Objective To investigate the influence of different diluents(physiologic saline, dis-tilled water and human inactivated serum) on measurement of 15 items of biochemical parameters. Methods Fifteen items of biochemical parameters [alanine transaminase(ALT), aspartic transaminase (AST), alkaline phosphatase( ALP), gamma glutamyl transpeptidase ( GGT), creatine kinase ( CK),lactate dehydrogenase(LD), hydroxybutyrate dehydrogenase(HBDH), total bilirubin(TBIL), direct bilirubin(DBIL), total bile acid(TBA), ereatinine(Cr), uric acid(UA), eholesterol(CHO), glucose (GLU), and blood urea nitrogen(BUN)] were chosen. For each parameter, 45 serum samples with different eoncentrations of the parameter were collected. After diluted with different diluents(physio-logic saline, distilled water or human inactivated serum), the serum samples were detected by applying the fully automated biochemical analyzer. The mean value was calculated and statistical analysis was performed. Results There were some differences of detection results when the specimens were diluted with different diluents. ALT, AST, GGT, DBIL, and HBDH serum samples could be diluted by 10 times with physiologic saline, distilled water or human inactivated serums ALP and TBA serum sam-ples could only be diluted with inactivated serum, otherwise its result would be lower; GLU, TBIL samples could be diluted with distilled water and inactivated serums for BUN, CR, UA, CK, LDH,and CHO samples, physiologic saline or human inactivated serum might be optimal; if distilled water was chosen, the results of other parameters tented to decline except UA. It was BUN was improper to dilute the BUN samples with distilled water. In addition, there was no significant difference between the items diluted by 5 times and 10 times with physiologic saline. All the 15 items could be diluted with inactivated serum. Conclusion The inactivated serum should be the first choice of diluents to e-nure the accurate results of biochemical parameters. If the prepared inactivated serum is absent, we may choose other diluents according to the above-mentioned results.

Objective To investigate the effects and related mechanism of iNO on erythrocyte deformability in septic acute lung injury(ALI).Methods Thirty anesthetized and mechanically ventilated young piglets(3-4weeks old)were allocated to 4 groups:(1)normal(group C,n=6):intraperitoneal injection of normal saline(NS).(2)normal+iNO(group CNO,n=4):NS followed by mechanical ventilation for 4 h,and then treated with iNO at 10 parts per million in volume(ppm).(3)model(group M,n=10):i.p.Escherichia coli at 5 × 109 cfu/ml,5 ml,and(4)NO(group MNO,n=10):i.p.Eschefichia coli at 5×109 cfu/ml lollowed by iNO-at 10 parts per million after ALI.Blood viscosity, malondialdehyde(MDA)in erythrocyte,Ca2+-Mg2+-ATPase and Na+-K+-ATPase of erythrocyte membrane were measured at basdine, establishment of ALI,12 and 24 h during the treatment. Results For normal age-matched piglets, there was little change of whole blood viscosity at high shear rate(110 s-1),which is a measure of erythrocyte deformability,so was the level of MDA in erythroeyte and the activity of erythroeyte membrane Ca2+-Mg2+-ATPase,Na+-K+-ATPase when iNO was provided for 12 or 24 h.At the time ALI was established, whole blood viscosity at high shear rate and MDA levels in erythroeyte increased significantly compared to the baseline level(P<0.05),while the activity of erythrocyte membrane Ca2+-Mg2+-ATPase and Na+-K+-ATPase decreased significantly(P<0.01,vsbaseline).Exposure of iNO for 12 h resulted in decreased whole blood viscosity at high shear rate and MDA levels in erythroeyte(P<0.05,vs group C),and higher activity of erythrocyte membrane Ca2+-Mg2+-AT-Pase and Na+-K+-ATPase.When iNO was administered for 24 h,there was little difference of whole blood viscosity, activity of erythrocyte membrane Ca2+-Mg2+-ATPase,Na+-K+-ATPase and MDA levels in erythroeyte between the C and NO groups. Conclusion In the piglets with septic ALI,there was deterioration of erythrocyte deformability.Inhaled NO for 12 h may alter the deterioration, however, this effect is transient.

Microbiology 1992;0(02):-
Isocitrate lyase (ICL) which plays a pivotal role in the glyoxylate cycle of mycobacterium tuberculosis(MTB) could be a potential target against mycobacterium tuberculosis.Deoxyribozyme(DRz) is a small single-strand,enzymatic DNA molecule.Assistant by sketch of secondary structure, appropriate and not appropriate target sequences of MTB ICL mRNA were predicted. Two DRzs were designed targeting above sequences respectively. To affirm feasibility of this kind of prediction,in vitro cleavage of MTB ICL mRNA segment by DRz-ICLcf and DRz-ICLcj were performed. The results showed that sketch of secondary structure could indeed predict potential target of DRz.This research will provide basis for developing DRz as a novel antimicrobial agent against MTB.

7.Mechanism of Spontaneous Rupture of Hepatocellular Carcinoma
Chinese Journal of Bases and Clinics in General Surgery 2003;0(02):-
Objective To study the mechanism of spontaneous rupture of hepatocellular carcinoma(HCC).Methods Articles have been reviewed to find out the theory of spontaneous rupture of HCC.Results Researchful results suggested that the injury of small arteries was usually followed in patients of spontaneous rupture of HCC.In this review, the immune complex, which composed of hepatitis B virus e antigen, complement C1q and immunoglobulins, was found deposited in the elastic membrane of arteries.Likely as a result of immune complex deposition, vascular injury occurs mainly in the small arteries where the deposition of immune complex was present. The small arteries in which immune complex deposited are readily injuried and cause hemorrhage and rupture of HCC during vascular load increase. Conclusion We would conclude that immune complex deposition in vessel wall led to the small arteries injury may be the factor involved in the pathogenesis of spontaneous ruptured HCC.
