3.Polymorphism study of six miniSTR loci in Han population from Northeast China

OBJECTIVE:Allele frequencies and forensic parameters for six miniSTR loci(D10S1248,D14S1434,D22S1045,D4S2364,D2S441 and D1S1677)were analyzed in a population of 173 unrelated Chinese Han population individuals from Northeast China.METHODS:The six miniSTR loci were preformed in two multiplex fluorescent PCR systems.ABI 310 Genetic Analyzer was utilized in capillary electrophoresis and the lengths of allele fragments were analyzed.Genetic data collection and analysis software were used for data collection and genotyping.Statistical analysis was performed to the data.RESULTS:The six miniSTR loci showed a moderate degree of polymorphism in Han population from Northeast China.The observed allele sizes were from 67 bp to 115 bp,and the observed heterozygosity ranged from 0.728 to 0.827.The combined power of discrimination and the combined power of exclusion for the six miniSTR loci in Northeast China were 0.999 99,and 0.993 31,respectively.CONCLUSION:The six miniSTR showed a moderate genetic polymorphism in Han population from northeast of China.Due to their small size of PCR amplicon,the six miniSTR could be useful supplements to the CODIS STRs,and they would be useful in population genetics and forensic analysis.
4.Impact of setup errors on dose distribution of three dimensional conformal radiotherapy for patients with esophageal carcinoma

Objective To measure the setup errors of patients with esophageal carcinoma during the treatment of three dimensional conformal radiotherapy (3DCRT), and to analyze the impact of setup errors on dose distribution of GTV,CTV and normal tissues around. Methods Forty-two patients with esophageal cancer treated by 3DCRT were included. The setup errors of each patient were measured once a week for 6 times by electronic portal imaging device (EPID). The setup errors were integrated into the treatment plan-ning system by moving the isocenter. Then the dose distribution of GTV, CTV and normal tissues were recal-culated. Results The systematic setup errors of the 42 patients were - 2.31 mm, - 0.55 mm and - 0.16 mm, and the random errors were 4.42 mm, 4.35 mm and 4.48 mm in the directions of lef-fight, anterior-posterior,and superior-inferior, respectively. The dose covered 95% GTV( D95 ) was reduced by 32 cGy and by 88 cGy for CTV D95. The lung V20 in the original plan and the integrated plan was 22.49% and 22.02%, respectively. The average dose of the heart in the two plans was 2077.62 cGy and 2036.23 cGy, respectively. In the original plan, no patient had maximum dose of spinal cord over 4500 cGy; While in the intergrated plan there were 18 patients had the spinal cord dose more than 4500 cGy, with a maximum dose of 5503.90 cGy. Conclusions The setup errors cause significant dose reduction of GTV and CTV, but not of the lung and heart . The maximum dose of the spinal cord may exceed 4500 cGy due to the setup errors.
5.Effect of phosphorylated-P38 MAPK on caspase-3 expression in substantia nigra of the MPTP mouse model of Parkinson's disease

Objective To investegate the effect of phosphorylated-P38 MAPK(mitogen-activated protein kinase) on the expression of caspase-3 in the substania nigra (SN) of MPTP-induced mouse model of(PD). Methods Mice were randomly divided into MPTP model group, which were treated with MPTP and inhibitor group. Once a day for 5 days; control group was treated with saline and DMSO as much as the model group received per day for 5 days. The behavioral were observed, immunohistochemistry and Western blot for TH, caspase-3 and phosphorylation of P38 MAPK were used to observe the change of positive cell number and the expression level in the SN of midbrain. Results Compared with the mice in control group, the model group showed typical symtoms of PD with decreased numbers of TH-positive neurons and the protein level of TH in SN of the midbrain by about 60% and 65% respec-tively(P<0.01) , the numbers of caspase-3 and phosphorylation of P38 MAPK immunoreactive cells and their protein level in the SN of the midbrain increased markedly (P<0.01). After giving SB203580, the above changes were reduced obviously (P <0. 01). Conclusion In the mouse model of subacute Parkinson's disease induced by MPTP, phosphorylated-P38 MAPK regulated caspase-3 in the SN of midbrain, the specific P38 MAPK inhibitor SB203580 is neurologically oprotective to the mouse model.
6.Verification of the Origin and Development of the Nomenclature and Location of Point Dingchuan (Ex-B1)

Point Dingchuan (Ex-B1) pertains to extraordinary points and often produces a good therapeutic effect on respiratory system diseases. Ancient Chinese medical books do not record point Dingchuan. Modern Chinese medical textbooks describe the location and efficacy of and indications for this point but do not state their provenances. In order to further popularize the clinical application of point Dingchuan, this article verifies the origin and development of the name and location of point Dingchuan so as to provide a certain reference for clinically correct selection and use of this point. The results show that point Dingchuan followed the course of development from ashi point to new point and then to extraordinary point. The development of point Dingchuan to the same acupoint name and location as now international use went mainly through four stages, that is, similar name and different location, similar name and location, same name and different location, and same name and location.
7.Epidemiological investigation of porcine pseudorabies virus and its coinfection rate in Shandong Province in China from 2015 to 2018

Background:
Pseudorabies, also known as Aujeszky's disease, is caused by the pseudorabies virus (PRV) and has been recognized as a critical disease affecting the pig industry and a wide range of animals around the world, resulting in great economic losses each year. Shandong province, one of the most vital food animal-breeding regions in China, has a very dense pig population, within which pseudorabies infections were detected in recent years. The data, however, on PRV epidemiology and coinfection rates of PRV with other major swine diseases is sparse.
Objectives:
This study aimed to investigate the PRV epidemiology in Shandong and analyze the current control measures.
Methods:
In this study, a total number of 16,457 serum samples and 1,638 tissue samples, which were collected from 362 intensive pig farms (≥ 300 sows/farm) covered all cities in Shandong, were tested by performing enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR).
Results:
Overall, 52.7% and 91.5% of the serum samples were positive for PRV-gE and -gB, respectively, based on ELISA results. In addition, 15.7% of the tissue samples were PCR positive for PRV. The coinfection rates of PRV with porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus, and classical swine fever virus were measured; coinfection with PCV2 was 35.0%, higher than those of the other two viruses. Macroscopic and microscopic lesions were observed in various tissues during histopathological examination.
Conclusions
The results demonstrate the PRV prevalence and its coinfection rates in Shandong province and indicate that pseudorabies is endemic in pig farms in this region. This study provides epidemiological data that can be useful in the prevention and control of pseudorabies in Shandong, China.
8.Excessive copper in feed not merely undermines animal health but affects food safety

Background:
Blackened intestines in slaughtered pigs have been commonly observed in China in recent years. However, no cause has been reported.
Objectives:
We attempted to determine whether the blackening of the pig intestine was related to an excess of copper (Cu) in their feed.
Methods:
In this study, we observed and collected porcine intestines in small- and large-scale pig slaughterhouses in Shandong province from May to October 2018. Twelve types of metal ions were detected in the black intestinal samples.
Results:
The Cu level in the intestine samples was mostly higher than the Chinese national limit for food. Further study showed that Cu supplementation in most commercial porcine feed also exceeded the national standard. An animal model (mouse) that could mimic the intestinal blackening in pigs was established. Compared to control mice, Cu accumulated in the liver and intestines of mice fed an excessive Cu level, confirming the excessive Cu in the feed may be considered the major cause of blackened porcine intestines. Microscopic examination revealed that black intestines had many particles containing Cu in the lamina propria of the intestinal mucosa, and the intestinal mucosal epithelial cells showed degeneration and necrosis.
Conclusions
In conclusion, overuse of Cu in animal feed can lead to animal poisoning and Cu accumulation in animal products. Such overuse not only harms the health of livestock but can also affect public health.
9.Influences of epidural labor analgesia on maternal postpartum cognitive function after vaginal delivery:a prospective cohort study

Objective To investigate the incidence of cognitive dysfunction in postpartum women who underwent epidural or non-drug labor analgesia and the influence factors.Methods A prospective cohort study was performed in 1 618 uneventful singleton pregnancies in International Peace Maternity and Child Health Hospital from January 2017 to January 2018.Women who received epidural labor analgesia were assigned to the epidural group (n=803),and those who received Doula technique non-drug labor analgesia rather than labor analgesia to the control group (n=815).Cognitive function was assessed using symbol digit modalities test (SDMT90) and Montreal cognitive assessment (MoCA) 1 d and 42 d after delivery.Incidence of maternal cognitive dysfunction,SDMT90 scores and pain intensity measured by visual analogue score (VAS) at the cervical dilatation of 3,6 and 10 cm between the two groups were compared using independent sample t-test,Chi-square test or logistic regression analysis.Results MoCA and SDMT90 scores of the epidural group were significantly higher than those of the control group 1 d after delivery (27.1 ± 1.5 vs 26.2± 1.8,49.1 ±2.4 vs 42.5 ± 3.6;t=3.775 and 16.956,both P<0.05),but the incidence of postoperative cognitive dysfunction (POCD) in the epidural group was remarkably lower comparing to the control group [13.9% (112/803) vs 21.2% (173/815),x2=14.769,P=0.002].The VAS scores of the epidural group at the cervical dilatation of 3,6 and 10 cm were all lower than those of the control group (2.3 ± 0.6 vs 6.9± 1.3,3.3 ± 0.9 vs 8.7± 0.9,5.7± 0.9 vs 9.7± 0.4;t=0.013,0.011 and 0.015;all P<0.001).Logistic regression analysis indicated that VAS scores ≤ 3 at the cervical dilatation of 3,6 and 10 cm were protective factors against the incidence ofPOCD 1 d after delivery [OR(95%CI):0.238 (0.198-0.287),0.180 (0.145-0.222) and 0.112 (0.088-0.142),all P<0.001],while the absence of epidural labor analgesia was a risk factor (OR=4.698,95%CI:1.812-11.321,P<0.001).Conclusions Epidural labor analgesia can reduce the incidence of postpartum cognitive dysfunction in women 1 d after delivery.The incidence of POCD has close relationship with the VAS scores at the cervical dilatation of 3,6 and 10 cm and epidural labor analgesia.
