1.CXCL12/CXCR4 and breast cancer

CXCL12 and its receptor CXCR4 can express in breast cancer,and are regulated by several factors in the genesis and development of breast cancer. Lots of researches have proved that CXCL12 and CXCR4 can be used as new independent prognostic makers of breast cancer.Treatment targeted for CXCR4 can be seen as a new kind of combination therapy,which may provides patients a more ideal treatment.
2.FURTHER STUDY ON HEPATITIS B VACCINATION STRATEGY

With decision tree model, both net utility and cost-benefit were analysed to comprehensively assess hepatitis B vaccination strategy in China. The result showed that neonates shoulUbe taken as the most favourable vaccinated population, infants aged 0~3 years the next, and immediate vaccination scheme was recommended. Although vaccination would result in negative benefit in the population aged 7 and over, certain net utility was also gained, Therefore we suggest that the subjects to be vaccinated be extended in due course.
3.Extended application of routine ophthalmic technology in dry eye

Dry eye is a most common external eye disease in clinic and it can cause ocular discomfort and visual disturbance. The conventional examinations for dry eye have some shortcomings, for example, high variability and insufficient association with symptoms. Recently, routine ophthalmic technologies play more extending roles in evaluation of dry eye, including corneal topography, wavefront aberrometer, optical coherence tomography (OCT) and confocal microscope. Corneal topography and wavefront aberrometer could be used to evaluate the stability of tear film and visual function. OCT could image tear meniscus directly, and confocal microscope could observe histopathology of ocular surface in vivo. The studies on ophthalmic technologies in dry eye make us deepen the understanding of pathophysiology of dry eye and also provide new diagnostic approaches.
4.Identification of a novel transcriptional factor HMGB34367 for activation of ?-smooth muscle actin gene in relation to pulmonary fibrosis

Objective: To demonstrate the influence of HMGB34367,a subfamily member of the high-mobility group protein B1(HMGB1),as a transcriptional factor of the ?-SMA gene expression.Methods: We cloned HMGB34367 cDNA from the fibrotic lung tissue of the Balb/C mouse treated with BLM by RT-PCR,sub-cloned it into a eukaryotic expression vector pcDNA 3.0 or pEGF-N2,and constructed a report plasmid,pDsRed-SMA,encoding red fluorescence(RFP) driven by ?-SMA promoter.Following the co-transfection of pDsRed-SMA and HMGB34367-pcDNA3.0/ pEGF-N2-HMGB34367 in the cultured 16HBE cells,we tested the expression of the RFP in the absence and presence of TGF?1 by fluorescence microscopy.After the transfection of HMGB34367-pcDNA3.0,the nucleus extracts from the transfected cells were subjected to electrophoretic mobility shift assay(EMSA) for the detection of the binding activity of HMGB34367 with ?-SMA promoter CarGB motif.RT-PCR was performed for the evaluation of the ?-SMA gene expression in the cells.Results: The over expression of HMGB34367 activated the ?-SMA promoter and enhances the expression of the ?-SMA gene.An increased binding activity of HMGB34367 with CarGB motif was detected by EMSA in the transfected cells.Conclusion:HMGB34367,a member of HMGB1 family,could act as a transcription factor for the transcriptional activation of the ?-SMA gene,which may play an important role in the development of lung fibrosis.
5.Screening of probiotic properties of twelve strains of lactobacilli by in vitro techniques

Objective: To study the probiotic properties of 12 selected strains of lactobacilli.Methods: The probiotic properties of 12 selected strains of lactobacilli were investigated for their resistance to pH 2.5 and 0.3% oxgall,their adhesion to Caco-2 cells and antimicrobial activity against enteric pathogenic bacteria.Results: The 12 strains all showed relatively high resistance to acid and bile salt and varied antimicrobial activities against pathogenic bacteria of intestinal infection.Lactobacillus johnsonii JCM1022 exhibited a broad inhibitory effect on enteric pathogenic bacteria,especially on enteropathgenic E.coli and Salmonella enteritidis,while Lactobacillus reuteri JCM1081 possessed a high activity of adhesion to Caco-2 cells(495.07?80.03).Conclusion: Among the 12 strains,Lactobacillus johnsonii JCM1022 and reuteri JCM1081 are qualified candidates for clinical application.

7.Role of epithelial-myofibroblast transdifferentiation under injury in the development of airway remodeling

Objective:To investigate if injury-related myofibroblast induction could occur in bronchial epithelial cells and the role of TGF-?_1(rhTGF-?_1) in epithelial-myofibroblast transdifferentiation. Methods:Cultured bronchial epithelial cells, 16HBE-14o, were exposed to poly-L-arginine(PLA). Using a immunohistochemical method, ?-smooth muscle actin (?-SMA) expression was detected in those cells after being treated with PLA . Relative activity of chloramphenicol acetyltransferase(CAT) was assessed in 16HBE-14o cells, which were transiently transfected with VSMP8 plasmid containing ?-SMA promoter linked to CAT gene, at different time points after rhTGF-?_1(10 ?g/L) treatment. Results:①?-SMA positive, spindle-shaped cells emerged among the regenerated 16HBE-14o epithelial cells following damage. The number of ?-SMA positive 16HBE-14o cells increased and cell repair was delayed under rhTGF-?_1 stimulation.② CAT activity increased significantly in VSMP8 plasmid transfected 16HBE-14o cells 24 h after TGF-?_1 stimulation. 8.Effect of lidocaine and bupivacaine on Na~+ current in the dorsal horn neurons of spinal cord

Objective The effect of lidocaine and bupivacaine on the Na+ current of dorsal horn neurons was observed to further evaluate the mechanism of local anesthetics . Methods The dorsal horn neurons of the SD neonates(0-7 d) were isolated acutely. Under the condition of holding voltage -80mV , and testing voltage -30mV with duration of 20 ms , the whole-cell patch-clamp technique was applied to recording the changes of voltage-gated Na+ currents following the administration of lidocaine or bupivacaine at 50-1000?mol/L.Results The voltage-gated Na+ currents ranged from 0.5-8nA peak amplitude , was inhibited by lidocaine and bupivacaine at clinical concentrations, the inhibitory degree was parallelly correlated with the concentration of local anesthetics(r=0.949 and 0.847 ,P

Objective To study the effects of desflurane on Ca 2+-ATPase activity isolated rat cardiac myocyte plasma membrane ,for the mechanisms of its inhibitory myocardial function.Methods The effects of different concentration desflurane(0%-11%) on Ca 2+-ATPase activity were studied at different calcium concentration(04-20?mol/L) and at 25℃ or 37℃Results Ca 2+-ATPase activity wan depressed by desflurane,the greater inhibitory effect,the higher desflurane concentration,and more severely at low calcium concentration or at 37℃ than at high calcium concentration or at 25℃Conclusions At clinical concentration desflurane produces tha inhibitory effect on rat cardiac myocyte plasma membrane Ca 2+-ATPase activity dose-dependently,which may in part explain its depression on myocardial function.
10.Effects of droperidol on sodium currents in isolated rat dorsal root ganglion neurons

Objective To investigate the effects of droperidol on the Na+ currents in rat dorsal root ganglion neurons.Methods The rat dorsal root ganglion neurons were enzymatically dissociated. Whole-cell patch-clamp technique was applied to record Na+ current. Results 3-300?mol?L-1 Droperidol inhibited the sodium currents by 14.12%-78.46% (P0.05, n =7).Conclusions Droperidol inhibits Na+ currents in rat dorsal root ganglion neurons in a concentration- and voltage-dependent manner. The results suggest that the concentration of epidural droperidol clinically applied during epidural patient control analgesia may enhance the analgesic effects.
