1.A mouse model of congenital human cytomegalovirus infection that induces liver damage in fetus

Objective To define that human cytomegalovirus (HCMV) can cross the placenta of the Balb/c mice and induce liver damage in developing fetus. Methods HCMV AD169 (6.0 logTCID 50 , 3.0 logTCID 50 , 1.5 logTCID 50 per mouse respectively) was injected into the peritoneum of mice (half of mice were female) when they were about 10 weeks old (weight 25 30g). Then, these mice were paired to mate. Fetus on day about to give birth was removed from the uterus and liver was obtained for virus isolation, pathological studies, examination of the viral DNA positive cells by in situ hybridization using digoxigenin labelled HCMV DNA oligonucleotide probe. Results HCMV could be isolated from the supernatant of tissues; HCMV DNA was found by PCR in supernatant of cell culture with CPE; the presence of viral DNA sequence in hepatocytes was confirmed by in situ hybridization; pathological changes in liver consist of swollen cytoplasm and destroyed nuclei of hepatocytes and distinct intranuclear inclusion in hepatocytes with a cellular infiltrates of predominantly phagocytic cells. All above were found more obviously in fetal mouse liver tissues from the group inoculated with 6.0 log TCID 50 HCMV AD169 as compared with 3.0 log TCID 50 group. In contrast, these positive results couldn't be well found in 1.5 log TCID 50 group. Nothing could be found in normal controls. Conclusions Our research suggests that primary maternal HCMV infection during pregnancy could induce congenital infection in fetus by transplacental transmission and induce fetal liver damage. The mouse model will provide the basis for the study on pathogenesis of congenital HCMV infection in liver and the development of prevention, diagnosis and antiviral agents for congenital HCMV infection in human being.
2.The study effects of the material of buildup cap to the measured value of output factor

To study the effects of the material of a buildup cap to the determined value of Output FactorSc.Material and MethodsUsing two buildup caps made of plastic A-150 and copper respectively.The ionizing values of ionization chamber were measured with Farmer 2570 electrometer in the fields of all kinds of sizes respectively and calibrated to reference field.Two groups of values of Sc were obtained and compared. Results The ionizing values measured with the buildup cap of copper is evidently larger than with the buildup cap of plastic A-150 and the maximum is 14% but the difference between two groups of the values of Sc is not much evident and all is in the range of 1%. Conclusions Though the material of high density is not equivalent with airthe effects of that to the measured value of Sc is less.So using the buildup cap made of the material of high density for the measurement of Sc is also a quite good method specially in the fields of the small sizes.
3.Clinical characteristics and prognosis of invasive lobular carcinoma of breast: a report 125 cases

Objective To analyze clinicopathologic characteristics and prognostic factors of the patients with invasive lobular carcinoma of breast. Methods Clinical data of 125 patients with invasive lobular carcinoma of breast treated at Cancer Center of Sun Yat-sen University from Jan. 1990 to Dec. 2008, were analyzed. The clinical characteristics, recurrence and survival of the patients were summarized. Results Median age of 125 patients was 45 years old (range, 27 to 76 years old). The patients with large tumor mass (≥ 3cm), positive local lymph node, more than Ⅱ stage and positive hormone receptor at diagnosis were 77 cases(61.6 %), 64 cases(51.2 %), 101 cases(80.8 %) and 112 cases(89.6 %), respectively. The median time of follow-up was 58 months (range, 11-222 months). Of the 125 patients, 32 had local recurrence and metastasis, and 18 died. The 5-year disease-free and overall survival rates were 82.2 % and 87.3 %, respectively. Multivariate COX regression analysis showed that whether endocrine therapy or not was only a prognostic factor of patients with invasive lobular carcinoma of breast. Conclusion There is no difference in media age of patients with invasive lobular carcinoma of breast at diagnosis from other pathologic type of breast cancer. These patients are usually with larger tumor masses, more lymph node metastasis and a higher proportion of positive hormone receptor. The prognosis of patients is not affected by clinicopathologic parameters.
4.pORF5 plasmid protein of Chlamydia trachomatis induces IL-1βand IL-18 production through NALP3 inflammasome activation

Objective:To investigate whether pORF5 plasmid protein of Chlamydia trachomatis(Ct) induces 1L-1βand 1L-18 production in THP-1 cells,and its potential molecular mechanism.Methods:pORF5 plasmid protein was used to stimulate THP-1 cells at different concentrations(0,3,6,12,24,36 μg/ml),then the inflammatory cytokines IL-18 and IL-1βwere detected by ELISA at the time of 0,8,16,24,36 h;The mRNA expression of NALP3 inflammasome were detected by Realtime-PCR,and Caspase-1 activity was determined by Western blot analysis.THP-1 cells were transfected with siRNA targeting NALP3 and ASC gene for 24 h or pretreated with Caspase-1 inhibitor(Z-YVAD-FMK) for 30 min,and subsequently stimulated with pORF5(24 μg/ml) for 24 h,then secretion of IL-1βand IL-18 were analyzed by ELISA.Results: The pORF5 plasmid protein induced THP-1 cells to secrete IL-1βand IL-18 by dose-and time-dependent manners,production of IL-1βand IL-18 reached their peaks(491 pg/ml and 186 pg/ml) at concentration of 24 μg/ml,and the peak amount of IL-1βand IL-18 occurred at 24 h and 16 h post-stimulation respectively.pORF5 plasmid protein in-creased mRNA expression of NALP3 inflammasome and activated Caspase-1 in THP-1 cells.NALP3 siRNA,ASC siRNA and Z-YVAD-FMK reduced pORF5-induced IL-1βand IL-18 production when compared with control groups(P<0.05).Conclusion:pORF5 plasmid protein could induce THP-1 cells to produce IL-1βand IL-18 through NALP3 inflammasome activation,which may play an important role in the pathogenesis in Ct infection.
5.Predictive value of NT-proBNP combined Lp (a) detection for coronary in-stent restenosis

Objective: To explore the relationship among levels of N terminal pro brain natriuretic peptide (NT-proBNP), lipoprotein (a) [Lp (a)] and in-stent restenosis (ISR) in patients after stent implantation. Methods: A total of 625 pa-tients, who received drug-eluting stent implantation in our department and underwent coronary angiography one year after surgery, were continuously collected. According to presence of ISR or not, patients were divided into no restenosis group (n=532) and ISR group (n=93). Plasma levels of NT-proBNP and Lp (a) were measured and compared between two groups. Single-factor and multi-factor Logistic regression analysis were used to analyze the relationship among levels of NTproBNP, Lp (a) and coronary ISR. Results: Compared with no restenosis group, there were significant rise in plasma levels of NT-proBNP [(749. 43±154. 82) ng/L vs. (856. 12±132. 45) ng/L]and Lp (a) [(299. 23±29. 07) mg/L vs. (321. 12±31. 05) mg/L]in ISR group, P=0. 001 both; multi-factor Logistic analysis indicated that plasma levels of NTproBNP (OR=2. 162, 95%CI 2. 002~2. 333) and Lp (a) (OR=2. 903, 95%CI 2. 648~3. 184) were independent risk factors for ISR, P<0. 05 both. Conclusion: N terminal pro brain natriuretic peptide and lipoprotein (a) are independent risk factors for in-stent restenosis in patients undergoing stent implantation.
6.Exploration of training mode for postgraduate education in local universities under the Background of "double first-class" construction

First-class education for postgraduate is the foundation for the construction of "double first-class" education. Universities play an important role in postgraduate education. This paper explored the measures for the reform and innovation of the construction of "double first-class" education for postgraduate students in our university, which include perfecting the supervisor's responsibility and authority mechanism, deepening the reform on curriculum system, strengthening the construction of sharing platform, and improving the evaluation mechanism of training quality and so on. In conclusion, initial achievement from the reform and innovation of training mode was observed, which provides a useful reference for the construction of "double first-class" education for local universities.
7.Effect of mentalization-based family therapy on depressive symptoms and non-suicidal self-injury behavior in adolescents with major depressive disorder

BackgroundThe major depressive disorder has high prevalence among adolescents, and non-suicidal self-injury (NSSI) behaviors frequently occur among patients, therefore, major depressive disorder in adolescents has become the researching focus. ObjectiveTo explore the effect of mentalization-based family therapy (MBFT) on depressive symptoms and NSSI behavior in adolescents with major depressive disorder, and to provide references for the rehabilitation of major depressive disorder in adolescents. MethodsA total of 90 adolescent patients with major depression disorder who met the diagnostic criteria of International Classification of Diseases, 10th edition (ICD-10) for depressive disorders and attended Wuhan Mental Health Center from January to December 2022 were selected, and were assigned into study group (n=44) and control group (n=46) using random number table method. All participants received routine intervention, based on this, study group added a 60-minute MBFT intervention once a week for 8 weeks. Before the intervention and at the end of 1st, 2nd,4th and 8th week,the two groups were assessed using Hamilton Depression Scale-24 item (HAMD-24), General Self-Efficacy Scale (GSES), Pittsburgh Sleep Quality Index (PSQI) and Ottawa Self-injury Inventory (OSI). ResultsThe repeated measures analysis of variance reported a statistical main effect of time, main effect of group, and interaction effect between time and group at the baseline and the end of 1st, 2nd, 4th and 8th week of treatment in HAMD-24 score (F=69.621, 15.428, 29.623, P˂0.05), OSI score (F=176.642, 37.682, 21.873, P˂0.05), GSES score (F=215.236, 57.421, 27.857, P˂0.05) and PSQI score (F=268.541, 61.863, 33.867, P˂0.05). Individual effect analysis discovered a statistical difference between study group and control group at the end of 2nd, 4th and 8th week of treatment in HAMD-24 score (t=5.567, 8.645, 6.233, P˂0.01), OSI score (t=3.675, 11.817, 9.632, P˂0.01), GSES score (t=23.462, 31.709, 12.750, P˂0.01) and PSQI score (t=9.664, 22.457, 9.333, P˂0.01). ConclusionMBFT may improve depressive symptoms, NSSI behavior, sleep quality and self-efficacy in adolescents with major depressive disorder. 8.Effects of Zigui yichong formula on premature ovarian insufficiency in mice through glycolytic metabolic pathway

OBJECTIVE To study the effects of Zigui yichong formula on premature ovarian insufficiency (POI) in mice through glycolysis metabolic pathway. METHODS Eighty SPF C57BL/6N female mice were divided into normal group, model group, Zigui yichong formula group (14.175 g/kg), Zigui yichong formula+2-deoxy-D-arabino-hexose (2-DG) group (Zigui yichong formula 14.175 g/kg + glycolysis inhibitor 2-DG 100 mg/kg), with 20 mice in each group. Except for the normal group, POI model mice were induced by intraperitoneal administration of cyclophosphamide in the other groups. After the model was successfully established, each group was given corresponding drugs. HE staining was employed to observe the pathomorphological changes in ovarian tissue and to count follicles at all developmental stages; radioimmunoassay was conducted to measure the serum levels of estradiol (E2), anti-Müllerian hormone (AMH), and follicle-stimulating hormone (FSH); TUNEL assay was employed to detect apoptosis in ovarian granulosa cells of mice; the activities of hexokinase (HK), pyruvate kinase (PK) and lactate dehydrogenase (LDH) were detected by colorimetry; Western blot and real-time fluorescence quantitative PCR were employed to analyze the protein and mRNA expressions of B-cell lymphoma-2 (Bcl-2), Bcl-2 associated X protein (Bax), caspase-3, HK2, pyruvate kinase M2 (PKM2), and lactate dehydrogenase A (LDHA). RESULTS Compared with model group, the number of primordial follicles, growing follicles, antral follicles and granulosa cells were increased significantly(P<0.05), and granulosa cells arranged neatly, but the number of atretic follicles and granulosa cells apoptosis were decreased significantly in Zigui yichong formula group (P<0.05); the serum levels of E2 and AMH, the activities of HK, PK and LDH, protein and mRNA expressions of Bcl-2, HK2, PKM2 and LDHA were increased significantly (P<0.05); the serum levels of FSH, the protein and mRNA expressions of Bax and caspase-3, Bax/Bcl-2 ratio were decreased significantly (P<0.05). 2-DG could reverse the improvement effects of Zigui yichong formula on the above indexes of POI model mice. CONCLUSIONS Zigui yichong formula may inhibit the apoptosis of ovarian granulosa cells, reduce follicle atresia and improve ovarian reserve function by promoting glycolysis levels in POI model mice.
