracture, and there was no significant difference between the two irradiation groups.CONCLUSION: Both CO2 laser and He-Ne laser irradiations can accelerate the process of fracture healing, and the curative effect of CO2 laser is better than that of He-Ne laser on the 15th day after fracture, but there is no significant difference clinically between the CO2 laser and the He-Ne laser irradiations on the 35th day after fracture.
4.The effects of pravastatin on serum adipocytokines in patients with IGT and metabolic syndrome

Objective To investigate the effects of pravastatin on the blood biochemical index,the levels of serum resistin,tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in patients with impaired glucose tolerance (IGT) and metabolic syndrome (MS).Methods 60 patients with IGT and MS were random treated with pravastatin or remedial life-style intervention,and the levels of lipids profiles,insulin resistance index (HOMA-IR),serum resistin,TNF-α and IL-6 were measured before and after treatment.Results The levels of total cholesterol [ (4.45 ±0.60)mmol/L vs (5.58 ±0.96) mmol/L,t =-5.42,P <0.01],HOMA-IR [3.22 ±0.64 vs 3.58 ±0.71,t =-2.05,P <0.05) ],resistin[ (1.97 ±0.72) μg/L vs (2.76 ±0.73) μg/L,t=-4.26,P <0.01 ],TNF-α[ (9.36 ±2.03) μg/L vs (13.87 ±2.30)μg/L,t =-8.06,P <0.01] and IL-6[ (3.50 ±0.99) μg/L vs (6.32 ±1.17) μg/L,t =-10.06,P <0.01 ] in pravastatin group were significantly lower than those in life-style intervention group.Conclusion Pravastatin could improve insulin sensitivity of the patients with IGT and MS,significantly decreased the serum levels of resistin,TNF-α,IL-6,and it had anti-inflammatory effect.
5.Advances in Surgical Management of Primary Venous Reflux Disease of Lower Extremity

Objective To evaluate the effect of surgical management of primary venous reflux disease in lower extremity. Methods Literatures about the surgical management of venous reflux disease in lower extremity were collected and reviewed. Results In patients with primary venous reflux disease,comprehensive surgical management of concomitant superficial,deep and perforating veins might greatly improve the clinical effect.Conclusion Comprehensive surgical management will be the direction of surgical management of primary venous reflux disease in lower extremity.
6.EFFECT OF FLAVONOIDS FROM SEED RESIDUES AND OIL OF HIPPOPHAE RHAMNOIDES L.ON SERUM LIPID OF FEMALE AGED RATS WITH OBESITY

Objective: To study the effect of flavonoids from seed residues of Hippophae rhamnoides L.( FSH) and Hippophae rhamnoides L. oil(HRO)on serum lipid in female rats with obesity. Methods Female aged rats with obesity were given FSH and HRO by oral administration for 6 w. Serum triglyceride(TG) and cholesterol(TC) were determined every 2 w. The serum free fatty acid (FFA), high-density lipoprotein cholesterol (HDL-C),body weight and Lee’s index were measured after 6 w. Results: Serum TG and FFA level decreased significantly in rats given FSH. At the same time, FSH could inhibit the increase in body weight and Lee’s index. HRO could regulate the lipid metabolism and decrease serum TG level significantly in female aged rats with obesity. Conclusion: Both FSH and HRO can improve lipid metabolism in female aged rats with obesity.
7.Factors Influencing Hospital Pharmaceutical Care

METHODS: We analyzed and summarized the problems appeared in the practical work and the factors influencing pharmaceutical care. RESUL-TS & CONCLUSION: The factors that influence pharmaceutical care mainly consist of policy, idea, specialty and method. Att-aching great importance and having a clear understanding to the above factors is the key to facilitate the successful development of hospital pharmaceutical care.
8.Production and characterisation of monoclonal antibodies against 19-Nortestosterone.

OBJECTIVETo produce anti-19-Nortestosterone (NT) monoclonal antibodies and identify their immunological characteristics.
METHODSHybridomas were prepared by fusing NS0 mouse myeloma cells with splenocytes isolated from immunized BALB/c mice. Noncompetitive and competitive indirect ELISA were employed to screen positive cell clones. A caprylic acid ammonium sulphate (CAAP) method was used to purify NT mAb, and the Batty saturation method was used to determine the affinity constant (Kaff).
RESULTSFive hybridoma cell lines, named NT-1, NT-2, NT-3, NT-4, and NT-5, were identified and their corresponding mAbs were of the IgG(1) isotype with a k light chain. The Kaffs of all mAbs were between 2.6 and 4.7 × 10(9) L/mol. The titers and IC(50) values of purified ascite fluids were in the range of (0.64-2.56) × 10(5) and (0.55-1.0) ng/mL, respectively. Of all the cross-reacting steroids, (-NT was the most reactive with the mAbs at 62% with NT-1 mAb and 64% with NT-2 mAb. Negligible cross-reactivity (<0.01%) with other steroids was observed.
CONCLUSIONThe establishment of these hybridomas allows the potential development of a rapid test kit, and may provide an alternative method for the detection of NT residues in food producing animals.

9.Recent progress in fusion enzyme design and applications.

Engineering and redesign of enzymes are important to industrial biocatalysis. Fusion enzyme technology, based on fusion protein design, is frequently used in multifunctional enzyme construction and enzyme proximity control. Here, we reviewed the recent progress in molecular design strategy and application studies of fusion enzymes. The concept and features of fusion enzymes were introduced, followed by a systematical summary of the design strategy of fusion enzymes. In particular, the effects of different linker properties on fusion enzymes and their possible mechanisms were discussed. In addition, recent studies on fusion enzyme applications were also discussed. Finally, based on our own studies on fusion enzymes and the current research progress, the key problems in fusion enzyme technology and perspectives of this field were discussed.

10.Recanalization for symptomatic chronic internal carotid artery occlusion: a preliminary study

Objective:To evaluate the value of endovascular recanalization and hybrid recanalization for chronic internal carotid artery occlusion(COICA), and to evaluate its feasibility, safety, success rate, and clinical outcomes.Methods:Totally 35 patients who received endovascular recanalization or hybrid recanalization with symptomatic COICA were enrolled from January 2019 to December 2019 in Department of Cerebrovascular Disease,Zhengzhou University People′s Hospital. The clinical characteristics, treatment strategies, success rate, and major events of the patients were analyzed retrospectively.Results:Thirty of 35 patients were successfully recanalized. Among them, hybrid recanalization was performed in 3 cases, carotid endarterectomy was performed in 1 case, and endovascular recanalization was performed in 26 cases, 5 patients failed because they could not reach the distal true cavity. Among the successful patients, 5 patients had operation-related complications, 3 patients had embolism cerebral infarction, 1 patient had hyperperfusion cerebral hemorrhage, 1 patient still had transient ischemic attack after operation. All patients were followed up clinically, 2 patients had reoccurrence of obstruction, 2 patient had restenosis, the remaining patients had no hemodynamic stenosis or reocclusion.Conclusion:In highly-selected cases, intracavitary recanalization for symptomatic COICA is feasible, relatively safe and effective.
