3. The angiogenesis patterns of human glioma transplanted in nude mice
Tumor 2007;27(6):437-440
Objective: To investigate the expression of molecular marker ABCG2 of stem cells and tumor angiogenesis-related gene products including vascular endothelial growth factor (VEGF), CD34 and very late antigen alfa-2 (VLA-2α) in human glioma-bearing nude mice and observe the patterns of angiogenesis in transplanted tumors by CD34-PAS double staining. Methods: The human glioma cells were subcutaneously transplanted in 30 nude mice. Six normal brain tissues were as controls. The hematoxylin and eosin staining and immunohistochemical examination were performed to determine the positive rate and the hot-spot of gene product expression. The patterns of angiogenesis were observed under light microscope. Results: The positive rates of ABCG2, VEGF, and VLA-2α expression were all 100% in transplanted tumors. The average percentage of positive cells was (3. 93 ± 1. 438)%, (34. 84 ± 6.212)% (30.33 ± 4.428)%, respectively. The value of micro-vascular density (MVD) labeled by CD34 averaged at 29. 78 ± 5.88. All the three microcirculation patterns, endothelium-dependent vessels, mosaic vessels and vasculogenic mimicry, were present in xenografted human glioma tissue samples. Conclusions: The ABCG2-overexpressed cells tended to be located close to blood vessels. VEGF and VLA-2α play an important role in angiogenesis of transplanted tumors in nude mice. There exist three different microcirculation patterns in high malignant transplanted tumors.

Objective To study the effect of blood glucose variation on gastric emptying and ghrelin expression in diabetic rats, and to explore the relationship between the delayed gastric emptying and the ghrelin expression in different diabetic stages. Methods Sixty Wistar rats were randomly divided into three groups:a normal control group(NC group),a diabetes mellitus group (DM group) induced by intra- peritoneal injection of streptozotocin (STZ),and an insulin-treated group (INS group). After one and four weeks the gastric emptying was measured by intragastric administration of phenol red and the expression of gastric ghrelin was determined by immunohistochemistry and semi-quantitative RT PCR.Results After one week of STZ injection, the gastric emptying, the ghrelin integral optical density and the ghrelin mRNA expression decreased significantly in DM group compared to those in NC group and INS group(P0.05).Conclusion Short term hyperglycemia delay gastric emptying through the reduction of gastric ghrelin expression, while long-term hyperglycemia may enhance the expression and release of gastric ghrelin to stimulate food intake and maintain energy balance.

Objective To develop the human colon carcinoma(HT-29) liver metastasis model in nude mice, and to detect the expression of human telomerase reverse transcriptases(hTERT) mRNA in liver of nude mice. Methods Liver metastases were established in 30 Balb/c nude mice by intrasplenic injection of colonic cancer cells(HT-29),and the spleens were resected. After being sacrificed, the tumor growth was observed, and the pathological examinations were performed. The expression of hTERT mRNA in the livers of nude mice was detected by RT-PCR technique. ResultsHuman colon carcinoma(HT-29) liver metastasis model was developed in all the 30 nude mice, with the liver metastasis rate of 100%.The pathological results revealed the occurrence of liver metastases, and the expression of hTERT mRNA was positive in the liver tissues. Conclusion Intrasplenic injection of HT-29 cell is a reliable way for producing colonic cancer liver metastasis model, and the positive expression of hTERT mRNA in liver tissues indicates the significance of hTERT in the early diagnosis and treatment of colon carcinoma liver metastasis.
