Objective:To predict the gastrointestinal side effects of chemotherapeutic drugs using healthy murine colon organoids. It aimed to identify safer alternative treatments for patients intolerant to certain chemotherapy regimens and demonstrate the potential clinical ap-plications of organoids in predicting gastrointestinal side effects. Methods:Healthy mouse colonic crypt cells were cultured in 3D.Paraffin sections of colon tissues and organoids were subsequently prepared, followed by haematoxylin and eosinand immunohistochemical staining(CDX2,Ki67,and CK19).The colonic organoids were treated with five chemotherapeutic drugs, and cell activity was assessed to determine their intestinal toxicity. The consistency of the incidence of gastrointestinal side effects observed in this study and in clinical practice were analyzed by comparing the results to the published literature. Results:The histological characteristics of the colon organoids were highly consistent with those of the original colon tissues. The tolerance of normal colon organoids to different chemotherapeutic drugs was vari-able.Capecitabine had the least cytotoxic effect on mouse colon organoids, whereas paclitaxel liposomes showed the strongest cytotoxic ef-fect when IC50 was the only consideration.Considering clinical drug concentrations,a significant difference was observed in the organoid in-hibition rates between albumin paclitaxel and liposomal paclitaxel.Statistical analysis of clinical trial data showed that the incidence of gradeⅢ/Ⅳ diarrhea caused by albumin paclitaxel, epirubicin, capecitabine, and cyclophosphamide was consistent with the corresponding or-ganoid inhibition rates. Conclusions:Combining clinical drug doses,we recommend prioritizing albumin paclitaxel and avoiding the use of liposomal paclitaxel to improve chemotherapy tolerance. This study demonstrates that normal colon organoids can effectively predict the occurrence of severe diarrhea associated with most chemotherapeutic drugs.

OBJECTIVETo study the gene mutation and streptomycin, isoniazid or rifampicin resistance of Mycobacterium isolated from silico-tuberculosis patient's sputum so as to find a more effective therapy for this disease.
METHODSMycobacteria tuberculosis were separated from 96 coal worker with silico-tuberculosis firstly. Then rpsL, KatG and rpoB fragments of genome were copied with PCR and compared their SSCP profiles with standard strains.
RESULTS67 strains of streptomycin, isoniazid or rifampicin resistant Mycobacteria tuberculosis were found in routine drug resistance test, with the percentages of 80.5% (54/67), 58.2% (39/67) respectively. PCR-SSCP showed that out of 67 drug-resistant strains, 66(98.5%) of rpsL, 47(70.1%) of rpoB and 42(62.7%) of KatG appeared abnormal.
CONCLUSIONMost of the resistant strains appeared gene mutation. The mution rates were higher than the results from routine drug resistance test.

The standard treatment for locally advanced rectal cancer is neoadjuvant chemoradiotherapy(nCRT) followed by surgery. The therapeutic efficacy of patients after nCRT differs greatly. Effective use of nCRT can accurately predict the efficacy and help patients avoid damage caused by excessive treatment. This article describes the main methods of current nCRT and newly proposed concepts, such as totally neoadjuvant therapy, summarzies its impact on the efficacy of locally advanced rectal cancer, introduces the potential predictive biomarkers of efficacy evaluation for nCRT and the latest advances in clinical, histological and molecular predictors, and discusses the potential value of efficacy prediction in nCRT .

Results:Compared with group C, the survival rate was significantly decreased, apoptosis rate was increased, the MDA level and amount of LDH released were increased, SOD levels were decreased, the expression of LncRNA-MALAT1, BNIP3 mRNA, Bax, and cleaved-caspase-3 was up-regulated, and miRNA-145 and Bcl-2 expression was down-regulated in the other four groups ( P<0.05). Compared with group H/R, the cell survival rate was significantly increased, apoptosis rate was decreased, the MDA level and amount of LDH released were decreased, SOD levels were increased, the expression of LncRNA-MALAT1, BNIP3 mRNA, Bax, and cleaved-caspase-3 was down-regulated, and miRNA-145 and Bcl-2 expression was up-regulated in S and pcDNA groups ( P<0.05). Compared with group S, the survival rate was significantly decreased, apoptosis rate was increased, MDA level and amount of LDH released were increased, SOD levels were decreased, the expression of LncRNA-MALAT1, BNIP3 mRNA, Bax, and cleaved-caspase-3 was up-regulated, and miRNA-145 and Bcl-2 expression was down-regulated in group MALAT1 ( P<0.05). Conclusion:The mechanism of sufentanil preconditioning-induced cardioprotection is related to inhibiting Lnc-MALAT1/miRNA-145/BNIP3 signaling pathway in rats.

Mitochondrial autophagy is a process to clear dysfunctional mitochondria in the cytoplasm to maintain the integrity of mitochondrial function and cell homeostasis. Mitochondrial autophagy is a complex physiological process, which can maintain the balance of mitochondrial quality and quantity, cell survival under starvation and harsh conditions, and the stability of the intracellular environment. Its molecular mechanism involves a variety of proteins. Many factors can induce mitochondrial autophagy, such as starvation, oxidative stress, hypoxia, depolarization, and other stresses. The accumulation of unfolded proteins can also induce mitochondrial autophagy. In recent years, as a research hotspot, the abnormality of mitochondrial morphology and function is closely related to the occurrence of a variety of diseases. The research on mitochondrial autophagy and the pathogenesis of clinical diseases has attracted more attention, such as tumors, cardiovascular diseases, liver diseases, nervous system diseases, and glucose metabolism disorders. It has been found that regulating mitochondrial autophagy may inspire the treatment of some diseases. Meanwhile, clinical researchers have paid more attention to traditional Chinese medicine (TCM). As revealed by in-depth research, Chinese medicine has a certain value in regulating mitochondrial autophagy. The research on the pathogenesis of mitochondrial autophagy in related diseases and the intervention of Chinese medicine has found that there are many reports on the regulation of mitochondrial autophagy by Chinese medicine in tumors, cardiovascular diseases, and nervous system diseases. However, the mechanism of mitochondrial autophagy, the balance of mitochondrial autophagy, and the difference in the activation or inhibition of mitochondrial autophagy by Chinese medicine remain unclear. The regulation of mitochondrial autophagy has become a new research target strategy of Chinese medicine in the prevention and treatment of diseases. This paper reviewed the available literature in recent years to provide reference materials for the regulation of mitochondrial autophagy by Chinese medicine and ideas for the follow-up research of Chinese medicine in mitochondrial autophagy.
