【Objective】 To study the location effect of the Mc Ab G9 to human esophageal carcinoma in tumor bearing nude mice. 【Methods】 125 I-G9 was prepared by Chloramine-T method. The distribution of 125 I-G9 was detected at different time (24, 48, and 72 h) after peritoneal injecti on. The percentage of the injected dose per gram of tissue(%ID/g) and the ratio of Tumor/non-Tumor were calculated. 【Results】 The distribution of 125 I -G9 in tumor at the third day was showed by autoradiography obviously higher th an in other organ/tissue (except blood) and the highest is at the 48 h. The T/NT values are 2-7. The autoradiography indicated that radioactivity concentrated in tumor tissue. The concentration in tumor edge is more obvious than in the ce nter. 【Conclusion】 125 I-G9 has a considerable targeting activity and can well locate in esophageal carcinoma tissue in tumor bearing nude mice.
3.Primary study on the method for separating mature sperms from human testicular tissue.

This study was aimed to explore a simple and applicable method of separating mature sperms from human testicular tissue for intracytoplasmic sperm injection and embryo transfer. The suspension of human testicular tissue was cultured in 10% human serum albumin and human tubule fluid with different concentrations (0 u/ml; 50 u/ml; 100 u/ml; 150 u/ml; 200 u/ml) of hyaluronidase for 24 h, and then the Percoll gradient centrifugation was processed to separate the sperms; meanwhile the sperms were counted and graded according to their motility. The difference in quality and quantity among the groups and the difference between the groups and the zero-hour culturing group were detected. It was shown that the four hyaluronidase-treated groups contained large quantity and high quality of sperms as compared with the two contrast groups (P<0.01). The groups in the solution of 50 u/ml, 100 u/ml and 150 u/ml concentrations of hyaluronidase had almost the same amount of sperms that displayed higher motility as compared against the sperms in the group treated with 200 u/ml concentration of hyaluronidase (P<0.01). There was no difference between the two contrast groups (P>0.05), or among the groups treated with 50 u/ml, 100 u/ml, and 150 u/ml of hyaluronidase concentration (P>0.05). This method of adopting hyaluronidase with Percoll gradient centrifugation in the process for separating mature sperms from human testicular tissue is applicable. It can increase the quantity and quality of sperms separated from testicular tissue suspensions when adequate concentrations of hyaluronidase is used.

4. Effect of apicidin on glioblastoma U87 cells and its regulation of OCT-4 gene expression

Objective:
To investigate the effect of histone deacetylase inhibitor apicidin on the glioblastoma U87 cells and its regulation of OCT-4 gene expression.
Methods:
Glioblastoma U87 cells were treated with different concentrations of apicidin, and dimethyl sulfoxide instead of apicidin was negative control. Methyl thiazolyl tetrazolium (MTT) assay was used to detect the proliferative ability of U87 cells treated by apicidin. The cell apoptosis was observed under the fluorescence microscope, and the cell cycle was detected by using flow cytometry. Reverse transcription-polymerase chain reaction and Western blot was used to detect the expression of mRNA and protein of U87 cells, respectively relative to the expression of GAPDH.
Results:
MTT assay results showed that apicidin inhibited U87 cells proliferation in a dose-dependent and time-dependent manner, and half of the inhibitory concentration of cell proliferation at 48 h was (1.74±0.13) μmol/L. The cell proportion of U87 cells in S-phase of the negative control, 0.2, 0.5, and 1.0 μmol/L apicidin was (32.68±0.49)%, (33.73±0.76)%, (42.92±0.56)%, and (56.95±0.53)%, respectively after 48 h apicidin administration (
5.Value of Doppler ultrasound detection of blood flow parameters in venous catheter and middle cerebral artery for predicting fetal distress

Objective To investigate the value of Doppler ultrasound detection of blood flow parameters in venous catheter and middle cerebral artery for the prediction of fetal distress. Methods All 102 pregnant women who received antenatal examination in our hospital from March 2013 to January 2014 received Doppler ultrasound detection and were divided into the normal group and the fetal distress group according to the fetal status. Results The two groups had sta tistically significant differences in the fetal DV parameters(QDV/UV and PLI),MCA parameters(S/D and RI)and Apgar score(P<0.05). Conclusion Through detecting the indicators of venous catheter and fetal cerebral artery, Doppler ultrasound can reflect the fetal hypoxia situation and enables more accurate understanding of fetal distress, thus re-duces neonatal morbidity and mortality.
6.Construction of content framework for transcultural nursing health education in hypertension patients

Objective To construct the items framework for transcultural nursing health education in hypertension patients .Methods Twenty-one experts received the two rounds of questionnaire consultation on the items framework for transcultural nursing health education in hypertension patients through Delphi method . The items framework for transcultural nursing health education in hypertension patients was preliminarily constructed by getting experts ’ conforming opinion .Results The items framework for transcultural nursing health education in hypertension patients preliminarily constructed included 10 level one entries , 34 level two entries and 101 level three entries, and the authority coefficient reached 0.910, and the judgment coefficient was 0.924, and the familiarity coefficient was 0.895.The effective response rate of questionnaires in the two rounds of expert consultation was more than 95%.Conclusions The items framework for health education in hypertension patients constructed under the guidance of transcultural nursing theory has good practicability and maneuverability , and provides the reference for the implementation of individualized health education .
7.Correlation between interleukin-17 and coronary atherosclerotic plaque load in type 2 diabetes mellitus patients and its predictive value for adverse cardiovascular events

Objective:To investigate the correlation between interleukin-17(IL-17) and non-calcified coronary plaque load in patients with type 2 diabetes mellitus.Methods:From January 2022 to November 2022, 150 patients with type 2 diabetes mellitus in the Fifth Affiliated Hospital of Xinjiang Medical University were retrospectively collected as the observation group, and 150 healthy adults were collected as the healthy control group at the same time. The levels of IL-17 and coronary non-calcified plaque load between the two groups were compared. The correlation between serum IL-17 level and non-calcified plaque load was analyzed by Pearson test. The patients with type 2 diabetes mellitus were followed up for 1 year, the predictive value of IL-17 and coronary non-plaque load on adverse cardiovascular events in type 2 diabetes patients were analyzed by receiver operating characteristic (ROC) curve.Results:The levels of IL-17, fasting blood glucose, body mass index, non-calcified plaque volume, non-calcified plaque load and carotid intima-media thickness in the observation group were higher than those in thehealthy control group: (60.34 ± 15.94) ng/L vs. (36.30 ± 14.29) ng/L, (9.38 ± 1.47) mmol/L vs. (5.98 ± 0.61) mmol/L, (26.85 ± 3.42) kg/m 2 vs. (24.84 ± 3.00) kg/m 2, (165.20 ± 27.37) mm 3 vs. (107.33 ± 36.72) mm 3, (27.82 ± 4.68)% vs. (17.98 ± 6.17)%, (1.40 ± 0.16) mm vs. (1.22 ± 0.16) mm; the left ventricular ejection fraction was lower than that in the normal control group: (56.58 ± 7.73)% vs. (67.07 ± 7.95)%, there were statistical differences ( P<0.05). The results of Pearson test showed that the level of IL-17 were significantly positively correlated with non-calcified plaque volume, non-calcified plaque load and carotid intima-media thickness in type 2 diabetes mellitus patients ( r = 0.161, 0.180 and 0.178, P<0.05). The results of ROC curve analysis showed that the level of IL-17, non-calcified plaque load, and non-calcified plaque volume had high predictive value for adverse cardiovascular events in type 2 diabetes mellitus patients within 1 year, the area under the curve were 0.735(95% CI 0.637 - 0.832, P = 0.001), 0.766 (95% CI 0.636 - 0.896, P<0.001), 0.774(95% CI 0.648 - 0.901, P<0.001). Conclusions:The increase of IL-17 in type 2 diabetes mellitus patients is related to the increase of non-calcified plaque load of coronary arteries, and has a high value in predicting adverse cardiovascular events.
8.A comparison study of prevalence,awareness,treatment and control rates of hypertension and associated factors among adults in China and the United States based on national survey data

Objective:This study compared the prevalence,awareness,treatment and control of hypertension and associated factors in China and the United States(US).Methods:Adult data from nationally representative samples were derived from the Chronic Disease and Risk Factors Surveillance in 2010 and 2013 in China and the National Health and Nutrition Examination Survey in 2010 and 2013 in the US.Multivariable logistic and Poisson regression analysis were conducted to assess associations of the four outcomes with body weight status and behavioral factors.Results:Age-standardized prevalence rates of hypertension was 35.7%(95%confidence interval[CI]:35.4%to 36.1%)in 2010 and 29.8%(95%CI:29.4%to 30.2%)in 2013 in China,and 35.3%(95%CI:33.6%to 37.1%)in 2010 and 37.9%(95%CI:36.0%to 39.7%)in 2013 in the US.Among hypertensive participants,the age-standardized rates of treatment were 18.4%(95%CI:17.9%to 18.9%)in 2010 and 23.8%(95%CI:23.1%to 24.6%)in 2013 in China and 54.5%(95%CI:50.3%to 58.7%)in 2010 and 50.9%(95%CI:46.5%to 55.3%)in 2013 in the US;the age-standardized hypertension control rates were 3.2%(95%CI:3.0%to 3.5%)and 5.7%(95%CI:5.3%to 6.0%)in 2010 and 2013 in China and 50.6%(95%CI:46.2%to 55.0%)and 55.3%(95%CI:50.3%to 60.3%)in the US.Obesity was significantly associated with prevalence,awareness and control rates in both countries.Different from the US,obesity was negatively associated with hypertension control in China.Conclusion:Hypertension prevalence in China is similar to that in the US,but the control rate in China was significantly lower.Obesity was a critical risk factor for poor hypertension control in China.
