Objective To synthesize 99Tcm- (hydrazinonictinamide- [Lys3] -bombesin) (tricine)(trisodium triphenylphosphine-3,3',3"-trisulfonate) ((HYNIC-[Lys3]-BBS) (tricine) (TPPTS)) and evaluate its biodistribution and binding capability with tumor tissue in Balb/c nude mice bearing human pancreatic cancer xenografts. Methods HYNIC was conjugated to the [Lys3] -BBS at pH = 9.0 with SnCl2 as reducing agent and both tricine and TPPTS as coligands for 99Tcm-labeling. 99Tcm-HYNIC-[Lys3]-BBS)(tricine) (TPPTS) was purified by Sep-Pak C18 cartridge and was analysed by HPLC. The radiochemical purity and radiolabeling yield were measured. The stability of 99Tcm-(HYNIC-[Lys3]-BBS) (tricine)(TPPTS) in serum, biodistribution (% ID/g) in the normal mice and imaging of the Balb/c nude mice bearing human pancreatic cancer xenografts in vivo were studied. Results The radiolabeling yield was (90 ±2)% and the radiochemical purity was over 95%. The radiochemical purity after 4 h in serum was over 85%. The distribution in normal mice showed rapid clearance from blood (the uptake was (0.07 ±0.01) %ID/g at 2 h postinjection). 99Tcm-(HYNIC-[Lys3]-BBS) (tricine) (TPPTS) was excreted mainly via the kidney with little radioactivity accumulation in the liver and gastrointestinal tract (the uptake of liver, stomach, intestine was (0.27 ±0.03), (0.06 ±0.03), (0.04 ±0.00) %ID/g at 2 h postinjection). Marked uptake of radioactivity was found in tumor tissue of the Balb/c nude mice bearing human pancreatic cancer with maximum T/NT ratio of 3.71 ± 0.57 at 2 h postinjection. Conclusions 99Tcm-(HYNIC-[Lys3]-BBS)(tricine) (TPPTS) can be easily prepared with high radiolabeling yield and radiochemical purity. The stability in serum and good biodistribution charateristics make it useful for the diagnosis of human pancreatic cancer with over-expression of the gastric-releasing peptide(GRP) receptor.

OBJECTIVETo investigate the effects of polysaccharide 2-1 from Gastrodia elata (PGE2-1) on blood coagulation and thrombosis.
METHODClotting time (CT) and bleeding time (BT) of mice were measured by glass method and tail-cutting method. Bleeding capacity (A540) was measured by cutting tail in 5 min. Plama recalcificatic time (RT) were measured in mice. Platelet aggregation was caused by adenosine diphosphate (ADP). Activated partial thromboplastin time (APTT), prothrombin time (PT), and thrombin time (TT) were measured by reagent boxes. During thrombosis in vitro, their lengths, wet and dry weights were measured by instrument; wet weights of arteriovenous experimental thrombosis were measured and the impressive rates were analyzed.
RESULTCT and BT of groups PGE2-1 (60, 120 mg x kg(-1)) were remarkably prolonged, and bleeding capacity (A540) were significantly increased (P < 0.05 or P < 0.01). RT of groups PGE2-1 (30, 60, 120 mg x kg(-1)) were remarkably prolonged, and platelet aggregation (PAG) were inhibited (P < 0.05 or P < 0.01). Human serous TT and APTT of groups PGE2-1 (10, 20, 40 mg x mL(-1)) were remarkably prolonged (P < 0.05 or P < 0.01), but the difference of effect on PT had no statistic significance. PGE2-1 (30, 60, 120 mg x kg(-1)) could make the mice obviously eliminate thrombus symptom and reduce the time of restoring independent activity (P < 0.05 or P < 0.01); thrombosis in vitro: Lengths, wet and dry weights of groups PGE2-1 (30, 60, 120 mg x kg(-1)) were significantly decreased (P < 0.05 or P < 0.01); wet weights of arteriovenous experimental thrombosis were dramatically decreased (P < 0.01), and impressive rates were respectively 32.5%, 49.0% and 61.5%.
CONCLUSIONPGE2-1 has remarkable effects of anticoagulation and antithrombosis, so it may be the main component of the isolation from G. elata in the field of antithrombosis.

Current study was carried out to optimize the priming condition of Oldenlandia diffusa seeds, and improve germination rate and seed vigor of 0. diffusa seeds under drought conditions. Uniform design was used to optimize the concentration and priming time of three priming materials (PEG, KNO3, GA3). Different concentrations of polyethylene glycol (PEG) was used to simulate drought stress. The seedling was cultured in 1/4 Hoagland medium for 30 d. The results showed that seed priming treatment with 366 mg x kg(-1) GA3 for 1h resulted in significant increase in germination rate, germination index, vigor, root length, plant height and biomass of O. diffusa seeds under drought stress (15% PEG), while seed priming with 3.0% KNO3 for 1 h showed little effect on germination and growth of O. diffusa seeds under drought stress. Seed priming treatment with appropriate GA3 concentration and priming time could enhance seed germination and drought resistance of O. diffusa in seedling stage.

Objective To observe the effects of five flavonoids include rutin(RU),dihydromyricetin(DMY),hesperetin(HP),daidzein(DA)and hydroxysaffor yellow A(HYSA)on myocardiocyte apoptosis induced by H2O2 and to explore their relationships with Keshan disease and the possible mechanism. Methods Primary cultured cardiocytes of neonatal rats were randomly divided into control group, model group, and flavonoids preincubation group. The cardiocyte apoptosis was examined by fluorescent staining, the rates of apoptosis were detected by flow cytometry, the expression of Bcl-2 family proteins associated with apoptosis were observed:by Western blot. Results Compared with model group[(24.33±6.51)%],RU[(13.95±3.80)%],DA[(11.82±3.50)%],HYSA[(12.33±3.78)%]could decreased the rate of apoptosis(P<0.05).The five flavonoids could up-regulate Bcl-2 expression,down-regulate Bax expression, and increase the Bcl-2/Bax ratio[RU(0.989±0.094),DMY(0.931±0.280),HP(0.980±0.095),DA(1.049±0.092),HYSA(1.031±0.039),vs model(0.490±0.046),the difference had statistical significances(P<0.05)],but the Bcl-xl did not significantly changed(P>0.05).Conclusions RU,DMY,HP,DA and HYSA have antiapoptotic effects on cardiomyocyte via regulating Bcl-2 and Bax,which gives us a hint in prevention and treatment of Keshan disease.
