1.Effect of betulinic acid on proliferation,apoptosis,and cell cycle of human lymphoma cell line Raji

Objective To investigate the anticancer effects and molecular mechanism of betulinic acid (BA)on Raji cells in vitro.Methods The effects of BA on the growth of Raji cells were studied by MTT assay.Apoptosis was assessed by Annexin-V/PI double-labeled cytometry.The influence on cell cycle was studied by flow cytometer.The cyclin D3 mRNA expression was checked by Western blotting and RT-PCR techniques.Results BA showed obvious inhibition on proliferation,as well as induction potency of apoptosis on Raji cells in vitro in a time-and dose-dependent manner by Annexin-V/PI double-labeled method.With the IC_(50)value for 24 h being(39.44?0.65)?g/mL,Raji cells treated with BA showed ac- cumulation in G_0/G_1 phase and reduction in the percentage of cells in S phase.The cyclin D3 mRNA ex- pression and protein were sharply decreased in Raji cells treated with BA.Conclusion BA could inhibit the proliferation of Raji cells by regulating the cell cycle that arrests cells at G_0/G_1 phase and induces apop- tosis of Raji cells.The antitumor effects of BA may be related to down-regulation of the expression of cy- clin D3.
2.Surgical management of traumatic false aneurysms in the extremities in 17 cases

Objective To review the surgical managements of patients with traumatic false aneu- rysms in the extremities.Methods From January 1990 to April 2006,17 patients with traumatic false aneurysms in the extremities were admitted into our hospital.Fourteen patients were treated by vascular repair including vascular repair in seven cases,end to end anastomosis in one,synthetic grafting in one, autogenous vein grafting in one,and direct ligation in four.Three patients were treated nonoperatively, but with local compressive dressing.Results There were no deaths or gangrenes in all cases.The clinical manifestations vanished after the treatment.The mean follow-up period was 13.2 months.The function of the injured extremities recovered satisfactorily.Conclusion Different types of traumatic false aneurysms should be managed by different therapeutic procedures after the diagnoses is made.
3.Correlation between the serum ferritin levels and the post-stroke depression at 3 months after ischemic stroke

Objective To investigate the correlation between the serum ferritin levels and the post-stroke depression (PSD). Methods From July 2014 to October 2015, the inpatients with the first-ever acute ischemic stroke were colected consecutively. Chemiluminescence microparticle immune assay was used to measure the serum ferritin levels within 24 h after admission. Depressive symptoms were screened by using the 17-item Hamilton depression scale (HAMD-17) at 3 months after onset. In patients with a HAMD-17 score ≥7, the depression was further diagnosed according to The Diagnostic and Statistical Manual of Mental Disorders, 4th edition. Results A total of 200 patients with the first-ever acute ischemic stroke were enroled, 55 (27. 5% ) of them were diagnosed as PSD. There were significant differences in the body mass index (BMI), years of education, waist circumference, high sensitive-C-reactive protein, homocysteine, National Institutes of Health Stroke Scale score (at baseline, discharge, and day 90), mRs score (at discharge and day 90), BI (at discharge and day 90), and the proportions of widowed or solitary patients between the PSD group and the non-PSD group (al P < 0. 05 ). The serum ferritin level in the PSD group was significantly higher than that in the non-PSD group ( median [ interquartile range], 261. 90[142. 10-364. 90] μg/L vs. 164. 40[132. 50- 195. 10] μg/L; Z = - 4. 814, P < 0. 001 ). Multivariate logistic regression analysis adjusted for confounding factors showed that the baseline serum ferritin level >136. 375 μg/L was an independent risk factor for PSD (odds ratio 1. 041 per 1-quartile increase, 95%confidence interval 1. 009-1. 239; P = 0. 045). Conclusions The elevated baseline serum ferritin level is associated with PSD.
4.Modified tubo-uterine implantations for proximal tubal occlusive infertility after femal sterilization with mucflago phenol

Objective To explore the effects of modified tubo-uterine implantations performed on women with proximal tubal occlusive infertility after femal sterilization with mucilago phenol.Methods Two hundred and eight infertile women who were admitted to the Second Affiliated Hospital of Sun Yat-sen University between 1986 and 2004 were included.They all accepted modified tubo-uterine implantation after occlusion of fallopian tubes with mucilago phenol.Results It was found that the occlusions were all located in the interstitial portion or isthmic portion of the fallopian tubes.Different degrees of pelvic adhesions were found in 65 cases.Fifty-seven cases were slightly adhesive,seven cases were of moderate degree and one case was severe.One hundred and ninety-nine cases were followed up after operations(95.7%).One hundred and ninety-three women accepted hydrotubation in the following month just after the operation and 185 women were found to be unobstructed(95.8%).One hundred and forty-three women became pregnant, the pregnant rate being 71.9%(143/199).One hundred and twenty-five women had term deliveries (87.4%),three women were in early pregnancy and two in midtrimester pregnancy.Eleven women had spontaneous abortion(7.7%).Two women had tubal pregnancy(1.0%).None of the 199 cases had any signs of endometriosis.Conelusions Modified tubo-uterine implantations are quite effective for proximal tubal occlusive infertility.It may be a favorable method for such kind of tubal occlusions.
5.Effects of Nd:YAG laser on micro-tensile bond strength of single bond adhesive system for non-carious sclerotic dentin.

OBJECTIVETo investigate the effect of Nd:YAG laser on microtensile bond strength of single bond adhesive system for non-carious sclerotic dentin.
METHODSTen human molars with occlusal wearing were cut into equal halves (nearly 12 mm(2)), and randomly divided into experimental group and control group. The teeth in experimental group were processed with Nd:YAG laser (1 W, 10 Hz), and then applied with Scotchbond and filled with Z350 resin. In control group, the teeth were processed with single bond and filled with Z350 resin. The specimens were sectioned, and the microtensile bond strengths of each sample was tested by a universal testing machine.
RESULTSThe bond strength of the experimental group [(26.11 ± 1.62) MPa] was significantly higher than that of the control group [(22.27 ± 2.16) MPa], P < 0.05. Stero-microscope examination indicated that most of the fractures occurred in dentin-resin interface.
CONCLUSIONSNd:YAG laser can increase the microtensile bond strength of single bond adhesive system in non-carious sclerotic dentin.

6.The different characteristics of ABL kinase domain mutation in the Chinese Han nationality imatinib resistant Philadelphia chromosome-positive acute lymphoblastic leukemia and chronic myeloid leukemia.

OBJECTIVETo identify the distribution and differentiation of ABL kinase domain mutation in the Chinese Han nationality imatinib resistant chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph(+)ALL).
METHODSBone marrow or peripheral blood samples of 112 imatinib resistant CML patients and 21 Ph(+)ALL patients were obtained from the first affiliated hospital of Soochow university according to local law. Total RNA was extracted from the mononuclear cells using a TRIzol reagent. ABL kinase domain (KD) mutation was detected by direct sequencing.
RESULTSOf the 112 imatinib resistant CML patients, 54.46%(61 cases) had ABL KD mutation. Twenty-three mutants were identified in 20 amino acid sites and 23.21% (26 cases) ABL KD mutations were in P-loop region. ABL KD mutations were also detected in 71.43% (15 cases) imatinib resistant Ph(+)ALL patients, with 10 mutations in 8 amino acid sites. The most frequent mutation was T315I (28.57%), followed by E255K/V (19.05%) and Y253F/H (14.29%). The frequency of T315I was much higher in imatinib resistant Ph(+) ALL than that in imatinib resistant CML (P = 0.001). Ph(+)ALL with additional chromosomal aberrations also had a higher rate of ABL KD mutation than that of CML (P = 0.010). Ph(+)ALL gained ABL KD mutation faster than CML (P < 0.010).
CONCLUSIONChinese imatinib resistant CML and Ph(+)ALL patients had different characteristics in ABL KD mutation. The rate of ABL KD mutation in Ph(+)ALL with additional chromosomal aberrations was much higher than that of CML with additional chromosomal aberrations.

OBJECTIVETo investigate the anticancer effects of betulinic acid (BA) on Jurkat cells in vitro and its molecular mechanism.
METHODSThe effects of betulinic acid on the growth of Jurkat cells were studied by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5diphenyl-2H-tetrazolium (MTT) assay. Apoptosis was assessed by Hoechst33258 staining and annexin-V/PI double-labeled cytometry. The effect of betulinic acid on the cell cycle of Jurkat cells was studied by propidium iodide staining. RT-PCR and Western blot were used to analyze the changes of cyclin D3, bcl-xl mRNA and protein levels in Jurkat cells after treatment with betulinic acid.
RESULTSThe proliferation of Jurkat cells was decreased in betulinic acid-treated group at a 24 h IC50 value of 70.0 micromol/L. The effect of betulinic acid to induce apoptosis in Jurkat cells was in a time- and dose-dependent manner. Jurkat cells treated with betulinic acid showed an increase of G0/G1 phase and decrease of S phase. The Jurkat cells treated with 0, 20, 60, 100 micromol/L betulinic acid for 24 h, showed an increased G0/G1 phase from 31.0% to 58.8%, whereas decreased S phase from 61.5% to 35.8%, respectively. PBMC was less sensitive to the cytotoxic effect of betulinic acid than Jurkat cells. The expression of cyclin D3, bcl-xl mRNA and protein were decreased sharply in Jurkat cells treated with betulinic acid.
CONCLUSIONBetulinic acid can inhibit the proliferation of Jurkat cells by regulating the cell cycle that arrests cells at G0/G1 phase and induces apoptosis in Jurkat cells. The antitumor effects of betulinic acid may be related to down-regulation of the expression of cyclin D3 and bcl-xl.

9.Urethroplasty with tunica vaginalis flap for treating hypospadias with hydrocele: a report of 9 cases.

OBJECTIVETo explore the treatment of hypospadias with hydrocele.
METHODSNine patients with hypospadias complicated by hydrocele underwent urethroplasty with tunica vaginalis flap.
RESULTSSatisfactory results were achieved in 7 cases, all with a normal anatomic position of urethral meatus, perfect urination and no infection. Local infection and meatal stricture occurred in 1 case, which was restored to normal by urethral sounding. Scrotal urinary fistula developed in another.
CONCLUSIONWith minimum surgical trauma, conveniently available material and fast tissue repair, urethroplasty with tunica vaginalis flap is a desirable method for the treatment of hypospadias with hydrocele.

10.Preliminary study on quantitative assessment of nonalcoholic fatty liver disease by MRI

Objective To discuss the feasibility for quantitative assessment of nonalcoholic fatty liver disease by sequence of diffusion weighted imaging and hydrogen proton magnetic resonance spectroscopy.Methods Thirty patients with NAFLD and twenty-six normal volunteers were enrolled in an experiment group,and 25 healthy volunteers were selected into a control group.All the patients and volunteers underwent CT and MRI examinations.With the ratio of liver-to-spleen CT value as reference standard,the correlation and difference of ADC value,value of water peak,the area under water peak,values of lipid peak and area under lipid peak and fat fraction were measured and compared between the two groups.Results There was positive significant correlation between ratio of liver-to-spleen CT value and ADC value (r=0.92,P<0.05),and negative significant correlation between ratio of liver-to-spleen CT value and lipid peak and area of lipid peak (r=-0.91 and r=-0.96,respectively;all P<0.05),there was significant negative correlation between ADC value and lipid peak and area of lipid peak (r=-0.91,P<0.05),and ADC value,the values of water peak,lipid peak and area under peak and fat fraction were different between the two groups (P<0.05).Conclusion There is significant correlation and difference between DWI and 1H-MRS in evaluating NAFLD,which has the potential to quantitatively diagnose the severity and degree of fatty liver.
