1.Case report of 16p11.2 microdeletion only manifested as paroxysmal kinesigenic dyskinesia

The clinical data of a child with paroxysmal kinesigenic dyskinesia (PKD) and being diagnosed and treated in the Department of Pediatrics of the First Affiliated Hospital of Zhengzhou University in October 2018 were analyzed retrospectively.The male patient was 13 years old.The clinical manifestation was the change of body position, and the temporary movement cannot appear.The manifestations included the turning of head to one side, the falling back of neck, head shaking, swinging, the tightly hugging of hands in front of the chest, the touching of two tiptoes to the ground, numb sole, and ache.Gene detection: chromosome 16p11.2 (chr16: 29594293-30189789) had about 595.5 kb heterozygosity deletion.A total of 8 cases of 16p11.2 microdeletion in children with PKD were reported in details.16p11.2 microdeletion is another form of gene expression that causes PKD.16p11.2 microdeletion should be screened for genetic evaluation in patients with PKD.
3.Immune tolerance induced by human Stro-1 positive mesenchymal stem cells in mouse skin graft.

This study was aimed to evaluate whether human mesenchymal stem cell (MSC) and the Stro-1 positive subgroup have inducing immune tolerance effect in mouse skin graft model. Human MSC were isolated and cultured from bone marrow-derived mononuclear cells of healthy adults, and Stro-1 positive cells were sorted out. Female C57BL/6 mice and female BALB/c mice were respectively used as donors and recipients in skin allogenic graft model. The recipients were divided randomly into 4 groups: (1) Stro-1(+) MSC group: 2×10(6) Stro-1(+) MSC were injected into the irradiated recipient mice before skin graft. (2) MSC group: 2×10(6) MSC were injected into the irradiated recipient mice before skin graft. (3) Irradiation control group: the recipient mice were just irradiated before skin allogenic graft. (4) Congenic control group: the irradiated BALB/c mice received the skin from the congenic mice. The survival time and pathologic changes of skin grafts were observed by macro- and microscopy with HE staining. The transforming growth factor β1 (TGF-β1) concentration in plasma of recipient mice was measured by ELISA before and after grafting. The results indicated that the survival time of skin grafts in the MSC group was (12.13 ± 3.34) d, which was not notably longer than the irradiation control group (11.38 ± 1.01) d. The survival time of skin grafts was significantly prolonged in the Stro-1(+) MSC group (30.68 ± 5.89) d, as compared with the irradiation control group and the MSC group, respectively; the pathologic examination of skin grafts showed a clear structure. After grafting, the TGF-β1 concentration in plasma of recipient mice was almost the same as before grafting in the irradiation control group and the congenic control group, but it significantly increased in the MSC group and the Stro-1(+) MSC group. It is concluded that the Stro-1(+) MSC induce greater immune tolerance than the unsorted MSC, and significantly prolong the survival time of skin grafts in vivo, while TGF-β1 does not contribute to the immune tolerance.

4.Status quo and influencing factors of disabilities caused by injury in Guangdong province.

OBJECTIVETo analyze the epidemiology status and risk factors of disabilities caused by injury in Guangdong province.
METHODSStratified multi-stage cluster sampling was used to identify 1530 subjects with disabilities (among them, 415 disabilities caused by injury), in the villages or districts in the four regions of Guangzhou, Qingyuan, Zhaoqing and Huizhou in 2010. Subjects were interviewed by trained staff with self-made questionnaires about the status of the disabled, then 415 disabled caused by injury were interviewed about the process of the injury causing disability. The variables in this survey included gender, regions, rural (urban) residences, occupation, age and injury style. The difference of injury were analyzed through χ(2) test, and the influencing factors were explored through classification tree model.
RESULTSThe top five causes of disability were road traffic injury, fall, work-related injury, medical accident, scald or burn, accounting for 32.53% (135/415), 21.45% (89/415), 12.77% (53/415), 8.43% (35/415) and 5.06% (21/415), respectively. The proportion of male was higher than female in all disabilities caused by injury (295 male, 117 female, 3 unknown). Female had higher proportion 8.55% (10/117) than male 3.05% (9/295) with the cause of scald or burn (χ(2) = 6.302, P = 0.012), the proportion of rural residents 38.89%(84/216)was also higher than urban residents 25.63% (51/199) in disabilities caused by traffic accident (χ(2) = 43.850, P < 0.001); and the proportion of rural residents 25.46%(55/216)was also higher than urban residents 8.55% (34/199) in disabilities caused by the fall (χ(2) = 8.257, P = 0.004). Different age groups and regions had different types of injury causing disability, 53.08% (69/130) traffic accident and 51.92% (27/52) work-injury mainly caused disability in 25 - 44 age group, 85.71% (30/35) medical accident and 76.20% (16/21) scald or burn mainly caused disability in 0 - 14 age group, the injury caused by the fall was 62.50% (5/8) in 65- age group. The proportions of fall in Qingyuan 29.09% (16/55) and Huizhou 28.23% (37/131) were significantly higher than that in Guangzhou 13.38% (21/157) and Zhaoqing 21.13% (15/71) (χ(2) = 11.904, P = 0.008). The proportions of work-injury in Guangzhou 19.11% (30/157) and Zhaoqing 19.72% (14/71) were significantly higher than that in Qingyuan 0.00% (0/55) and Huizhou 6.09% (8/131) (χ(2) = 22.309, P < 0.001). The proportions of disabled by scald or burn in Guangzhou 10.83% (17/157) and Qingyuan 0.00% (0/55) were significantly higher than that in Zhaoqing 1.41% (1/71) and Huizhou 2.27% (3/131) (χ(2) = 17.826, P < 0.001). Classification tree model revealed that the age group, occupation and urban-rural were major factors influencing intended harm, the highest proportion of the intended harm was in 25 - 44 age group (59.21%, 151/255).
CONCLUSIONThe road traffic injury, fall, work-related injury, medical accident and scald or burn were main causes of disability in Guangdong province. Gender, age, and urban-rural factors were associated with disabilities caused by injury.

5.Expression of CD44v6 and Livin in gastric cancer tissue.

BACKGROUNDCD44v6 plays an important role in invasion and metastasis of tumor, Livin has anti-apoptotic effects. The present study aimed to explore the expression and clinical significance of CD44v6 and Livin in gastric cancer tissue.
METHODSStreptavidin-peroxidase linked immunohistochemical method was used to determine the expression of CD44v6 and Livin in gastric cancer tissue and adjacent normal gastric tissues from 59 patients with histopathologically confirmed gastric cancer, and in gastric tissue specimens of 15 patients with gastric polyps, and 15 patients with chronic non-atrophic gastritis. The chi-square test was used for comparison of the relevant factors, Spearman's rank correlation test was applied for relationship among positive expression of the proteins.
RESULTSThe expresion of CD44v6 was positive in 64.4% of the gastric cancer patients; 5.1%, 0 and 13.3% in specimens of normal tissues adjacent to the cancer tissues, in gastric tissue specimens of patients with gastric polyps, and patients with chronic non-atrophic gastritis, respectively. The expression of Livin was positive in 52.5% of the gastric cancer tissues, 6.8%, 0 and 6.7% in the adjacent normal gastric tissue, specimens of patients with gastric polyps and chronic non-atrophic gastritis, respectively. The expression of CD44v6 was significantly correlated with the depth of invasion, the degree of differentiation, and lymphnode metastasis of gastric cancer (P < 0.05). The positive expression rate of Livin protein was also significantly correlated with degree of differentiation of gastric cancer cells and metastasis to lymphnodes (P < 0.05), but not correlated with the depth of invasion and pathological types (P > 0.05). The expression of CD44v6 and Livin in the gastric cancer tissue was positively correlated (r(s) = 0.286, P = 0.028).
CONCLUSIONSThe increased expression of CD44v6 and Livin in gastric cancer tissue may be closely related with development and progression of gastric cancer. CD44v6 and Livin may be new biological markers of gastric cancer.

6.Effect of body position intervention on sleep quality of patients undergoing video-assisted thoracic surgery

Objective To explore the body position intervention on the sleep quality of patients undergoing video-assisted thoracic surgery (VATS).Methods Totals of 148 patients were randomly divided into control group (n =75) and observation group (n =73),control group received the routine nursing,while observation group received special body position intervention nursing.Then,sleep quality of two groups in one week before the surgery and three days after the surgery were compared.Results No significant difference was found in the score of sleep quality of VATS patients between two groups before one week of thoracic surgery (t =0.8623,P>0.05).Three days after the thoracic surgery,the score of sleep quality of observation group was statistically higher than that of control group [(9.82 ± 2.81) vs (13.29 ± 3.32),t =6.8 591,P < 0.05)].Conclusions Body postural intervention can promote to improve sleep quality of VATS patients after thoracic surgery,also promote the recovery of the patients,and improve nursing quality.
7.Efficacy and safety of Saw Palmetto Extract Capsules in the treatment of benign prostatic hyperplasia.

OBJECTIVETo assess the efficacy and safety of Saw Palmetto Extract Capsules in the treatment of benign prostatic hyperplasia (BPH).
METHODSWe conducted a multi-centered open clinical study on 165 BPH patients treated with Saw Palmetto Extract Capsules at a dose of 160 mg qd for 12 weeks. At the baseline and after 6 and 12 weeks of medication, we compared the International Prostate Symptom Scores (IPSS), prostate volume, postvoid residual urine volume, urinary flow rate, quality of life scores (QOL), and adverse events between the two groups of patients.
RESULTSCompared with the baseline, both IPSS and QOL were improved after 6 weeks of medication, and at 12 weeks, significant improvement was found in IPSS, QOL, urinary flow rate, and postvoid residual urine. Mild stomachache occurred in 1 case, which necessitated no treatment.
CONCLUSIONSaw Palmetto Extract Capsules were safe and effective for the treatment of BPH.

9.Effects of TIGAR on oxidative stress in adipose tissue of ketotic cows

This experiment was divided into two parts:in vivo and in vitro.In the in vivo experi-ment,10 healthy and 10 ketotic cows were selected,and adipose tissues were collected.ELISA re-sults showed that malonaldehyde(MDA)content and reactive oxygen species(ROS)activity were higher in adipose tissues of cows with clinical ketotic compared with healthy cows,and glutathione peroxidase(GPP)activity was higher than that of cows with clinical ketotic.Western blot results showed that compared with the healthy group,adipose tissue of the ketotic group showed a signifi-cant increase in the expression level of TP53 induces glycolysis and apoptosis factors(TIGAR)protein,a significant up-regulation in the expression level of Nrf2-HMOX1 signaling pathway pro-tein,and a significant increase in the expression level of oxidative stress protein in adipose tissue of the ketotic group.The expression level of TIGAR protein was significantly up-regulated,the pro-tein level of Nrf2-HMOX1 signaling pathway was significantly up-regulated,and the protein levels of oxidative stress-related indexes SOD1,catalase(CAT)and glutathione S-transferase(GST)were significantly increased.It indicated that oxidative stress occurred in the adipose tissue of ketotic cows in vivo.In vitro experiments were carried out to detect the effect of TIGAR on oxida-tive stress in bovine adipocytes by adenoviral silencing or overexpression of TIGAR in isolated and cultured bovine primary adipocytes as well as by the addition of hydrogen peroxide in vitro for 2 h.The Western blot results showed that the hydrogen peroxide group showed enhanced oxidative stress compared with the control group,and the nuclear correlation factor 2 was significantly in-creased.correlation factor 2(Nrf2)pathway,decreased expression of Nrf2 and hemeoxygenase-1(HMOX1),and decreased expression of related oxidative stress proteins in the hydrogen peroxide group;compared with the hydrogen peroxide group,the protein expression levels of Nrf2-HMOX1 and related oxidative stress proteins were up-regulated in the group with overexpression of TIGAR and hydrogen peroxide,and the expression levels of related oxidative stress proteins were up-regu-lated in the group with overexpression of TIGAR and hydrogen peroxide.Expression level was up-regulated,and the expression of related oxidative stress proteins SOD1,CAT and GST increased.This indicates that overexpression of TIGAR alleviated hydrogen peroxide-induced oxidative stress in adipocytes.Protein expression of Nrf2-HMOX1 signaling pathway and oxidative stress-related proteins SOD1,CAT and GST were down-regulated in the silencing TIGAR plus hydrogen perox-ide group compared to the hydrogen peroxide group.It indicates that silencing TIGAR exacerbated the oxidative stress caused by hydrogen peroxide to adipocytes.
10.Application of intraoperative magnetic resonance imaging and multimodal navigation in surgical resection of glioblastoma.

OBJECTIVETo evaluate the efficacy of intraoperative magnetic resonance imaging (iMRI) and multimodal navigation in surgical resection of glioblastoma.
METHODSBetween February 2009 and July 2010, 76 glioblastoma patients underwent surgical resection guided by iMRI and multimodal navigation. The cohort consisted of 43 male and 33 female patients, with a mean age of 49 years (range: 14-79 years). Rates of gross total resection (GTR) and extent of resection (EoR) were calculated at first and final iMRI scans.Pearson χ(2) test was used to compare the rates of GTR.
RESULTSiMRI and multimodal navigation were successfully implemented in all cases. Rates of GTR were misestimated by neurosurgeons in 24 cases (31.6%), which were confirmed by first iMRI. Total tumor resection were achieved in 20 cases (26.3%) as a result of iMRI scan, increasing the rates of gross total resection from 52.6% to 78.9% (χ(2) = 11.692, P = 0.001). Extent of resection in 28 patients who underwent further tumor resection were increased from 81.5% to 98.1%, leading to the overall extent of resection improved from 92.3% to 98.4%. At 3-month follow-up, 3 cases (3.9%) developed permanent neurologic deficits. The mean clinical follow-up was 15.6 months (range 3.0-45.0 months). The 2-year overall survival rate was 19.7%. The median progression-free survival of gross total resection group was 12 months (95% CI: 10.1-13.9 months), compared with 9 months (95%CI: 7.9-10.1 months) of the subtotal resection group (χ(2) = 4.756, P = 0.029). The overall survival of gross total resection group was 16 months (95% CI: 13.7-18.3 months), compared with 12 months (95% CI: 9.7-14.3 months) of the subtotal resection group (χ(2) = 7.885, P = 0.005).
CONCLUSIONCombined with multimodal navigation, iMRI helps maximize surgical resection of glioblastoma, preserving neurological function while increasing progression-free survival and overall survival.
