1.Post- traumatic growth and related factors in patients with breast cancer

Objective To investigate the posttraumatic growth(PTG)in female patients with breast cancer after operation and the possible influencing factors related to the PTG of these patients. Methods 300 female patients with breast cancer from Oct 2014 to Jan 2015 in 7 third-level grade A hospitals were selected by random number table. The posttraumatic growth inventory (PTGI)was used to study PTG status in these patients. Clinical data including age, degree of education, marital status, underlying disease, clinical stage, surgical method, adjuvant therapy and postoperative complications were univariate analyzed, then the positive factors were logistic regression analyzed. Results The mean PTG score was 59.83±12.90, univariate analysis showed that degree of education (F=15.80, P=0.000), underlying disease (t=3.90,P=0.000), clinical stage (F=10.17, P=0.000), surgical method (t=5.12, P=0.000) and postoperative complications(t=4.85,P=0.000) were influencing factors, while multivariate analysis by logistic regression showed that clinical stag(OR=6.635, P=0.000), surgical method (OR=2.858, P=0.000) and postoperative complications (OR=2.731, P=0.035) were independent influencing factors for female patients with breast cancer after operation. Conclusions The posttraumatic growth status in breast cancer patients undergoing operation declined dramatically. The level of metal change dimension are main clinical features of breast cancer patients. There are close associations between PTG and clinical stag, surgical method and postoperative complications.
2.Progress in the study of anti-vascular endothelial growth factor application for the treatment of neovascular age-related macular degeneration and the mechanism of poor and non-response

According to the best corrected visual acuity and the morphological changes of the macular fovea,responses to the neovascular age-related macular degeneration (nAMD) who receive anti-vascular endothelial growth factor (VEGF) therapy show large variability,including poor and non-responders.Various factors will be reviewed to account for poor and non-response to anti-VEGF therapy,such as the related susceptibility genes,factors related with the development of choroidal neovascularization and morphologic parameters,pharmacokinetics and tachyphylaxis.The future research should focus on comprehensive assessment of factors affecting the efficacy of anti-VEGF therapy to improve the therapeutic outcome of nAMD.
4.Study on transvaginal two-dimensional and three-dimensional ultrasound in the eveluation of endometrial receptivity duringin vitro fertilization-embryo transfer treatment

ObjectiveTo determine endometrial thickness, endometrial volumeand, endometrial and subendometrial blood flows, by using transvaginal two-dimensional (2D) and three-dimensional (3D) ultrasound examination and to compare the value of all the parameters in the prediction of endometrial receptivity during in vitro fertilization-embryo transfer (IVF-ET) cycles.MethodsOne hundred and twenty patients undergoing IVF cycles were recruited for this study. Transvaginal 2D and 3D ultrasound examination was performed to determine endometrial thickness, endometrial volume, pulsatility index (PI), resisitance index (RI) , systolic/diastolic ratio (S/D) of endometrial, subendometrial blood flows , vascularization index (VI), flow index (FI) and vascularization flow index (VFI) of endometrial and subendometrial regions by experienced sonographers on the day of human chorionic gonadotropin (HCG) injection. Statistical comparison was carried out by Mann-Whitney test. The receiver operator characteristic (ROC) curve analysis was applied and the area under the curve (AUC) was calculated. Statistic analyses were performed using SPSS 13.0. The two-tailed value of was considered statistically significant.ResultsThere were no significant differences between the pregnant group and non-pregnant group in endometrial thickness, endometrial volume (P>0.05). Patients in the pregnant group had lower PI, RI and S/D of endometrial and subendometrial blood flows than those in the non-pregnant group (Z=-3.811,-3.097,-2.071, P=0.001, 0.001, 0.038), with statisticalsignificant difference. Patients in the pregnant group had higher VI, FI and VFI of endometrial and subendometrial regions than those in the non-pregnant group (Z=-1.970,-2.698,-1.981, P=0.490, 0.007, 0.048;Z=-2.098,-2.090,-2.115,P=0.036, 0.037, 0.034), with statistical significant difference. ConclusionsEndometrial and subendometria blood flows measured by transvaginal 2D and 3D ultrasound can be as a reliable indicator to assess endometrial receptivity and predict the pregnancy outcome. Among all the endometrial and subendometrial blood flow parameters, VI of endometrial and subendometrial are better predicators.
5.Effect of octreotide combined with compound coenzyme on Th17/CD4+CD25+Foxp3+ Treg imbalance in patients with purulent appendicitis

Objective To analyse the effect of octreotide combined with compound coenzyme on Th17/CD4 +CD25 +Foxp3 + regulatory T cells imbalance in patients with purulent appendicitis.Methods 150 patients with acute suppurative appendicitis were randomly divided into experimental group and control group, 75 cases in each group.Patients of two groups underwent appendicitis laparoscopic resection, then the control group were given conventional treatment and experimental group were given subcutaneous injection of octreotide on the basis of conventional treatment.Th17, CD4 +CD25 +Foxp3 +regulation T cells ( Treg) , and Th17 /Treg balance were analyzed.ResuIts Compared with control group, Th17/Treg imbalance of experimental group recovered better: the proportion of Th17, Treg, and Th17/Treg ratio decreased (P<0.05); the expression levels of related transcription factor, RORγt and Foxp3 mRNA decreased (P<0.05); serum cytokine (IL-4, IL-6, IL-10, IL-17, IL-23 and TGF beta) and C-reactive protein (CRP) content decreased (P<0.05);serum interferon gamma (IFN-gamma) content increased (P<0.05).ConcIusion Octreotide combined with compound coenzyme could correct Th17/Treg imbalance in suppurative appendicitis patients, improve the immune function of patients and affect the prognosis of patients.
6.Immunomodulatory effects of polysaccharopeptide on EAC tumor-bearing mice through MyD88-dependent signaling pathway

Objective:To detect the effects of polysaccharopeptide(PSP) on MyD88-dependent signaling pathway in EAC tumor-bearing mice,and explore the immunomodulatory mechanism of PSP.Methods: Ehrlich′s ascites carcinoma(EAC) C57BL/6 mice were used to establish the animal model for solid tumor.Mice were divided into two groups:WT group and MyD88-/-group.After 22 days of treatment,quantitative real-time PCR( Q-PCR) ,Western blot and were used to detect the related gene and protein expression of TLR4 pathway in spleens,ELISA were used to detect the terminal effect factors secretion eyeball blood from each group.Results:Related genes and proteins of TLR4 pathway in spleen were up-regulated significantly from two groups.Compared with WT group, related genes and proteins in MyD88-dependent pathway(MyD88,TRAF6,NF-κB,AP-1)was down-regulated in MyD88-/-group(P<0.05).Meanwhile,There was no significant change of the related genes and proteins of MyD88-independent pathway(TRAM,TRIF)in MyD88-/-group( P>0.05 ) .The terminal effect factors secretion of IL-2, IL-6, IL-10, TNF-αand IFN-γin MyD88-/-group were decreased significantly compared with WT group(P<0.05).Conclusion:The immunoregulatory effect of PSP on EAC tumor-bearing mice may be implement through the regulation of MyD88-dependent signaling pathway.
7.Effect on Anti-inflammatory Efficacy and Bitter Cold Properties of Coptis Processed by Evodia Juice

Objective To explore the influence on anti-inflammatory efficacy and bitter cold properties of Coptis processed by Evodia juice. Methods Auricle swelling model induced by croton oil in mice was used in the anti-inflammatory efficacy experiment. The mice were assigned randomly into distilled control group, dexamethasone group, crude drug group, 3 processed drug groups that Euodia juice which plant origin was Euodia rutaecarpa (Juss.) var. bodinaieri (Dode) Huang (SMYHL), Evodia rutaecarpa (Juss.) Benth and Euodia rutaecarpa (Juss.) Benth var. officinalis (Dode) Huang, respectively. The crude drug group and the 3 processed drugs groups were orally administrated one time and the dosages were all 6 g/kg. The control group was orallly administrated the same volume water. Dexamethasone group was intraperitoneally injected dexamethasone 30 g/kg. The swelling degree of each group was observed. In the bitter cold properties experiment, rats were assigned randomly into control group, crude drug group and 3 processed drug groups. The crude drug group and the 3 processed drug groups were orally administrated for 14 days and the dosages were all 3 g/kg. The control group was orally administrated the same volume water. The body weight, capacity for eating and drinking, body temperature, heart rate and blood viscosity of rats were measured. Meanwhile, TSH, IL-2, IL-8 in serum and plasma were determined by radioimmunoassay. Results The degree of mouse auricle swelling was reduced by the crude and processed drugs. Fourteen days after medication, the weight, capacity of eating and drinking of rats were declined to different degrees in crude drug group and in processed drugs group as compared with control group. The blood viscosity was increased and IL-2 was decreased in SMYHL group as compared with control group (P<0.05). TSH was decreased in crude drug group as compared with control group (P<0.05). Conclusion There is no obvious change in anti-inflammatory function of Coptis before and after processed. The cold properties of Coptis can be reduced after processed by Evodia Juice.
8.Effect of anti-vascular endothelial growth factor therapy on wet age-related macular degeneration and its recurrence factors

Objective:To evaluate the effect of anti-vascular endothelial growth factor (VEGF) in patients with wet age-related macular degeneration (wAMD) and investigate the recurrence factors.Methods:In this case-control study, 40 eyes of 38 patients with wAMD treated by anti-VEGF therapy in Xijing Hospital from October 2015 to April 2017 were included.Ranibizumab or conbercept was consecutively intravitreally injected monthly under 3+ pro re nata (PRN) regimen.All included patients were followed-up at least 12 months.The best corrected visual acuity (BCVA) was expressed in logarithm of the minimum angle of resolution (LogMAR) units.Optical coherence tomography (OCT) was used to observe the morphologic characteristics of macula and measure the central retina thickness (CRT). Patients were divided into recurrence and recurrence-free group based on whether recurrence occurred during follow-up.Potential associated factors with recurrence after treatment were analyzed by multivariate logistic regression analysis.Meanwhile, the morphological and functional response were identified at 1 month after loading phase as well as at last follow-up visit.This study protocol was approved by the Ethics Committee of Shanghai General Hospital (No.2016KY243) and written informed consent was obtained from each patient prior to receiving injection.Results:All the operative eyes showed retinal effusion absorption at least once, and 21 (52.5%) eyes had relapse during follow-up.The patients with lower BCVA (LogMAR) value at the first absorption time had higher risk of recurrence ( OR=16.7, 95% CI=1.22-100, P<0.05). The eyes in the recurrence group received more times of treatment than eyes in the recurrence-free group (5.8±2.3 vs 3.5±1.4, t=3.64, P<0.05). However, there was no significance between the two groups in BCVA change before and after treatment ( t=-1.52, P>0.05). In the recurrence group, the BCVA (LogMAR) and CRT at last visit were 0.73±0.34 and (187.2±81.7)μm respectively, while were significantly improved in comparison with 0.81±0.37 and (234.2±74.5)μm at the relapse time ( t=2.14, 2.62; both at P<0.05). There were 87.5% (35/40) eyes showed good morphological responses at 1 month after the loading phase while 12.5% (5/40) eyes presented poor morphological responses, the retinal effusion was absorbed after additional (3.2±2.2) times of injections.The BCVA of eyes with a good morphological response at 1 month following loading phase was better than that at the first retinal effusion absorption time ( t=-2.23, P<0.05). Average CRT at last visit was significantly decreased in comparison with the baseline CRT ( Z=-4.62, P<0.01). Conclusions:For wet AMD patients, the anti-VEGF treatment is safe and effective in the short term, while there is still a high rate of recurrence.More attention should be paid to eyes with better vision at the first time of retinal effusion absorption which may have more chance to experience recurrence.Patients need long-term follow-up after treatment to prevent recurrence.In addition, the complete loading treatment is needed for patients who have a good morphological response after the first retinal effusion absorbed.
9.Eukaryotic expression of human arresten gene and its effect on the proliferation of vascular smooth muscle cells.

The eukaryotic expression of human arresten gene and its effect on the proliferation of in vitro cultured vascular smooth cells (VSMCs) in vitro were investigated. COS-7 cells were transfected with recombinant eukaryotic expression plasmid pSecTag2-AT or control plasmid pSecTag2 mediated by liposome. Forty-eight h after transfection, reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of arresten mRNA in the cells, while Western blot assay was applied to detect the expression of arresten protein in concentrated supernatant. Primary VSMCs from thoracic aorta of male Sprague-Dawley rats were cultured using the tissue explant method, and identified by immunohistochemical staining with a smooth muscle-specific anti-alpha-actin monoclonal antibody before serial subcultivation. VSMCs were then co-cultured with the concentrated supernatant and their proliferation was detected using Cell Counting Kit-8 (CCK-8) in vitro. The results showed that RT-PCR revealed that the genome of arresten-transfected cells contained a 449 bp specific fragment of arresten gene, suggesting the successful transfection. Successful protein expression in supernatants was confirmed by Western blot. CCK-8 assay showed that the proliferation of VSMCs were inhibited significantly by arresten protein as compared with control cells (F=40.154, P<0.01). It was concluded that arresten protein expressed in eukaryotic cells can inhibit proliferation of VSMCs effectively in vitro, which would provide possibility to the animal experiments.
10.Role of endometrial and subendometrial blood flows measured by three-dimensional power Doppler ultrasound in the prediction of endometrial receptivity during IVF-ET treatment
