1.Surgical complications of orthotopic liver transplantation

Objective To study the prevention and treatment of postoperative complications of liver transplantation (LT). Methods According to experience of Thomas E. Starzl Transplantation Institute and the Departments of Surgery University of Pittsburgh Medical Center, we introduce the prevention and treatment of surgical complications. Results and Conclusions Intra-abdominal hemorrhage, hepatic artery thrombosis, portal vein thrombosis, bile leaks, biliary obstruction, intestinal perforation, gastrointestinal bleeding are the postoperative complications of LT which are related to the surgical technique. Rapid improvement of diagnostic assessment of the alterations from normal postoperative course, and timely intervention are very important to minimize morbidity of surgical complications and mortality .
2.Mechanism of 5-fluorouracil in the treatment of acute pancreatitis

Objective To study the mechanism of 5 fluorouracil(5 FU) in the treatment of severe acute pancreatitis(SAP). Methods SD rats were randomly divided into three groups:acute pancreatitis group (AP group,n=12), 5 FU treatment group( 5 FU group,n=10),and control group (n=10). In 5 FU group,5 FU(1mg/100g) were injected immediately after AP was induced.12h after the AP was induced, blood samples were taken to determine nitric oxide(NO) and interleukin 6(IL 6). Results Compared with AP group,in 5 FU group, the survival rate increased,mean survival time prolonged,ascites volume decreased significantly,and the inflammatory mediators subsided. Conclusions NO and IL 6 may play an important role in the pathogenises of acute pancreatitis. The treatment effect of 5 FU for AP may through inhibit the inflammatory mediators.
3.THE CYTOTOXIC EFFECTS OF CRUDE BILE ON HUMAN PANCREATIC CANCER CELL LINES

Objective To identify effects of bile acids on pancreatic cancer, The ultrastructure and growth of PANC-1 and MIA PaCa-2 cell lines in crude bile modified medium were studied. Methods The growth of PANC-1 and MIA PaCa-2 cells in RPMI 1640 with or without 1%, 2% and 4% of the purified crude bile (containing total bile acids 10.17mmol/L) was assessed for 2, 4, 6, 8d by using MTT assay to determine inhibitory rate. The cell surface and intracellular ultrastructure of PANC-1 cells was investigated by SEM and TEM at 24h and 48h, respectively. Re sults The proliferation of both cell lines in bile treated medium were greatly retarded (P <0.001). The inhibitory rate of 1%, 2% and 4% bile on Panc-1 cells in 4d were 38%, 60% and 66%, respectively (P <0. 05), on MIA PaCa-2 cells at 4d were 28%, 39% and 52%, respectively (P <0. 05). The cells grown in bile for 48h lost their mi crovilli, their mitochondria and other organelles became vacuolated. Conclusion The bile acids in bile has cytotoxicity on PANC-1 and MIAPACA-2 cells, which may inhibit pancreatic cancer progress in patients clinically.
4.The open field test as a method for ethology in Kunming mice: test-retest reliability

Objective To explore the test-retest reliability of the open field test,as a method for ethology in Kunming mice. Methods The behavior of adult male Kunming mice in the open field test was recorded twice,once for five minutes, with a one-week inter-trial interval. The following parameters were evaluated:number of central squares; number of peripheral squares; percent of central squares; number of horizontal squares; number of rearings; total score; and number of fecal boli. Subsequently,the intraclass correlation coefficient (ICC) and the agreement index (Kappa) were calculated for each of these parameters. Results Both ICC and Kappa for total score( ICC =0.902, P<0.01; Kappa =0.680, P<0.01; r=0.815, P<0.01 ) showed a good reliability and correlation between the test trials. Only ICC for number of central squares ( ICC = 0. 578, P < 0.01 ), number of peripheral squares( ICC =0.615, P<0.01 ) ,percent of central squares(ICC=0.523, P<0.01) ,number of horizontal squares( ICC = 0. 607, P < 0.01 ) and number of rearings ( ICC = 0. 606, P < 0.01 ) indicated a fair reliability between the test trials. Neither ICC nor Kappa for number of fecal boli displayed a significant reliability between the test trials. The difference of these parameters between test and retest, such as number of peripheral squares( t =3.01, P<0.01 ),number of horizontal squares( t = 3.34, P < 0.01 ) and total score( t = 7.86, P < 0.01 ) were significant. Conclusion The parameter,total score for Kunming mice in the open field test,is found to be stable over time, and indicates a good reliability with a one-week inter-trial interval.
