1.Evaluation of skin barrier-related parameters in 116 healthy children

Objective To assess and compare physical and chemical properties of skin among different body areas of healthy children at different ages from Beijing.Methods A total of 116 healthy children were recruited from Beijing,China,and classified into four age groups.Tewameter TM300 was used to measure transepidermal water loss (TEWL) value,Corneometer CM825 to estimate stratum corneum hydration,and skinpH-Meter PH905 to determine skin surface pH,in the forehead,cheek and forearm.The Mexameter MX 18 probe to determine melanin index of the face.Analysis of variance was carried out to assess differences in these parameters among different measurement sites and age groups.Results TEWL value did not differ among different age groups or measurement sites (both P > 0.05).The stratum corneum hydration value of forehead,cheek and forearm was 51.53 ± 15.70,39.88 ± 10.48 and 50.33 ± 17.54 respectively in the age group < 1 year,49.95 ± 17.88,32.51 ± 12.09 and 36.10 ± 7.43 respectively in the age group 1-3 years,51.37 ± 10.60,31.65 ± 9.01 and 34.41 ± 8.21 respectively in the age group 4-6 years,49.74 ± 10.64,39.99 ± 50.43 and 29.35 ± 8.10respectively in the age group 7-12 years,with significant differences among different measurement sites (P < 0.05) but not between different age groups (P > 0.05).The cheek and forearm showed lower stratum corneum hydration value than the forehead.No statistical difference was observed in pH value at the same measurement sites between the age group < 1 year (forehead 5.27 ± 0.60,cheek 6.12 ± 0.51 and forearm 5.48 ± 0.45),1-3years (forehead 4.68 ± 0.58,cheek 6.80 ± 0.55 and forearm 5.07 ± 0.58),4-6 years (forehead 4.58 ± 0.37,cheek 5.70 ± 0.48 and forearm 5.09 ± 0.49),and 7-12 years (forehead 4.87 ± 0.51,cheek 5.72 ± 0.49 and forearm 5.09 ± 0.51),but the cheek had significantly higher pH value than the forearm and forehead (both P < 0.05).Melanin value on the face did not differ between different age groups (P > 0.05).Conclusion Physical and chemical properties of skin vary with body sites in healthy children.
2.Aggrecanases and osteoarthritis

3.Clinical efficacy of Qingpeng ointment for the treatment of chronic eczema

ObjectiveTo evaluate the efficacy and safety of Qingpeng ointment in the treatment of chronic eczema.MethodsTotally,95 patients with chronic eczema were enrolled and divided into 3 groups to receive topical Qingpeng ointment (n =33),0.2% hydrocortisone cream (n =32),10% urea ointment (n =30) respectively twice a day for 4 weeks.All the patients were followed up once a week for the evaluation of therapeutic and side effects.ResultsAfter 4-week treatment,the response rate was 78.8%,84.4% and 46.7% in the Qingpeng ointment-,hydrocortisone cream- and urea ointment-treated patients respectively.Significant differences were observed in the response rate between the Qingpeng ointment- and urea ointment-treated patients (P < 0.05 ),but not between the hydrocortisone cream- and Qingpeng ointment-treated patients (P >0.05).Skin flushing with a sensation of burning was observed at the application sites of 2 patients treated with Qingpeng ointment,and tingling sensation at the application sites was reported in 1 patient treated with hydrocortisone cream.All of these adverse reactions were tolerable and had no influence on the treatment.Conclusion Topical Qingpeng ointment is effective and safe for the treatment of chronic eczema.
4.Factors associated with the relapse of propranolol-treated infantile hemangioma

Objective To analyze factors associated with the relapse of propranolol-treated infantile hemangioma.Methods The clinical data were collected from 235 children with infantile hemangioma who had discontinued propranolol for 6 months,and retrospectively analyzed.Factors for the relapse of propranolol-treated infantile hemangioma were analyzed by univariate and multivariate unconditional logistic regression analyses.Results Of 235 patients followed for 6 months after drug withdrawal,66 (28.1%) were identified to have recurrence of infantile hemangioma,of whom,15 (22.7%) had severe recurrence.The risk of recurrence was significantly increased in patients taking propranolol at a daily dosage of 1.5 mg/kg compared with those taking propranolol at a daily dosage of 2 mg/kg (OR =3.566,95% CI:1.306-9.739),in patients aged > 8 weeks at the initial drug treatment compared with those aged ≤ 8 weeks at the initial drug treatment (OR =5.043,95% CI:1.248-20.376),in patients with the course of medication ≤ 6 months compared with those with the course of medication > 6 months (OR =17.661,95% CI:4.899-63.665),as well as in patients aged < 1 year at drug withdrawal compared with those aged ≥ 1 year at drug withdrawal (OR =6.089,95% CI:1.835-20.204).Conclusion There are many risk factors associated with the relapse of propranolol-treated infantile hemangioma,so some measures aimed at these risk factors should be taken to reduce the recurrence rate.
5.Expression and significance of cannabinoid receptor 2 in malignant melanoma

Objective To investigate the expression of cannabinoid receptor 2 (CB2) in pigmented nevus and malignant melanoma and its significance. Methods The protein and mRNA expressions of CB2 were measured in tissue samples of 20 patients with pigmented nevus and 20 patients with malignant melanoma using immunohistochemical staining and RT-PCR, respectively. Results CB2 was expressed in both pigmented nevus and malignant melanoma. In tissue of pigmented nevus, CB2 was chiefly expressed in nevocytes and basal cell layer, and only a small quantity of CB2 was expressed in subcutaneous tissue. Statis-tical differences were observed between samples of malignant melanoma and pigmented nevus in the expres-sion intensity of CB2 (P < 0.05). Conclusions Increased protein and mRNA expressions of CB2 are observed in tissue of malignant melanoma, and the expression intensity of CB2 is higher in tissue of malig-nant melanoma than in that of pigmented nevus.

7.Influence of multidrug resistance gene 1 C3435T genetic polymorphism on the eradication of gastric ulcer with Helicobacter pylori infection

Objective To study the influence of multidrug resistance gene (MDR)1 C3435T genetic polymorphism on the eradication of gastric ulcer with Helicobacter pylori (Hp) infection.Methods A total of 106 gastric ulcer patients with positive Hp were randomly divided into two groups by lot with 53 cases each.One group was assigned with 20 mg esomeprazole,0.5 g clarithromycin,1.0 g amoxicillin twice one day(EAC group),and the other group was assigned with 20 mg omerprazole,0.5 g clarithromycin,1.0 g amoxicillin twice one day (OAC group).The therapy of two groups was one week.Hp was detected at least 4 weeks after the end of treatment.MDR1 C3435T genetic polymorphism was determined by polymerase chain reaction-restriction fragment length polymorphism assay.The influence of MDR1 C3435T genetic polymorphism on the eradication of Hp was recorded and analyzed.Results The Hp eradication rate was 84.9% (45/53) and 77.4% (41/53) in EAC group and OAC group,and there was no significant difference between two groups (P > 0.05).There was no significant difference in the Hp eradication rate in patients with different MDR1 C3435T genotypes in two groups (P > 0.05).The Hp eradication rate was 66.7%(16/24),86.3%(44/51),83.9%(26/31) in TT,CT,CC genotype,and there was significant difference(P< 0.05).The Hp eradication rate in patients with TT genotype was lower than that in patients with CT,CC genotype,and there was significant difference (P< 0.05).Conclusion There is significant relationship between the effect of gastric ulcer with Hp eradication and MDR1 C3435T genetic polymorphism,and the Hp eradication rates of patients with TT genotype are more lower.

Objective To discuss the teaching effects of PBL teaching approach applied in clinical practice of medical oncology.Methods Throush clinical practice of medical onclolgy with the PBL teaching approach and the traditional one.A contrast between two teaching modes was made.And the teaching results were investigated based on the questionaic.Results The PBL teaching approach is superior to the traditional one.Conclusion The PBL teaching approach has an obvious advantage in the teaching of clinical practice of medical oncology and can improve the teaching quality
9.Oral negative pressure in amelioration of tension headache in 20 patients with microcirculation disturbance

BACKGROUND: Tension headache means contraction headache or oppressive headache in bilateral occipital-cervical part or whole head. Its pathogenesy bas not been completely known yet, which may be related to many factors and the conventional therapy is drug heteropathy but the effects are different, especially on patients with chronic disease and side effects will be induced by long-time intake of drugs.OBJECTIVE: To explore the curative effect of oral negative pressure on patients with tension headache and its mechanism of ameliorating microcirculation disturbance.DESIGN: Case-control observation.SETTING: Department of Pathophysiology of Hebei North University.PARTICIPANTS: Twenty patients with tension headache in the Department of Neurology, First Affiliated Hospital of Hebei North University from December 2001 to June 2002 including 12 females and 8 males,aged 18-28 (mean 23.4) with the disease course of 1-6 years, were randomly divided into therapeutic group and control group with 10 patients in each group.METHODS: The oral negative pressure instrument was applied. Put the negative pressure exerting equipment in the optimal position of mouth, adjusted the negative pressure to (0.05±0.01) MPa. The therapeutic time was 10 minutes each time (5 minutes for the first time) once a day with the time fixed. There were 5 days in one course and totally 3 courses. Put the negative pressure exerting equipment into mouth of patients in the control group for 10 minutes each time without exerting negative pressure. Quantitative evaluation on pain was performed with visual analogue scale (VAS)method, excellent as the VAS score decreased above 70%, effective as the VAS score decreased between 30% and 70%, invalid as the VAS score decreased below 30%. XTL- Ⅱ type microcirculation micro-television system was used to observe the changes of nailfold microcirculation by magnifing 260 times. The first row nailfold of left ring figure was checked conventionally, state of microvessel, micro-bloodstream and peri-loop were recorded and degree of microcirculatory disturbance was quantitatively analyzed according to TianNiu 's weighing integral method.MAIN OUTCOME MEASURES: The therapeutic effect after three courses and results of microcirculation observation.RESULTS: Totally 20 included patients were involved in the analysis of results. VAS scores of 7 patients in the therapeutic group decreased above 70% (70%), while no patient in the control group; VAS scores of 2 patients in the therapeutic group decreased from 70% to 30% (20%) and one patient in the control group (10%); VAS scores of one patient in therapeutic group decreased below 30% (10%) and nine patie nts in the control group (90%). Negative pressure could obviously ameliorate the microcirculation disturbance and enable it to recover from moderate before treatment to nearly normal with the total score decreased from (4.18±0.68) points to (1.97±0.41) points (P < 0.01).CONCLUSION:Oral negative pressure has a significant curative effect on patients with tension headache. Its mechanism may relate to the improvement of microcirculation and the adjustment of nervous functional disturbance.
10.Prediction of the secondary structure and B cell epitopes for the Izumo protein of Homo Sapiens

Objective:To predict and analyze the secondary structure and B cell epitopes of Izumo protein.Methods: The secondary structure and flexible regions of Izumo protein were predicted by the methods of Chou-Fasman,Gamier-Robson and Karplus-Schulz.Moreover,hydrophilicity plot,surface probability plot and antigenic index of Izumo protein were predicted by the methods of Kyte-Doolitde,Emini and Jameson-Wolf,respectively.Results: Izumo protein contained moreαhelix regions.There were several centers ofαhelix in the regions of 6-17,30-40,88-99,103-120,153-160,173-188,249-260,283-297,334-338 and 339-346 of Izumo protein,and several centers of βsheet in the regions of 21-25,198-200,245-248 and 320-323.Moreover,many distinct B cell epitopes in Izumo protein possibly localized in the regions of 3642,62-66,94-99,118-122,129-132,151-154,161-164,173-177,205-208,212-216,256-265,271-276,283-288,314-318 and 336-350.Conclusion:These results are helpful for identification of the dominant B cell epitopes and the functional domains of Izumo protein.
