1.Research Advances in Mechanism of Bioactive Tanshinones in Salviae Miltiorrhiae Regulating Angiogenesis

[Objective]To explore the mechanism of bioactive tanshinones in Salviae Miltiorrhiae regulating angiogenesis and lay the foundation for the prevention and treatment of cancer, atherosclerosis, ischemic heart disease and other angiogenesis-related diseases. [Method]Based on angiogenesis mechanisms, infer to recent 10 years of articles from home and abroad, analyze and summarize the angiogenesis regulatory mechanisms of various bioactive ingredients in Danshen on endothelial cells, in vitro tumour cells and in vivo xenograft tumor. [Result] Among various bioactive ingredients of Salvia, Salvianolic acid B, Tanshinone IIA, Cryptotanshinone, Dihydrotanshinone I can promote or inhibit angiogenesis. Furthermore, Salvianolic acid B and Tanshinone IIA are considered to be the most important bioactive ingredients in Danshen and exhibit a dual angiogenic regulating activity through regulating various approaches, such as pro-angiogenic factors, MMPs, HIF-1α, the PI3K/AKT/eNOS signal pathway and so on. [Conclusion]Various bioactive ingredients of Salvia can serve as a regulator of angiogenesis, and it may provide new ideas for the prevention of angiogenesis-related diseases.
2.Effects of midazolam on N-type calcium currents of rat sympathetic neurons

ve To investigate the effects of clinical relevant concentrations of midazolam on whole-cell N-type calcium currents of sympathetic ganglion neurons trying to assess the involvement of sympathetic ganglion in circulation depression produced by midazolam. Methods 7-10 d SD rats were decapitated and bilateral superior cervical ganglions were rapidly removed and kept in 4℃, oxygen saturated incubation solution. TTX 1?mol/L and nifedipine 10 ?mol/L were added to the solution to block the Na+ and L-type calcium currents. Midazolam was also added to the solution. The final concentration of midazolam were 0.1, 0.3, 3, 10, 50, 100?mol/L.The effects of different concentration of midazolam on the whole-cell N-type calcium channel currents were assessed by patch-clamp technique. 3.Effect of Different Processing Methods on Paeoniflorin Content in White Peony Root

Objective To study the effect of different processing methods on paeoniflorin content and to provide a basis for stabilizing the processing technology of white peony root.Methods RP-HPLC method was used to determine the content of paeoniflorin on Agilent Eclipse XDB-C18(150 mm?4.6 mm,5 ?m),with mobile phase of acetonitrile-0.1% phosphoric acid(14∶86),flow rate of 1.0 mL/min,column temperature of 25 ℃,and detective wavelength of 230 nm.Results The average recovery rate was 99.195%,RSD=0.8%(n=6).The content of paeoniflorin was 0.55%,2.70%,3.40%,3.25%,respectively.Conclusion In the processing of white peony root,heating,water immersion and sulfur smoked have greater effect of paeoniflorin content in white peony root.
4.An investigation on prevention of epidural scar adhesion after microendoscopic discectomy

Objective To investigate the use of sodium hyaluronate and the preservation of epidural fat in the prevention of epidural scar adhesion after microendoscopic discectomy (MED). Methods A total of 300 patients receiving MED from July 2000 to October 2001 were randomly divided into 2 groups: patients in group A were given sodium hyaluronate with the preservation of epidural fat, while those in group B were on the contrary. Results The mean follow-up duration was 42 months in the group A and 44 months in the group B, respectively. The rate of excellent or good results was 98.6% in the group A (144/146), while 93.8% in the group B (135/144) ( ? 2=4.731, P =0.030). Postoperatively, the pain reappeared in 4 cases in the group A and in 6 cases in the group B, respectively, in which a re-operation was required. Re-operations found the epidural scar adhesion was grade 0 (3 cases) and grade 1 (1 case) in the group A, and grade 2 (2 cases) and grade 3 (4 cases) in the group B ( P =0.005). Conclusions Use of sodium hyaluronate with the preservation of epidural fat can improve the efficacy of MED and effectively prevent epidural scar adhesion.

METHODS: Whole cell patch clamp recordings were made from enzymatically isolated rat (7- 10 d ) superior cervical sympathetic ganglion neurons. Isobolographic analysis was applied to evaluate the potency of combinations of propofol and midazolam on Na + channel currents. RESULTS: Under V h= 80 mV and V t= 0 mV . AIM: To explore the influence of different concentration midazolam on the macroscopic voltage gated potassium currents and to discuss the relationship between potassium currents and inhibitory effect of clinical relevant concentration midazolam on sympathetic nervous system. METHODS: Superior sympathetic ganglion neurons were dissociated enzymatically from 7 to10 day old rat. Experiments were performed about 5 h after plating at room temperature (20- 24 ℃ ). Appropriate solution was chosen to separate the K + current from the other transmembrane currents. 1 ?mol?L -1 TTX was applied to the extracelluar solution to block the Na + current. Midazolam was also resolved in extracelluar solution to get various concentration ( 0.1 , 0.3 ,3,10,50,100 ?mol?L -1 ). Currents were recorded with the patch clamp technique in whole cell configuration using glass electrodes with a tip resistance of 2- 4 M . Potassium currents were evoked by test pulse from -100 mV to +30 mV with holding potential -80mV. Data were analyzed using Clampfit 6.0 and Oringih 5.0 software. Whole cell current records were corrected for leakage and capacitance by using the P/5 protocol. RESULTS: Midazolam dose dependently inhibited the whole cell potassium currents. Clinical relevant concentration midazolam ( 0.3 ?mol?L -1 ) only reduced the peak currents by 3.89 %(P= 0.88 ). The concentration required to produce 50% current inhibition(IC 50 ) was 76.065 ?mol?L -1 . CONCLUSION: Midazolam inhibits the whole cell potassium current significantly and dose dependently, but clinical relevant concentration midazolam has minor effect on the potassium currents, indicating that the inhibitory effect of midazolam on potassium current is not related to the suppression of activity of sympathetic system.
10.Effect of IFN-? inhalation on the cellular immunity of the lungs

AIM: To study the effect of IFN-? inhalation on the anti-infection ability of the lungs in the immunocompromised host. METHODS: The immunological factors in the immunocompromised rats and the immunocompromised rats administrated IFN-? via aerosol were investigated after 1, 3, 7 days when they were injected Candida albicans via tracheal The Canidda albicans count of the left lung was also determined after 7 days when injecting pathogen. RESULTS: The Canidda albicans count of the left lung in IFN-? group was significantly less than that of control group. The phagocyting and bactericidal percentages, Ia antigen expression percentages, the levels of TNF-?, IL-1? and IL-6 in the culture supernatant of the AM, the activity of IFN-? and TNF-? in BALF (except the TNF-? on 7 th day) in IFN-? group were markedly higher than those in control group. The expression of IFN-? and IL-1? pulmonary tissues in IFN-? group was higher than that in control group. The expression of TNF-? in IFN-? group was less than that in control group. The expression of IL-6 was no changes between two groups. The levels of IFN-?, IL-1? and IL-6 in the blood (except IL-1? on 3 rd day), and the killing ability of the lymphocytes in blood had no difference between two groups. CONCLUSION: Administration of IFN-? via aerosol obviously enhanced the anti-infection ability of the lungs in the immunocompromised host, but has no influence on the whole body cellular immunity.
