1.The preliminary report of the device and clinical application of continu ous and constant pressure skin expansion perfusion apparatus(CCPSEPA)

Objective To search for a simple and rapid skin expansion. Methods CCPSEPA-A kind of mutual feedback type continuous normal saline perfusion system was assembled with the use of conventional transfusi on apparatus, sphygmomanometer, three-way stopcocks, plasma bag etc.14 cases ( 17 expanders) were carried out with such a device, controlled by patients themselves. Results The volume of expanders was 50~300 ml and the duration of expansion was 7~14 days. All the cases proved a wonderful effect that the expansion was finished by only one or two times of water filling of storage water bag, and without any complications.Conclusion CCPSEPA is a new skin expansion apparatus of new type and practical ,simple and low price .The general goal of safe,simple, rapid and useful was realized basically by the device,and worth popularizing.
2.Clinicopathological analysis of solid-pseudopapillary tumor of the pancreas

Purpose:To study clinical and pathological features of solid-pseudopapillary tumor(SPT) of the pancreas. Methods:The clinical data were reviewed in four patients with SPT. Immunohistochemistry for Vim, CK8, CK18, Chromogranin, Sy, AAT, NSE, S-100 was performed in all four cases using EnVision. Results:All four patients were female. The mean age was 30 years. The tumors were composed of papillary and solid structures. The uniform cells were strongly diffuse positive for Vim, and strongly positive for focal AAT in four cases. Three cases were positive for NSE. All four cases were negative for Sy , Chromogranin and S-100. One case was poorly positive for CK8 and CK18. Conclusions:SPT of the pancreas, preferentially in young women, is a tumor with low malignant potential. Immunohistochemistry is very important in the diagnosis and differential diagnosis of SPT of the pancreas..
3.The Clinical Observation on the Treatment of Chronic Pharyngitis with Point Application of Chinese Medicinal Bread

Objective To explore the clinical effects of treating chronic pharyngitis with point application of Chinese medicinal bread.. Methods Retrospective analysis was adopted in the study. All patients were divided into a point application group (212 cases)and a dissolved medicine group (174 cases). The point application group was treated with point application of Chinese medicinal bread on Tiantu point for successive 4 therapeutic courses (one therapeutic course constitutes 10 days), while the dissolved medicine group was taken pharyngitis dissolved medicines orally for successive 6 therapeutic courses (one course constitutes 6 days). Results The total therapeutic effect of the point application group and the dissolved medicine group was 99.06% and 93.68% respectively. The difference between the two groups has statistical significance. Conclusion Point application of Chinese medicinal bread on Tiantu point is effective in treating chronic pharyngitis.
4.Association of vitamin D deficiency with pediatric sepsis and septic shock: a systematic review

Objective:To evaluate the relationship of vitamin D deficiency(VDD) between sepsis and septic shock among critical ill children, and investigate the influence of VDD on etiology and prognosis of sepsis.Methods:The observational cohort studies, involving vitamin D and critically ill children, were searched in Medline, Web of Science, EBSCO host and China National Knowledge Infrastructure, Wanfang Data Knowledge Service Platform.The quality assessment was conducted on the basis of the Newcastle-Ottawa Quality Assessment Scale.Consistent definitions of sepsis and septic shock were applied and standardized data were obtained following the study design.We retrieved the clinical outcomes related to septic shock and serum 25-hydroxyvitamin D 3concentrations and made a meta-analysis for the relation between VDD and clinical outcomes at two cut-offs. Results:In the analysis of collected data including severe VDD as a risk factor for incidence of sepsis and septic shock, the pooled RRwas 1.71(95% CI1.11 to 2.63, P=0.01) and 2.05(95% CI1.35 to 3.10, P<0.001) respectively, which were all higher compared with VDD.And severe VDD was also associated with cv-SOFA score( RR0.68, 95% CI0.45 to 0.91, P<0.001). However, no significant differences in mortality or the duration of vasopressor was found at severe VDD or VDD cut-off levels among septic shock patients. Conclusion:Among pediatric critical illness, severe VDD is more likely related to the incidence for sepsis and septic shock compared with VDD.And comorbidity with severe VDD could increase the cv-SOFA score of septic children.
5.Effect of antisense oligonucleotides of telomerase on the growth of RCZ cells derived from renal cell carcinoma in culture

Objective To investigate the effect of antisense inhibitor of telomerase on growth of tumor cells.Methods:Phosphorothioate antisense oligonucleotides (PS-ASON) with sequence TAGGGTTAGACAA which can recognize the RNA template region of telomerase and PS-random primer with 13-nucleotides were incubated with RCZ cell line derived from renal cell carcinoma (RCC).The number of cells was counted from 1 to 43 days,doubling time was calculated and inhibiting rate was determined by MTT method.Results ASON group reduced the number of cells (P<0.01), lengthened doubling time (P<0.005),and elevated the inhibiting rate (P<0.01) compared with random primer control and empty control.The efficacy depended on the dose of ASON (P<0.05) and it was of sequence-selective specificity.Conclusion The ASON targeted to the RNA template has an inhibition effect on the growth of RCZ cell and may have potential significance in tumor therapy.
6.The effects of probiotics on the stabilization of blood glucose and the mechanism in children with severe infectious pneumonia

Objective:To investigate the effects of probiotics on the stabilization of blood glucose and its mechanism in children with severe infectious pneumonia.Methods:Eighty children with severe infectious pneumonia admitted to PICU were enrolled in the clinical trial prospectively.They were divided into two groups by random.Forty patients given probiotics were regarded as treatment group,the others not given probiotics were considered to be controlled group,in addition to the regular therapy.Blood glucose,plasma H2 S,D-lactate and acetic acid,propionic acid and butyric acid concentrations in stools were measured respectively at the beginning of the treatment and on day 8 after the treatment.At the same time the changes of blood glucose,fluctuation of them and prognosis were examined.Results:There was no significance in all the parameters of the two groups at the beginning of the treatment (P > 0.05).The levels of plasma H2S and acetic acid,butyric acid in the stools of the probiotics group was (24.6 ± 7.4) μmol/L,(32.8 ± 8.6) μmol/g,(15.2 ± 5.2) μmoL/g on the day 8 respectively;but the concentration of these parameters in the control group was (20.9 ± 7.4) μ mol/L,(28.6 ± 7.9) μmol/g,(12.9 ± 5.0) μmoL/g.Meanwhile the level of D-lactate in patients given probiotics (1.25 ± 0.39) μg/ ml was significantly lower than that the control group (1.45 ± 0.4) μg/ml.The blood glucose (6.1 ± 2.7) mmol/L was lower than than in the other group (7.4 ± 2.7) mmol/L on day 8.And a higher recov ery rate was found in probiotic group.Furthermore,blood glucose and H2S,acetic acid levels expressed negative correlation at 8 day;but positive correlations were found between blood glucose and D-lactate (P < 0.05).Conclusion:Application of probiotics to children with severe infectious pneumonia can moderate the structure of enterobacteria,reinforce the intestinal barrier and elevate the levels of plasma H2S and had positive effects on the stabilization of blood glucose and prognosis.
7.Chromosome aberration and micronucleus formation in human peripheral blood lymphocytes induced by 12C heavy ions

Objective To investigate the biological effects of 12C heavy ions with different LETs,and to construct the dose-effect curves of chromosomal aberrations and micronucleus.Methods Human peripheral blood lymphocytes were irradiated with 0.5 - 5.0 Gy of 60Co γ-rays,29 and 148 keV/μm 12C with a dose rate of 0.5 Gy/min,respectively.The colchicine method and cytokinesis-blocking method were applied to measure chromosome aberrations and micronuclei formation,respectively. Results The chromosomal aberrations induced by 29 and 148 keV/μm of 12C ion had a linear dose response,while the dose-response of micronucleus induction followed to a linear-quadratic model.Chromosome aberration and micronucleus frequency increased with the LET of 12C ions. However, the micronucleus frequency increased slowly when the radiation doses exceeded 3.0 Gy..Chromosomal aberrations induced by 148 keV/μm 12C ions increased significantly when the time of colchicine treatment was prolonged.Conclusions Chromosome aberration and micronucleus induced by 12C ions increased with the LET,and the micronucleus frequency increased with dose less than 3 Gy.The biological effects of 12C ions were stronger than those of low LET irradiation of γ-rays.
8.Differential expression profile in the progeny of normal human liver cells irradiated by 60Co γ-rays

Objective To investigate the differential expression profile in the progeny of human liver cells surviving from ionizing radiation.Methods Complemental DNA gene chip was used to measure the transcriptional profile in progeny of HL-7702 cells exposed to 0, 2, 4, and 6 Gy of 60Co γ-rays, and the differentially expressed genes HAVCR2 and RAN were further identified by real-time PCR.Results The transcription level of 262 genes, 2746 genes and 3406 genes changed in the progeny of survival cells at 2, 4 and 6 Gy, respectively.A total of 71 common differentially expressed genes were screened, most of which were associated with transduction, cell cycle regulation, cellular immunity, cytoskeleton and movement, cell replication and repair mechanism.Conclusions Ionizing radiation could induce the expression changes of many genes, which might reveal the molecular mechanisms of gene expression in radiation induced genomic instability.
9.Proteomics study of progeny of normal human liver cells irradiated by 60Coγ-rays

Objective To characterize the differential protein expression in the progeny of human liver ceils surviving from ionizing radiation by the proteomic analysis.Methods Two-dimensional electrophoresis gel coupled with mass spectrometry was used to explore the specific protein expression in the progeny of 7702 human liver cells surviving from ionizing radiation.Alterations in expression level of protein spots between the control and the progeny groups were statistically analyzed by ImageMaster 2D Platinum software and mass spectrometry was used to identify the protein spots with significantly altered expression-level.Results The progeny of irradiated ceils were derived from human liver cell line exposed to 0,2,4,6 Gy of 60Co γ-irradiatian.A total of 42 differentially expressed proteins between the control and the progeny of the irradiated cells groups were screened,of which 17 were identified by matrix assistant laser desorption ion-top off light-mass spectrometry(MALDI-TOF MS)analysis,including 4 up-regulated and 13 down-regnlated proteins.Conclusions The differentially expressed proteins profile could be significantly altered in the progeny of irradiated cells.The proteomics approach has the potential to detect the protein changes relevant to radiatian-induced genomic instability(RIGI).Further study of differentially expressed proteins would likely reveal the molecular mechanisms of gene expression in RIGI.
10.Radiological protection effect of vanillin derivative VND3207 against radiation-induced cytogenetic damage in mouse bone marrow cells

Objective To study the protection of vanillin derivative VND3207 on the cytogenetic damage of mouse bone marrow cell induced by ionizing radiation.Methods BALB/c mice were randomly divided into five groups:normal control group,2 Gy dose irradiation group,and three groups of 2 Gy irradiaiton with VND3207 protection at doses of 10,50 and 100 mg/kg,respectively.VND3207 was given by intragastric administration once a day for five days.Two hours after the last drug administration,the mice were irradiated with 2 Gy γ-rays.The changes of polychromatophilic erythroblasts micronuclei (MN),chromosome aberration (CA) and mitosis index (MI) of mouse bone marrow cells were observed at 24 and 48 h after irradiation.Results Under the protection of VND3207 at the dosages 10,50,100 mg/kg,the yields of poly-chromatophilic erythroblasts MN and CA of bone marrow cells were significantly decreased(t = 2.36-4.26,P < 0.05),and the marrow cells MI remained much higher level compared with the irradiated mice without drug protection (t = 2.58,2.01,P < 0.05).The radiological protection effect was drug dose-dependent,and the administration of VND3207 at the dosage of 100 mg/kg resulted in reduction by 50% and 65% in the yields of MN and CA,respectively.Conclusions VND3207 had a good protection effect of on γ-ray induced cytogentic damage of mouse bone marrow cells.
