Object To solve the problem of low growth rate an d metabolism level in Taxus chinensis var. mairei (Lem?e et L?vl.) Cheng et L. K. Fu Methods Cells on logar it hmic phase (20 d) and stationary phase (30 d) were subcultured individually. Th euptake of carbohydrate, nitrogen and phosphate from the medium was examined, th e growth rate of Taxus cell and the synthesis of taxol were anal y zed too during the whole growth period Results The Taxus cell subcultured at logarithmic phase uptook carbohydrate and nit rate earlier than that subcu ltured at stationary phase. The special growth rate of the former was 1.5 folds of that of the latter. The production rate of taxol was increased by nearly 4 ti m es Conclusion Subculturing at logarithmic phase will benefi t the biomass accumulation and the taxol synthesis

OBJECTIVETo investigate the effect of intensive rosuvastatin therapy on adhesion molecules in patients with peripheral atherosclerosis and explore the possible upstream mechanism.
METHODSTwenty asymptomatic patients with peripheral atherosclerosis were enrolled and given 5-20 mg/day rosuvastatin for 3 months. Before and after the treatment, the lipid profile and plasma vascular cell adhesion molecule-1 (VCAM-1) levels were examined. The expression of intercellular adhesion molecule-1 (ICAM-1) in the mononuclear cells was measured using flow cytometry, and the mRNA and protein expressions of peroxisome proliferator-activated receptor γ (PPARγ) were detected using RT-PCR and Western blotting, respectively.
RESULTSCompared with the baseline levels, ICAM-1 expression decreased and PPARγ protein expression increased in the lymphocytes. Rosuvastatin therapy did not produce obvious effects on plasma VCAM-1 level or ICAM-1 expression in the monocytes in these patients.
CONCLUSIONRosuvastatin produces anti-inflammatory effects by decreasing the expression of ICAM-1 in mononuclear cells, and its upstream mechanism may involve the PPARγ pathway.

4.Optimization of Preparation Technology of Celastrol Nanostructured Lipid Carrier and Its Characterization

OBJECTIVE: To optimize the preparation technology of Celastrol nanostructured lipid carriers (Cel-NLC), and to characterize it. METHODS: Cel-NLC was prepared by melt-emulsification ultrasonic method. Based on single factor test, using encapsulation rate of Cel as index, the ratio of liquid lipid (the ratio of total mass), the amount of compound emulsifier and the dose of main drug were optimized by central composite design-response surface methodology. Validation test was conducted. Zeta potential and particle size of Cel-NLC that prepared by optimal prescription were determined by using granularity and Zeta potential analyzer. The morphology of liposome was observed by TEM. RESULTS: The optimal prescription included that the ratio of liquid lipid was 39%;the amount of compound emulsifier was 196 mg;the dose of main drug was 8 mg. The average encapsulation efficiency of 3 batches of Cel-NLC was 87.22%; average particle size was (41.2±1.1) nm,and average Zeta potential was (-18.4±0.2) mV (n=3). It was spherical under electron microscopy. CONCLUSIONS: The optimized technology is simple, stable and feasible, and it is suitable for the preparation of Cel-NLC.
5.Prevalence and correlates of HIV infection among cross-border families in Dehong Dai and Jingpo Autonomous Prefecture

Objective:To study the prevalence and correlations of HIV infection among cross-border couples in the Dehong prefecture.Methods:A cross-sectional mass screening study with questionnaire interview and HIV testing was conducted among 17 594 registered cross-border couples from May 2017 through June 2018.Results:Among 32 400 participants, the overall prevalence of HIV infection was 2.27% (736/32 400), 2.44% (375/15 372) for Chinese citizens, and 2.12% (361/17 028) for foreign spouses. Among all the 13 853 couples with both spouses receiving HIV testing, 13 415(96.84%) were seroconcordant-negative couples, 142(1.03%) were serocondordant-positive couples, and 296(2.13%) were serodiscordant couples, including 167(1.20%) couples with positive husband and negative wife and 129(0.93%) couples with positive wife and negative husband. Multiple logistic regression analyses indicated that HIV infection was associated with drug use and risky sexual behaviors for male spouses. In contrast, HIV infection was associated with risky sexual behaviors for female spouses.Conclusion:The prevalence of HIV among cross-border couples in Dehong prefecture is high, underscoring the urgent need to scale up HIV testing, prevention, and behavioral intervention.
6.A study on the psychological health status of migraine patients

Objective To investigate the clinical symptoms and psychological health status of migraine patients.Methods A total of 120 migraine patients admitted to Xingtai People's Hospital from September 14,2022 to July 30,2023 were selected and divided into mild pain group(0-3 points,51 cases),moderate pain group(4-6 points,48 cases),and severe pain group(7-10 points,21 cases)according to numerical rating scale.General clinical data,headache impact test-6(HIT-6),patient health questionnaire-9(PHQ-9),generalized anxiety disorder(GAD-7)and Pittsburgh sleep quality index(PSQI)were collected.Results The scores of HIT-6,PHQ-9,GAD-7,and PSQI of migraine patients were(56.33±11.30)points,(3.38±1.51)points,(3.01±0.58)points,and(4.73±0.78)points,respectively.There were statistically significant differences in the duration and total number of headache attacks among three groups(P<0.05).With the aggravation of headache degree,the duration of headache attacks increased and the number of attacks increased.There were statistically significant differences in the scores of HIT-6 and GAD-7 among three groups(P<0.05),and the scores of HIT-6 and GAD-7 increased with the aggravation of headache.Conclusion There are differences in the daily life impact,anxiety,pain duration and frequency of migraine in patients with different degrees of migraine.
7.Study on Preparation and in vitro Release Characteristic of Ursolic Acid/PF 127/TPGS-doxorubicin Mixed Nanomicelles

OBJECTIVE: To prepare Ursolic acid (UA)/Pluronic F127 (PF127)/TPGS-doxorubicin (DOX) mixed nanomicelles, and to characterize it and study its in vitro release behavior. METHODS: UA/PF127/TPGS nanomicelles were prepared by thin film hydration method. Using encapsulation efficiency of UA as index, combined with the results of single factor tests, L9(34) orthogonal test was used to optimize drug dosage of UA, molar ratio of PF127 to TPGS, hydration temperature and hydration volume, validation test was performed. On the basis of succinylated TPGS, TPGS-DOX was synthesized and mixed with UA/PF127/TPGS to prepare UA/PF127/TPGS-DOX mixed nanomicelles, the appearance, particle size and critical micelle concentration (PF127/TPGS) were investigated. The drug release behavior was examined by dialysis bag diffusion method. RESULTS: The optimal preparation technology of UA/PF127/TPGS nanomicelles was as follows as drug dosage of UA 8 mg, molar ratio of PF127 to TPGS 3 ∶ 7, hydration temperature 50 ℃, hydration volume 4 mL. Average encapsulation efficiency of UA in nanomicelles was 89.00% (RSD=0.43%, n=3). The prepared UA/PF127/TPGS-DOX mixed nanomicelles solution was clear with opalescence. The nanomicelles were spherical and uniform in size; average particle size was (115.00±9.42) nm; critical micelle concentration of PF127/TPGS (molecular ratio 3 ∶ 7) was 0.001 3%. The in vitro drug release of UA and DOX in the mixed nanomicelles was significantly slowed down, compared with raw materials or substance control. The drug release process of the two drugs in the nanomicelles conformed to Weibull equation. CONCLUSIONS: UA/PF127/TPGS-DOX mixed nanomicelles are successfully prepared with uniform particle size, good stability and good sustained-release effect.

Cashmere, also known as soft gold, is produced from the secondary hair follicles (SHFs) of cashmere goats. The number of SHFs determines the yield and quality of cashmere; therefore, it is of interest to investigate the transcriptional profiles present during cashmere goat hair follicle development. However, mechanisms underlying this development process remain largely unexplored, and studies regarding hair follicle development mostly use a murine research model. In this study, to provide a comprehensive understanding of cellular heterogeneity and cell fate decisions, single-cell RNA sequencing was performed on 19,705 single cells of the dorsal skin from cashmere goat fetuses at induction (embryonic day 60; E60), organogenesis (E90), and cytodifferentiation (E120) stages. For the first time, unsupervised clustering analysis identified 16 cell clusters, and their corresponding cell types were also characterized. Based on lineage inference, a detailed molecular landscape was revealed along the dermal and epidermal cell lineage developmental pathways. Notably, our current data also confirmed the heterogeneity of dermal papillae from different hair follicle types, which was further validated by immunofluorescence analysis. The current study identifies different biomarkers during cashmere goat hair follicle development and has implications for cashmere goat breeding in the future.
