Arsenic trioxide(As2O3)effectively triggers apoptosis in acute promyelocytic leukemia (APL) cells and induces complete remission and cure in most APL patients ,if they can refrain from early mortality .In the last few years,it has been under investigation as a potential treatment for non -APL leukemia and a variety of sol-id tumors.In the meantime,it has been found that As2O3 also inhibits proliferation of non -APL leukemia cell and several solid tumor cell such as liver ,esophageal ,and gastric cancer cell in vitro or in vivo in the more and more studies .The molecular mechanisms that As 2 O3 induces cellular apoptosis in non APL leukemia cell and solid tumor cells have been discussed ,which also includes building new targeting preparation in order to obtain much more satisfying therapeutic effect and relieve its cellular toxic and side effect and so on.Here, we give a summary which is connected with the research progress above .

ObjectiveTo explore the effects of optional cesarean section on the level of plasma motilin and serum gastrin in umbilical blood and their relationship with gastroenteric symptoms of neonates. MethodsWe measured the concentrations of plasma motilin and serum gastrin in umbilical blood from neonates undergoing optional cesarean operation, emergency cesarean section and vaginal delivery by radioimmunoassay(RIA). ResultsFull-term deliveries undergoing optional cesarean section were with significant gastrointestinal symptoms. Plasma motilin and serum gastrin were significantly lower in neonates undergoing cesarean operation without birth initiate group(optional cesarean section)than that in cesarean section with birth initiate group and vaginal delivery group (P<0.05). ConclusionCesarean operation without birth initiate influences neonatal gastroenteric symptoms.Breast feeding which begins as early as possible will help to relieve those symptoms.

Objective To explore the performance of serum AFP-L3 and PIVKA-Ⅱ in differential diagnosis of benign and malignant liver disease in high risk population.Methods The serum levels of AFP-L3 and PIVKA-Ⅱ in 48 patients with primary hepatic carcinoma,43 patients with cirrhosis and 81 patients with chronic hepatitis B were analyzed retrospectively.Results There was no statistically different significance among the median levels of serum AFP in primary hepatic carcinoma patients,cirrhosis patients and chronic hepatitis B patients (x2=4.014,P=0.134).Both median level of AFP-L3 and PIVKA-Ⅱ in primary hepatic carcinoma patients were higher than cirrhosis patients and chronic hepatitis B patients (x2 =33.93,52.33,both of P values were below 0.001).The specificity (92.74％) of AFP-L3 and the sensitivity (79.17％) of PIVKA-Ⅱ were all higher.The accuracy (84.88％) of combined detection in series was the highest,with its 47.92％ of sensitivity and 99.19％ of specificity.Conclusion Combined detection PIVKA-Ⅱ and AFP-L3 series will help to differential diagnosis of benign and malignant liver disease in high risk population.

Objective To investigate the optimum techniques for processing Yuhuanlian with microwave. Methods Nine samples were obtained by orthogonal design. Thin-layer chromatography was used to determine berberine in Yuhuanglian. The content of berberine was taken as the index for the evaluation on the superiority of processing techniques. Objective To monitor the function of infection on myelination in white matter damage,neonatal Wistar rats of postnatal day 2 (P2) and postnatal day 7 (P7) were injected intraperitoneally with the same doses of lipopolysaccharides (LPS),and 2',3 '-cyclic nucleotide phosphodiesterase (CNPase) and myelin basic protein (MBP) were labeled in immature oligodendrocytes and mature oligodendrocytes.To investigate the function of tumor necrosis factor(TNF)-α according to test the change of TNF-α expression in the brain.Methods Ninty-six neonatal Wistar rats were randomly divided into four groups (each group 24 rats):group A:LPS (5.0 mg/kg) was injected intraperitoneally on P2 ; group B:LPS (5.0 mg/kg) was injected intraperitoneally on P7 ;group C1 and C2 were control groups in which equal amount of normal saline was injected intraperitoneally on P2 or P7.The expression of CNPase at 24 h after injection and MBP at P14 in brain tissue of each group were measured by immunohistochemistry and express of TNF-α mRNA at 4 h after the injection was measured by RT-PCR.Results Punctate hemorrhage in the corpus callosum,external capsule and intraventricular hemorrhage were seen in group A.Periventricular leukomalacia appeared in the corpus callosum and glial cells hyperplasia could be seen periventricular in P14 rat brains,but not found in the group B and any of the saline-injected rat brains.Compared with group C1 and C2 respectively,CNPase-positive cells showed obvious decrease in the area of white matter in periventricular in group A(106.93 ± 2.62 vs 113.67 ± 2.69,P ＜ 0.01) and group B (96.37 ± 1.82 vs 101.65 ± 2.01,P ＜ 0.01).Following LPS treatment in group A,the protein expression of MBP in neonatal brain decreased evidently compared with group C1 at P14 (128.21 ± 2.99 v s 134.81 ± 2.98,P ＜ 0.01),while no significant change was found between group B and group C2(134.77 ±3.68 vs 134.81 ±2.98,P ＞0.05).After 4h of the LPS treatment,the level of TNF-α mRNA was greatly increased in group A,it was significantly higher than that in group B (1.79 ± 0.04 vs 1.18 ± 0.04,P ＜ 0.01).Conclusion Intraperitoneal injection of LPS to the development neonatal rats can lead to dysmyelination and white matter damage.The expression of TNF-oα mRNA increased significantly in these immature neonatal rats,while only myelination delay occurred in those of mature neonatal rats without dysmyelination.

Objective To observe the curative effect of warm needling therapy combined with kidney-supplementing and blood-activating Chinese herbal medicine on patients with scapulohumeral periarthritis and its effects on immunoglobulin IgA, IgG and IgM;To explore the mechanism. Methods Ninety patients with scapulohumeral periarthritis were randomly divided into needle with medicine group, needle group and medicine group, 30 cases in each group. Patients in the needle with medicine group were treated with needle warming therapy combined with kidney-supplementing and blood-activating Chinese herbal medicine, patients in the needle group were treated with needle warming therapy, patients in the medicine group were treated with kidney-supplementing and blood-activating Chinese herbal medicine, 30 days for medicine and every other day for acupuncture. Five time acupuncture was one course of treatment, and patients were treated for 3 courses. Shoulder pain index was evaluated, and the levels of IgA, IgG and IgM was determined before and after 3 courses treatment. The total curative effect of the three groups were compared. Results The total effective rates for needle and medicine group, needle group and medicine group were 93.33% (28/30), 73.33% (22/30), 66.67% (20/30), respectively, with statistical significance between needle and medicine group and the other two groups (P<0.05),without statistical significance between needle group and medicine group (P>0.05). There was statistical significance between the shoulder indexes before and after treatment of the three groups (P<0.05). Before treatment, the levels of IgA, IgG and IgM of all three groups increased significantly compared with the normal reference values (P<0.05). After treatment, the levels of IgA, IgG and IgM were reduced significantly in three groups after treatment (P<0.05), the levels of IgA, IgG and IgM in the needle and medicine group were significantly lower than those in the other two groups (P<0.05). Conclusion The curative effect of needle warming therapy combined with kidney-supplementing and blood-activating Chinese herbal medicine was better than alone needle warming therapy or kidney-supplementing and blood-activating Chinese herbal medicine. This combined therapy could significantly improve the symptoms of patients, reduce the levels of IgA, IgG and IgM, regulate the body immune function, which is probably one of the mechanisms in the treatment of scapulohumeral periarthritis.

Objective To develop neural stem cells(NSCs) which can stably express exogenous brain-derived neurotrophic factor(BDNF) in vitro. Methods NSCs from the subependymal zone of embry-onic day 14.5(E14.5) rat brain were purified by limiting dilution assay and then infected with supernatant of recombinant retrovirus pLXSN-BDNF and retrovirus pLXSN. The original copy numbers of exogenous gene templates from three groups NSCs(pLXSN-BDNF viral infection group, pLXSN viral infection group, control group) were detected by fluorescent quantitative PCR(FQ-PCR). ELISA assay was used for determining the protein contents of BDNF of supernatant from three groups NSCs for six days continually after seeded in 24-well plates in the same cell density. Results NSCs were purified successfully by limiting dilution assay.The original copy numbers of exogenous BDNF gene templates from pLXSN-BDNF viral infection group by FQ-PCR were (19.57±0.65) × 10~3 copies/μl, higher than those of another two groups(P < 0.05). The protein contents of BDNF of supernatant from NSCs of pLXSN-BDNF viral infection group was highest among three groups and compared with another two groups had statistical significance (P <0.05) . Conclusion The purified NSCs can be transduced exogenous BDNF successfully with supematant of recombinant retrovir-us pLXSN-BDNF which provide experimental evidences and laying foundations for further research of retinal transplantation and quantization investigation of gene therapy for optic nerve injury.
