2.The influencing factors of psychological health status and social disability in hashimoto thyroiditis

Objective To analyze of the influencing factors of mental health status and social disability for hashimoto thyroiditis(HT) patients.Methods The research employed Symptom Checklist 90 (SCL-90) and Social Disability Screening Schedule (SDSS) to evaluate the change and the influencing factors of mental health status and social disability of Hashimoto Thyroiditis patients.Result The scores of SCL-90 share,body symptom,sensitiveness relationship,depression,anxiety and social disability [(1.7±0.6),(2.0±0.7),(1.9±0.6),(2.0±0.60),(1.8±0.5) and (5.2±0.9),respective] in HT were increased compared with normal group[(1.4±O.4),(1.3±O.5),(1.6±O.5),(1.5±0.6),(1.4±0.4),(1.3±0.8) respective](P<0.01).In HT subjecte receiving levothyroxine(L-T4) therapy for 3 months,the scores of SCL-90 share score,body symptom,sensitiveness relationship.depression,anxiety and social disability were reduced respectively by(1.3±0.5),(1.5±0.5),(1.6±0.4),(1.6±0.4),(1.4±0.6) and(1.8±0.3)(all P<0.01).At the same time,the score of social disabilitv after therapy was increased compared with normal group (t=5.78,P<0.01).With the Logistic analyses.the score of social disability(F=1.16,P<0.01),TSH (F=2.05,P<0.01) and TP0(F=3.48.P<0.01)were risk factors of SCL-90 share score in HT subjects.Conclusion There were psychological health symptoms and social disability in HT case.TSH and TPO were risk factors of psychological health.L-T4Wag good for returning to normal mood.
3.Optimization of expression conditions of an induction strategy for improving liver targeted interferon (IFN-CSP) production in E. coli.

Expression conditions of induction strategies for the cytoplasmic inclusion bodies (IBs) production of liver targeted interferon IFN-CSP by recombinant Escherichia coli (E. coli) BL21 (DE3) were optimized in shake-flask cultures in this study. The factors of the optimized protocol included in the present study were pH, inducer IPTG (isopropyl beta-D-thiogalactoside) concentration, culture growth temperature, incubation time and induction point. The effects of those factors were investigated by 'single variable at a time' method, aimed to analyze characterization of the recombinant strain. Orthogonal experimental design was further used to optimize the above critical factors for IFN-CSP production. According to the expression optimization result, it was confirmed that the main influence factors were cell density and induction temperature. The IFN-CSP gene expression optimized conditions were: pH value of the culture medium was 6.0, culture temperature 37 degrees C, adding IPTG to final concentration 0.4 mmol/L when the recombinant strain growth density OD600 achieved 0.8 and induction time 4 h. At this point, the IBs represented 74.3% of the total cellular protein. Compared with the non-optimized condition, IFN-CSP production obtained in optimized induction strategies were increased by approx. 1.2-fold. The optimized induction strategy yielded 688.8 mg/L of IFN-CSP, providing experimental data to study the biology activity and productive technology of IFN-CSP.

4.Research on hepatitis C virus entry inhibitor.

Hepatitis c virus (HCV) infection has become one of the global public health problem,while there is no vaccine to prevent HCV infection, the so-called "cocktail" therapy that use a combination of drugs targeting multiple steps in the HCV infection cycle could achieve better curative effect. the process of HCV entering into host cell is the important step of drug intervention, in which HCV envelope protein El and E2, Host cell factors including Heparan sulfate(HS), CD81, scavenger receptor class B type I (SR-BI), Occludin (OCLD), Claudin (CLDN), low densitity lipoprotein receptor (LDLR), dendritic cell-specific ICAM-3-grabbing nonintegrin (DC-SIGN), Liver/lymph node specific ICAM-3-grabbing integrin(L-SIGN), trans- ferrin receptor 1 (TfR1) and so on play a important role. The virus and the host factors can be used as targets of hcv entry inhibitors many studies have shown that as novel and promising compounds, HCV entry inhibitors combinating with other drugs can be more effective in the treatment of HCV, this paper have re- viewed targets and inhibitors of HCV enterring into host cell since 1990s.

5.Related risk factors for injury severity of non-motor vehicle crashes in Hefei

Objective To explore the related risk factors of injuries caused by non-motor vehicle crashes in Hefei,Anhui Province. Methods From June 2009 to June 2011,the wounded from nonmotor vehicle crashes in Hefei were admitted to our hospital.The data of the wounded were collected to perform hypothesis test to screen out the related risk factors,which were then analyzed with multiple-factor non-conditional Logistic regression analysis. Results A total of 205 cases who were wounded in the non-motor vehicle crashes were admitted to our hospital from June 2009 to June 2011.Among all the cases,146 cases were wounded in electric bicycle crashes and 59 cases in bicycle crashes.There were 147cases (71.7%) of mild to moderate injuries ( ISS ≤ 15 ),and 58 cases (28.3%) of severe injuries (ISS > 15 ) including six deaths (2.9%).Single factor of different categories of the hypothesis test suggested that age,category,driving behavior,crash model,impact type,vehicle type were all related with the severity of injuries caused by non-motor vehicle crashes in Hefei.The multiple-factor non-conditional Logistic regression analysis showed four risk factors for the injury severity in non-motor vehicle crashes in Hefei,ie,vehicle type,crash model,violations of rules and regulations as well as age,according to the degree of correlation.Conclusion With the study of non-motor vehicle crashes in Hefei,primary identification of the risk factors for the traffic injuries is obtained,which provides scientific basis for the decrease of casualty and establishment of intervention measures.
6.Interleukin 2 promotes the expression of immunoglobulin G in a cervical cancer cell line HeLaS3

Objective:To analyze the effect of interleukin 2(IL-2) on immunoglobulin G(IgG) expression in cervical cancer cell line HeLaS3.Methods:By immunofluorescence-flow cytometry(FCM),we detected membrane and cytoplasmic IgG expression variation of HeLaS3 after IL-2 stimulation.By western blot,we detected IgG expression variation of HeLaS3 after IL-2 stimulation.Meanwhile,by RT-PCR with primers designed to amplify four subclasses of IgG,we detected ? transcript variation in HeLaS3 after IL-2 stimulation at mRNA level.Results:The result of immunofluorescence-FACS showed that after IL-2 stimulation for 48 hours,membrane and cytoplasmic IgG expression elevated in HeLaS3.The result of Western blot showed after IL-2 stimulation for 48 hours,IgG expression elevated in HeLaS3.Meanwhile,the result of RT-PCR showed that ?1 transcript mildly elevated after IL-2 stimulation for 48 hours.Conclusion:IL-2 can promote IgG expression in cervical cancer cell line HeLaS3.
7.Preliminary study of liver targeting interferon on the immune toxicity of mice

Objective To evaluate the immunotoxicity effect of Liver targeting interferon (IFN -CSP) on mice.Methods Mice were randomly divided into five groups:low, middle and high dose of IFN-CSP, solvent control group(saline) and Positive control group (cyclophosphamide).They were injected subcutaneously for 2 weeks.Delayed hypersensitivity test was used to determine the cell immunefunction and plaque forming cell assay was used to determine the humoral immune function.Results There was no significant difference of the the index of immune organ and the ear swelling degree between IFN-CSP groups and control group.There was also no significant difference on hemolytic plaque test between them.Conclusion IFN-CSP has no significant effect on both cellular immunity function and humoral immune function of mice, this results will provides the basis for further safety evaluation.

9.Effect of IFN-CSP on gene of JAK-STAT signaling pathway in HBV transgenic mice

Objective To investigate the anti-HBV molecular mechanisms of liver targeting interferon ( IFN-CSP ) in Balb/c-HBV transgenic mice.Methods Balb/c-HBV transgenic mice were randomly divided into 3 groups.Control group (treated with physiological saline), IFN α2b group (treated with 103 U/g IFN α2b), IFN-CSP group (treated with 102 U/g IFN-CSP).Another group of the non-transgenic mice were used as the Normal group.Each mouse was intramuscular injected with 50 μL dose once a day for 4 weeks.Total RNA of mice liver were extracted, and STAT1, STAT2, IRF-9, OAS1 gene expression of JAK-STAT signaling pathway were analyzed by real-time PCR.Results IFN α2b and IFN-CSP can significantly up regulate the expression of STAT1, STAT2, IRF-9, OAS1 gene of JAK-STAT signaling pathway (P<0.01).The induce effects of IFN-CSP on STAT1, STAT2, IRF-9, OAS1 were significantly better than that of IFN α2b (P<0.05).Conclusion The anti-HBV molecular mechanisms of liver targeting interferon (IFN-CSP) in Balb/c-HBV transgenic mice maybe related to regulate the expression of STAT1, STAT2, IRF-9, OAS1 gene of JAK-STAT signaling pathway.These results will lay a basis for the application of recombinant liver-targeting interferon.
10.Efficacy of Medical Therapy after Conservative Operation for Endometriosis

Objective To investigate the efficacy of medical therapy after conservative operation for endometriosis.(Methods)Endometriosis was confirmed in 89 patients by laparoscopic inspection,and conservative operations were performed to remove the adhesion,excise or destroy all the endometriotic tissues,including single excision of endo-(metriotic) masses or plus unilateral salpingoophorectomy,and to restore pelvic anatomy to the best possible condition.All the patients were divided into two groups randomly: treatment group(n=44),three months drug therapy of go-(nadotropin-)releasing hormone analogs(GnRHa) after operation;control group(n=45),no drug therapy after operation.Clinical symptoms,concentrations of CA-125,manual and ultrasonic pelvic check-up,as well as pregnancy rate to those who had a desire for future fertility(n=28)were followed up.
