1.Changes of plasma endothelin, thromboxame B2and 6-keto-PGF1αin patients with coronary heart disease after PTCA

Objective To explore the regularity of changes of plasma endothelin (ET), throm-boxame B2(TXB2) and 6-keto-prostaglandin F1α(6-keto-PGF1α) and restenosis after percutaneous transluminal angioplasty (PTCA) in patients with coronary heart disease. Methods Radioimmunoas-say was applied to measuring plasma levels of ET, TXB2and 6-keto-PGF1αat 0,30 min, 1 day and 3 days after PTCA in 41 patients with coronary heart disease. Results The level of ET in the patients with coronary heart disease was significantly decreased in 30 minutes after PTCA (P<0.05), but no significant difference was observed in 1 day and 3 days after PTCA (P>0.05). The level of plasma TXB2has no statistical difference after PTCA in 30minutes, 1 day and 3 days (P>0.05). The level of 6-keto-prostaglandin F1α(6-keto-PGF1α) of the patients with coronary heart disease was significantly declined in 30 minutes after PTCA (P<0.05) ,but no significant difference was observed in 1 day and 3 days after PTCA (P>0.05). Conclusion PTCA may lead to fluctuation of plasma levels of ET, TXB2and 6-keto-PGF1αThe clarification of changing regularity of these vasoactive substances contrib-utes to prevention of acute artery occlusion or restenosis after PTCA.

Based on amino acid score of food, this article presents an "Amino Acid Score Difference (AASD) " for protein quality evaluation, the minus value of AASD representing the limiting amino acid and its number re- presenting the degree. The minus value in one food can be compensated by the positive value in another when we directly select the rational couple for amino acid complementation in diet. In addition, a calculation formula is introduced for amino acid complementation in two kinds of food or in mixed diet including many foods. It gives an effective means in evaluating the nutritional value of protein in foods and planning an ideal diet by selecting foods on the basis of amino acid complementation
4.cDNA cloning and sequencing of MH2 domain of Smad1 from human dental papilla cells

Objective:To clone and sequence cDNA of MH2 domain of Smad1 gene from human dental papilla cells. Methods:Total RNA was isolated from primarily cultured human dental papilla cells and reversely transcribed into single stranded cDNA.The desired DNA product was obtained by PCR with two gene specific primers.The segment was inserted into PUC19 vector and the plasmid was transformed into E.coli JM109.The double stranded cDNA of positive clone was sequenced.Results:The sequence of MHz domain of Smad1 cloned from human dental papilla cells was consistent with that reported by Hoodless et al. Conclusion:Smad1 exists in human dental papilla cells,BMP signaling may be mediated by smad1 in human dental papilla cells.

Objective: To investigate the antitumor effects of arsenic trioxide (As 2 O 3 ) on solid tumor in vivo , the transplanted hepatic carcinoma of mice were treated with various dosages of As 2 O 3 . Methods: After 50 mice were injected intraperitoneally with 0,2, 4, 6, 8 ?mol/g As 2 O 3 respectively for observation of acute toxicity, 1, 2, 3 ?mol/g were selected for this experiment. Eighty mice were transplanted with hepatic carcinoma cells (1.5?10 6 cells/each side) into both subaxillae. As 2 O 3 in 0, 1, 2 and 3 ?mol/g were injected into right subaxilla respectively once every day for 10 days. At the 30th day after transplanted hepatic carcinoma cell, mice were killed and the size of solid tumors was measured. The chronic cytotoxicity of As 2 O 3 was observed in these mice.Results: In right subaxillae tumors, which were injected directly with As 2 O 3 , were apparent smaller than that in the left ( P

Objective To optimize conditions for extraction of alkaloids from Evodia rutaecarpa by means of 732 cation exchange resin and performe its anti-breast cancer bioactivity.Methods The optimum processing route on extraction of alkaloids from Evodia rutaecarpa was investigated by means of 732 cation exchange resin.The performance of 732 cation exchange resin was compared with tranditonal process ( aqueous extraction-ethanol precipitation in extraction-purification process).The alkaloid reagent-potassium mercuric iodide test and MTT test were performed on the crudes.Results Compared with traditional purification process, it was much better to use 732 cation exchange resin approach for extraction of alkaloids from Evodia rutaecarp with high yield(16.81%) The best eluent should be saturated brine with excellent purification.No obvious correlations were found between the toxin of breast cancer cell and concentration of total alkaloids.When the concentration of total alkaloids was 50μmol/mL, cellular survival rate was 68%afterward 24 h.When the concentration of total alkaloids comes to 100 μmol/mL and 150 μmol/mL, cellular survival rates were slightly decreased by 66% and 60%.Conclusion 732 cation exchange resin performes much higher than traditional process in purification of total alkaloids extracted from Evodia rutaecarpa.Simultaneously, the extracted total alkaloids showe remarkable inhibition in breast cancer cell.
7.The Auditory Brainstem Response of Guinea Pigs Recorded at Different Electrode Positions

Objective To observe auditory brainstem response evoked at different electrode positions adopting electrode slices .Methods The ABRs of 12 normal guinea pigs (24 ears)were recorded by two methods using clicks as stimulation .Active electrode was positioned at the vertex while the ground electrode at the nose .The reference e‐lectrode was placed on bilateral pinnas or mastoids .Then observe waveforms ,response thresholds (RT) ,peak la‐tencies(PL) and inter-peak latencies (IPL)of ABR .Results No significant difference were observed in the RT ,Ⅰ ,Ⅱand Ⅲ PL ,Ⅰ - Ⅲ IPL of ABR between the two methods(P>0 .05) .When stimulation was decreasing and with the reference electrode slices placed at bilateral pinnas ,waveⅡpeak was stable and discernable of ABR record‐ings across twenty -four ears of guinea pigs .The amplitude decreased more slowly than any other waves ,and dis‐appeared at the last .While reference electrode slices were placed on bilateral mastoids ,the amplitude of wave Ⅲ de‐creased more slowly than any other waves except wave Ⅳ ,and disappeared at lost .Ⅱ - Ⅲ ,Ⅲ - Ⅳ IPL became shorter .These two positions both recorded amplitude higher and PL advanced for wave Ⅳ as the stimulation de‐creased by 20 to 40 dB nHL ,and Ⅲ - Ⅳ IPL became shorter .This phenomenon was more obvious when reference electrode was placed at mastoid .Conclusion Wave Ⅱwas more stable and discernible with reference electrode placed at the bilateral pinnas .Therefore wave Ⅱ recorded by this way can be a new method to record the ABR threshold of g uinea pig s .
8.CYTOGENETIC STUDIES ON HPV-18-IMMORTALIZED CELLSFROM ESOPHAGEAL EPITHELIA OF HUMAN EMBRYO

Objective To study the cytogenetic changes in HPV-immortalized cells(SHEE)for further investigating the esophageal carcinogenesis. Methods Giemsa staining was used to show the modal number of chromosomes.G-banding was done to observe the chromosomal aberrations.Fluorescence in situ hybridization(FISH)of the interphase nuclei with the centromere probes of chromosome 1,7 and 8 were performed to determine the numerical abnormalities. Results The modal numbers at passage 10 and 20 were 58-63 and 57-64, respectively.Passage 61 had a bimodal distribution 58-61 and 63-65.The structural aberrations could almost be found in all of karyotypes.The following structural aberrations were identified:1.del(1)(q12);2.del(1)(p32);3.der(4),t(4;?)(q31;?);4.der(5),t(5:?)(q31;?).FISH results showed that trisomy 1 and tetrasomy 7 markedly increased with the increase of passages and disomy 8 kept in faint change in all passages.Conclusion The modal number moved right slowly in the graph of the modal distribution and the increases of chromosomal numbers were in imbalance with increases of passages of SHEE.The structural aberrations of chromosome were found in most of karyotypes.Our results suggest that the cell immortalization is a multistep process involved in the multiple chromosomes.;

Objective:To conduct clinical psychological intervention for the perioperative patients through solu-tion focused approach, excavate potential and improve the adaptation ability. Methods: On the basis of establis-hing a good nurse-patient relationship, the application of the solution focused approach was conducted in the peri-operative patients ( description of the problem, the construction of specific aims, and discussion on the exception, giving feedback, and evaluation of progress) . Results:After the intervention of solution focused approach, the pa-tients′negative emotion such as nervousness, anxiety, fear and depression decreased significantly (P <0. 05). Conclusion:Psychological intervention could effectively eliminate the negative mood of patients, improve the doc-tor-patient relationship, dissolve the contradiction between doctors and patients, reduce the incidence of medical disputes, and thereby enhance mutual understanding, trust and respect between doctors and patients, which makes the doctor-patient relationship more harmonious.
10.Survey on age of menarche in 56 924 women recruited from Pudong district of Shanghai

Objective To survey age of menarehe in Pudong district in Shanghai. Methods Data in this study were derived from 56 924 women at age of 20 -81 years in screening for cervical cancer between January 2007 and July 2008 in Pudong district. The age of menarche were recorded in a questionnaire. To investigate the trends in age at menarehe in different socioeconomic status, the subjects were divided into 12 groups in 5-year birth cohorts. The mean menarche age in each group was analyzed by analysis of variance(ANOVA). The percentage of menarche age at 10- 12 years and more than 18 years was analyzed by χ2 method. Results (1 ) The minimum age of menarcbe recorded is 10 years old, and the maximum is 28 years old, with average age of menarche at 15.7 years. In all groups, the smallest average age of menarcbe is 14. 6 years in 26 - 30 years old age group, while the biggest average age of menarche age is 16. 5 years in > 75 years old group; The difference showed statistical significance (P < 0. 01 ). (2) The percentages for participants with early menarehe age (10 - 12 years old) or late menarehe age (> 18-year-old menarche) were 1.82% (1034/56 924 ) and 5.20 % (2959/56 924 ) respectively. However, the maximum percentage for early menarche was recorded in 31 -35 years old group (4. 45% ,197/4431 ), only 0. 84% (10/1191 ) of participants in >75 years old group was classified as early menarebe. Meanwhile, the lowest percentage for late menarehe was 0. 38% (17/4431 ) in 31 - 35 years old group, and the highest percentage was 14. 70% (91/619) in > 75 years old group. The changes in the percentages for early menarche or late menarche are significantly associated with age differences (P < 0. 01 ). Conclusion The study suggested that the average of onset age of menarche in Pudong district has declined over the past decades in an age-based way, accompanied with the increase of the percentage for early menarche and the decrease of percentage for late menarehe.
