1.Influence of propofol on the expression of iNOS after spinal cord injury in rat

Objective To investigate the influence of propofol on the induction of inducible nitric oxide synthase (iNOS) expression in spinal cord after spinal cord injury (SCI) in rats. Methods SCI model was reproduced according to Allen′s method with modification. Seventy-four SD rats were randomly divided into three groups: normal group (n=4), SCI with propofol treatment group (n=35), and SCI with saline treatment group (n=35). Immunohistochemistry and Western blot were employed to detect iNOS expression in spinal cord at different time (1h, 3h, 8h, 1d, 3d, 7d, 14d). 2.Preventive effects of aldose reductase inhibitor AL-1576 on galactose cataract in rat

Background Sugar cataract is one of the major diabetic complications in the eye,but there is not effective medicine to prevent or delay development of cataract. Objective The goal of this study was to investigate the effects and the potential mechanism of aldose reductase (AR) inhibitor,AL-1576 on prevention of galactose cataract in rats. Methods Forty-two SD rats were randomly and equally divided into 7 groups.The cataracts were induced by feeding with 50% galactose.At the day of feeding galactose and the day 5,10 and 15 after feeding galactose,AL-1576 was added into the feeds.The rats were divided AL-1576 prevention group and early-,intermediate-or late-stage intervention groups.For another group,the withdrawing AL-1576 group,AL-1576 was added into the feeds at the day of feeding galactose,then was removed after 10 days.The lenses of the rats were examined under the slit-lamp microscope before and after given AL-1576 every 5 days.At the day 35,the lenses were obtained.The wet and dry weight of the lenses were weighted,respectically,to calculate the water content of the lenses.Activities of AR and superoxidedismutase (SOD) and contents of glutathione (GSH) of the lenses were measured by their commercial detecting kits.The care and use of the animals complied with the Regulations for the Administration of Affairs Concerning Experimental Animals by State Science and Technology Commission. Results In AL-1576 prevention group,all lenses maintained clear.Opacification of the lenses were significantly attenuated in all three AL-1576 intervention groups and withdrawing AL-1576 group compared with the cataractous model group ( P<0.05),but the inhibiting role was weaken with late intervention.The water contents and the activities of AR of the lenses were decreased,the contents of SOD and GSH were dramatically increased in all different AL-1576 treated groups compared with the cataractous model group (P<0.05).Moreover,AL-1576 prevention group showed the best effect on all indexes (P<0.05). Conclusions The activity of AR can be inhibited by AL-1576 at the different stages of development of cataract induced by galactose.By blocking and attenuating formation of the edema and elevating antioxdative capacity in the lenses,AL-1576 prevents and delays the formation of galactose cataract.

4.Source data management in clinical researches.

Source data and its source documents are the foundation of clinical research. Proper source data management plays an essential role for compliance with regulatory and GCP requirements. Both paper and electronic source data co-exist in China. Due to the increasing use of electronic technology in pharmaceutical and health care industry, electronic data source becomes an upcoming trend with clear advantages. To face new opportunities and to ensure data integrity, quality and traceability from source data to regulatory submission, this document demonstrates important concepts, principles and best practices during managing source data. It includes but not limited to: (1) important concepts of source data (e.g., source data originator, source data elements, source data identifier for audit trail, etc.); (2) various modalities of source data collection in paper and electronic methods (e.g., paper CRF, EDC, Patient Report Outcomes/eCOA, etc.); (3) seven main principles recommended in the aspect of data collection, traceability, quality standards, access control, quality control, certified copy and security during source data management; (4) a life cycle from source data creation to obsolete is used as an example to illustrate consideration and implementation of source data management.

5.Study on blood uric acid,lipid levels and perceived stress in Kazakh hypertensive patients

Objective To study the relationship between Kazakh hypertensive patients with blood uric acid ,lipid levels and psy-chological factors .Methods 90 Kazak patients with hypertension in our hospital from January to August 2012 were randomly se-lected as the hypertension group and contemporaneous 171 non-hypertension patients as the healthy control group .Blood uric acid (UA) ,triglycerides(TG) ,total cholesterol(TC) ,low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein choles-terol(HDL-C) were detected and the perceived stress scale(PSS) was adopted to assess the mental status .Results The mean val-ues of TG ,TC ,HDL and LDL in males of the hypertension group and the mean values of UA ,TG and LDL in females of the hyper-tension group were significantly higher than those in the normal control group after adjusting age and BMI (P<0 .05);in the hyper-tension group ,UA was positively correlated with TC (r=0 .221 ,P<0 .05) .The level of perceived stress was positively correlated TC and LDL(r=0 .473 ,P<0 .05;r=0 .321 ,P<0 .05) and negatively correlated with TG and HDL (r= -0 .461 ,P<0 .05;r= -0 .412 ,P<0 .05) after adjusting age ,sex and BMI .Conclusion The blood lipid level in Kazakh hypertensive patients has certain correlation with the perceived stress .
6.The Effect of Tiaozhixin NO/ET、6-PGF_2/TXB_2 Hemorrhedoge of Rats with Hyperlipoidemia

Objective: To study the influences of Tiaozhixin(TZX) on NO/ET, 6 keto PGF 1a /TXB 2 and hemorrheology rats with hyperlipoidemia. Methods: The rat hyperlipoidemia and early atheroscleorsis models were established by feeding high lipid diet for 40 days. Meanwhile TZX was taken by oral administration at the dosages of 40、80g/kg. The levels of NO、ET 1、6 keto PGF 1a and TXB 2 in serum were determind, and the hemorrheology markers were observed. Results: TZX could raise the levels of NO in serumobviourly ET 1 level of the normal and model rats. The large dose of TZX could increase 6 keto PGF 1a content remarkably which benefits maintenance of the balances of 6 keto PGF 1a /TXB 2. It could lower the whole blood specific viscosity, whole blood reduction specific viscosity, plasma specific viscosity; aggregation index of RBC; shorten RBC electrophoresis time; also decrease fibrinogen content; inhibit the platelet aggregation of normal rats induced by ADP. Conclusion: TZX can improve the abnormal hemorrheology and recover the balance of TXB 2/6 keto PGF 1a and NO/ET of rats with hyperlipoidemia, which might be one of mechanisms of antiatherosclerosis action.
7.Comparison of Different PBL Effecting on the Clinical Teaching of Pediatrics

Objective To compare and evaluate the effect of PBL in clinical teaching of Pediatrics.Methods Among students of Grade 2002 in our university,two types of PBL,pre-learning and case-discussion,were used in their clinical learning of Pediatrics. And then,their effects were evaluated and compared with those of traditional learning method.Results More than 60% of the students agreed with PBL methods,and they considered PBL favorable to practice scientific logical thinking of clinical affairs,to increase their capabilities of learning,oral expression,communication and cooperation.The teachers agreed with PBL methods too for the better learning effect resulting from PBL.Conclusion PBL fits the needs of medical learning reformation.To train new type of doctors in century 21st,it is necessary to use kinds of new learning methods,including PBL methods and standardized patient (SP)in clinical teaching.
8.Nerve Stem Cells Orientation Differentiation in Neonatal Rat Hippocampus Induced by Brain-Derived Neurotrophic Factor in Vitro

Objective To explore the influence of brain-derived neurotrophic factor (BDNF) on the differentiation of nerve stem cells (NSCs) from neonatal rat hippocampus in vitro and to find new revulsant of NSCs,which can improve the percentage of NSCs differentiating into neurons.Methods Twenty-four hours neonatal rats were selected to obtain hippocampus tissue to culture NSCs in serum-free culture medium by suspending culture.The high pure NSCs were obtained after passing 2 generations.The culture cells were identified as NSCs by staining of nestin,which was NSCs special marker.After passaged three generations,the NSCs were randomly classified into 2 groups:test group and control group.There were 15 pieces per group.There was 2 mL per piece,which contains 1?105 cells.50 g/L fetal bovine serum(FBS) and 20 ?g/L BDNF were added into foundational culture medium in test group;only 50 g/L FBS was added into foundational culture medium in control group.The neurons and their percentage were tested using the immunofluorescence labeling and flow cytometer after 7 days of differentiated cultivation.Results The hippocampus tissue cells grew in globular in serum-free culture medium by suspending culture,which expressed highly positive by nestin immunofluorescence staining.Its purity was above 90%.The percentage of neurone specific enolase(NSE)-positive cells in test group was 60.45%,which was obviously higher than that of control group (23.67%).The difference was significant between 2 groups(?2=27.75 P

Torpor refers to a state in which the metabolic activity in the body of the living animal is greatly reduced during the period of reduced food supply, which is manifested as a substantial decrease in body temperature, metabolic level, and exercise level. Mammals have a strict body temperature regulation system to maintain a constant body temperature. When the energy supply is insufficient for a long time, some mammals will enter a hibernation state. Torpor is very similar to the hibernation state. The research on the mechanism of torpor state is of great significance in aerospace, military medicine and other fields. This review summarizes the specific mechanisms regulating the occurrence of torpor from four aspects: adenylate cyclase activating polypeptide (adcyap) neurons, leptin, pyroglutamylated RFamide peptide (QRFP) neurons, and sympathetic nervous system, aiming to provide ideas for further research on the mechanism of torpor.
