1.Effect of Xuebijing on lung fibrosis in a rat model of oleic acid-induced lung injury

ObjectiveTo investigate the effect of Xuebijing (Chinese herbal medicine preparation) on lung fibrosis in a rat model of oleic acid - induced lung injury.MethodsSixty SPF level of male Wistar rats were randomly ( random number) divided into three groups ( n =20 ):normal control group ( group A),oleic acid model group (group B),oleic acid and Xuebijing group (group C).Rats in group A were injected with normal saline (0.13 ml/kg) ; acute lung injury (ALI) model rats were injected with oleic acid (0. 13 ml/kg) intravenously in B and C groups.Twelve hours after administrations of saline or oleic acid into rats of three groups separately,samples of arterial blood from all rats were taken for blood gas analysis.By analysis of blood oxygen partial pressure,the acute lung injury model successfully made was confirmed.Then the rats in group C were treated with xuebijing (4 ml/kg) intravenously,once a day from the 1st to 7th day.The rats in groups A and B were treated with saline (4 ml/kg).Five rats in each group were randomly sacrificed on the 1st,7th,14th,28th day,separately.The bronchoalveolar lavage (BAL) of left lung of all rats was carried out after weight of lung weighed.The lefi lungs of rats were weighed in wet and dry in order to get the wet/dry ratio.Venous blood and BAL fluid (BALF) were obtained for detection of procollagen Ⅲ ( PC Ⅲ ) concentrations with enzyme - linked immunosorbent assay (ELISA).Right upper lobe of lung specimens was taken for histopathological examination after paraffin - embedded sections with hematoxylin-eosin (HE) staining and Masson staining.Multiple comparisons were analyzed by using oneway analysis of variance (SNK-q test).Statistical analyses were performed by using SPSS 13.0 software.ResultsOxygenation index of groups B and C ( PaO2/FiO2 < 300) significantly decreased compared with the group A ( P < 0.01 ).Levels of serum PC Ⅲ on the 14th day and 28th day in group C decreased significantly in comparison with group B ( P < 0.05 ).Compared with group A,the levels of BALF PC Ⅲ and serum PC Ⅲ in group B and group C on the 7th day,14th day and 28th day increased ( P < 0.01 ).Levels of PC Ⅲ in BALF on the 7th day,14th day and 28th day in C group decreased in comparison with group B ( P <0.01 ).Compared with group A,the levels of PCⅢ in BALF in group B and group C on the 7th day,14th day and 28th day increased ( P < 0.05 ).ConclusionsThe result suggests that treatment with xuebijing may be useful in inhibiting pulmonary fibrosis during the course of ALI in rots.
2.The levels of serum myocardial enzymes and high density lipoprotein cholesterol in children with septic shock

ObjectiveTo investigate the changes of serum myocardial enzymes and high density lipoprotein cholesterol(HDL-C) in children with septic shock.MethodsFifty-two children with septic shock in our hospital from Jan 2006 to Mar 2011 were divided into severe septic shock group( n =27 ) and mild septic shock group ( n =25 ).The serum creatine kinase-MB ( CK-MB ),α-hydroxybutyrate dehydrogenase (α-HBDH),lactic dehydrogenase(LDH) and HDL-C were analysed in both two groups on 1,2,4,7,10,15 d,and compared with 32 normal children as control group.Meanwhile,the children's condition,death cases and cured time were recorded.ResultsThe levels of serum CK-MB [ ( 32.084 ± 4.595 ) U/L,( 61.481 ±5.639) U/L],α-HBDH/LDH(0.694 ±0.080,0.884 ±0.079) in mild and severe septic shock groups were higher than those of normal control group[ (21.675 ± 3.453) U/L ] and (0.443 ± 0.065 ) ( P < 0.01 ) ;the levels of serum HDL-C[ (0.646 ±0.067) mmol/L,(0.310 ± 0.124) mmol/L ] in the two septic shock groups were decreased significantly than that of control group[ ( 1.012 ±0.156) mmol/L] (P <0.01 ).Five cases died in the severe septic shock group while no cases died in mild septic shock group.The times of CK-MB recovered[ (9.82 ±1.76) d vs (4.68 ± 1.22) d],α-HBDH/LDH recovered[ (7.23 ± 1.38) d vs (3.76 ± 0.83 ) d] and HDL recovered[ ( 12.14 ± 2.21 ) d vs ( 6.48 ± 1.33 ) d ] were more slowly in severe septic shock group than those of mild septic shock group( P < 0.01 ).ConclusionThe higher of the serum CK-MB and α-HBDH/LDH,and the lower of HDL-C in septic shock children,the disease is more severe,the recovery time is longer and a worse prognosis may occur.
3.Efficacy of palace production technique of puerpera with gestational hypertension by ropivacaine combined with sufentanil and epidural anesthesia

Objective To observe the efficacy of palace production technique of Puerpera with gestational hypertension by ropivacaine combined with sufentanil and epidural anesthesia.Methods77 cases with hypertensive disorders in pregnancy from January 2014 to January 2015 hypertensive, 39 cases were randomly divided into observation group and the control group of 38 patients, the control group received ropivacaine anesthesia, the observation group received ropivacaine combined with sufentanil and epidural anesthesia, observed two groups of mothers onset time of anesthesia, operative time, pain duration, neonatal apgar score, postoperative pain scores and adverse reactions.ResultsThe onset time of anesthesia, analgesia duration, and postoperative pain scores were significantly better than the control group, the difference was significant (P<0.05);two groups of mothers in the operation time, score and adverse neonatal apgar, the total incidence the difference was not statistically significant.ConclusionRopivacaine combined with sufentanil CSEA anesthesia for hypertensive disorders in pregnancy cesarean exact effect is better than alone ropivacaine spinal anesthesia program, and no obvious side effects and is worthy of further promotion and application.

5.The clinical study of dynamic detection of lymphocyte subsets in children with septic shock

Objective To investigate the dynamic changes of lymphocyte subsets (lymphocyte count, CD3 +T,CD4 +T,CD8 +T,CD19 +B,CD56 +NK)in children with septic shock.Methods Peripheral blood lymphocyte subsets were analysed in 25 cases with severe septic shock and 24 cases with mild septic shock on day 1 ,3,8,and compared with those of 25 healthy volunteers.Children with severe septic shock were divided in-to survival group and dying group according to the outcome.The detection data were compared.Results In the first day of admission,peripheral blood lymphocyte subsets in children with septic shock decreased significantly compared with the normal children,and there was significant difference in the three groups(P <0.05 ).In the third day of admission,children with mild septic shock had statistical increase of CD4 +T,CD3 +T and lympho-cyte count(P <0.05),and children with severe septic shock had statistical decrease of lymphocyte subsets ex-cept CD56 +NK.In the eighth day of admission,children with mild septic shock had no significant difference in CD4 +T lymphocyte count compared with control group(P >0.05),but children with severe septic shock had statistical increase of CD4 +T,CD3 +T and lymphocyte count(P <0.05).There were 5 cases died who had se-vere septic shock in day 2 to day 5 after admission.The count of lymphocyte subsets were detected significantly lower in dying group than survival group in the third day of admission(P <0.05).Conclusion The continuous low level of lymphocyte subsets indicates bad outcome.The patientˊs condition improved with increase of CD4 +T,CD3 +T,lymphocyte count.CD4 +T is the most sensitive.Dynamic detection of lymphocyte subsets in the pe-ripheral blood of children with septic shock is of great clinical significance in judging the severity of disease and cura-tive effect.
6.Relationship between Calpain10 gene polymorphisms and diabetes in familial type 2 diabetes mellitus of Uygur in Xinjiang

Objective To analyse the relationship between Calpain10 gene UCSNP43 and UCSNP44 poly morphisms and diabetes in familial type 2 diabetes mellitus of Uygur in Xinjiang.Methods Sixty-two Uygur type 2 diabetic families were enrolled.The polymorphisms of Calpain10 gene UCSNP43 and UCSNP44 were measured by SNP-stream of gene chip.Results (1) The heritability of the first degree relative and the second degree relative in Uygur familial type 2 diabetes were 0.578 and 0.032.(2)There was no difference between UCSNP43 and UCSNP44 polymorphisms of the first and second degree relatives in familial type 2 diabetes mellitus.(3)The carriers of USCNP43 AA genotype had higher body mass index and fasting insulin than GG and GA genotypes carriers.Conclu sion With the downgrading of the familial relationship,the heritability of type 2 diabetes mellitus was decreasing. The carriers of UCSNP43 AA genotype might have higher insulin resistance than those with GG and AG genotypes.

To explore the relations of Morphology Immnuophe-notype Cytogenetics Molecular biology(MICM) detection on diagnosis andtreat,emt of leukemia. Methods: 68 cases of leukemia patients had beenanalyzed by morphology(FAB). Immunohistochemistry(Flow Cytometry, FCM). chromosome G banding technique and dual-color fluorescence insitu hybridization (D-FISH).Technique:All patients were treated bychemotherapy. T test and X2 test of significance. Results: 7 cases have acute leukema aberration antigen expression. 5 out of 47 cases acutemyeliod leukemia patients accompany lymhocytic interrelated antigenexpression. 2/l5 cases acute lymphoid leukemia accompany myelocyteinterrelated antigen expression. 2 cases acute lmphoid leukemia are T cell and B cell interrelated antigen mingle expression. had been examined46,XY,t(8,2l) translocation of chromosome and bcr/abl fusion genes inthe acute leukemia patients. 16 out of 20 chronic myeloid leukemia patientshad philadelphia chromosome. 7 out of 20 patients had complicate karyotype. 5 out of 20 patients had bcr/abl fusion gene, l out of 4 patient had bcr/abl fusion gene that Ph chro mosome showed negative in CML. 3/4 cases patients had complicate chromosome. The ratio of CR use l time chemotherapy and the total ratio of CR using 2 times chemotherapywith aberration antigen expression in acute leukemia was significantly lessthan those of the acute leukemia patient had single system antigenexpression(P<0.05). The time of CML-BC with complicate c hromosome karyotype was significantly short than those of Ph showed negative in CML(P<0.05). Conclusion: The MICM classification is more acc urate for diagnosis of leukemia and more significant in guiding the leukemiatreamen.
8.Anticoagulant chitosan nanoparticles:synthesis, characterization and biological safety

BACKGROUND:Studies have shown that chitosan and other natural polysaccharides have heparin-like anticoagulant function after sulfonated modification. Sulfonated chitosan has good anticoagulant property because the sulfonate group formed by sulfonated chitosan is similar with the active group of heparin. OBJECTIVE: To prepare the anticoagulant chitosan nanoparticles and to detect its morphology, physical and chemical properties and biological security. METHODS: Chitosan nanoparticles were synthesized by emulsion-chemical cross link. Sulfonated chitosan nanoparticles were synthesized by sulfonation reaction. Its morphology was described by transmission electron microscope. The peak-value change of its specific groups was observed by infrared spectroscopy. (1) Coagulation experiment: Heparin, chitosan nanoparticles and 10, 30 and 50 mg of sulfonated chitosan nanoparticles were added into the blood of Spraque-Dawley rats. The coagulation indicators were detected. (2) Hemolysis experiment: deionized water, physiological saline and 10, 30, 50 g/L sulfonated chitosan nanoparticles extracts were added into 2% red blood cel suspension of rabbits. The hemolysis rate was detected. (3) Cytotoxicity experiments: DMEM medium containing fetal bovine serum and 10, 30, 50 g/L sulfonated chitosan nanoparticle extracts were used to culture human umbilical vein endothelial cels. Cel relative growth rate and toxicity grading were detected after 72 hours. RESULTS AND CONCLUSION: Scanning electron microscopy showed that sulfonated chitosan nanoparticles had good morphology, with a diameter of 50 nm. Infrared spectroscopy showed that the sulfonated replacement occurred.In vitro coagulation experiments showed that sulfonated chitosan nanoparticles had significant anticoagulant effects in a dose-dependent manner. Sulfonated chitosan nanoparticles meet the national safety standard for hemolysis rate of less than 5%, non-induced hemolysis property. Cytotoxicity assays showed that sulfonated chitosan nanoparticles extracts had no significant cytotoxicity, and its biological safety was in line with the national standards.
9.The effects of Saccharomyces boulardii powder on preventing antibiotic-associated diarrhea in children with pneumonia

Objective The aim of this randomized,double-blind,placebo-controlled trial was to evaluate the efficacy of the Saccharomyces boulardii (S.boulardii) power in preventing antibiotic-associated diarrhea (AAD) in hospitalized children with pneumonia.Methods Two hundred and forty-five hospitalized children with pneumonia were randomly divided into two groups,group 1 treated with antibiotics and S.boulardii power (n =104),and group 2 with antibiotics and placebo,along with S.boulardii power and zinc when suffered from diarrhea (n =141).All children received montmorillonite powder and fluid replacement therapy for diarrhea.The stool frequency and consistency were observed.Diarrhea incidence,diarrhea and pneumonia duration of treatment,and therapeutic effect were recorded.Results AAD is more prevalent in group 2 than in group 1 (P <0.05).A 55.7% relative risk reduction in AAD was seen in group 1 compared with group 2 (RR =0.443,95%CI 0.233 ~ 0.845,P =0.012).Patients with preventive treatment of S.boulardii power in broup 1 had lower AAD incidence compared with those in group 2 (P < 0.05).Both the diarrhea and pneumonia duration of treatment in group 1 were shorter than that in group 2(P =0.001 for diarrhea duration,P =0.030 for pneumonia duration,respectively).No drug related adverse reactions were found during the observation period.Conclusion Preventive use of S.boulardii power may effectively reduce the risk of AAD,improve therapeutic effect,and shorten hospital stays for hospitalized children with pneumonia.
10.Protective effect of glucocorticoidson on rat lung in a rat model of oleic acid-induced acute respiratory distress syndrome lung injury

Objective To investigate the effect of glucocorticoid on rat lung in a rat model of oleic acid-induced acute respiratory distress syndrome(ARDS)lung injury. Methods Sixty SPF level of healthy male Wistar rats were randomly divided into normal control group(group A),oleic acid model group(group B)and oleic acid and Methylprednisolone group(group C),and each group with 20 cases rats. ARDS model rats were established by injection with oleic acid(0. 14 ml/ kg)intravenously. Twelve hours after injection,aterial blood was drawn for blood gas analysis of blood oxygen partial pressure in order to confirm that acute lung injury model was successful. Then the rats in group C were treated with injection of Methylprednisolone( 4 mg/ kg) intravenously,once a day,from the 1st to 7th day. The rats in A and B groups were given saline(4 ml/ kg). Five rats in each group were randomly sacrificed on the1st,7th,14th,28th day respectively. The left lung of all rats was underwent bronchoalveolar lavage after weighed. Bronchoalveolar lavage fluid( BALF)were obtained for detection of procollagen III(PCⅢ)concentrations with enzyme-linked immunosorbent assay( ELISA). Right lobe of lung specimens was used for histopathological examination. Results Compared with the group A,PaO2 in group B and C were decreased significantly(P < 0. 01),but there was no significant difference between group B and group C(P > 0. 05). The levels of PCⅢ concentrations of BALF in Group A,B and C group were(30. 61 ± 0. 52)μg/ L,(30. 14 ± 0. 41)μg/ L and(30. 71 ± 0. 74)μg/ L respectively on the first day,(29. 15 ± 0. 17)μg/ L,(34. 35 ± 0. 73)μg/ L and(31. 72 ± 0. 46)μg/ L respectively at the 7th day,(30. 23 ± 0. 41)μg/ L, (38. 61 ± 1. 21)μg/ L and(33. 32 ± 0. 57)μg/ L respectively at 14th day and(31. 12 ± 0. 22)μg/ L,(37. 53 ± 2. 46)μg/ L and(34. 45 ± 0. 27)μg/ L respectively at 28th day. Levels of PCⅢ concentrations of BALF at 7th, 14th and 28th day time point in group B and C were decreased in comparison of A group(P < 0. 05),and there was significant difference between B and C group(P < 0. 01). Conclusion A treatment with Methylprednisolone may be useful in inhibiting pulmonary fibrosis in the process of ARDS in rats.
