1.Optimizing preparation of arsenic trioxide-loaded polylacticco-glycolic nanoparticles

Objective To study the preparation of polylactic-co-glycolic acid(PLGA) nanoparticles carrying arsenic trioxide(As_(2)O_(3)).Methods Polymer PLGA was used to prepare arsenic trioxide nanoparticles with w/o/w double-emulsion evaporation technique,which was optimized by uniform design test.Results The shape of As_(2)O_(3)-loaded PLGA nanoparticle was round,the size was well-distributed,the mean diameter was 178.2 nm,the average encapsulation ratio reached 53.19%,and the average drug loading was 0.64%.Conclusion The technique to prepare nanoparticles is simple and the quality can be easily controlled.
2.Detecting, cloning and sequencing of Mycobacterium tuberculosis IS6110 in paraffin-embedded tissues

Objective To evaluate the sensitivity and specificity of a nested polymersase chain reaction (nested PCR) for detection of the Mycobacterium tuberculosis IS6110 in paraffin embedded tissues. Methods 31 samples from tuberculosis patients and 5 biopsy specimens from patients with hepatitis were subjected to detection of Mycobacterium tuberculosis IS6110 by nested PCR. PCR products of two randomly selected samples were cloned and sequenced. Results Mycobacterium tuberculosis IS6110 was positive in 28 of 31 samples of tuberculosis. The sensitivity and specificity of nested PCR for detection of IS6110 were 90.3% and 100%, respectively. The predictive value of nested PCR was 100%. The sequences of two samples were compared with known sequence of H37Rv isolate (reported by Thierry D) and with nucleotide homology of 97% and 95.3%, respectively. Conclusions Nested PCR is sensitive and specific in the diagnosis of Mycobacterium tuberculosis infection in tissues of routinely paraffin embedded. We propose the diagnostic application of nested PCR for the identification of Mycobacterium tuberculosis infection, especially in the cases which can not be distinguished with certainty from other diseases by histopathology and Ziehl Neelsen's staining.
3.Influence of vascular activator on the expression of VEGF gene in human umbilical vein endothelial cells under hypoxia.

Objective To investigate the regulation and expression of VEGF gene in human umbilical vein endothelial cells under hypoxia and the influence of vascular activators,such as endothelin(ET),nitric oxide(NO)and NO synthesis inhibitor N-nitro-L-Arginie(L-NNA),on the expression of VEGF.Methods Culture the human umbilical vein endothelial cells and then treat by hypoxia and vascular activators.The expression on VEGF mRNA level and protein level were assayed by northern blots,ELISA and computer figure analysis system respectively.Results When human umbilical vein endothelial cells were subjected to hypoxia for 6 hours,the expression of VEGF began to increase.ET could increase the expression and NO inhibited it,and LNNA affected the expression of the VEGF.VEGF protein concentration in the cell culture media matches the mRNA result.They are 6 hours hypoxia (8^2?1.1)?g/L,ET+6 hours hypoxia(9^37?1^02)?g/L,NO+6 hours hypoxia(2^86?0^91)?g/L,LNNA+6 hours hypoxia(14^75?1^87)?g/L respectively.Conclusion (1)Hypoxia can induce the expression of VEGF in human umbilical vein endothelial cells.(2)ET can increase the inducible effect of hypoxia,NO can inhibit it and LNNA stimulated the expression of VEGF in human umbilical vein endothelial cells.
4.Correlation between coronary artery disease and levels of glycosylated hemoglobin and endothelin

Objective To analysis the correlation between coronary atherosclerosis and levels of glycated hemoglobin (HbA1c) and endothelin(ET ) ,furthermore to explore the clinical significance of the two indexes in the department of cardiology .Methods 572 inpatients were divided into four groups according to the blood glucose level and coronary angiography results :negative control group with 40 patients ,simplex DM group with 50 patients ,simplex CHD group with 262 patients ,CHD+DM group with 220 pa-tients .The HbA1c and ET levels in the four groups were monitored ,and the correlations between the two indexes and coronary ath-erosclerosis were analysed by Logistic regression analyses .Results HbA1c level :control group and CHD group < DM group
5. Relationship between of COX-2 expression and radiosensitivity of the irradiated human glioma SHG-44 cells

Objective: To investigate the changes of radiosensitivity and the expression of cyclooxygenase 2 (COX-2) in the surviving progeny from the irradiated human glioma SHG-44 cells and to provide a theoretical basis for application of COX-2 inhibitors in clinic. Methods: Radiosensitivity of mother SHG-44 cells and that of surviving progeny from the irradiated human glioma SHG-44 cells were studied by measuring the colony-forming rate. The mRNA transcription and protein expression of COX-2 were detected by RT-PCR and immunohistochemical staining, respectively. Results: The radiosensitivity was decreased and the mRNA and protein expressions of COX-2 increased in the surviving progeny of the irradiated SHG-44 cells compared with mother SHG-44 cells. The expression levels of COX-2 were negatively correlated with the radiosensitivity in the irradiated SHG-44 cells. Conclusion: Radiation induced upregulation of COX-2 expression and downregulation of the radiosensitivity in the the surviving progeny from the irradiated human glioma SHG-44 cells. The elevated expression of COX-2 may be one of the reasons for the radioresistance of irradiated SHG-44 cells.
6.Analysis of paraplegia during transarterial chemoembolization in liver cancer

Objective To evaluate the pathophysiology of paraplegia during transarterial chemoembolization in liver cancer and investigate effective management and prevention for improving clinical situation and relieve major symptoms.Method 2758 patients accepted TACE procedure because of liver cancer(and/or combined with remote metastasis),demonstrated 4 cases suffering from paraplegia(3 males,1 female)since Mar.2003 to Feb.2005,with mean age of(51?14)years old,The operative records and the clinical features after chemoembolization were summarized in detail.Results The incidence of paraplegia was 0.145%,with major symptoms of dysesthesia and hypokinesise emerging within 4 hours after TACE,and most symptoms aggravating gradually within 24-48 hours untill paraplegia appeared,and then turned to stabilization about 5-7 days,with their body functions partial recovery in 2 months.Conclusion More attention should be paied to prevent ectopic embolization of spinal cord vessels originating from extrahepatic collateral arteries during TACE or TAE.
7.Effects of vascular endothelial growth factor receptors on endothelial cells in hypoxia

Objective In order to study the important role of two specific VEGF receptors(Flt 1 and KDR) in angiogenesis, we design the experiment to detect the gene expression of Flt 1 and KDR in human umbilical vein endothelial cells cultured in hypoxium. Methods We cultured the human umbilical vein endothelial cells for different time and treated it with vascular activate factor, such as endothelin (ET), Nitric oxide (NO), LNNA (inhibitor of NO). The expressive level was analyzed by Northern blot or RT PCR. Results Hypoxia increases flt 1 mRNA in endothelial cells at 6 hours. Administration of ET stimulated an increase in flt 1 mRNA. NO inhibited expression of flt 1 mRNA. LNNA increased its expression. Hypoxia partly affected the expression of KDR. LNNA enhanced the VEGF transcripts. Conclusion Hypoxia up regulated flt 1 gene expression, partly KDR. ET enhanced and NO inhibited flt 1 gene expression.
8.Diagnostic Value of L-homocysteic Acid and Lysophosphatidylcholine as Markers of Ovarian Cancer

Objective To study whether L-cysteine and lysolecithin (16:0) could be as a serum markers in the detection of OVC for overcoming the OVC defects of early detection .Methods Totally 142 cases of healthy check -up patients ( control group ) 100 cases from First Affiliated Hospital of Wenzhou Medical University in January 2012 to January 2014 were extracted patient specimens .These pa-tient specimens were used to detect by MALDI -TOF-MS mass spectrometer and obtain the peak m/z by the method of principal compo-nent analysis for screening expression difference between the two groups in the metabolites and the correlation between metabolites and pathological grade.Results The most difference substance between two groups were 184.05 and 496.30m/z which identified as LPC (16:0) and HCA;On HCA average level and detection rate , OVC group was significantly higher than control (P<0.01); On 184.05 and 496.30m/z peak area, control group was significantly higher than OVC group (P<0.01);HCA positively was correlated with patho-logic grade (P<0.05);184.05 and 496.30 m/z peak area were negative correlation with pathologic grade (P<0.05).Conclusion L-cysteine and lysolecithin (16:0) mechanism in the pathogenesis of OVC is unclear , but it can be used for the detection of serum OVC markers.
9.Treatment of big aera burn accompanying inhalation injury

Objective To review our clinical experiences in the treatment of big area burn accompanying in-halation injury to explore the optimal preventive measures and therapy. Methods To analyze the clinical cases con-ducted of 20 such admitted the big area burn aeeornpanying inhalation injury from 2004 to 2007. Results 18 patients (90%) were cured and 2 patients(2%) died. Conclusion The curative rate of big area burn accompanied by inha-lation injury can be enhanced by combining various treatments,including early treat,early presentive tracheotomy,ap-propriate tracheobronchial lavage,and clear traeheoobronchial secretion.
10.Multifocal VEP in patients with optic nerve disease

· AIM:To determine whether the multifocal visual evoked potential (mfVEP) can be used as a clinical method to assess the patients with optic nerve disease.· METHODS: Fifteen patients with optic nerve disease were examined in this study. All patients underwent visual acuity examination, slit-lamp inspection, ophthalmoscopy, Goldmann perimeter, fundus fiuorescein angiography, visual field and mfVEP. Although these patients with unilateral optic nerve damage,data from both eyes were included in the analysis.· RESULTS:In all patients the visual fields defect was demonstrated on the mfVEP and showed good correspondence in location of the scotoma. However, we also found some slight difference between visual field and mfVEP. In some locations, when the local visual fields were normal, mfVEP showed that its amplitude reduced. In reverse, when the local mfVEP seemed normal, visual field showed abnormity.· CONCLUSION:Multifocal visual evoked potential could be used as a clinical diagnosis option in patients with optic nerve disease. Local monocular damage to the optic nerve can be measured by an interocular comparison of the mfVEP.
