1.Differential proteomics research on exosomes derived from tongue squamous cell carcinoma cells and normal mucosa cells.

OBJECTIVEThis study aimed to explore further the mechanisms of tongue squamous cell carcinoma (TSCC) cell recurrence, metastasis, and diffusion, as well as to establish the experimental basis for the molecular biology research on TSSC. We intend to complete our objective through differential proteomics and preliminary analysis protein expression of exosomes derived from TSCC and normal mucosa cells.
METHODSWe acquired cultured supernatant fluid in vitro in the laboratory by culturing TSCC (tongue cancer Tca8113 cell line) and human normal mucosa cells (HOK cell line). The exosomes were separated and purified through differential centrifugation. Furthermore, the different protein expressions were identified through dielectrophoresis and mass spectrometry. The functions of the different protein expressions were identified through an online database search.
RESULTSTSCC and human normal mucosa cells secrete a large amount of capsule bubble structure substances in vitro, as confirmed by electron microscopy and surface markers heat shock protein-70 and major histocompatibility complex class 1. A total of 16 oral cancer cell-derived exosomes that expressed quantity more than two times, twelve that increased their expression levels, and four that cut their expressions were identified through the differential proteomics research on the two groups.
CONCLUSIONDifferential proteins that were verified through the online database serve an important function in exosome formation and in the progress of cancer.

2.Analysis on Distribution and Drug Susceptibility of Pathogens in Bronchoalveolar Lavage Fluid of Hospital-acquired Pneumonia Patients

OBJECTIVE:To study the value of bronchoalveolar lavage fluid(BALF) bacteria culture and drug sensitivity test in the hospital-acquired pneumonia(HAP) therapy.METHODS:The bacteria types in BALF samples and drug sensitivity test in 516 HAP cases were analyzed retrospectively.RESULTS:The positive rate of BALF culture was 46.9% with the main pathogenic bacteria of Candida albicans,Acinetobacter baumannii,Pseudomonas aeruginosa,Klebsiella pneumoniae.Of total nosocomial pneumonia,single bacteria infection accounted for 58.9% and mixed infection 41.1%.Candida had no drug resistance against 5-fluorocytosine,amphotericin B.Extended spectrum ? lactamase was found in 75.9% of Escherichia coli and 35.7% of Klebsiella pneumoniae with severe drug resistance.CONCLUSIONS:BALF bacteria culture and drug sensitivity test can provide reference for rational use of drug in the clinic.

4. Progress in managing pulmonary arterial hypertension associated with congenital heart disease

Pulmonary arterial hypertension(PAH) is the most common complication of congenital heart disease(CHD). Because of the complexity and diversity of cardiac anatomy and pathophysiology, as well as a variety of adaptive mechanisms that are not fully understood, little attention is paid on pulmonary arterial hypertension associated with congenital heart disease(CHD-PAH). This article aims to summarize the clinical status, disease characteristics, outcomes, and prognosis of CHD-PAH in Pediatric Cardiology, Beijing Anzhen Hospital, proposing current unresolved issues and future challenges.
5.Observation on Improving Anorectal Routine Postoperative Pain and Edema with Traditional Chinese Medicine Fumigating and Washing

Objective:To explore the effect of the traditional Chinese medicine fumigating and washing on improving anorectal routine postoperative pain and edema. Methods:From Mar 2013 to Dec 2014,126 cases of anorectal surgery patients were randomly divided into the observation group and the control group. Two groups of patients were given intravenous injection of antibiotics. The control group was treated with Potassium Permanganate bath, sooner or later each time, each time 20 min, continuous bath for 3 days for a course of treatment. The observation group was treated with the use of pain relief, Chinese medicine, sooner or later each time. Fumigation sitz bath for 20 min, each fumigation end in advance disinfection tampons and medication during fasting spicy stimulation food,to prevent the fire caused by dry stool. Satisfaction survey,visual analogue scale (VAS) was used,edema score, postoperative complications and clinical efficacy was analyzed. Results:Before treatment, there was no difference between the two groups of patients with pain and edema (P>0.05) . After treatment with 1-2, the pain and edema of the two groups significantly decreased, but the observation group was significantly lower than that in the control group (P<0.05) . Observation group patients appeared pain, edema, hemorrhage, the number of burning and itching was significantly lower than that in the control group (P<0.05) . The total effective rate in the observation group was significantly higher than that in the control group (P<0.05) . Conclusion:Anorectal diseases surgery patients using traditional Chinese medicine fumigating and washing, can significantly improve pain and edema,reduce the incidence rate of adverse reactions,improve the treatment efficiency.
6.Optimization of β-cyclodextrin Inclusion Process of Cinnamomi Ramulus Formula Granules Volatile Constituents Based on Standard Relation and Information Entropy Method

OBJECTIVE To optimize β-cyclodextrin(β-CD)inclusion process of Cinnamomi Ramulus Formula Granules vola-tile constituents by orthogonal test,based on standard relation and information entropy method.METHODS On the basis of single factor experiments,the ratio of β-CD to aromatic aqueous solution,the inclusion temperature,and the inclusion time were selected as the investigating factors;the inclusion rate,drug loading,and standard relation of cinnamic aldehyde in inclusion complex were used as the evaluation index.The information entropy method was used to determine the weight coefficient of each index,then the comprehen-sive score was calculated,the inclusion process conditions were optimized by orthogonal experiment.The inclusion complex was charac-terized by thin layer chromatography,ultraviolet absorption spectroscopy,Fourier infrared spectroscopy,and X-ray diffraction.RE-SULTS The best inclusion process was that the ratio of β-CD to aromatic aqueous solution was 3∶100(g·mL-1),the inclusion temperature was 50℃,and the inclusion time was 1 h.The average inclusion rate of the obtained inclusion compound was 80.84%,the drug loading was 8.63%,and the standard relation was 0.91.The results of thin-layer chromatography,ultraviolet,infrared spec-troscopy and other characterization experiments showed that the volatile components in the aromatic aqueous solution successfully en-tered the β-CD cavity,and the inclusion complex was successfully prepared.CONCLUSION The optimum inclusion process is sta-ble and feasible,which can provide references for the preparation process of Cinnamomi Ramulus Formula Granules.
8.Downregulation of ceramide synthase 1 promotes oral cancer through endoplasmic reticulum stress.

C18 ceramide plays an important role in the occurrence and development of oral squamous cell carcinoma. However, the function of ceramide synthase 1, a key enzyme in C18 ceramide synthesis, in oral squamous cell carcinoma is still unclear. The aim of our study was to investigate the relationship between ceramide synthase 1 and oral cancer. In this study, we found that the expression of ceramide synthase 1 was downregulated in oral cancer tissues and cell lines. In a mouse oral squamous cell carcinoma model induced by 4-nitroquinolin-1-oxide, ceramide synthase 1 knockout was associated with the severity of oral malignant transformation. Immunohistochemical studies showed significant upregulation of PCNA, MMP2, MMP9, and BCL2 expression and downregulation of BAX expression in the pathological hyperplastic area. In addition, ceramide synthase 1 knockdown promoted cell proliferation, migration, and invasion in vitro. Overexpression of CERS1 obtained the opposite effect. Ceramide synthase 1 knockdown caused endoplasmic reticulum stress and induced the VEGFA upregulation. Activating transcription factor 4 is responsible for ceramide synthase 1 knockdown caused VEGFA transcriptional upregulation. In addition, mild endoplasmic reticulum stress caused by ceramide synthase 1 knockdown could induce cisplatin resistance. Taken together, our study suggests that ceramide synthase 1 is downregulated in oral cancer and promotes the aggressiveness of oral squamous cell carcinoma and chemotherapeutic drug resistance.
