Objective To evaluate the clinical efficacy and safety of orlistat in Chinese overweight and obese patients. Methods In six research centers, 444 overweight and obese patients (body mass index 25～40 kg/m 2) were divided into orlistat group (296 patients) and placebo group (148 patients) with a low-energy diet for 24 weeks by a randomized, double-blinded, placebo-controlled study. Results For intent-to-treat analysis, 286 orlistat-treated and 142 placebo-treated subjects were evaluated. Objective To discuss whether acupuncture at Neiguan (PC 6) inhibits myocardial hypertrophy via monoamine neurotransmitters.Method Sixty female Sprague-Dawley rats were randomized into a control group, a model group, a sham electroacupuncture (EA) group, and an EA group, 15 cases in each group. The rat models of myocardial hypertrophy were developed by subcutaneous injection of Isoprenaline hydrochloride (ISO) for 14 d. The EA group was intervened by EA at Neiguan (PC 6), and the sham EA group was by EA at a sham acupoint [5 mm away lateral to Neiguan (PC 6). The Electrocardiogram (ECG) of each group was recorded, electron microscope was used to observe the microstructure of the rats’ myocardial tissues, and high performance liquid chromatography (HPLC) was adopted to detect the contents of catecholamines in myocardial tissues.Result The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) in the model group and sham EA group were significantly different from that in the control group (P<0.05). The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) of the EA group were significantly different from that of the model group (P<0.05).Conclusion EA at Neiguan (PC 6) can improve myocardial hypertrophy, which is related to the regulation of the topical neurotransmitters; it has provided laboratory evidences for the relationship between Neiguan (PC 6) and heart, i.e. the relationship between acupoints and internal organs.

This study was aimed to evaluate impacts on immune regulation with spleen-strengthening and kidney-tonifying method among advanced malignant tumor patients. The enrolled advanced malignant tumor patients were randomly divided into the control group and the treatment group. The control group was treated with conventional treatment plan. The treatment group was treated with spleen-strengthening and kidney-tonifying Chinese herbs on the basis of conventional treatment plan for three months. Clinical effect, integral of clinical symptom (ICS), T-lym-phocyte subsets, and drug safety index were observed and detected before and after treatment. The results showed that the effective rate and response rate in the control group were 31.1% and 71.1%, respectively. And the effective rate and response rate in the treatment group were 54.2% and 85.4%, respectively. The treatment group was better than the control group (P< 0.05). Compared to the control group, the treatment group had obvious regulation on T-lymphocyte subsets. The improvement on ICS in the treatment group was better than the control group (P < 0.05). There was no obvious adverse reaction with the application of Chinese herbs. It was concluded that the spleen-strengthening and kidney-tonifying method can improve clinical effect, life of quality, and reduce adverse reaction a-mong advanced malignant tumor patients. Its mechanism may be through the regulation of immune function.

The aim of this study was to the prepare the standard decoction of paeoniae radix rubra for its investigation of quality standard.The standard decoction of paeoniae radix rubra was dissolved by water using standardization method and the content of peoniflorin was determined by UPLC-DAD.Then the transfer rate and the rate of extractum were calculated,and the fingerprint analysis was carried out.As a result,the content of the standard decoction of paeonia lactiflora ranged from 3.79 to 5.68 mg·mL-1 from 15 paeoniflorin batches,while the transfer rate ranged from 56.58％ to 90.14％,and the average transfer rate was 73.97％ and the standard deviation was 10.91％.The rate of extractum ranged from 16.74％ to 29.24％,the average yield was 21.82％ and the standard deviation was 3.55％.There were eleven common peaks,four of which were identified,including oxypaeoniflorin,albiflorin,paeoniflorin and benzoylpaeoniflorin.In conclusion,it was demonstrated that the method was simple with good repeatability for the preparation and quality standard studies of the standard decoction of paeoniae radix rubra.

Objective To compare the efficacy and tolerability of nateglinide with those of acarbose in Chinese type 2 diabetes mellitus (T2DM) patients.Methods This multi-center,randomized,double-blind,parallel-arm study compared the efficacy and tolerability of nateglinide( 120 mg,3/d,n = 119) and those of acarbose( 100 mg,3/d,n = 118) during a 12-week treatment in T2DM patients uncontrolled by diet with glycosylated haemoglobin (HbA1c) 6.5% - 11.0% .Results Monotherapy with nateglinide (120 mg,3/d)or acarbose (100 mg,3/d)decreased HbA1c to a similar extent during 12-week treatment.The mean change from baseline to end-point in HbAlc was ( -0.90±0.98)% and ( -0.83±0.81 )% in patients receiving nateglinide and acarbose,respectively,with no significant difference between the two groups (P>0.05).The decrease in fasting plasma glucose (FPG)was similar between nateglinide and acarbose (P > 0.05).The mean change in 2-hour postprandial plasma glucose ( PG2h ) was ( - 1.45 ± 2.74) mmol/L and ( -2.20±2.21 ) mmol/L in patients receiving nateglinide and acarbose(P =0.0017).Body weight was significantly decreased in both groups at the end-point ( P < 0.05 ),although the decrease was more with acarbese than nateglinide [( -0.66±1.79)kg vs (-2.06±2.00) kg,P=0.0000].And the proportion of patients experiencing any presumed drug related adverse events was not significantly different between the two groups.Conclusions Nateglinide ( 120 mg,3/d) is effective and well tolerated in T2DM patients uncontrolled by diet,demonstrating similar HbA1c reductions as compared with acarbose (100 mg,3/d).

Objective To explore whether major depressive disorder（MDD）and the therapeutic effect of fluoxetine are related to a functional polymorphism-1019C/G in the promoter region of the 5-HT1A receptor（HTR1A）gene.Methods Genotype and allele frequencies of HTR1A receptor gene-1019C/G polymorphism in MDD patients and healthy subjects（control）were examined by PCR-RFLP technique.Before and after the MDD patients accepted fluoxetine treatment for 6 weeks,17-item Hamilton depression rating scales（HAMD）were made to determine the severity of the symptoms,the outcome and remission status.Results There were significant differences in-1019C/G gene genotypes and alleles distribution between the patients and the healthy control,G allele frequency of the MDD patient was higher than that of the healthy control（P 0.05）.There were significant differences in HAMD scores among the patients with different genotypes in MDD group（P 0.05）,the score of C/C genotype patient was especially higher than that of C/G genotype（P 0.05）and G/G genotype patient（P =0.008）.There was no statistical difference in the therapeutic effect of fluoxetine among the patients with different genotypes in MDD group（P =0.761）.Conclusion HTR1A gene-1019C/G genetic polymorphism might related to MDD,especially G allele might be the possible risk factor of MDD.C allele might be correlated with the degree of pathogenetic severity,especially patients with the-1019C/C carriers.-1019C/G genetic polymorphism was not related to the clinical outcome of MDD patients treated with fluoxetine.

Objective To provide a practical device and protocol to hold conscious rats for subsequent operations which can overcome the disadvantages of existing methods .Users can complete the experiment more efficiently , with or without prior experience .Methods Using transparent plastic film , plastic sealing machine and sponge to make a simple device for holding rats , by taking advantage of their escaping nature .To compare the performance of the new method and existing methods for holding and injecting rats .Results Compared with existing methods , the new device and method can reduce the time-consuming to hold rats by 44.7%, from 18.13 seconds to 10.03 seconds.For holding and injecting , the new method can reduce the time-consuming by 55.3%, from 139.33 seconds to 52.26 seconds .Conclusions The new device and method is good for holding and injecting rats or drawing blood from the caudal veins .It can shorten the time of operation and reduce the stress reaction in the animals .It’ s especially helpful for inexperienced experimenters such as students in teaching and research tasks .

Objective: To construct a lentiviral vector with overexpression of endoplasmic reticulum protein 29 ERp29), and to explore the effects of ERp29 overexpression on the proliferation, migration and invasion of gastric cancer MGC803 and SGC7901 cells. Methods: The lentiviral plasmid with ERp29 pCDH-ERp29) tagged with red fluorescentce protein and control plasmid pCDH-Vector) were constructed, and then the MGC803 and SGC9901 cells were infected with these lentivital vectors. Under fluorescence microscope, the infection of these cells were observed. CCK-8 assay and clone formation assay were performed to test the proliferation abilities of MGC803 and SGC7901 cells infected with ERp29. Transwell chamber assay was used to test the abilities of migration and invasion of MGC803 and SGC7901 cells infected with ERp29. Real-time PCR and Western blotting methods were used to detect the expression levels of ERp29 mRNA and protein in MGC803 and SGC7901 cells which were stably infected by ERp29, respectively. Results: The enzyme digestion identification result showed that the pCDH-ER29 overexpression vector was successfully constructed. The fluorescence microscope result showed the successful infection in the gastric cancer cells. The CCK-8 assay and clone formation assay results showed that compared with pCDH-Vector group, the proliferation ability of the cells in pCDH-ERp29 group had no marked changes (P > 0.05). Transwell chamber assay revealed that compared with pCDH-Vector group, the number of migration and invasion cells in the MGC803 and SGC7901 cells in pCDH-ERp29 group were significantly decreased (P < 0 . 05). The expression levels of ERp29 mRNA and protein in the cells in pCDH-ERp29 group were significantly increased compared with pCDH-Vector group (P < 0 . 05). Conclusion: The lentiviral vector with overexpression of ERp29 is constructed successfully, and the overexpression of ERp29 in MGC803 and SGC7901 cells can significantly inhibit their abilities of migration and invasion.

Objective To compare the clinical efficacy of hemiarthroplastyand and proximal femoral intramedullary nailing treatment (PFNA) in treatment of elderly unstable intertrochanteric fracture. Methods A total of 368 cases of unstable intertrochanteric fracture were selected in our hospital. They were divided intoⅠgroup (184 cases) andII group (184 cases) by using the method to random number table. TheⅠgroup was treated with hemiarthroplasty, while the IIgroup was treated with PFNA. Results The operative time, postoperative and postoperative bleeding, operation of drainage, the incidence of complications inIIgroup were significantly lower than those inⅠgroup, the differences were statistically significant(t=-16.58,-9.38,-7.26,χ2=9.125, P<0.01). The limb weight bearing time, excellent and good rate of hip function and late complication incidence rate inⅠgroup were better than those of IIgroup. There were statistical differences between two groups (Z=11.11,P<0.05). Conclusion Hemiarthroplasty is suitable for patients who have good body condition, while there is a high early complication incidence rate. Those patients who have poorly body condition and more basic diseases, PFNA would be a better choice, while more attention should be paid to later complication in-cidence rate.
