1.Clinical Observation of Habitual Abortion Patients Treated by Yishen Tianjing Zhuyun Prescription Together with Active Lymphocyte Immunotherapy

Objective To investigate the therapeutic effect of Yishen Tianjing Zhuyun Prescription(a Chinese herbal recipe consisting of Xianqi Yizhen Capsules and Xianzi Yizhen Capsules, having the actions of strengthening kidney-essence and promoting gestation) together with active lymphocyte immunotherapy for habitual abortion (HA)patients. Methods A total of 139 HA outpatients with negative blocking antibodies(BA)were randomly divided into treatment group(70 cases) and control group(69 cases). The control group was treated only with active lymphocyte immunotherapy, while the treatment group was treated with Yishen Tianjing Zhuyun Prescription and active lymphocyte immunotherapy . Results (1) After treatment, the BA-positive rate was 82.86% in the treatment group and was 60.87% in control group, and there existed significant difference between the two groups (P < 0.05 ). (2) The total anti-abortion effective rate was 87.14% in the treatment group and was 66.67% in control group, the difference being significant between the two groups(P<0.05 ). Conclusion Yishen Tianjing Zhuyun Prescription combined with active lymphocyte immunotherapy is more effective than active lymphocyte immunotherapy alone for the treatment of HA patients with negative BA.

3.Evaluation of cefoxitin disk diffusion test for the detection of methicillin resistant Staphylococcus aureus

Objective To establish and evaluate the method for detection of methicillin-resistant Staphylococcus aureus (MRSA) by the cefoxitin disk diffusion test.Methods The disks with 30 microgramme of cefoxitin and oxacillin recommended by NCCLS were used for the detection of MRSA with PCR by using mecA as the reference.The MIC of cefoxitin to Staphylococcus aureus was also determined.Results Of the 145 test isolates of Staphylococcus aureus,83 isolates showed the mecA-positive by PCR,62 isolates were negative.In cefoxitin disk diffusion tests,82 isolates showed MRSA-positive and 63 isolates were negative.The sensitivity was 98.8% and the specificity was 100%.In oxacillin diffusion tests,the sensitivity was 95.2% and the specificity was 96.8%.Conclusions For the detection of MRSA,the cefoxitin disk diffusion test was consistent with the results by PCR for mecA.The cefoxitin disk diffusion test is easy to be performed and can be used in routine diagnostic works.
4.Experimental study of osthole on inhibition of thrombosis and platelet aggregation

Aim To investigate the effect of osthole against thrombosis and platelet aggregation.Methods Rat model of artery-vein bypass thrombosis and mouse model of thrombosis by injecting collagen-adrenaline to the vein of the tail were used to measure the thrombus weight and to observe the number of dead mice in 5 min and the recovery mice from hemiplegia in 15 min. ADP,thrombin and arachidonic acid were used to induce platelet aggregation on human and the rate of platelet aggregation in 1,3 and 5 min and the maximum rate of platelet aggregation were detected.Results Osthole inhibited artery-vein bypass thrombosis and reduced thrombus weight in rats to defferent degrees,inhibited the thrombosis induced by collagen-adrenaline and reduced the death rate in 5 min and increased the recovery rate in 15 min. Osthole also inhibited human platelet aggregation induced by ADP,thrombin and arachidonic acid in vitro.The values of IC_(50 )(dose of the drug giving 50% inhibition) were 0.444 g?L~(-1) for ADP,0.186 g?L~(-1) for thrombin, 0.421 g?L~(-1) for arachidonic acid, respectively.Conclusion Osthole exert remarkable effects against thrombosis and platelet aggregation.
5.Effects of Panax notoginseng saponins(三七总皂苷) on extra-vascular lung water and respiratory dynamics in dog with oleic acid induced acute lung injury

Objective To investigate the effects of Panax notoginseng saponins(PNS,三七总皂苷) on extra-vascular lung water(EVLW) and respiratory dynamics in dog with oleic acid induced acute lung injury(ALI).Methods Eighteen Beagle dogs,intubated and mechanically ventilated with intermittent positive pressure ventilation(IPPV) mode(tidal volume(VT) 10 ml/kg, positive end-expiratory pressure(PEEP) 0,inspiratory oxygen concentration(FiO2) 1.00),were randomly assigned into three groups(each n=6): normal control group,ALI model group(induced by intravenous injection of oleic acid) and PNS group(received PNS after the ALI model was constructed).PNS 10 mg/kg being dissolved in 100 ml 5% glucose solution(GS) was pumped into central vein (2.5 ml/min) after ALI model was formed in the PNS group.Similar amount of glucose solution was given to the normal control and model groups.Respiratory dynamics and arterial blood gas(ABG) were monitored every hour.Four hours after the establishment of ALI,the dogs were sacrificed and extra-vascular lung water index(EVLWI) was quantified by a gravimetric measurement.Results In ALI dogs,PNS significantly decreased the index of EVLWI((14.10?1.45) ml/kg vs.(17.97?0.85) ml/kg,P0.05).Conclusion PNS has certain protective effect on dog with oleic acid induced ALI,it may lower EVLW and elevate the Cst total,that is beneficial to the improvement of hypoxemia.
6.Study on the Blood Concentration of Fluorouracil Administered into Guinea Pigs by Arterial&Intravenous Infusion and Celiac Infusion

OBJECTIVE:To compare and analyze blood concentration changes of fluorouracil administered to guinea pigs by different paths.METHODS:The peripheral vein and portal vein blood samples of guinea pigs were taken at different time after administered with fluorouracil in3different ways:group A(by arterial infusion),group B(by intravenous infusion)and group C(by celiac infusion).The blood concentration changes were determined and analyzed by HPLC.RESULTS:The portal vein and peripheral vein blood concentrations in group A and B reached peak value5min after administration with flu?orouracil while group C reached peak at10min;the portal vein blood concentrations in group A reached the highest in the first20min while group C showed the highest value in the last30min.CONCLUSION:Arterial infusion combining celiac infusion path was recommended for local chemotherapy;arterial infusion combining intravenous infusion or intravenous infusion com?bining celiac infusion was recommended for the local therapy while giving consideration to systemic chemotherapy.
7.Establishment and genetic checkup of an inbred strain of DXB/c mouse

An inbred strain of DXB/c mouse has been established by hybridization between DBA/2 female mice and C57BL/6 male ones and subsequently by sibmating their offsprings beginning from the F2 generation.Now DXB/c mouse has been passed for 28 generations of full sibmating since 1979.Genetic checkup by means of skin grafting,mandibular morphology analysis,mixed lymphocyte cultivation,coat colour gene testing,and biochemical marker gene examination confirmed that the full homozygosity of alleles has been achieved in DXB/c mouse and DXB/c mouse comforms to the criteria of an inbred strain of mouse.In addition,the genetic background of DXB/c mouse is composed of the genes of its progenitors DBA/2 and C57BL/6 as shown by coat colour gene testing and biochemical marker gene examination.
8.Study on correlation between homocysteine and diabetes mellitus

Objective To explore the correlation between homocysteine(Hcy),lipids amd type 2 diabetes mellitus (T2DM). Methods The laboratory test results of 533 cases of patients with T2DM(T2DM group)and 362 cases of healthy individuals (healthy control group)were retrospectively analyzed.Results The serum levels of Hcy and rates of abnormal serum cholesterol (TC),triacylglycerol(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C)levels in the T2DM group were higher than those in the healthy control group,had statistically significant differences(P <0.05).Conclusion The high serum level of Hcy and abnormal lipid metabolism are correlated with T2DM,which might be risk factors of diabetes mel-litus and cardiovascular disease.
9.Perceptual Learning and Transfer Study of First-and Second-order Motion Direction Discrimination

In order to investigate the interactions between first- and second-order motion perception systems, 14 adult subjects with normal or correct to normal visual acuity were recruited and divided into two groups. Then these two groups were trained, in their parafovea, to discriminate the direction of first- and second-order motion gratings respectively. The spatial frequency of the gratings used in study was fixed at 2 cycles/degree and the temporal frequency was fixed at 8 Hz during training. Contrast sensitivity of subjects in these two training groups for first- and second- order motion was measured before and after 7 days' training to estimate the effects of training. In addition, the differences between the two training groups(14 subjects) and another control group (11 subjects) were studied. The results showed that: 1) the training with first-order motion gratings can improve subjects' performance in first-order motion direction discrimination but the improvement can't be transferred to the performance in second-order motion task; 2) the training with second-order motion gratings can improve subjects' performance in both first- and second-order motion direction discrimination tasks. In conclusion, an " asymmetric transfer" occurred between the training effect of first- and second-order motion gratings. These results indicate that there are two mechanisms for perceiving first- and second-order motion respectively. However, they are not completely different from each other but only partly separated.
10.Rapid identification of common clinical organism by matrix-assisted laser desorption/ionization time of-fright mass spectrometry

Objective To estimate the application for the rapid identification of common clinical bacteria by MALDI-TOF MS. MethodsFour hundred and twenty-six bacteria, including Salmonella spp strains collected from Zhejiang center for disease control and prevention were collected from blood, sputum,secretion and urine in 2nd Affiliated Hospital of Zhejiang University during December 2008 to August 2009. The isolates included 76 gram positive coccus and 350 gram negative bacilli. Species identification was performed with the Vitek system, and serotypes of Salmonella and Shigella were determined by serum agglutination test. 16s rDNA gene of 91 bacteria were amplified by PCR. The RCP products were sequenced. Then the results were compared with the reported sequences from GenBank. All strains were identified by MALDI-TOF MS. Results of three identification methods were compared with each other. Results Among 426 tested isolates, identification results from Vitek system and MALDI-TOF MS for gram positive coccus and 323 out of 350 gram negative bacilli (exception for Salmonella and Shigella spp.),were identical. For 23 Salmonella and Shigella spp. , only 2 Salmonella enterica subsp, enterica serovar Typhimurium were identified the same results by the three methods. Besides, results from Vitek system and serum agglutination test for 1 Salmonella enterica subsp, enterica serovar Typhi, 3 Salmonella enterica subsp.enterica serovar Paratyphi A, 1 Salmonella enterica subsp, enterica serovar Paratyphi B, 1 Salmonella enterica subsp, enterica serovar Enteritidis, and 1 Salmonella enterica subsp, enterica serovar Bovis-morbificans were consistent with that from 16S rDNA gene sequence. Four isolates which were confirmed as S. flexneri by Vitek system and serum agglutination test were identified as Escherichia coli by both 16S rDNA gene sequence and MALDI-TOF MS. ConclusionMALDI-TOF MS could be used for rapid and accurate identification of common clinical bacteria with good repeatabihty, excepting for the Salmonella and Shigella spp.
