Journal of Huazhong University of Science and Technology (Medical Sciences) 2014;34(3):387-92
A surgical technique of lateral crus strut graft to correct the deformity of nostril contour and improve asymmetric nostril shape by autologous rib cartilage was reported. Thirty-five patients (20 males and 15 females), with a mean age of 19 years (range of 16-26 years) were included in this study. All of the patients suffered moderate to severe unilateral cleft lip nostril deformity and underwent surgical technique of lateral crus strut graft by autologous rib cartilage to correct the asymmetric nostril contour from 2010 to 2012. The views of the nostril contour on both sides were reviewed and some parameters were measured at different time points (preoperatively, and 3 months and one year postoperatively) including the long axis and short axis of the nostrils. The differences in nostrils including long axis, short axis, elliptical area and eccentricity on the both sides were compared at different time points. The differences in the long axis and eccentricity of the nostrils on the two sides were decreased significantly between the preoperative view and postoperative view (P<0.05). The surgical technique of lateral crus graft with autologous rib cartilage is effective to correct the abnormal nostril contour and improve the asymmetry for unilateral cleft clip patients.

Objective:To investigate the prevalence and influencing factors of amnestic mild cognitive impairment of rural elderly in Guizhou province, which aims to provide scientific basis for the prevention and control of cognitive impairment in the elderly. Methods:Adopting a multi-stage cluster sampling method, a total of 1 535 rural Han and Bouyei elderly people aged 60 and above were selected from Guiyang city and Qiannan prefecture in Guizhou province as the survey subjects for the current situation survey, including demographic sociological characteristics, such as social was utilized behavior, social behavior disease history, height and weight and so on.Mini-mental state examination was utilized to measure cognitive function and SPSS 26.0 statistical software was used to perform χ 2 inspection and multivariate unconditional Logistics regression analysis to calculate odds ratio ( OR) and 95% confidence interval (95% CI). Results:A total of 242 elderly patients with aMCI were detected (15.8%). The results of univariate analysis showed that ethnicity(χ 2=4.333, P<0.05), gender(χ 2=18.367, P<0.01), marital status(χ 2=9.721, P<0.01), occupation(χ 2=7.786, P<0.01), annual family income(χ 2=28.085, P<0.01), current smoking(χ 2=11.873, P<0.01), specific hobbies(χ 2=25.968, P<0.01), physical exercise(χ 2=11.871, P<0.01), living style(χ 2=13.190, P<0.01), and activity participation(χ 2=13.004, P<0.01) all had an impact on aMCI. Multivariate Logistic regression analysis showed that Bouyei nationality( P<0.05, β=0.288, OR=1.333, 95% CI=1.002-1.775) and the women( P<0.05, β=0.516, OR=1.676, 95% CI=1.233-2.278)were risk factors for aMCI, and high annual family income( P<0.05, β=-0.839, OR=0.432, 95% CI=0.308-0.606), specific hobbies( P<0.05, β=-0.580, OR=0.560, 95% CI= 0.394-0.795), physical exercise( P<0.05, β=-0.410, OR=0.664, 95% CI=0.493-0.894), participation in activities( P<0.05, β=-0.424, OR=0.654, 95% CI=0.488-0.877), and non-living alone( P<0.05, β=-0.563, OR=0.569, 95% CI= 0.374-0.866) were the protective factors.Comparison of the prevalence of the disease between the Han and Bouyei nationalities, the detection rate of aMCI for the Bouyei elderly (18.0%) was higher than that of the Han (14.1%) (χ 2=4.333, P<0.05). After stratification according to gender, family annual income, specific hobbies, physical exercise, participation in activities and living style, the detection rate of elderly female subjects of Bouyei nationality was higher than that of Han nationality, whose difference was statistically significant (χ 2=5.562, P<0.05). The detection rate of Bouyei elderly was higher than that of Han when the annual household income was less than 30 000, and the difference was statistically significant (χ 2=8.570, P<0.01). Conclusion:The incidence of aMCI among the elderly of Bouyei nationality is higher, and the incidence of aMCI among females is higher than that of males, which should be paid more attention to.It is of vital importance to strengthen health education and publicity, guide the formation of knowledge-belief-behavior health-related behavior patterns, so as to improve the quality of life and reduce the risk of cognitive impairment.

AIM: To investigate the effect of selective silencing of SLC7a8 on uptaking L-dopa in renal tubular epithelial cells of rat (NRK-52E). METHODS: The three siRNAs targeting SLC7a8 (siRNA-1, siRNA-2, siRNA-3) were designed and synthesized. A siRNA with nonspecific coding sequence (siRNA-con) was used for control. All siRNAs were transfected into NRK-52E cells. The siRNA-con transfected group, blank control group and gene-specific silencing SLC7a8 group were set up. The efficiency of transfection was estimated by flow cytometry. The efficiency of RNA interference was detected and screened by RT-PCR preliminarily, and was followed by Western blotting at protein level. The concentrations of L-dopa uptake into the NRK-52E cells were detected by ultraviolet spectrophotometry at different time points (6, 12, 24, 36, 48, 60, 72 and 120 min). RESULTS: The transfection efficiency was 94% detected by flow cytometry. The initial screening of RT-PCR showed that the efficiencies of RNA interference of siRNA-1 and siRNA-3 were higher, and siRNA-3 was the highest at protein level determined by Western blotting. No distinctive change was found between siRNA-con treated NRK-52E and blank control cells. The L-dopa uptake at different time points (6, 12, 24, 36, 48, 60, 72 and 120min) in siRNA-interference group was lower than that in siRNA-con transfected group and blank control group. No significant difference of L-dopa uptake between siRNA-con group and blank control group was observed. CONCLUSION: RNA interference technology selectively down-regulates SLC7a8 expression in rat renal tubular epithelial cells. The L-dopa uptake is also decreased after specifically silencing the slc7a8 expression.

OBJECTIVETo study DNA damage, Bcl-2 and Bax expression, and ultrastructure change in spermatogenic cell of mice by cadmium exposure.
METHODSTwenty-four male mice were divided into 4 groups: 3 groups treated with cadmium chloride of 1, 5, 10 micromol x kg(-1) x d(-1) i.p. respectively for 5 days, and one normal saline control group. The DNA damage of spermatogenic cell by single-cell gel electrophoresis technology was detected. The expression positive rate of Bcl-2, Bax protein in spermatogenic cell by the immunohistochemical method was assayed, and the ultrastructural change of spermatogenic cell by the transmission electron microscope was observed.
RESULTSDNA damage rates of of spermatogenic cell in 1, 5, 10 micromol/kg cadmium chloride groups were higher than that of normal group (P < 0.001). Bcl-2 protein expression positive rates were lower than that of normal group (P < 0.001). Bax protein positive expression rate in 5 micromol/kg group was higher than those in normal group, and 1, 10 micromol/kg groups. The ultrastructure of karyotis, karyotheca, mitochondria, endoplasmic reticulum in three treated groups had different degree of damage and the degree of ultrastructural change was increasing with rising concentration of cadmium.
CONCLUSIONCadmium exposure will cause the DNA break, Bcl-2 and Bax protein abnormal expression and ultrastructural change in spermatogenic cell.
