3.Role of JNK signaling pathway in paclitaxel-induced apoptosis in hippocampal neurons of rats: the relationship with NF-κB pathway

Objective To evaluate the role of c-Jun N-terminal kinase (JNK) signaling pathway in paclitaxel-induced apoptosis in hippocampal neurons of rats, and the relationship with nuclear factor kappa B (NF-κB) pathway.Methods The primarily cultured hippocampal neurons were seeded in 96-well plate at a density of 1×106 cells/ml (200 μl/hole) , and were randomly divided into 4 groups (n=8 each) using a random number table: control group (C group), paclitaxel group (P group), JNK inhibitor SP600125 group (S group), and SP600125 + paclitaxel group (S+P group).Paclitaxel 2 ml (1 μmol/L) was added to group P.SP600125 2 ml (10 μmol/L) was added to group S.In group S+P, SP600125 2 ml (10 μmol/L) was added, the cell were then incubated for 1 h, and then paclitaxel 2 ml (1 μmol/L) was added.The cells were then incubated for 24 h.At 24 h of incubation, the apoptosis in hippocampal neurons was detected by flow cytometry, and the expression of NF-κB p65 was measured by Western blot.The apoptosis rate was calculated.Results Compared with group C, the apoptosis rate was significantly increased, and the expression of NF-κB p65 was up-regulated in P and S+P groups, and the apoptosis rate was significantly decreased, and the expression of NF-κB p65 was down-regulated in group S (P<0.05).Compared with group P, the apoptosis rate was significantly decreased, and the expression of NF-κB p65 was down-regulated in group S+P (P<0.05).Conclusion JNK signaling pathway mediates paclitaxel-induced apoptosis in hippocampal neurons of rats, and the mechanism is likely related to inhibition of NF-κB pathway activation.

Allele-specific polymerase chain reaction (AS-PCR) is a technique based on allele-specific primers, which can be used to analyze single nucleotide polymorphism (SNP) effectively including the transition, transversion and insertion/deletion polymorphism and has been exploited in the study of diseases research, molecular diagnosis, and forensic biological evidence. The article systematically reviews the principle, the detection methods, improvement of AS-PCR, and its research updates in the fields of autosome, Y chromosome and mitochondrial SNP, as well as its application in forensic science.

5.Therapeutic effect of cardiac rehabilitation combined routine treatment on ischemic cardiomyopathy

Objective :To study influence of cardiac rehabilitation combined routine treatment on cardiac function and ECG in patients with ischemic cardiomyopathy (ICM).Methods :A total of 108 ICM patients treated in our hospital from Jul 2013 to May 2017 were randomly and equally divided into routine treatment group and cardiac rehabilitation group (received cardiac rehabilitation therapy based on routine treatment group ) ,both groups were treated for eight weeks .LVEF ,left ventricular fractional shortening (LVFS) ,QT dispersion (QTd) ,T peak‐T end interval (Tp‐e) before and after treatment ,and therapeutic effect were observed and compared between two groups .Results :Total effective rate of cardiac rehabilitation group was significantly higher than that of routine treatment group (87. 0%vs.75.9%) , P=0.043. Compared with before treatment ,there were significant rise in LVEF and LVFS ,and sig‐nificant reductions in QTd and Tp‐e in two groups after eight‐week treatment (except Tp‐e of routine treatment group) , P<0.05 or <0.01. Compared with routine treatment group after treatment ,there were significant rise in LVEF [(42.3 ± 5.8)% vs.(49.8 ± 6.5)%] and LVFS [(25.6 ± 6.1)% vs.(35.2 ± 6. 9)%] ,and significant reduc‐tions in QTd [ (52. 3 ± 6. 3) ms vs .(45. 2 ± 7. 1) ms] and Tp‐e [ (129.7 ± 12. 5) ms vs.(118. 5 ± 13.0) ms] in car‐diac rehabilitation group ,P=0.001 all.Conclusion :Cardiac rehabilitation combined routine treatment possesses sig‐nificant therapeutic effect on ICM patients .It can significantly improve cardiac function .
6.Metabonomics-a useful tool for individualized cancer therapy.

Metabonomics has developed rapidly in post-genome era, and becomes a hot topic of omics. The core idea of metabonomics is to determine the metabolites of relatively low-weight molecular in organisms or cells, by a series of analytical methods such as nuclear magnetic resonance, color spectrum and mass spectrogram, then to transform the data of metabolic pattern into useful information, by chemometric tools and pattern recognition software, and to reveal the essence of life activities of the body. With advantages of high-throughput, high-sensitivity and high-accuracy, metabolomics shows great potential and value in cancer individualized treatment. This paper introduces the concept,contents and methods of metabonomics and reviews its application in cancer individualized therapy.

7.Differential display of homeobox gene expressions in the normal, wounded human fetal and adult skins by DNA microarray.

OBJECTIVETo explore the differential expression of homeobox genes in the normal, wounded human fetal and adult skins and its significance in fetal scarless healing.
METHODSGene chips containing 14 000 human genes were used to investigate homeobox gene expressions of the normal, wounded human fetal and adult skins.
RESULTSThere were significant differences between the expression of homeobox genes, especially for PRX-2, HOXB13, HOXB6 and HOXB7.
CONCLUSIONSThe homeobox gene is in close relation to developmental biology. The different expressions and changes of homeobox genes in the normal, wounded human fetal and adult skin may be a primary cause of different wound healing between fetal and adult skin.

8.Preliminary clinical observations on autologous cultured skin fibroblasts transplantation to treat the facial soft tissue deficiencies.

OBJECTIVETo observe the effect and safety of autologous cultured skin fibroblasts transplantation for treating depressed facial skin defects.
METHODSA total of 19 patients were treated from Jan, 2010 to Oct, 2010. Autologous skin fibroblasts were separated from postauricular skin biopsy or resected skin tissue in other surgeries such as blepharoplasty. They were cultured and expanded with exclusive method. Cells (2 x 10(7)/ml) within three passages were injected intradermally at the site of skin depression three times at one-month interval. Adverse events were observed and recorded. Clinical effects were evaluated and graded by two unrelated physicians before and 6 months after the first injection.
RESULTSCells from 16 patients were successfully cultured at the first time. The other 3 patients underwent a second harvest. A total amount of 6 x 10(8) cells could be reached within three passages in 45 days. 16 out of 19 patients accomplished the whole course of this study. Minor adverse events were observed in two patients including small ulcer caused by over injection in one patient and slightly redness and swelling in the other. The redness disappeared after a week without any treatment. No serious complications were observed. Significant difference was noticed between the scores obtained before and after the treatment.
CONCLUSIONSFrom this study, neither serious complications nor excessive cell proliferation or scar formation was found after cell injection. The effect of using autologous fibroblast transplantation was obvious and long-lasting, which provides a new choice for the treatment of depressed facial skin defects.

9.Determination of three constituents in Radix Astragali by HPLC-MS.

AIMTo determine calycosin-7-O-beta-D-glucoside, astragaloside IV and formononetin in Radix Astragali and other relative samples by HPLC-MS.
METHODSHPLC was carried out with Agilent 1100LC/MSD, equipped with Agilent Zorbax SB C18 column (250 mm x 4.6 mm ID, 5 microm) and mass spectrum detector. The mobile phase (CH3CN-H2O) was eluted in gradient mode.
RESULTSThe calibration curves of calycosin-7-O-beta-D-glucoside, astragaloside IV and formononetin were linear in the range of 0.03 - 1.21 microg x mL(-1), 0.35 - 13.86 microg x mL(-1) and 0.38 - 15.22 microg x mL(-1), respectively. These recoveries of samples were from 95% to 105% with RSD less than 1.5%.
CONCLUSIONThe method was employed to analyse 25 samples of Radix Astragali and other relative samples, including Radix Astragali slice, Radix Astragali Preparata, Hedysarum polybotrys Hand. -Mazz, Astragalus ernestii Comb. The contents of three constituents vary greatly because of the species, place of collection and season of harvesting. This method could apply to evaluate the quality of Radix Astragali and it is simple, sensitive and reliable.

10.Advance of exosomes in ischemic stroke

Stroke is one of the major causes of death and disability in China and even worldwide.At present, treatment of stroke has been traditionally focused on reducing death of ischemic cells.However,clinical trials have shown that none of neuroprotective drugs tested achieve clinical benefit after acute stroke.Exosomes are 30~100 nm extracellular vesicles derived from cells with cell membrane structure.Many studies suggest exosomes play essential roles in intercellular communication by transferring their cargo between source and target cells in brain.Emerging data show that exsomes also make a contribution to brain recovery via regulating highly interactive process pathway after stroke.Here,we review these advances and highlight the potential therapeutic functions of exosomes in brain remodeling after stroke.
