1.Application of graphic method in external quality assessment of iodized salt

Objective To evaluate the detection capability for salt iodine test by using graphic method,and to provide reliable detection data for prevention of iodine deficiency disorders.Methods The examination results of external quality assessment for disease control mechanism in Liaoning Province in 2013 were analyzed by using standard Z score and Youden plot.The level of reproduction and type of error were analyzed by Z score sequence diagram in order to find the outliers and results need to be reviewed by the Youden plot.Salt iodine was detected by the general test method in salt industry for determination of iodine (GB/T 13025.7-2012).Results According to single sample evaluation of the Z score,111 laboratories were qualified,the qualified rate was 97.4% (111/114).Three labs were not qualified,unqualified rate was 2.6% (3/114).According to double sample evaluation of the Z score,112 laboratories were qualified,the qualified rate was 98.2% (112/114).Two laboratories were not qualified,unqualified rate was 1.8% (2/114).According to the Youden plot,5 laboratories were possible outliers with results need to be reviewed.Conclusion Statistical graphic method has displayed the examination results directly,which is helpful in searching targeted improvements,and improving the detection capability.
2.Clinical observation on biifco combined with triple therapy for eradication of Helicobacter pylori

Objective To investigate the clinical efifcacy of biifco combined with triple therapy for eradication of Helicobacter pylori. Methods 84 cases with Helicobacter pylori infection were selected as the observation objects, and were randomly divided into treatment group and control group, 42 cases in each group. Control group used standard triple (omeprazole, amoxicillin, clarithromycin) oral;treatment group took biifco on the basis of control group use. Hp eradication rate, gastrointestinal symptoms and the incidence of adverse reaction in two groups after treatment were compared. Results Hp eradication rate in treatment group was 92.9%, higher than 75.0%in control group, the difference was statistically signiifcant(P<0.05);the incidence of adverse reaction in treatment group was 2.4%, signiifcantly lower than 10.0%in control group, the difference was statistically signiifcant (P<0.05);the total symptom relief rate in treatment group was 90.5%, signiifcantly higher than 75.0%in control group, the difference was statistically signiifcant(P<0.05). Conclusion The eradication rate of biifco combined with triple therapy for Helicobacter pylori is high and signiifcant, which can effectively reduce adverse gastrointestinal reactions.

Objective To investigate the effect of radix paeoniae rubra (RPR) on expression of heme oxygenase (HO-1) and inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-induced acute lung injury (ALI) and explore its protective mechanism. Methods Forty Wistar rats were randomly and equally divided into five groups, ie, control group, LPS group, RPR treatment group, RPR prevention group and Hemin group. Arterial blood was drawn for blood gas analysis. Models of endotoxin-induced ALI were used to observe the protein content, the ratio of neutrophiles in bronchoalveolar lavage fluid (BALF), the malondialdehyde (MDA) content in lung and the activities of serum NO. 4.Mechanism of protective effect of sodium nitroprusside against endotoxin- induced acute lung injury in rats

Objective To investigate the effect of sodium nitroprusside (SNP) , an exogenous NO donor, on expression of hemeoxygenase-1 (HO-1) and inducible NO synthase (iNOS) in the lung tissue in a rat model of lpopolysaccharide (LPS)-induced acute lung injury and the underlying mechanism.Methods Thirty-two Wistar rats of either sex weighing 190-210 g were randomly divided into 4 groups ( n = 8 each) : Ⅰ sham operation (S) ;Ⅱ LPS;Ⅲ hemin + LPS and Ⅳ SNP + LPS. The animals were anesthetized with intraperitoneal (IP) 7% chloralhydrate 5 ml?kg-1. LPS 750 ?g?kg-1 in 300 ?l was instilled into the lungs via trachea in group Ⅱ . In group Ⅲ hemin (a HO-1 inducer) 40 ?mol?kg-1 was injected IP 12 h before LPS. In group Ⅳ SNP 25 ?g?kg-1 + LPS 750 ?g?kg-1 in 300 ?l was instilled into the lungs via trachea. In group S normal saline 300 ?l was instilled via trachea instead of LPS. The animals were killed by (1) exsanguination from carotid artery 8h (2) after LPS instillation and lungs were removed for broncho-alveolar lavage (BAL) and detennination of protein concentration in BALF and the expression of HO-1 and iNOS protein in the lung tissue by immune-histochemical technique and HPIAS-2000 image analysis system and MDA content in lung tissue and (3) light microscopic examination (4) wet/ dry lung weight ratio.Results The 4 groups were comparable with respect to body weight and sex (M/F ratio) . Objective To obtain sufficient recombinant multiepitope peptide of ESAT-6.Methods DNA encoding ESAT-6 peptide was amplified by PCR using a pair of primers which were designed in accordance with the reported ESAT-6 gene sequence.The ESAT-6P gene was inserted into the prokaryotic expression vector pET-30a with the N-terminal 6His-tag.The recombinant peptide vector pET30a-ESAT-6P was transformed into E.coli BL21(DE3) via chemical transformation.The positive clone was screened by means of PCR.The target peptide was expressed in E.coli after induction with IPTG and analyzed by SDS-PAGE.The immunogenicity of the peptide was evaluated by dot immunobinding assay.Results The target peptide was successfully expressed and purified.The solubility analysis showed that the recombinant peptide existed as inclusion body in the E.coli.DIBA indicated that the ESAT-6P was specifically reactive to sera from TB patients.Conclusions The recombinant multiepitope peptide of ESAT-6 engineering bacteria has been obtained,which will be quite helpful to develop novel specific diagnostic reagents and effective vaccine.
7.Progress in mesenchymal stem cells as a novel nano-targeted antitumor drug carrier

In recent years,targeted drug delivery system as one of the hottest areas in pharmaceutical research has received more and more attention.However,the targeted anti-tumor drug delivery system has many shortcomings including low targeted efficiency which limit its clinical application.Elevating tumor targeting efficiency has become the bottleneck of nano-drug development and clinical application.Therefore,it is urgent to design and develop novel targeted drug delivery vehicle.Recent studies showed that mesenchymal stem cells (MSCs) have characters of tumor tropism and migration,which make them as ideal targeted anti-tumor drug delivery carriers for the treatment of solid and metastatic tumors.This paper reviews the progress in using MSCs as targeted anti-tumor drug delivery carrier,recapitulates the problems and challenges of the system,and proposes a solution for these problems.
8.N-acetylcysteine attenuates myocardial hypertrophy in streptozotocin-induced diabetic rats

Objective To investigate the protective effect of N-acetylcysteine(NAC) on the development of myocardial hypertrophy in diabetic rats induced by streptozotocin. Methods Male SD rats were randomly divided into normal control (NC), control treatment (NCT), diabetic (DM) and diabetes treatment (DMT) groups. NAC was administered at dose of 1.4~1.5g·kg~(-1)·d~(-1) to NCT and DMT groups in the drinking water for 8 weeks. At termination the rats were surgically prepared for hemodynamic measurement, such as systolic blood pressure, heart rate, the rate of left ventricle relaxation (-dp/dt) and the time to maximum relaxation (T). Subsequently, the hearts were removed to assay relative ratio of left ventricle weight versus body weight (LVW/BW) and cardiomyocyte cross sectional areas.Plasma glucose, insulin, 15-F_(2t)-isoprostane(15-F_(2t)-Isop) concentration and superoxide dismutase (SOD) activity were analyzed. Results The LVW/BW ratio and cardiomyocyte areas were significantly attenuated by NAC treatment as compared to DM group (P< 0.05). Increases in -dp/dt and SOD activity and the decreases in glucose, 15-F_(2t)-Isop T (P< 0.05) were found at the same time. Conclusions NAC can effectively prevent the development of myocardial hypertrophy and improve diastolic function in diabetic rats, which may be related to its antioxidant capacity.
9.Analysis of the monitoring results on iodine deficiency disorders in Liaoning Province in 2011

Objective To understand the condition of iodine deficiency disorders,and the iodine nutritional status of population before adjustment of iodine level in edible salt in Liaoning Province.Methods Thirty countries (cites,districts) which were divided into coastal,inland,city,rural areas according to location and population characteristics were sampled by population probability sampling method in the whole province; one township (town,street office) was sampled from each country (city,district); one village (neighborhood) and one school were chosen from each township(town,street office).Forty children aged 8-10 were selected from each school to measure their thyroid volumes and household salt samples were collected to detect their iodine content; fourteen of those 40 children were selected to detect their urinary iodine content.In each village (neighborhood),five drinking water samples were collected in the east,the west,the south,the north and the center positions.Two tap water samples were collected to detect their iodine content if the water supply was centralized.Around each school; three townships were selected,in each township,random urine samples were collected from 5 pregnant women and 5 lactating women to detect their iodine content.Thyroid volume was examined by ultrasound method; the salt iodine was tested by the method of direct titration; iodine content of urine and drinking water was tested by arsenic cerium catalytic spectrophotometry.Results Totally one thousand two hundred and nineteen children aged 8-10 were examined; twenty-nine children were diagnosed goiter,and the goiter rate was 2.4%(national standards:< 5%).One thousand two hundred and nineteen edible salt samples were tested,and the median salt iodine level was 30.1 mg/kg.The iodized salt coverage rate was 99.3%(1 211/1 219),and the consumption rate of qualified iodized salt was 97.9%(1 194/1 219).Four hundred and eighty urine samples were tested,and the median urinary iodine level was 189.0 μg/L(suitable content of salt iodine was 100-199 μg/L) ; urinary iodine < 20 μg/L accounted for 0.6%(3/480).Ninety-nine drinking water samples were selected,and the average iodine content of the drinking water was (5.9 ± 5.7)μg/L.Four hundred and fifty-one urine samples of pregnant women and four hundred and fifty urine samples of lactating women were selected,and the median urinary iodine level of pregnant women and lactating women was 163.2,151.0 μg/L(suitable contents of urinary iodine in pregnant women and lactating women were 150-249 μg/L,> 100 μg/L).The median urinary iodine of pregnant women was 135.4 μg/L in coastal city,138.0 μg/L in coastal rural,168.0 μg/L in inland city,171.1 μg/L in inland rural.The difference of urinary iodine between coastal region and inland region was significant(H =14.287 6,P < 0.05).Conclusions The iodine nutrition conditions of pregnant women,lactating women and children are adequate in Liaoning Province,but the iodine nutritional status is insufficient in pregnant women from the coastal areas.
