3.Assay and Analysis on Three Virus Pathogen of Pneumonia in Children

Objective To explore the child pneumonia in infection circumstances of Coxsackie B virus(CVB), Cytomegalovirus(CMV),Epstein-Barr virus (EBV).Methods Indirect ELISA were used to detect the specific IgM antibody of CVB, CMV, EBV. Results The total positive rate,which suffer from the three kinds of viruses in 140 children wich pneumonias was 38.6%.The positive rates of the CVB- IgM, CMV- IgM, EBV- IgM were 24.3%,8.6%,6.5%.Conclusions CVB has the higher infection rate in the pneumonia, is one of the main causes in the infection of child pneumonia.CMV,EBV also has the certain infection rate, although the infection rate is not high, they can cause damage of many organs and systems .They should be watched out for and valued.
4.Advances in theoretical studies on coagulating substances secreted by vascular endothelial cells

BACKGROUND:Vascular endothelial cel s participate in the coagulation cascade reaction or contraction of blood vessels by secreting abundant coagulating substances that promote coagulation. OBJECTIVE:To overview the effects of different coagulating substances secreted by vascular endothelial cel s, and provide theoretical basis for the screening of coagulant biomaterials. METHODS:A computer-based research in CNKI and PubMed databases was performed for relevant literatures addressing vascular endothelial cel s and its secreting coagulating substances published from 1988 to 2016 using the keywords of“vascular endothelial cel s, endothelin, Ang II, TXA2, tissue factor, col agen, fibronectin, von wilbrand factor, thrombospondin, platelet activating factor, plasminogen activator inhibitor, proaccelerin, antihemophlic factor”in English and Chinese, respectively. Final y 36 articles were enrol ed for result analysis. RESULTS AND CONCLUSION:Vascular endothelial cel s can secrete numerous coagulation factors that play important roles in the process of coagulation, inflammation reaction and thrombosis fol owing vascular injury. Among them, coagulation factor V and VIII are directly involved in the coagulation cascade reaction and promote thrombosis. In the meanwhile, the vasoconstrictors narrow the lumen, thereby assisting coagulation and promoting thrombosis indirectly. 5.The effect of lactulose oral solution in the treatment of patients with chronic functional constipation and its influence on the level of MTL and NO

Objective To explore the effect of lactulose oral solution in the treatment of patients with chronic functional constipation and its influence on the level of MTL and NO.Methods 54 cases of patients were selected with primary glaucoma who were treated in our hospital from June 2013 to October 2014 as research objective,and they were randomly divided into research group(29 cases)and control group(25 cases)according to the number table method.All of the patients were given treatment as diet adjusting,defecating exercise and so on.The research group got extra treatment as taking lactulose oral solution 10 g,2 to 3 times per day,for 6 weeks.Results MTL in the research group before treatment was 203.71ng/mL,and it was 371.03ng/mL after treatment for 6 weeks,there was statistically significance on difference of MTL levels between before treatment and after treatment(P <0.05).NO in the research group before treatment was 120.52ng/mL,andit was 69.01ng/mL after treatment for 6 weeks,there was statistically significance on difference of NO levels between before treatment and after treatment(P <0.05).MTL in the control group before treatment was 206.21ng/mL,which was 279ng/mL after treatment for 6 weeks,there was statistically significance on difference of MTL levels between before treatment and after treatment(P <0.05).NO in the control group before treatment was 123.92ng/mL,which was 98.75ng/mL after treatment for 6 weeks,there was statistically significance on difference of NO levels between before treatment and after treatment(P <0.05 ).MTL levels in the research group after treatment was higher than that in the control group when NO levels was lower than that in the control group,the difference was statistically significant(P <0.05).Conclusion Lactulose can improve the secretion of gastrointestinal hormones in plasma such as promoting the secretion of MTL and reducing the synthesis of NO.
6.Identification of Dendrobii Caulis basing on ITS sequence.

Isolation of high-quality genomic DNA from dried and processed crude drug is the key for the DNA identification of Dendrobii Caulis. The DNA extract of Dendrobii Caulis was firstly compared using different method to isolate genomic DNA from dried and processed crude drug, including commercial DNA extracted kit and CTAB method. Using modified CTAB method (extracted from large samples), the genomic DNA was successfully isolated from Dendrobii Caulis, including Huangcao and Fengdou. The ITS regions were amplified using the purified DNA as template, and then cloned and sequenced. These ITS sequences were compared with data from Genbank database and our lab, 14 Dendrobium species and five similar species were identified from "Huangcao" and "Huangcao" slice, while six species and three similar species from "Fengdou" according to their sequence similarity. The study demonstrated that the dried Dendrobii Caulis could be identified using DNA molecular method, which could overcome deficiencies and limitations of traditional identification method and has a certain application prospects.

OBJECTIVETo investigate the clinical significance of matrix metalloproteinase-9 (MMP-9) and tissue factor (TF) secreted by cultured monocyte-derived macrophages (HMDM) from patients with coronary heart disease (CHD) in vitro, and to evaluate the intervenient effect of puerarin (Pur) on them.
METHODSA total of 40 patients were enrolled, including 12 patients with acute myocardial infarction (AMI), 16 patients with unstable angina pectoris (UAP), 12 patients with stable angina pectoris (SAP). Besides, 8 healthy subjects with normal coronary arteriograph were set as controls. Monocytes acquired from their peripheral blood were incubated for 48 h and induced to differentiate into macrophages by phorbolester 12-myristate 13-acetate (PMA), the contents of MMP-9 and TF in supernatant were assayed, and the relationship of them with patients' age, risk factors of CHD and coronary artery lesion scores were analyzed. HMDMs randomly from selected 12 patients with acute coronary syndrome (ACS) were arranged for observing the intervenient effects of different concentrations of Pur on the levels and activity of MMP-9 and TF.
RESULTSThe levels of MMP-9 and TF in UAP and AMI patients were significantly higher than those in SAP patients and healthy subjects (P < 0.01), but no statistical correlation was found between levels of MMP-9 and TF with different CHD risk factors, as well as patients' age and coronary artery lesion scores. The levels and activity of MMP-9 and TF in the 12 ACS patients were significantly decreased in a dose-dependent manner after Pur intervention when compared with the controt group.
CONCLUSIONThe levels of MMP-9 and TF secreted in vitro by HMDM from CHD patients could be taken as indexes for evaluating patient's condition of ACS. Pur can inhibit the expression and the activity of MMP-9 and TF secreted by HMDM, stabilize the plaque and improve the vulnerability of blood to certain extent.

9.Evaluation of CT-guided percutaneous transthoracic biopsy in the diagnosis of mediastinal masses

Objective To evaluate the clinical application and safety of CT-guided pereutaneous transthoracic biopsy in the diagnosis of mediastinal masses.Methods Thirty three cases were undertaken CT- guided percutaneous transthoraeie biopsy with automatic biopsy gun and then the sampling specimens were undergone histological examination.The accuracy of puncture,diagnostic correctness and complications were analyzed.Results The operations were performed successfully in all 33 cases(100%),the definite pathologic diagnosis were made in 28 out of 33 cases(85%)and no complications occurred.Conclusion As for midiastinal masses,CT-guided percutaneous transthoracic biopsy is a feasible,successful,efficient interventional diagnostic method with high accuracy in localization,puncture,diagnosis and few complications, which should he recommended in clinical use more widely.(J Intervent Radiol,2007,16:852-854)
10.Clinical outcome of therapeutic cervical cerclage in short cervix syndrome

Objective To investigate the clinical effect of therapeutic cervical cerclage on short cervix syndrome for anti-premature birth in the second trimester. Methods Totally 44 singleton pregnant patients were diagnosed as short cervix syndrome, which was cervical length ≤2.5 cm without cervical dilatation,and received treatment from January 2008 and July 2015 in Peking University Third Hospital were collected. Among them, 30 patients who received therapeutic cervical cerclage were defined as cerclage group and another 14 cases who received conservative treatment were defined as un-cerclage group. The days of conservative treatment, delivery rate of different gestational weeks, birth weight of newborns, neonatal survival rate within 7 days of birth were analyzed between the two groups. Results There were no significant differences between the two groups in days of pregnancy conservative treatment [103(84-141)vs 105(85-114)days], delivery weeks [38.0(35.5-39.4)vs 38.5(37.3-39.5)weeks], birth weight of newborns [3120(2750-3400)vs 3130(2760-3545)g], and survival rate of newborns [100%(30/30)vs 13/14]. The fetuses of both groups were all delivered after 28 weeks. There was no significant difference in accumulated delivery rate between the two groups after 32 weeks, 34 weeks, and 37 weeks, respectively(all P>0.05). Conclusions The treatment of cervical cerclage is not superior to conservative means in single pregnancy of cervical length ≤2.5 cm without cervical dilatation. For such patients with short cervix syndrome, the treatment of cervical cerclage may not be necessary, but dynamic monitoring and search for the causing factors and prompt treatment are more important.
