International Journal of Laboratory Medicine 2009;30(1):16-19
Objective To investigate the status of AmpC β-lactamase and extended spectrum β-lactamases (ESBLs) production and drug-resistance characteristic of Enterobacter cloacae. Methods Enterobacter cloacae was isolated and cultured from variety of specimens from April 2005 to July 2007. VITEK32 automatic microbial analytical system was applied to performing identification of bacteria and susceptibility test. ESBLs were confirmed by NCCLS double disc assay, and three-dimensional test was used to detect AmpC.Results A total of 63 strains of Enterobacter cloacae were isolated, including 13(20.6%) single AmpC producing strains, 24(31.8%) single ESBLs producing strains, 6(9.5%) both AmpC and ESBLs producing strains, and 20(38.1%) neither AmpC nor ESBLs producing strains. The resistance of lactamases producing strains was significantly higher than that of lactamases non-producing ones. Especially in both AmpC and ESBLs producing strains, the drug resistance was serous. Conclusion The prevalence of AmpC β-lactamases and/or ESBLs producing strains is a higher level in Fuyang area. The drug resistance of AmpC β-lactamases producing Enterobacter cloacae isn′t identical to that of ESBLs producing one. Different categories of antibacterials should be chosen according to the different types of producing lactamases.
2.Effect of Valproate exposure-induced autism spectrum disorder model on neuroimmune system of infant mice at early pregnancy
Chinese Journal of Applied Clinical Pediatrics 2015;30(16):1265-1268
Objective To establish the Valproate (VPA) exposure-induced autism spectrum disorder (ASD) model at early pregnancy in rats, and to study the changes of social interaction test and neuroimmune system in autism model rats, and discuss the pathogenic mechanism. Methods Twelve-week-old rats were randomly divided into the non-exposed group (n =10) and the VPA-exposed group (n =10).Their babies were respectively just the normal control group (n =10) and the model group (n =10).The autism-like social behavior was evaluated via the social interaction test. The level of interleukin (IL)-1β,IL-4,IL-6,interferon-gamma (IFN-γ),transforming growth factor-beta (TGF-β) in mothers and offspring were detected by using enzyme-linked immunosorbent assay (ELISA).The histopathological damage in brain was observed with immunohistochemical staining. Results The score of social interaction test in the model group [(-163.16 ± 101.92) scores]was lower than that in the normal control group [(132.73 ± 114.63) scores] (t =-6.100,P < 0.05).The levels of IL-1β,IL-4,IL-6,IFN-γ in VPA-exposed group were higher than those in the non-exposed group (t =5.883,6.394,5.655,5.393,all P < 0.05),while the level of TGF-β was lower than that in the non-exposed group (t =-6.726,P < 0.05).The levels of IL-1β,IL-4,IL-6,IFN-γ in the model group were higher than those in the normal control group(t =3.058,3.048,6.670,5.486,all P < 0.05),while the level of TGF-β was lower than that in the normal control group (t =-6.516,P < 0.05).The change trend of the level of cytokines in the serum of ASD model rats was similar to the change trend of the level of cytokines in the serum of VPA exposed rats. The result of social interaction test the approach-avoidance score was inversely correlated with the levels of IL-1β and IL-4 (r =-0.802,-0.781,all P < 0.05).The result of immunohistochemical staining showed the expressions of glial fibrillary acidic protein (GFAP) and ionized calcium-binding adapter molecule 1 (IBA1) in the model group were higher than those in the normal control group. Conclusions The main neuroimmunological pathogenesis mechanism can be explained as follows:at early pregnancy, the maternal cytokines skewing influenced by VPA exposure can make immune activation, induce immune system dysfunction and affect the brain growth and development, which result in autism-like behavior in offspring.

5.Identification of Eight Novel Alternative Splicing Forms of CD72 and Their Differential Expression in a Mouse Model of SLE
Progress in Biochemistry and Biophysics 2006;0(11):-
CD72 is a B cell specific receptor that exists in multiple alternative splicing forms. Eight novel alternative splicing forms of CD72 were identified from the spleenocytes of BALB/C mice. Two very unique intron sequences were found in those alternative splicing forms. One kind of splicing variants retained the intron1 in the mRNA. This intron can be translated into 32 amino acid residues without changing the reading frame of the whole proteins. Another kind of splicing variants used an alternative 3' splice site in intron 3(3'AS) which led to premature termination of its encoded protein. The differential expression of the CD72 splicing variants were compared in BALB/C and NZB/W mice that were at different stage of systematic lupus erythematosis(SLE) disease development. It was found that 1) splicing forms containing 3'AS was rare in all samples examinated; 2) splicing forms containing two ITIM domains and transmembrane domains were more abundant in BALB/C mice than in NZB/W mice, even in some cases the two ITIM domains were separated by the intron 1; 3) a shorter splicing form with both exon2 and exon3 missing was expressed highly in terminally diseased NZB/W mice. These results suggested an important role of CD72 alternative splicing forms in B cell receptor signaling and in SLE.
6.Clinical application and progress of Tei index
International Journal of Pediatrics 2016;43(2):101-104,108
Currently, heart failure is one common disease causing serious damage to children's health and life which has high missed diagnosis rate and mortality.Accurate and effective evaluation of pediatric cardiac function is of great importance in the treatment, surveillance and prognosis of disease.Tei index was first presen-ted by Japanese scholars as a new comprehensive indicator of global (combined systolic and diastolic)myocar-dial function. It was defined as a ratio between the sum of isovolumic times (ICT and IRT)and ejection time (ET).This index can be measured by a simple, reproducible and non-invasive method regardless of changes in the heart rate or ventricular shape and other external factors. The sensitivity for ventricular dysfunction could be high.Tei index has been used more and more often in the clinical evaluation of cardiac function especially in children. This paper reviews the measurement methods, related factors and pediatric clinical application progress of Tei index.
