Objective To explore the best serum-free cultural way in neural stem cells from the hippocampus of the newborn rats and observe the characteristics of growth,proliferation,and induced differentiation of neural stem cells.Methods The neonatal rats′ hippocampus tissues were dissociated mechanically.The cells were cultured in the serum-free cultural medium which were added separately basic fibroblast growth factor(bFGF),epithelium growth factor(EGF),bFGF+EGF by suspended cultural way.The neural stem cells were identified by nestin immunofluorescence.After induced differentiation of embryonic cow serum,the differentiated cells were identified.Results The serum-free medium added bFGF and EGF could perfectly induce neural stem cells to proliferate and the differentiated cells expressed the specific antigen of neurons,astrocytes and oligodendrocytes.Conclusion The serum-free medium with bFGF and EGF can be used to culture neural stem cells from hippocampus tissue of rats in vitro.

Objective To investigate the mental health and its correlated factors for senile dementia caregivers in nursing homes in Shenyang. Methods Questionnaire survey was conducted among 174 senile dementia caregivers recruited from the Nursing Home of Shenyang and Shenyang Social Welfare Institute by using Symptom Check-List-90 (SCL-90), Social Support Rating Scale (SSRS) and self-designed general information questionnaire and to analyze the scores of self rating scale of nursing staff, the total score of social support scale, and the factors affecting the mental health status of nursing staff. Results The SCL-90 scores of senile dementia caregivers with education levels from illiteracy, primary school or junior high school, high school or secondary school, college or above were 130.50 ± 10.11, 132.70±12.55, 135.06±9.15, 129.24±8.83, the difference was statistically significant (F=15.05, P<0.01). The SCL-90 scores of senile dementia caregivers with poor, general and good perceived health status were 133.40±10.52, 132.60±11.32, 128.00 ±10.02, the differences were statistically significant (F=9.07, P<0.01). The SCL-90 scores of family monthly income of 1001 to 3000 yuan, 3001 to 5000 yuan and more than 5001 yuan were 132.90 ± 10.39, 132.80 ± 11.52, 131.50 ± 8.20, the difference was statistically significant (F=12.11, P<0.01). The multivariate linear regression analysis showed that the regression coefficient of perceived health status was-0.168 (t=2.880, P<0.01), monthly family income was-0.145 (t=-2.512, P<0.05), SSRS was 0.121 (t=2.112, P<0.05). Conclusions Mental health status of senile dementia caregivers in Shenyang is poor. The health sector should take some measures to improve the degree of social support, nursing staff wages and to ensure their health in order to promote physical and mental health of nursing staff and the nursing level so as to improve the quality of life of the senile dementia elderly.

The culture conditions of Saccharomyces cerevisiae sp.strain by 1.1 b were optimized for the production of D-(-)-mandelate dehydrogenase which is useful for the asymmetric bioreduction of benzoylformate to form D-(-)-mandelate.The optimum medium(per liter)consistes of 60 g peptone,30 g maltose, 0.5 g MgSO_4,0.01 g ZnSO_4,1.0 g KCl.After optimization of the culture medium,the enzyme production in shake flasks is enhanced from 2.56 to 20.21 U/L.The optimum fermentation conditions were determined as follows:medium volume 100 mL(i.e.,40%for a 250-mL shake flask),pH 6.5,inoculum size 10%,temperature 30℃,and cultivation time 25 h.

Lysins are efficient bacteria cell wall digesting enzymes encoded by DNA bacteriophage. Gram-positive bacteriophage lysins feature similar domain structure, high lytic efficiency, synergic antibacterial effect with antibiotics, rare neutralization by antibodies, less chance of developing drug-resistant strains, et al. The past decade has seen a considerable amount of research worldwidely focused on lysin, and lysins have been used successfully in a variety of animal models to control pathogenic antibiotic resistant bacteria found on mucosal surfaces and infected tissues. The great potential of lysins as an anti-infective agent prompted this review.

Objective To explore the effects of the autoantibody against the second extracellular loop of ?3-adrenoceptor(?3AR)from children with heart failure on isolated cardiomyocytes of adult rats.Methods The synthetic peptide corresponding to the sequence of the se-cond extracellular loop(176-202 amino acid residues)of the human ?3AR was used as the antigen to screen the anti-?3AR autoantibodies from the sera of the 50 children with heart failure and 50 healthy children by enzyme linked immunosorbent assay(ELISA);IgG in the children with heart failure of positive autoantibodies sera was purified.The effects of purified IgG per each group on contractile response of isolated cardiomyocytes of adult rats were observed using motion-edge detection system and double-provocation fluorescence photomultiplier system.Results Among the 50 children with heart failure,the anti-?3AR autoantibodies positive rate was 30%(15/50 cases),and ob-viously higher than that of healthy children[12%(6/50)cases,P

Objective To evaluate the value of ultrasound (US) in assessment knee joint inflammation in patients with juvenile rheumatoid arthritis(JRA).Methods US scans of the knees obtained in 30 children at clinically active stage; JRA was compared with those obtained in 30 healthy children and 10 JRA patients in clinical remission.Results Changes in synovial membrane thickness and presence of fluid in suprapatellar bursa showed statistically significant differences between JRA patients with active disease and the other subjects.Alterations in contour of the articular cartilage were demonstrated in 10 knees of patients with JRA.Conclusion US is a simple sensitive and reliable methods for the assessment and monitoring of knee joint involvement in JRA.

Objective To investigate the effects of essential trace elements selenium, zinc, copper, iron,cobalt, chromium and molybdenum upon arsenic poisoning caused by coal-burning. Methods Stratified random sampling method was used to conduct epidemiological investigation on 139 arsenic exposed residents(including nonpatient, light, moderate and severe patients) in an area polluted by coal-burning arsenic in Xingren county of Guizhou province as exposure group. Control group included 34 residents who lived about 13 km away from the endemic area of arsenic contamination. Inductively coupled plasma-optical emission spectrometry (ICP-OES) was used to analyze arsenic, selenium, zinc, copper, iron, cobalt, chromium and molybdenum in coal, soil, rice,corn, chilli, hair, blood and urine. Results Arsenic content in coal, soil, corn and chilli of polluted area were 4.894,146.551,0.522,1.440 mg/kg, respectively. These arsenic content were significantly higher than those in control area which were 1.980,50.167,0.296,0.948 mg/kg, respectively(P ＜ 0.05 or ＜ 0.01) . The content of selenium in soil of the diseased area(5.038 mg/kg) was significantly lower than that in soil of control area(8.948 mg/kg, P ＜0.05). The content of copper, iron, chromium in soil and iron in corn were 44.114,5731.500,98.323,89.996 mg/kg, respectively. These elements content were significantly higher than those in control area which were 13.473,1298.430,36.839,57.391 mg/kg, respectively (all P ＜ 0.05) . Hair and urine arsenic levels were 1.985mg/kg and 149.593 μg/g Cr in exposed group, respectively. These arsenic levels were significantly higher than those in control group which were 0.670 mg/kg and 49.853 μg/g Cr, respectively(all P ＜ 0.01) . Hair selenium level in exposed group(1.706 mg/kg) was significantly lower than that in control group(2.405 mg/kg, P ＜ 0.01). Hair levels of iron and chromium, blood level of eopper and the ratio between copper and zinc in exposed group were 88.295,8.933 mg/kg, 1.053 mg/L and 0.074, respectively. These element levels and elements ratio were significantly higher than those in control group which were 47.970,4.099 mg/kg, 0.934 mg/L and 0.065, respectively(P ＜ 0.01 or ＜ 0.05). Hair selenium level was negatively correlated with the progression of arsenism(r = - 0.414, P ＜ 0.01) .Hair levels of iron and chromium, the ratio between copper and zinc in blood were positively correlated with the progression of arsenism(r = 0.271,0.261,0.250, all P ＜ 0.01) . Conclusions Low selenium, high copper, high iron and high chromium coexists in arsenic polluted area. In exposed group, hair selenium is low, hair iron and chromium, blood copper and ratio between copper and zinc are high. These element changes with environment trend.These element changes are associated with the occurrence and development of the disease caused by coal-burning.
