To isolate the specific genes in protoscoleses (PSC) of Echinococcus granulosus under oxidative stresses from the SSH library constructed in the previous study, the gene expression in PSC under oxidative stresses was studied by using real-time PCR. The previously amplified library was sequenced and analyzed in GenBank with Blast research. Sequence analysis indicated that all clones in the SSH library contained the coding sequences, of which some clones showed homology in the GenBank and others were unknown. Differential expression of 4 genes randomly selected and the TPx gene in this library were studied with real-time PCR. It was demonstrated that the gene expression of S88 and H32-1 in oxidative tissues was 2.0 and 2.3 times higher than the un-oxidative stresses respectively. The TPx gene was up-regulated when PSC was induced with H_2H_2 of more than 0.8 mmol/L. These results implies that the up-regulated expression of the above-mentioned genes may be related with the related functions of anti-oxidative process in PSC and they may be used as the candidate genes for the study of anti-oxidation of E.granulosus.

Objective To provide accurate bone tunnel positioning reference for clinical individual anterior cruciate ligament (ACL) reconstruction by three-dimensional (3D) reconstruction of femoral and tibial original footprint of ACL.Methods Forty volunteers were selected for this study,including 20 males and 20 females,and aged 18 to 35 years old.3D CT scanning images of knee data were obtained.3D bone models of knee and footprint of ACL were reconstructed.On the 3D reconstruction models,the shape and position of original footprint of ACL were displayed.Statistical significance was compared for their difference.Results Femoral original footprint of ACL,long axis of footprint,distances to the distal margin of the articular cartilage (DDCM),distances to the posterior margin of the articular cartilage (DPCM),double bundle center distance,and tibial original footprint short axis of ACL were significantly different between genders (P ＜ 0.05).However,no significant difference was found between genders for tibial original footprint long axis of ACL (P ＞ 0.05).Conclusions The insertion mark shape and nature position of footprint of ACL could be observed by 3D reconstruction using CT scan images.

AIM:To assess the protective role of melatonin(MEL)in a rat model of oleic-induced acute lung injury.METHODS:Twenty-four rats were randomly allocated to three groups as follows:saline(NS)injection group,oleic acid(OA)injection group and MEL plus OA injection group,the lavage protein,lung wet-to-dry weight ratio,malondialdehyde(MDA)content,superoxide dismutase(SOD)activity and lung histopathology were examined.RESULTS:(1)Injection 0.15 mL/kg of OA led to a severe acute lung injury(ALI),characterized by significantly increasing in lavage protein,lung coefficient(P＜0.01),and by histopathological alterations which presented hemorrhage,edema.thickened alveolar septum and the existence of inflammatory cells in alveolar spaces;(2)Infusion of MEL(20 mg/kg,intraperitoneally for 60 min before the oleic acid)markedly alleviated above-mentioned symptom induced by OA,consistent with decrease of MDA level(P＜0.01) and the increase of SOD activty(P＜0.01).CONCLUSION:Pre-treatment with MEL can attenuate the OA -induced ALI in rats via cleaning and preventing the formation of free radicals and further lessening the increase of alveolocapillary membrane permeability,these data suggest that MEL may be effective in the prevention of ALI.

Objective To discuss the characteristics,growth and development of atlantoaxial pedicle in preschool children,and to provide the basic theoretical basis for the design,production and clinical application of pedicle screw technique in the treatment of atlantoaxial disease in preschool children.Methods Excluded patients with atlantoaxial fractures,deformities,tuberculosis and tumor,the clinical data of 60 children aged from 3 to 6 years old with cervical spondylolisthesis were collected.The original CT images were reconstructed in DICOM format and the subjects were reconstructed with Mimics software.The subjects were grouped by age to measure and analyze the atlantoaxial pedicle correlation.Results The pedicle width(PW),pedicle height(PH) and total length of pedicle bone channel (PL) increased gradually with age.The PW/PH ratio of the vertebrae(C1) is greater than 1.0,and that is, the width of the pedicle of the C1 pedicle is greater than the height.The PW/PH ratio of the vertebral (C2) is less than 1.0,which means the width of the pedicle of the C1 pedicle is less than the height.Conclusion The growth and development of spine in preschool children are fast, and the pedicle of atlantoaxial calculus in different age groups has different developmental characteristics.In this study,the diameter of the pedicle of the atlantoaxial pedicle in the 3 to 6 year old children is increasing with age.The pedicle screw with the smallest diameter of 3.5 mm can be implanted into the atlantoaxial pedicle with feasibility in theory.

This study aimed at exploring a fast method to accurately identify the medicinal insect Vespa mandarinia Smith from its adulterants using DNA barcode and COI sequences.The extracted DNAs from V.mandarinia and its adulterants V.soror were amplified by polymerase chain reaction (PCR) and sequenced bilaterally based on COI barcode sequence investigation.The information of the COI sequences of V.mandarinia and V.soror were gathered from GenBank.All the sequences were compared and analyzed,and their intraspecific and interspecific genetic distances were calculated using MEGA 6.06.In addition,the phylogenetic tree was established with neighbor-joining (NJ) method.As a result,the COI sequences of V.mandarinia and V.soror were successfully amplified.The minimum interspecific distance between V.mandarinia and its adulterants was 0.152 ± 0.017,being considerably larger than the maximal intraspecific distance between V.mandarinia,0.009±0.004.The constructed phylogenetic tree showed an independent branch for each species.It was concluded that the DNA barcode based on COI sequence can efficiently identify V.mandarinia and its adulterants.This study provided an innovative tool for the quality control and market regulation of Chinese materia medica,securing the safe medication of V.mandarinia.

Currently, lacking standards of data communication and storage has been becoming a huge problem in tertiary medical rehabilitation networks. Several rehabilitation management requirements need be met, such as integrating rehabilitation resources, sharing patient data, and augmenting efficiency of rehabilitation therapies. By summarizing existing standards within medical devices and data management, this paper proposed a novel standardized protocol for rehabilitation, which is composed of standards in data format, communication signaling and processing. To demonstrate it, an application in current tertiary medical rehabilitation networks was also proposed in this paper. As a result, the outcomes of this paper are expected to solve the 'information isolated island' problem in current rehabilitation medical rehabilitation networks.

Nano-fumed silica derivative with quaternary ammonium group was synthesized and the antimicrobial activity was investigated. The nano-fumed silica derivative was investigated by Fourier-transform infrared spectroscopy (FTIR). The zeta potential and the size of the nano-fumed silica derivative were measured. The antimicrobial properties of the nano-fumed silica derivative against selected microorganisms were tested by the quantitative suspension method. The zeta potential showed that the isoelectric points of nano-fumed silica and modified nano-fumed silica are pH=4. 8 and pH =10.5-10.8, respectively, and the shift of the isoelectric point is due to the quaternary ammonium salt. The obtained nano-fumed silica derivative inhibited the growth of Gram-negative (Escherichia coli), Gram-positive bacteria (Staphylococcus aureus), and fungus (Candida albicans). The inhibiting effect of nano-fumed silica derivative on the microorganisms varied with the time of exposure. The bacteriostatic rates were noted to be 99.99%, 99.99% and 94.12% after 15 minutes' exposure, respectively. Thus the results indicate that nano-fumed silica derivative with quaternary ammonium group has significant inhibitory effect on the growth of bacteria.

Objective To explore the diagnostic value of quantitative radiomics features from dual-modal ultrasound composed of elastography and B-mode for axillary lymph node metastasis in breast cancer patients. Methods We retrospectively analyzed 161 axillary lymph nodes (69 benign and 92 metastatic) undergoing real-time elastography and B-mode ultrasound from 158 patients with breast cancer. We extracted a total of 428 features, consisting of morphologic features from B-mode, and intensity features and gray-level co-occurrence matrix features from the dual modalities,and the optimal subsut of features was selected through least absolute shrinkage and selection operator (Lasso) under the condition of leave-one-out cross validation. We used SVM for the classification of benign and metastatic nodes. Results The sensitivity, specificity, accuracy and Youden's index of the 35 radiomics features selected with Lasso were 86.96%, 85.51%, 86.34% and 72.46%, respectively. Conclusion The radiomics features from dual-modal ultrasound (elastography and B-mode) have demonstrated good performance for classification and have potential to be applied to clinical diagnosis of axillary lymph node metastasis.

Echinococcus granulosus is an important zoonotic parasite globally causing cystic echinococcosis (CE) in humans and animals. In this study, prevalence of CE and variation of cox1 gene sequence were analyzed with isolates E. granulosus collected from different areas in northern Xinjiang, China. The survey showed that 3.5% of sheep and 4.1% of cattle were infected with CE. Fragment of cox1 was amplified from all the positive sheep and cattle samples by PCR. In addition, 26 positive samples across the 4 areas were included. The isolates were all E. granulosus sensu stricto (s.s.) containing 15 haplotypes (Hap1-15), and clustered into 2 genotypes, G1 (90.1%, 91/101) and G3 (9.9%, 10/101). Hap1 was the most common haplotype (48.5%, 49/101). Hap9 were found in humans samples, indicating that sheep and cattle reservoir human CE. It is indicate that E. granulosus may impact on control of CE in livestock and humans in the region.

Alveolar and cystic echinococcosis are important zoonotic diseases caused by the dog/fox tapeworms of Echinococcus granulosus and E. multilocularis, which are distributed in almost whole the world. The diseases remarkably impact on the people's health and economic development of communities. Echinococcus tapeworms need two mammalian animals to complete their lifecycle. The worms undergo different developmental stages (adult, egg/oncosphere, cyst, and protoscolex). Each stage has its own distinct physiological characteristics. We summarize the characteristics and emphasize that some features that strongly impact on design of control program. Based on the lifecycle of these worms, two stages are infectious, one is egg the only stage for primarily infecting humans and intermediate host, and another stage is protoscolex, the only stage for infecting the definite hosts of the worms. Eggs are produced by the adult worms parasitized in the intestine of definite hosts. It takes 45 days for eggs becoming mature in dogs. With this feature of the tapeworm, New Zealand and Tasmania in Australia designed control program for deworming 8 times annually with an interval between deworms at 45 days. E. multilocularis takes 28-30 days in dog/fox producing mature eggs. Given that co-existence of E. granulosus and E. multilocularis in western China, a control measure so called "prepatent deworming control measure" has been used in these endemic areas with dosing worms in dogs every month. It normally takes 12-15 months for producing protosocleces in E. granulosus cysts in sheep. If meat markets attract most lambs, that is an effective measure for controlling cystic echinococcosis given that there are no protoscoleces produced from those lambs. In addition, Echinococcus has its own unique biological characteristics, such as sexual reproduction and asexual reproduction, single-cell layer structure of cyst, bidirectional development of protoscoleces induced by bile salts, and long term of infection causing host asymptomatic reaction make the parasites a models for addressing some biological and biomedical issues. And more, hydatid cyst fluid is the antigen resource for identifying diagnostic reagents; the specific gene expressed in oncospheres has been developed as an effective vaccine used for control program. With the development and application of high-throughput omics, including genomics, transcriptomics and proteomics, we can use the physiological characteristics of Echinococcus for searching diagnostic reagents, developing preventive vaccines and identify new drug targets.
