3.Advances in theoretical studies on coagulating substances secreted by vascular endothelial cells

BACKGROUND:Vascular endothelial cel s participate in the coagulation cascade reaction or contraction of blood vessels by secreting abundant coagulating substances that promote coagulation. OBJECTIVE:To overview the effects of different coagulating substances secreted by vascular endothelial cel s, and provide theoretical basis for the screening of coagulant biomaterials. METHODS:A computer-based research in CNKI and PubMed databases was performed for relevant literatures addressing vascular endothelial cel s and its secreting coagulating substances published from 1988 to 2016 using the keywords of“vascular endothelial cel s, endothelin, Ang II, TXA2, tissue factor, col agen, fibronectin, von wilbrand factor, thrombospondin, platelet activating factor, plasminogen activator inhibitor, proaccelerin, antihemophlic factor”in English and Chinese, respectively. Final y 36 articles were enrol ed for result analysis. RESULTS AND CONCLUSION:Vascular endothelial cel s can secrete numerous coagulation factors that play important roles in the process of coagulation, inflammation reaction and thrombosis fol owing vascular injury. Among them, coagulation factor V and VIII are directly involved in the coagulation cascade reaction and promote thrombosis. In the meanwhile, the vasoconstrictors narrow the lumen, thereby assisting coagulation and promoting thrombosis indirectly. 5.Effect of Rhodiolae rosae on extracellular regulated kinases and angiogenesis in post myocardial infarction

AIM:To study the role of Rhodiolae rosae in post myocardial infarction angiogenesis and its effect on extracellular regulated kinases(ERK 1/2). METHODS: The experimental myocardial infarction was made by ligation of rat left anterior descending coronary artery,and then divided into Rhodiolae group and saline group.The sham operation group and normal group were also made as control one.On the first,the 4th,the 7th and the(14th) day after the operation,rats were sacrificed and infarcted border tissue or normal tissue were used to test VEGF receptor Ⅱ and CD34 expression level and ERK 1/2 activity level. RESULTS: ERK 1/2 activity level from the first to the 4th day,VEGF receptor Ⅱ and CD34 expression level from the 4th to the 14th day after the operation,were all higher in the Rhodiolae group than that in the saline group. CONCLUSION: Rhodiolae rosae can elevate ERK 1/2 activity which was related to and prior to its angiogenic effect.
6.Effect and Interaction of Total Flavonoids and Saponins from Huang-Qi Ge-Gen Decoction on IL-12 and IL-15 of Liver among Diabetes Rats

This study was aimed to explore the effect of total flavonoids and saponins from Huang-Qi Ge-Gen (HQGG) decoction on blood glucose (BG), serum lipid, interleukin-12 (IL-12) and interleukin-15 (IL-15) of liver in diabetes mellitus (DM) rats, in order to investigate their interactions in regulating DM processes. A total of 66 SD rats were randomly divided into 6 groups, which were the normal group, model group (A1B1), control group, total flavonoids group (A2B1), total saponins group (A1B2), and total flavonoids and saponins group (A2B2), with 11 rats in each group. Except the normal group, other groups were intraperitoneal injected with streptozotocin (STZ). And the experiment was according to 2×2 factorial design experiment scheme. The BG was determined before STZ injection and 7 days after the STZ injection. After 30 days, BG, serum lipid, IL-12 and IL-15 of liver were tested. Related indexes were calculated to the weighted composite score. Main and interactive effect of total flavonoids and saponins were studied according to the factorial design experiment scheme. The results showed that compared with the normal group, all indexes of model group showed statistical differences (P<0.05). Total flavonoids and saponins from HQGG decoction can effectively reduce BG, without any interactions between them. Both the total flavonoids and total saponins can reduce serum cholesterol (CHO), triglyceride (TG), liver IL-12 and IL-15. And there were interactive effects. The single use of herb achieved better effects than the combination. It was concluded that total flavonoids and saponins from HQGG decoction can reduce BG, CHO, TG, and liver IL-12 and IL-15 levels in rats. However, the regulation of total flavonoids and saponins on indexes mentioned above showed no additive effect.
7.Study on Diterpenoid Alkaloids from the Roots of Aconitum Straminiflorum

Objective:To study the chemical constituents of Aconitum straminiflorum. Methods:The air-dried roots of A. stra-miniflorum were powdered and extracted three times with 90% ethonal under reflux. After the removal of the solvent under reduced pressure,the crude extract was dissolved in 1. 5% HCl solution(7 L). The acidic solution was basified to pH 9. 0 with ammonia (25%)and extracted with CHCl3 to obtain crude extract of total alkaloids after the removal of CHCl3 . The compounds were isolated and purified by column chromatography and identified based on spectral analysis( MS,1 H-üMR and 13 C-üMR). Results:Totally 7 compounds were isolated from A. straminiflorum,and characterized as aconosine(1),13-deoxyludaconitine(2),dolaconine(3), ezochasmanine(4),talatisamine(5),scopaline(6)and cammaconine(7),respectively. Conclusion:Seven diterpenoid alkaloids are isolated from A. Straminiflorum for the first time.
8.Clinical significance of matrix metalloproteinase-9 and tissue factors secreted by cultured monocyte-derived macrophage of patients with coronary heart disease in vitro and the intervenient effect of puerarin on them.

OBJECTIVETo investigate the clinical significance of matrix metalloproteinase-9 (MMP-9) and tissue factor (TF) secreted by cultured monocyte-derived macrophages (HMDM) from patients with coronary heart disease (CHD) in vitro, and to evaluate the intervenient effect of puerarin (Pur) on them.
METHODSA total of 40 patients were enrolled, including 12 patients with acute myocardial infarction (AMI), 16 patients with unstable angina pectoris (UAP), 12 patients with stable angina pectoris (SAP). Besides, 8 healthy subjects with normal coronary arteriograph were set as controls. Monocytes acquired from their peripheral blood were incubated for 48 h and induced to differentiate into macrophages by phorbolester 12-myristate 13-acetate (PMA), the contents of MMP-9 and TF in supernatant were assayed, and the relationship of them with patients' age, risk factors of CHD and coronary artery lesion scores were analyzed. HMDMs randomly from selected 12 patients with acute coronary syndrome (ACS) were arranged for observing the intervenient effects of different concentrations of Pur on the levels and activity of MMP-9 and TF.
RESULTSThe levels of MMP-9 and TF in UAP and AMI patients were significantly higher than those in SAP patients and healthy subjects (P < 0.01), but no statistical correlation was found between levels of MMP-9 and TF with different CHD risk factors, as well as patients' age and coronary artery lesion scores. The levels and activity of MMP-9 and TF in the 12 ACS patients were significantly decreased in a dose-dependent manner after Pur intervention when compared with the controt group.
CONCLUSIONThe levels of MMP-9 and TF secreted in vitro by HMDM from CHD patients could be taken as indexes for evaluating patient's condition of ACS. Pur can inhibit the expression and the activity of MMP-9 and TF secreted by HMDM, stabilize the plaque and improve the vulnerability of blood to certain extent.

9.Evaluation of CT-guided percutaneous transthoracic biopsy in the diagnosis of mediastinal masses

Objective To evaluate the clinical application and safety of CT-guided pereutaneous transthoracic biopsy in the diagnosis of mediastinal masses.Methods Thirty three cases were undertaken CT- guided percutaneous transthoraeie biopsy with automatic biopsy gun and then the sampling specimens were undergone histological examination.The accuracy of puncture,diagnostic correctness and complications were analyzed.Results The operations were performed successfully in all 33 cases(100%),the definite pathologic diagnosis were made in 28 out of 33 cases(85%)and no complications occurred.Conclusion As for midiastinal masses,CT-guided percutaneous transthoracic biopsy is a feasible,successful,efficient interventional diagnostic method with high accuracy in localization,puncture,diagnosis and few complications, which should he recommended in clinical use more widely.(J Intervent Radiol,2007,16:852-854)
10.Effects of frequency on osteoblast apoptosis induced by tensile strain

Objective To investigate the effect of frequency on osteoblast apoptosis induced by tensile strain.Methods MC3T3-E1 cells were applied with 1% biaxial tensile strain at the frequency of 1,2,3,4,5 Hz,re spectively for 1 hour per day in 8 days.The survival rate of the cells was determined by activity of lactate dehydrogenase (LDH).Annexin V-FITC/ PI Flow cytometry was used to test cell apoptosis.Real-time RT-PCR was used to detect the gene level of apoptosis markers caspase-3,-9 as well as Bcl-2 and Bax,and Western blotting was used to test protein expressions of caspase-3,-9.Results Different loading frequencies had no effect on osteoblast activity of LDH.There was no significant difference in the total apoptosis rate of flow cytometry at different frequencies.However,the frequency of 2 Hz could induce early osteoblast apoptosis.Tensile strain at the frequency of 2 Hz could significantly increase the expression of caspase-3,-9 gene and protein,and induce cell apoptosis with the up-regulation of the Bax/Bcl-2.Conclusions Osteoblast apoptosis and death cannot be induced by 1% biaxial tensile strain at the frequency of 1-5 Hz,but the frequency of 2 Hz can induce the early apoptosis of osteoblasts by up-regulating the expression of Bax/BCI-2.
