Steroid-restant nephrotic syndrome (SRNS) is often continuously relapsed, and even develops into end-stage renal di-sease. It has been a great difficulty in the treatment of nephritic syndrome in children. In recent years, many researchers of children SRNS on diagnosis, pathology, treatment have been carried on domestically and in abroad. This paper reviews the latest update on the di-agnosis and treatment of SRNS in children.

BACKGROUND:Acidic fibroblast growth factor (aFGF) possesses good effects on vascularization and osteogenesis.But whether aFGF can promote the vascularization in animals with early-stage avascular necrosis of the femoral head (ANFH) remains unclear.OBJECTIVE:To investigate the vascularization of aFGF composited by partially depreteinized bone (PDPB) in repair of early-stage ANFH.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed in the College of Life Science,Nanhua University between January 2008 and January 2009.MATERIALS:Ribs from healthy,adult,New Zealand rabbits were prepared into PDPB by a series of physico-chernical methods including degreasing,deproteinization,partial decalcification and freeze drying,aFGF diluted with sterile distilled water was composited by PDPB particles to prepare artificial composite bone.METHODS:A bone window was made at the juncture of femoral head and femoral neck bilaterally in 24 healthy,adult,New Zealand rabbits.Rabbit models of bilateral ANFH were established by removing approximately 50% of cancellous bone and perfusion with 95% ethanol.Successful bilateral ANFH models were randomly divided into a blank group,a simple PDPB group,and an artificial composite bone group.PDPB and artificial composite bone were implanted into the PDPB and artificial composite bone group accordingly.The blank group did not receive any implantation.MAIN OUTCOME MEASURES:At 2,4,and 8 weeks after surgery,ink-injected specimens were prepared for microvessel count and microvessel area analysis.RESULTS:Microvessel number and microvessel area were least in the blank group,followed by simple PDPB group,and lastly the artificial composite bone group.There was significant difference in microvessel number and micrevessel area between artificial composite bone group and blank,simple PDPB groups (P＜0.05).CONCLUSION:Tissue-engineered artificial bone composited by aFGF and PDPB promotes vasculadzation in repair of earlv-staqe ANFH in rabbits.

Inhalation injury seriously threatens the survival and quality of life in burn and trauma patients. So far there is no breakthrough in the treatment of inhalation injury. A significant advance has been witnessed in the experimental study of the use of stem cells in the treatment of lung injury in recent years. In this paper, according to the results of our study in the systemic transplantation of bone marrow mesenchymal stem cells for the treatment of inhalation injury, the effect of mesenchymal stem cells on anti-inflammatory process and repair of lung tissues in inhalation injury, and its possible mechanisms are reviewed.

Purpose To explore the correlation of Noxa gene with clinical pathology and its effect on proliferation and apoptosis of HepG2 cells.Methods Noxa protein in 100 hepatocellular carcinoma tissues and 100 paracancerous tissues were detected by imnunohistochemical technique,clinicopathological parameters and follow-up data were statistically analyzed.The recombinant eukaryotic expression plasmid pIRES2-EGFP-Noxa was transiently transfected into HepG2 cells with lipofectamine.Both Noxa mRNA and protein were detected by RT-PCR and Western blot respectively.The inhibition of cell proliferation was evaluated by MTT assay.The cell apoptosis was detected by flow cytometry (FCM).Results Immunohistochemistry showed that in tumor tissue of hepatocellular carcinona the positive rate of Noxa protein expression was 50％,while it was 78％ in cancer adjacent normal mucosal tissue,with statistically significant difference (P ＜ 0.05).The expression of Noxa was related to TNM staging and the tumor differentiation.Exotic Noxa gene was expressed successfully in HepG2 cells after transfected with lipofectamine.The expression of Noxa mRNA and protein of HepG2 cells was significantly up-regulated after transfected 24 h,48 h and 72 h (P ＜ 0.05).MTT assay showed that the Noxa expression inhibited HepG2 cell proliferation in a time-dependent manner for 24 h,48 h and 72 h (P ＜ 0.05),which was significantly different from that of the control group (P ＜ 0.05).Apoptosis rate after transfected 24 h,48 h and 72 h was (15.5 ± 0.9)％,(24.6 ±0.8) ％ and (35.4 ±0.7) ％,respectively.There were significant difference between Noxa and the control group (P ＜ 0.05).Conclusion The expression of Noxa in hepatocellular carcinoma tissue is closely related to TNM stage and the tumor differentiation.Noxa gene overexpression is able to effectively inhibit the proliferation and promote the apoptosis of HepG2 cell line.

Objective To investigate the effect of pseudolaric acid B(PAB) on the expression of vascular endothelial growth factor (VEGF) in human hepatoma BEL-7402 cells. Methods BEL-7402 cells were cultured with PAB (0, 0.5, 1.0, 2.0, 4.0 and 8.0μmol · L-1) for 6 h and stained with 0.4%trypan blue. The cell viability was observed and calculated under the microscope. The cell proliferation was detected by WST-8 analysis. The expression of VEGF mRNA in BEL-7402 cells was detected using RT-PCR method. The expression of VEGF protein in BEL-7402 cells was detected using Western blot assay. Results There was no significant difference in the cell proliferation of BEL-7402 cells between different PAB treat-ment groups and control group. With the increased concentration of PAB, the expressions of VEGF mRNA were significantly decreased in different PAB treatment groups (1.90±0.02,1.31±0.03,0.86±0.04,0.60±0.03 and 0.34±0.02) compared with that of control group (2.53±0.07, P＜0.01). The expressions of VEGF protein were significantly decreased in different PAB treat-ment groups (2.60±0.09, 2.42±0.07, 1.85±0.05, 0.92±0.05 and 0.60±0.04) compared with that of control group (3.06±0.15, P＜0.01). Conclusion PAB can inhibit the expressions of VEGF mRNA and protein in BEL-7402 cells.

BFU-E growth is enhanced significantly after addition of either non-stimulated or LPS-stimulated peritoneal macrophage culture supernatants. This supernatant shows positive and negative control on the growth of CFU-E and CFU-GM, low concentration supernatant shows promoting activity and high concentration supernatant presents inhibition. The LPS-stimulated supernatant shows much stronger promoting activities than non-stimulated supernatant. This supernatant has no obviously action on the proliferation of CFU-S. The mechanism of the regulation of mouse hemopoiesis by the conditioned media of the peritoneal macrophage is discussed. Our results show that this supernatant of cultured peritoneal macrophages may contain EPO-like, BPA-like and CSF-GM-like factors.
