1.Correlation Between PCP in HIV/AIDS and CD_4 ~+T Lymphocyte Level

OBJECTIVE To explore the correlation between Pneumocystic carinii pneumonia(PCP) in HIV/AIDS and CD4+T-lymphocyte level.METHODS Totally 300 cases from more than 10 000 HIV/AIDS patients who were taken chest X-ray were sampled randomly.According to count of CD4+T-lymphocyte number,300 cases were divided into 2 groups: the count of CD4+T-lymphocyte number over than 200/mm3 was group A,while the other was group B.Analysis and comparison of lung complications in HIV/AIDS and follow-up survey of PCP curative effect in these 2 groups were done.RESULTS PTB,PCP,bacterial pneumonia Kaposi sarcoma(KS) and other lung ailmentd were the most common lung complications in HIV/AIDS.There were more lung complications and worse effect of PCP therapy in group B.There was significantly difference in group A and group B(P

Objective To investigate the role of anti apoptosis gene bcl 2 in the survival of cultured uveal melanoma UM cells. Methods Antisense oligodeoxynucleotides (AS ODN) bcl 2 were delivered with cationic lipid to primary cultured UM cells. The inhibitory effect of AS ODN bcl 2 on proliferation of UM cells was examined by 3, 4,5 Dimethyliazol 2,5 diphenyl tetrazolium bromide (MTT) method. Using DNA ladder to determine if the UM cells had been apoptotic. Bcl 2 expression was detected by RT PCR and Westernblot technics. Results The effect of AS ODN bcl 2 in inhibiting the proliferation of cultured UM cells had opposite relation to dosage. It down regulated the mRNA and protein level of bcl 2 gene, and the sense ODN didn′t have this effect. Conclusion AS ODN bcl 2 can down regulate bcl 2 expression, inhibits UM cells proliferation and induces apoptosis.
3.A missense mutation outside the large pocket of the retinoblastoma protein

AIM: To understand the effect of the RB1 gene mutation on the function of pRB (the protein product of the RB1 gene) in the patients with retinoblastoma (RB). METHODS: The genomic DNA from retinoblastoma patients was extracted. After amplification, the promoter and all 27 exons were screened by SSCP-heteroduplex method. The mutation was cloned and identified by sequencing. The effect of the mutation product on the function of pRB was analyzed. RESULTS: One missense mutations of the exon 4 of the RB1 gene was identified in the genomic DNA from RB patients. This mutation was outside the large pocket of the pRB. No mutation of the RB1 gene was found in the genome DNA of the patient's parents. This is the fourth report that there was a genome mutation located outside the large pocket of pRB in the RB patients. CONCLUSION: The amino-terminus of the pRB may be essential for growth suppression.
4.Relationship of the gene of multidrug and drug resistance and the patients′ prognosis in uveal melanoma

Objective To probe the relationship between the patients′ prognosis and the gene of multidrugs and drug resistance in uveal melanoma. Methods The gene expression of cyclin D1, epithelial growth factor receptor (EGFR), non metastasis gene 23 (nm 23), P glucose protein (P gp), multidrug resistance relation protein (MRP) and lung resistance protein (LRP) expression in 96 cases of uveal melanoma were detected by depigment immunohistochemistry. The patients with complete anamnesis data were observed continuously, and the follow up results were classified. Results Among the 96 cases of uveal melanoma, the epithelioid cell type was in 21, the mixed cell type in 56, and the spindle cell type in 19; including 76 at intraocular stage and 20 at extraocular stage. As the level of metastasis suppress gene nm 23 expression decreased and the level of cyclin D1 and EGFR expression increased, the expression level of drug resistance genes increased. The levels of LRP and MRP had negative correlation to the expressions of nm 23 and postive correlation to the expressions of nm 23, Cyclin D1 and EGFR. In 58 patients′ who were observed continuously, 19 died in 5 years and 26 survived over 5 years. Conclusion There are significant association between patients′ prognosis and multidrug and drug resistance gene in uveal melanoma.
5.Research on the relationship between pathological features of the uveal melanoma and prognosis.

OBJECTIVETo study the pathological features of uveal melanoma and to evaluate their influence on patients' prognosis.
METHODSParaffin embedded uveal melanoma tissues of 115 cases were examined using routine pathologic methods. Three histological types were classified according to the modified Callender system and patients were followed clinically. The data were done regression and survival analysis by SPSS statistic soft.
RESULTSThe patient with epithelial cell type, mixed type, and spindle cell type uveal melanoma have different life times, the average life time is 35.6 +/- 21.5 months, 63.7 +/- 37.0 months, 69.5 +/- 36.5 months in turn, patients with epithelial uveal melanoma had shorter survival time than other two types. The survival time was negatively related to the largest diameter of contact area with the sclera, the largest height and the depth of tumor invasion to the sclera.
CONCLUSIONSEpithelial uveal melanoma is more malignant than the other two types. Histological classification of this tumor combined with other pathologic features can indicate the patient's prognosis.

6.Expression and diagnostic value of 34βE12, Galectin-3 and HBME-1 in thyroid nodules

[ABSTRACT]OBJECTIVETo study the expressions of 34βE12, Galectin-3 and HBME-1 in thyroid nodules, and to explore its diagnostic value for papillary thyroid carcinoma (PTC).METHODSEn VisionTM immunohistochemical technique was used to detect the expression of 34βE12, Galectin-3 and HBME-1 in 352 thyroid lesions. The correlation between the expressions of the 3 protein markers and clinicopathological characteristics was evaluated. The receiver operating characteristic area under the curve (ROC-AUC) and their index for diagnosis evaluation were also calculated.RESULTSThe positive rates of 34βE12, Galectin-3 and HBME-1 in 246 PTC lesions were significantly higher than those in benign nodules (P<0.001). There was no relationship between the expression of the 3 protein markers and clinicopathological characteristics (eg. gender, age, numbers of lesions, tumor size, capsular invasion, lymph node metastasis, TNM staging). The ROC-AUC of 34βE12, Galectin-3 and HBME-1 for diagnosis of PTC was 0.936, 0.915 and 0.898 respectively. The sensitivity of 34βE12, Galectin-3 and HBME-1 for diagnosis of PTC was 94.3%, 95.5% and 91.1% respectively, while the specificity was 81.1%, 71.7% and 83.0% respectively, and the diagnostic accuracy rate was 90.3%, 88.4% and 88.6% respectively.CONCLUSION The expressions of 34βE12, Galectin-3 and HBME-1 are statistically different between PTC and benign lesions, but no associations are found with clinicopathological characteristics, indicating the three protein markers have important diagnostic value for PTC.
7.Optimization of the Extraction Technology of Senecio scandens by Orthogonal Design Combined with Informa- tion Entropy Weighting Method

OBJECTIVE:To optimize the extraction technology of Senecio scandens ,and to provide reference for the further development and utilization of the medicinal material. METHODS :The method of reflux extraction with 80 ℃ water bath was used to extract S. scandens . HPLC method was adopted to determine the contents of chlorogenic acid and hyperoside. The determination was performed on Diamonsil C 18 with mobile phase consisted of 0.2% glacial acetic acid water solution-methanol (82∶18,V/V)at the flow rate of 1.0 mL/min;the column temperature was set at 35 ℃;the detection wavelength was 327 nm,and the sample size was 5 μL. UV-Vis spectrophotometry was used to determine the contents of total flavonoids(by rutin )and total alkaloids (by matrine)and the detection wavelengths were set at 509 nm and 208 nm,respectively. Based on single factor tests ,using ethanol volume fraction (A,%),solvent folds (B,mL/g),extraction time (C,h),extraction times (D,times)as factors ,using the contents of chlorogenic acid ,hyperoside,total flavonoids and total alkaloids as indexes ,and t he weight coefficients of above indicator components were calculated based on information entropy weighting method so as to calculate their comprehensive scores , then L 9(34)orthogonal design was used to further optimize the extraction technology. The validation tests were also performed. RESULTS:The optimal extraction technology of S. scandens included that 8-fold 50% ethanol,extracting for 3 times,lasting for 1.5 h each time. Results of 3 times of validation tests showed that RSDs of the contents of chlorogenic acid ,hyperoside,total flavonoids and total alkaloids were all lower than 1.5%(n=3). CONCLUSIONS :The optimized technology is reproducible , stable and feasible ,and can be used for the extraction of S. scandens .
8.The improvement effect and metabolic mechanism of Echinops sphaerocephalus extract on osteoporosis model rat

OBJECTIVE To study the intervention effect and metabolic mechanism of Mongolian medicine Echinops sphaerocephalus extract on D-galactose-induced osteoporosis. METHODS Thirty-six 12-week-old male Wistar rats were selected and randomly divided into blank group, model group, Gushukang group, E. sphaerocephalus high-dose, medium-dose and low- dose groups, with 6 rats in each group. Except for blank group, other groups were intraperitoneally injected with D-galactose at 120 mg/kg per day. After 8 weeks of continuous injection, E. sphaerocephalus high-dose, medium-dose and low-dose groups were given drugs intragastrically at dose of 878, 439, 219.5 mg/kg, respectively. Gushukang group was given Gushukang 105.1 mg/kg intragastrically, once a day, for consecutive 8 weeks. After last administration, blood was collected from the abdominal aorta. Enzyme-linked immunosorbent assay was used to measure the contents of bone metabolism indexes [hydroxyproline (HYP), alkaline phosphatase (ALP)] and oxidative stress indexes [total antioxidant capacity (TAOC), superoxide dismutase (SOD), malondialdehyde (MDA)] in serum of rats. Positron emission tomography/computedtomography (PET/CT) was used to analyze the changes of bone microstructure in right tibia bone. Meanwhile, metabolomic technology was used to study the regulation effect of E. sphaerocephalus on osteoporosis model rats. RESULTS Compared with blank group, HYP, ALP, MDA, ratio of bone surface to bone volume (BS/BV), and trabecular separation (Tb·Sp) in model group were significantly increased (P<0.05), while TAOC, SOD, bone mineral density (BMD), bone volume fraction (BVF), trabecular E-mail:Xpfdc153@163.com thickness (Tb·Th) and trabecular number (Tb·N) were significantly decreased (P<0.05). Compared with model group, above indexes of administration groups were all reversed to different extents. The results of metabonomics study showed that after intervened with the extract of E. sphaerocephalus, 18 metabolites such as arachidonic acid, phenylalanine, tyrosine, tryptophan, isoleucine and uric acid in the serum of rats changed significantly, involving 15 metabolic pathways such as arachidonic acid, phenylalanine and tyrosine, of which arachidonic acid metabolism, phenylalanine metabolism and tyrosine metabolism were the main influencing pathways. CONCLUSIONS E. sphaerocephalus extract can effectively improve D-galactose-induced oxidative stress and the deterioration of bone microstructure, which interferes with metabolic pathways such as arachidonic acid metabolism and amino acid metabolism.
9.Analysis of Ongoing Change Characteristics of the Contents of Syringin and Total Flavonoids in Different Medicinal Parts of Toricellia angulata from Guizhou

OBJECTIVE:To study ongoing change characteristics of the contents of syringin and total flavonoids in different medicinal parts (root bark ,tree bark ,leaf)of Toricellia angulata from Guizhou ,and to provide reference for the development and application of T. angulata . METHODS :The root bark ,tree bark and leaf parts of T. angulata during different harvesting periods (Jan.-Dec.) were taken as the research samples. The content of syringin was determined by HPLC. The determination was performed on Agela Promosil C 18 column with mobile phase consisted of 0.5% phosphoric acid solution-acetonitrile (gradient elution)at the flow rate of 1.0 mL/min. The detection wavelength was set as 210 nm,and column temperature was 35 ℃. The sample size was 5 μL. The content of total flavonoids was determined by UV-visible spectrophotometry under detection wavelength of 510 nm. RESULTS :The linear range of syringin and total flavonoids were 0.095 9-1.150 8 mg/mL(r=0.999 6)and 0.072 2- 1.083 0 mg/mL(r=0.999 9),respectively. RSDs of precision ,stability and repeatability tests were all less than 3%(n=6). The average recoveries were 101.74%(RSD=2.36% ,n=6)and 99.63%(RSD=2.19% ,n=6),respectively. During different harvesting periods ,the contents of syringin in root bark ,tree bark ,leaf of T. angulata collected on Aug. ,May and Sept. were the highest,and the contents of total flavonoids in samples collected on Feb. ,Dec. and Sept. were the highest. The contents of syringin in different medicinal parts of T. angulata were in descending order as follows as tree bark >root bark >leaf,and the content of syringin was commonly relatively high in tree bark part ;the content of total flavonoids in different medicinal parts of T. angulata were in descending order as follows as root bark >tree bark >leaf,and the contents of total flavonoids in three medicinal parts was generally low. The content of total flavonoids in root bark was the highest in Feb. of that year ,and the content of syringin in root bark at same month was second only to Aug. of that year ;the content of syringin in tree bark was the highest in May ,and the content of total flavonoids in tree bark at same month was second only to Oct. and Dec. of that year ;the contents of total flavonoids and syringin in leaf were the highest in Sept. of that year. CONCLUSIONS :It is suggested that Feb. is the best time for harvesting root bark ,May for tree bark and Sept. for leaf of T. angulata .

Necrotizing enterocolitis (NEC) is one of the acquired intestinal inflammatory disease in neonate, with a high mortality rate.The morbidity and mortality rate of NEC remain at high level although recognition for NEC has been improved in last decades.Understanding of this disease has been furthered in latest years.The research advance of NEC is reviewed in present which is focus on the diagnosis and medical treatment to provide new evidence for further study.
