Objective To study the effects of fluvastatin on the proliferation of blood vessel smooth muscle cell (SMC) and the expression of platelet derived growth factor-A(PDGF-A) in experimental atherosclerosis(As) rabbits. Methods The expression of proliferating cell nuclear antigen(PCNA) of SMC was detected by immunohistochemistry. The expression of PDGF-A protein and mRNA was determined using immunohistochemical method and RT-PCR, respectively. 2.Craniofacial growth of chinese girls from 7 to 14 years of age

?Objective: To study the craniofacial growth in Chinese girls. Methods: The hand wrist radiographs of 282 chinese girls aged 7 to 14 years old were analysed by CHN scoring method to assess their skeletal age. Furthermore, their lateral cephalometric films were taken and analysed with percentile method. The craniofacial growth percentile curves of the girls were made. Results:The fastest craniofacial growth in the girls took place at the age of 11 to 12. The growth of SN was not significant,but the mandibular length(Co-Gn) increased by 17.3 mm.Conclusion: The data may be reference for the jaw growth prediction
3.Nursing and observation for patients with recurrent or refractory and aggressive non-Hodgldn's lymphoma by MINE regimen

Objective To report the clinical observation and nursing points for recurrent or refractory and aggressive non-Hodgkin's lymphoma treated with MINE regimen. Methods Patients (46 cases) with recurrent or refractory and aggressive non-Hodgkin's lymphoma were given mental nursing,preparation and evaluation of the condition of patients before chemotherapy. Complications especially extravasation of infusion fluid should be prevented during chemotherapy and myelosuppression and long-term complications should be prevented after treatment. Results No patient died after treatment but several complications such as infection(14 cases), nausea and vomiting (grade Ⅰ~Ⅱ, 22 cases, grade Ⅲ,2 cases), alopecia (grade Ⅰ~Ⅱ, 27 cases), mucositis (grade Ⅰ~Ⅱ, 4 cases), liver function lesion (grade Ⅱ, 1 case) were found and improved after proper treatment. No nephrotoxicity and hemorrhagic cystitis were found. Conclusions Selective mental nursing,proper chemotherapy preparation,evaluation of patients' condidtion and positively prevention of chemotherapy-related complications could increase the chemotherapy effect of patients with recurrent or refractory and aggressive non-Hodgkin's lymphoma.
4.Effects of antiplatelet drugs on proliferation and secretion of human bone marrow mesenchymal stem cells

BACKGROUND: Results from clinical trials suggested that clopidogrel and ticlopidine had side effects of granulopenia, and aspirin could inhibit endothelial progenitor cell proliferation. There is no report of effects of these drugs on human bone marrow mesenchymal stem cells (hBMSCs) in stem cell transplantation. OBJECTIVE: To investigate the effects of antiplatelet drugs including clopidogrel, ticlopidine and aspirin on hBMSC proliferation and secretion. DESIGN, TIME AND SETTING: The cytology in vitro observation was performed at the Laboratory of Toxicology, Shanghai Municipal Center for Disease Control and Prevention from March to December 2006.MATERIALS: The second passage of hBMSCs was kindly donated from Shanghai Tissue Engineering Research & Development Center, Shanghai Ninth People's Hospital. Clopidogrel (Lot number J20040006) and ticlopidine (Lot number H19980186) were obtained from Hangzhou Sanofi-Synthelabo Minsheng Pharmaceutical CO., Ltd. Aspirin (Lot number 20050059) was obtained from Bayer Vital GmbH. METHODS: The standard culture medium consisted of DMEM-LG, 10% heat-inactivated FBS, 100 U/mL penicillin and 100 μg/mL streptomycin. After being cultured in vitro expanded out to passage 6, hBMSCs were treated with antiplatelet drugs of different concentrations and compared with control group. MAIN OUTCOME MEASURES: Cell proliferation was assessed by 3- (4, 5-dimethylthiazol -2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay, level of vascular endothelial growth factor (VEGF) of culture medium was detected by enzyme-linked immunoadsordent assay (ELISA), and surface antigens of hBMSCs were analyzed by the flow cytometry. RESULTS: A570 values of hBMSCs treated by clopidogrel or ticlopidine of 0.02,0.1,0.4,2,10,40 μmol/L were higher than control group (P < 0.01), while A570 values of aspirin group of 60, 600, 2 000 μmol/L were lower than control group(P < 0.05). Antiplatelet drugs had no obvious effect on cell surface antigens(CD34, CD105, CD106)expressed by hBMSCs. Treated by high dose clopidogrel or ticlopidine (40 μmol/L), VEGF level from hMSCs was lower than that of control group(P < 0.01), but VEGF level of low dose (0.02 μmol/L) ticlopidine group was higher than control group(P < 0.01), and there was no significantly difference of VEGF level among low dose clopidogrel group (0.02 μmol/L), aspirin group (5, 2 000 μmol/L), and control group(P > 0.05). CONCLUSION: Clopidogrel and ticlopidine improve proliferation of hBMSCs, but aspirin inhibits proliferation of hBMSCs. High dose of clopidogrel and ticlopidine suppress VEGF secretion of hBMSCs, while low dose of ticlopidine promote it. Antiplatelet drugs have no obvious effect on hBMSCs differentiation.

6.Role of endothelin and nitric oxide in the pathogenesis of asthma

AIM:To explore whether the balance between endothelin(ET) and nitric oxide(NO) plays an important role in airway hyperresponsiveness of asthmatic rats. METHODS:The tension of isolated perfused rat tracheal rings was measured after ET-1 stimulation and incubation of JKC 302 and L-NAME. RESULTS:ET-1 constricted isolated rat tracheal ring, produced slowly developing and long-lasting contraction. The ET-1-induced contraction response of asthmatic rat tracheal ring was higher than that of normal control group (P<0.01). JKC 302, a selective ETA receptor antagonist, partly blocked ET-1-induced contraction in asthmatic rat trachea ring. L-NAME significantly augmented the constriction caused by ET-1. CONCLUSION:The effects of ET on bronchomotor tone may be modified by NO as this is a bronchomotor, and the imbalance between ET and NO may play an important role in asthma pathogenesis.
7.Analysis of insulin release pattern in patients with polycystic ovary syndrome and normal glucose tolerance

Objective To investigate the release pattern of insulin after the load of glucose in patients with polycystic ovary syndrome ( PCOS ) and normal oral glucose tolerance.Methods Sixty-three patients with PCOS were undertaken oral glucose tolerance test (OGTT) and insulin release test,while 34 women with normal menstrual cycle served as control.Results Among 63 patients with PCOS,33 cases were obese with body mass index over 25 kg/m2,including 5 with abnormal OGTT.All 30 non-obese patients with PCOS had normal OGTT.The prevalences of insulin resistance were 78.8%,16.7%,and 9.0% in obese PCOS,non-obese PCOS,and control groups,respectively.Abnormal insulin release curve were found in 84.5%,70.0%,and 14.7% of subjects in these 3 groups,respectively.In 58 PCOS patients with normal OGTT,the prevalence of insulin resistance was 44.8%,and 75.9%with abnormal insulin release curve. Among them,body mass index of 32 patients,whose homeostasis model assessment for insulin resistance (HOMA-IR) and fasting insulin remained in normal range,was similar to those of control group [ ( 20.52 ± 2.86 vs 20.01 ± 2.54 ) kg/m2,P>0.05].Conclusion These findings indicate that insulin release test is useful in detecting insulin resistance.Insulin release is elevated in PCOS patients even with normal OGTT.
8.Expression of ? Defensin Genes in Rat Airway Epithelium after Intraperitoneal Injection of Emulsified Isoflurane

Objective To examine the expression of ? defensin genes and inflammatory cytokines genes in rat airway epithelium after intraperitoneal injection of emulsified isoflurane.Methods Thirty-six SD rats were assigned into six groups(A-F) randomly.The tissue from rat airway epithelium were harvested at 1 h after intraperitoneal injection of normal saline,30% fat milk and 8% emulsified isofluranes of 4 different dosages(0.25,0.5,1.0 and 1.5 times of ED50).Then expressions of ? defensin(BD-1,BD-2),tumor necrosis factor-alpha(TNF-?),interleukin beta(IL-1?),IL-8 mRNA were measured by semiquantitative polymerase chain reaction(PCR) using GAPDH as an internal standard.Results Except BD-1 and IL-8 mRNA,marked increases in expressions of TNF-?,IL-1? and BD-2 genes were observed with a dosage-dependent manner.Conclusion The expressions of BD-2 genes and inflammatory cytokines genes(IL-1?,TNF-?)in rat airway epithelium increase significantly in a dosage-dependent manner after intraperitoneal injection of emulsified isoflurane.

Objective To edit Chinese version of the Minnesota living with heart failure questionnaire (MLHFQ) and to estimate its reliability and validity.Metheds Translated and edited the Chinese version of MLHFQ according to the standard procedures,and tested 83 inpatients which were recruited from three polyclinics of Beijing district in China.Results Cronbach's alpha consistency was>0.80 for the three MLHFQ scores.Reproducibility ranged from 0.48 to 0.84 for items of weighted kappa coefficient and from 0.88 to 0.94 for three do-mains of Spearman's correlation coefficient.MLHFQ scores varied significantly with NYHA functional class(P<0.05),and there were intermediate-to-high correlations with the assumed corresponding SF-36 domains(-0.73 to -0.59).Factor analysis revealed three-factor structure explained 49.34% of the total variance.The MLHFQ scores before and after the treatment combined TCM and western medicine were significant (physical domain (26.15±7.15,17.63±8.50),emotional domain (8.96±5.73,6.8l±5.31),overall score(56.38±16.88,39.77±15.69) all P<0.01).Conclusions The Chinese version of MLHFQ is translated and edited according to the standard procedures.Its reliability and validity is good,and could be used as a clinical outcome measurement.
