Haemophilus influenzae (Hi) is a pathogen exclusively found in humans. It causes a wide range of infections from the upper respiratory tract to serious invasive diseases. Such as pneumonia, septicemia and meningitis. Strains of Hi are usually classif ied into six serotypes a to f and nontypeable H. influenzae (NTHI) according to the antigenicities and compositions of their polysaccharide capsules. Hib was a common cause of serious infections in younger children. The polysaccharide-protein conjugate vaccines against Hib had almost eliminated H. influenzae as a cause of pediatric meningitis. However, NTHI remains an important pathogen, particularly in children and the elderly. Efforts to understand and control NTHI disease have been hampered by the diversity of these bacteria. This review introduced the study progress about pathogenic mechanism of NTHI. In order to provide the help for development of vaccine, clinic treatment and prevent the occurrence of diseases causing by NTHI.

3.Fe(CN)3-6 Chemiluminescence System for the Determination of Rutin

A novel chemiluminescence (CL) system combined with flow injection analysis for the determination of rutin is presented . It is based on the CL reaction of rutin and hexacyanoferrate*# in sodium hydroxide medium. The proposed method has a linear range of 1×10-4g/mL ~ 1×10-6 g/mL with a RSD of 3.7%(5×10-6g/mL,n=11) and a sample throughput of 60 h-1. The method was successfully applied to the determination of rutin in a pharmaceutical preparation.
4.Preparation of Ibuprofen Lysine Sustained-release Liquid Suppository and Its Evaluation in vivo and in vi-tro

OBJECTIVE:To prepare Ibuprofen lysine sustained-release liquid suppository,and conduct the evaluation in vivo and in vitro. METHODS:Ibuprofen lysine sustained-release liquid suppository was prepared by using Poloxamer solution as main matrix,carbomer as bioadhesive agent,laurocapram as penetrant. The ratios of poloxamer 407(P407)-poloxamer 188(P188)and carbomer content were screened by using gel temperature,gel intensity,biological adhesion and release rate in vitro as indexes, and the concentration-time curves and pharmacokinetic parameters of prepared liquid suppository and common solid suppository af-ter rectal administration in Beagle dogs in vivo were compared. RESULTS:When the P407-P188 ratio was 1:1.2,the gel tempera-ture of sustained-release liquid suppository was 30.4-38.1 ℃,which was the nearest to the rectal temperature;when the content of carbomer was 0.8%,the in vitro release of sustained-release liquid suppository was in zero-order model,with favorable correlation (R2=0.996). The tmax of common solid suppository was 3.206 h,then plasma concentration decreased significantly,release time did not exceed 12 h,AUC0-∞ was 501.826 mg·h/L;tmax of sustained-release liquid suppository was 8.814 h,then plasma concentration decreased,release time exceeded 24 h,AUC0-∞ was 715.489 mg·h/L. CONCLUSIONS:Ibuprofen lysine sustained-release liquid suppository is successfully prepared,which shows better sustained-release effect and excellent correlation in vivo and in vitro than common solid suppository.
5.A novel luminol-based chemiluminescence method for the determination of amikacin sulfate in serum by using trivalent copper-periodate complex

A novel chemiluminescence (CL) reaction was based on the oxidizing reaction of luminol by the trivalent copper-periodate complex (K5[Cu(HIO6)2], DPC) in alkaline medium. The CL intensity could be enhanced in the presence of amikacin sulfate (AKS). A new CL method was developed for the determination of AKS by coupling with flow injection (FI) technology. Because of the distinctive oxidative effect of DPC, the luminol-based CL reaction could occur at a low concentration of 10à7 M. The relative CL intensity was proportional to the concentration of AKS in the range of 4.0 ? 10à9-4.0 ? 10à6 g/mL with the detection limit of 1.2 ? 10à9 g/mL. The relative standard deviation was 2.1% for 8.0 ? 10à9 g/mL AKS (n?9). The proposed method was successfully applied to the direct determination of AKS at the level of ng/mL in serum samples. The recovery varied from 97.0% to 106.3%. A possible mechanism of the CL reaction was discussed in detail by relating to the CL kinetic characteristics and electrochemical activities of the oxidant DPC.

Objective To investigate the relation of morbidity law of upper gastronintestinal hemo rrhage and cycle of Heavenly Stems and Earthly Branches in Gansu province. Methods Investigative statistical forms were set up and analyzed by computer about twelve years in-patient of upper gastronintestinal hemorrhage in TCM hospital of these district. Results High morbidity corresponded with theory of YUNQI, in 1988 (year of the fifth of ten Hevenly Stems and the fifth of the twelve Earthly Branches, HUOYUN is high sufficient), 1995 (year of the second of ten Hevenly Stems and the last of the twelve Earthly Branches, JINYUN is insufficient), and Baiyin district in 1994 (year of the first of ten Hevenly Stems and the eleventh of the twelve Earthly Branches TUYUN is high sufficient), Cheng district in 1996 (year of the third of ten Hevenly Stems and the first of the twelve Earthly Branches, SHUIYUN is high sufficient), YUNQI was high sufficient or insufficient, which causing diseases conform to five internal organs and geographical position. Conclusion Morbidity law of upper gastronintestinal hemorrhage corresponds with cycle of Heavenly Stems and Earthly Branches.
7.The Relation between TCM Syndromes of Upper Gastrointestinal Hemorrhage and Solar Terms Change in Different Years

Objective To investigate the relation between TCM syndromes of upper gastrointestinal hemorrhage and solar terms change in different years. Methods Data of upper gastrointestinal hemorrhage in-patients in seven districts of TCM hospital of Gansu province in twelve years were filled in investigative statistical forms and analyzed by computer. Results There was obvious difference in TCM syndromes among different solar terms and different years. Conclusion Atmospheric pressure, atmospheric temperature, air humidity, diurnal temperature range of solar terms change in different years directly effect TCM syndromes of upper gastrointestinal hemorrhage.
8.A study of correlation of anti-Helicobacter pylori antibody and anti-aquaporin 4 antibody in centre neurological system demyelination disease

Objective:To investigate the relationship between anti-Helicobacter pylori antibody(Hp-IgG)and anti-aquaporin 4 antibody which are in neuromyelitis optica(NMO)and multiple sclerosis(MS).Methods: Serum specimens were collected from the 33 patients with MS,7 patients with NMO,and 35 health examination cases.Hp-IgG were detected by enzyme-linked immunosorbent assasy and anti-aquaporin 4 antibody were detected by cell based assay respectively.The positive rate of Hp-IgG and anti-aquaporin 4 antibody were analyzed,and the difference of Hp-IgG positive rate was compared between patients with Hp-IgG positive and negative.Results: Serum Hp-IgG positive rate of MS,NMO and normal control groups were 69.70%,85.71% and 42.86% respectively with a significant statistically difference of Hp-IgG(P<0.05).Positive rate of serum anti HP-IgG antibody in MS group,NMO group and normal control were significantly different(P<0.05);but there was no statistical significant difference of anti HP-IgG antibody positive rate between MS group and NMO group(P>0.05).Serum anti AQP4 antibody positive rate of MS,NMO and normal control groups were 4.2%,85.71% and 0% respectively with a significant statistically difference of anti AQP4 antibody(P<0.05).The positive rates of anti HP-IgG which were in MS patients and NMO patients with positive anti AQP4 antibody were 72.73%,the positive rates of anti HP-IgG which were in MS patients and NMO patients with negative anti AQP4 antibody were 79.31%,the difference was not statistically significant(P>0.05).Conclusion: HP infection is a risk factor for the occurrence of MS and NMO,but not associated with MS and NMO patients with anti AQP4 antibodies.
9.Correlation between activated circulating endothelial cells and efficacy of anti-angiogenic therapy in non-small cell lung cancer patients

Objective: This study aimed to examine the number of activated circulating endothelial cells (aCECs) in the peripheral blood of patients with non-small cell lung cancer (NSCLC), and investigate the relationship among aCECs, anti-angiogenic therapy, and prognosis of NSCLC patients. This study also aimed to identify novel predictive markers for anti-angiogenic therapy, and provide basic data and experimental basis for establishing an evaluation system for this therapy. Methods: A total of 142 NSCLC patients were randomly divided into the chemotherapy group (Group 1) and combined therapy group (i.e., chemotherapy plus endostatin, Group 2). The number of aCECs was measured using flow cytometry by detecting the expression status of CD105 and CD146 in the peripheral blood. The correlation between the changes in aCECs and efficacy of drug treatment was statistically analyzed using SPSS software. Results:The number of aCECs in Group 2 increased significantly at 8 and 29 d, two cycles, 50 and 71 d, and four cycles after treatment, respectively (P<0.05). In particular, aCECs amount in cases of progressive disease increased more significantly after combined therapy (P<0.05). A negative correlation was found between the treatment cycle and difference in aCECs amount before and after therapy (r=-0.970, P=0.001). A negative correlation was also observed between the difference in aCECs amount and time to tumor progression (TTP) (r=-0.351, P=0.039). Therefore, the difference in aCECs amount before and after therapy could serve as an important predictor for TTP in NSCLC patients. Conclusion:CD105 and CD146 reflected the activation status of endothelial cells, and responded to the drug treatment. Thus, CD105 and CD146 could act as ideal markers for aCECs. The number of aCECs increased during cancer progression, but significantly decreased after long-term treatment. Therefore, the change in aCECs amount may be a useful marker in predicting the efficacy of anti-angiogenic therapy.
10.Effect of Total Flavonoids of Hippophae Rhamnoides on Contractile Mechanics and Calcium Transfer in Stretched Myocyte

Objective To study the role of total flavonoids of hippophae rhamnoides in improving contactile function of stretched cardiac myocyte.Method Flavonoids were given to stretched myocytes which were proved their contractile function decline and then myocyte contractile mechanics characteristics and calcium transfer were measured.Result Flavonoids increased myocyte contractility,as indicated by myocyte shortening,velocity of shortening,peak+dL/dt and peak-dL/dt during shortening,in a concentration-dependent manner (r>0.9,P<0.001),and with no relation to the intracellular calcium transfer in the myocytes.Conclusion Flavonoids of the traditional Chinese drug hippophae rhamnoids is effective in improving the contractile function of stretched cardiac myocyte in low dosage.
