1.Clinical Study on Intensive Needling Stimulation at External Laogong (EX-UE 8) for Post-stroke Hand Hypermyotonia

Objective To evaluate the clinical efficacy of intensive needling stimulation at external Laogong (EX-UE 8) in treating post-stroke hand hypermyotonia.Method Sixty patients with post-stroke hand hypermyotonia were randomized into a treatment group and a control group, 30 cases in each group. The treatment group was intervened by ordinary acupuncture plus strong stimulation at external Laogong (EX-UE 8), while the control group was by ordinary acupuncture. After 8-week treatment, the modified Ashworth Scale (MAS) was used to assess the muscular tension of the affected limbs, Fugl-Meyer Assessment (FMA) was for assessing motor function of the affected hand, and Activities of Daily Living (ADL) was used to evaluate the activities of daily living before and after the intervention.Result The MAS, FMA, and ADL scores were significantly changed after the intervention in both groups (P<0.05). The MAS, FMA, and ADL scores of the treatment group were significantly different from that of the control group (P<0.05).Conclusion Acupuncture at external Laogong (EX-UE 8) with intensive stimulation is an effective approach in treating post-stroke hand hypermyotonia.
2.Influence of Acupuncture on HPA Axis in a Rat Model of Chronic Stress-induced Depression

To investigate the neurobiological mechanism of depression pathogenesis and reveal the mechanism of acupuncture treatment of depression. Methods: Wistar rats were selected for subjects. A rat model of depression was made by individually housing with unpredicted chronic moderate stimuli. Changes in behavior and hypothalamus-pituitary-adrenocortical axis were examined in rat models of stress-induced depression. Meanwhile, the intervening effect of acupuncture was evaluated and the curative effects of different acupuncture methods compared. Results: CORT and ACTH contents of serum were significantly higher in the model and normal saline groups than in the control group (P<0.05), significantly lower in the hand acupuncture and electroacupuncture groups than in the model group (P<0.05) and significantly lower in the medication group than in the normal saline group (P<0.05). There were no significant differences between the hand acupuncture, electroacupuncture and medication groups. Conclusion: Acupuncture of Baihui(GV 20) and Taichong (LR 3) has a marked antidepressant effect. Its mechanism may be related to the regulation of HPA axis by acupuncture.
3.Study on the protection of Yufeng Capsule on activity of ATPase in brain tissue of rat with ischemia-reperfusion

Objective: To observe the effect of Yufeng Capsule (Radix Astragali, Radix Salviae Mitiorrhizae, Rhizoma Gastrodiae, etc) on the moisture content, activity of Na +,K + AT Pase and Ca 2+ ATPase in brain tissue of rat with ischemia reperfusion.Methods:the model with ischemia reperfusion was established by line embolism method, the moisture content and activity of ATPase in brain tissue of model rat were determined respectively at ischemia 2h reperfusion 1h and ischemia 2h reperfusion 24h.Results:Yufeng Capsule could significantly decrease moisture content of rats brain, increase the activity of Na +,K + ATPase and Ca 2+ ATPase.Conclusion:Yufeng Capsule could prevent and treat the cerebral edema caused by ischemia reperfusion, its mechanism may be related to improving cytomembrane ionic transport after ischemia reperfusion.
4.Effects of Jiaji Electroacupuncture on Cortical Somatosensory Evoked Potentials in Rats with Spinal Cord Injury

Objective To explore the effects of Jiaji electroacupuncture on the cortical somatosensory evoked potentials(CSEP) and promotion of the function recovery in rats with spinal cord injury(SCI).Methods The experiment was performed in the Medical Experimental Center of Heilongjiang University of Chinese Medicine from November 2008 to February 2009. ①Allen's injury model of T10 spinal cord was established in SD rats with strike force of 50 g·cm. Fifty SD rats were divided into sham operated group (group A), only SCI group (group B), MP treatment group (group C), MP treatment+6 hours after SCI electroacupuncture treatment group (group D) and MP treatment+2 weeks after SCI electroacupuncture treatment group (group E) by means of random number table, ten rats in each group. For groups C, D and E first administration of MP (30 mg/kg) was taken within half hour after SCI, followed by 23 successive administration of MP(5.4 mg/kg·h) for every each hour. Equal amount of normal saline was given group B as that for group B. For groups D and E, electroacupuncture treatment was began 6 hours and 2 weeks after SCI respectively. ② Acupuncture method: The filiform needles of 0.25 mm×25 mm were vertically inserted into the acupoints 5 mm deeply, which were located at 4 mm away from the bilateralis of the lower margin of T8 and T12spinous process. KWD-808Ⅱ Electroacupuncture instrument was adopted with dilatational wave of AC PULSE current. The current intensity was 2 mA and the frequency was 2/100 Hz. Needles were kept in muscles for 30 minutes once a day, until the 8th week. ③Observation index: BBB score was used once a week at 1st, 2nd, 4th, 6th, 8th weeks and CSEP was observed once a week at 2nd, 4th, 6th, 8th weeks after SCI. Results 50 SD rats were all involved in the result analysis. The BBB score and the latency period of P1 in CSEP in each group were normal, there was no significant difference among them(P>0.05); The BBB score was less and the latency period was longer in SCI groups than that in group A, the difference was significant(P<0.05); At 1st week after SCI: The score was less than 7 in SCI groups (P>0.05); At 2nd week: the score in groups C, D and E was more than that in group B (P<0.05),but there was no significant difference among groups C, D and E (P>0.05); There was no significant difference among the latency period of SCI groups (P>0.05); From 4th to 8th weeks the score was the most and the latency period was the shortest in group D among SCI groups (P<0.05); At 4th week: The score was more and the latency period was shorter in groups E and C than that in group B (P<0.05), but there was no significant difference between groups C and E (P>0.05); At 6th week: The score was more and the latency period was shorter in group E than that in groups C and B (P<0.05), the score was the fewest and the latency period was longest in group B among SCI groups (P<0.05); At 8th week: The score was more and the latency period was shorter in group E than that in groups C and B (P<0.05), but there was no significant difference among group C and B (P>0.05).Conclusion The Jiaji electroacupuncture has promotion of the function recovery in rats with SCI and the effects of early electroacupuncture intervention are better than the later stage.
5.Experiment research of the function of human umbilical cord blood mesenchymal stem cells in the regeneration of rat's sciatic nerve

Objective To evaluate the efectiveness of using mesenchymal stem cells(MSCs)derived from HUCB(human umbilical cord blood)to a tissue engineered bioartificial nerve on bridging a 1 0 mm sciatic nerve gap.Methods The cord blood mononuclear cells were isolated by lymphocyte separation medium,purified and expanded with MesencultTM medium and acidic environment to produce adherent layer(MSCs).Thirty SD female rats were randomly divided into three groups.Group A:Human amnion tubes were seeded by HUCBMSCs together with fibrin sealant.Group B:Human amnion tubes were seeded only with fibrin sealant.group C:autografts.9 weeks later,a series of examinations were performed which included morphological,sciatic nerve function index,weight of gastrocnemius,histological staining and immunostaining of S100.Results The HUCB-derived mononuclear cells,when seeded in specific medium,gave rise to adherent cells (MSCs).At 9 weeks after the operations,all the examinations of group A was better than group B(P<0.05).Conclusion HUCBMSCs can be isolated,purified, cultivated and expanded Mesencult~(TM) medium.HUCBMSCs can promote the nerve to regenerate in reparing the sciatic nerve gap.
6.Clinical Observation of Electroacupuncture at Jiaji (EX-B2) Points for Primary Trigeminal Neuralgia

Objective With ordinary acupuncture as the control, to observe the feasibility and effectiveness of electroacupuncture at Jiaji (EX-B2) points in treating primary trigeminal neuralgia.Method Forty patients with primary trigeminal neuralgia were randomized into a group of electroacupuncture at Jiaji points and an ordinary acupuncture group, 20 cases in each group. In the electroacupuncture group, Jiaji of C2and T1on the affected side were selected, while the points were selected by following the Acupuncture-moxibustion Therapeuticsin the ordinary acupuncture group. Carbamazepine (CBZ) was taken as the basic treatment for the two groups. A treatment course (3 weeks) was observed, and follow-up study was conducted every 4 weeks for a total of 12 weeks. The dose of CBZ, Visual Analogue Scale (VAS) and Short-form McGill Pain Questionnaire (SF-MPQ) scores at each time point were recorded. The changes of the index in the two groups at each time point were compared.ResultAt the end of the treatment (the third week), the dose of CBZ, VAS and SF-MPQ scores all declined in the two groups, and the between-group differences were statistically insignificant (P>0.05). The follow-up study in the 7th week showed that the data of the observed indexes all decreased in the two groups, while the electroacupuncture group presented a more significant efficacy, and the between-group differences were statistically significant (P<0.05). The follow-up studies in the 11th and 15th weeks showed that the data of the observed indexes continued to decline in the electroacupuncture group but began to increase in the ordinary acupuncture group, and the between-group differences were statistically significant (P<0.001).Conclusion Electroacupuncture at Jiaji points and ordinary acupuncture both can reduce the dose of CBZ for patients with primary trigeminal neuralgia, improve pain and other discomforts as well as the negative emotions, but electroacupuncture at Jiaji can produce a more significant long-term efficacy compared to the ordinary acupuncture.
7.Establishment and application of a TaqMan real-time PCR assay for the detection of encephalomyo-carditis virus

Objective To establish a TaqMan real-time PCR assay for the detection of encephalo-myocarditis virus ( EMCV) .Methods Based on the conservative region of 3D gene of EMCV published in GenBank , a pair of primers and one TaqMan probe were designed and synthesized .Then a TaqMan real-time PCR assay was set up and the reactive system was optimized .The sensitivity and specificity of the assay was evaluated respectively .The TaqMan real-time PCR assay was then carried out to detect 98 randomly selected swine serum samples and the results were compared with those by using ELISA .Results The Ct value of the templates had a good linear relationship with the log starting quantity , with a correlation coefficient of 0.995.The TaqMan real-time PCR assay was only specific for EMCV and its sensitivity was 100 times higher than that of the ordinary PCR .The coincidence rate between the established assay and the ELISA assay was 98.0%in the detection of 98 blood samples.Conclusion The TaqMan real-time PCR assay for the detec-tion of EMCV was successfully established with advantages of high sensitivity and good specificity .It could be used for detection of EMCV and quantitative analysis .
8.The role of PDK in regulating cytokine secretion by extracellular matrixes

Objective To investigate the regulatory effects of extracellular matrixes, including fi-bronectin(FN), and collagen Ⅰ (ColⅠ ) on the immunologic function of human airway smooth muscle cells (HASMCs) passively sensitized with asthmatic serum, and the role of phosphoinosifide 3-kinase (PI3K). Methods Primarily cultured HASMCs were inoculated on the blank plates or on the plates coated with difference matrix proteins, added 10% asthmatic serum to passively sensitized non-asthmatic HASMCs and 10% non-asthmatic serum treated HASMCs as control, cell pretreated with PI3K inhibitor LY294002 for 30 min. The expressions of RANTES, Eotaxin, TGF-β1 mRNA were observed by RT-PCR and RANTES, Eotaxin, TGF-β1 protein in the ceil culture supernatunts was detected by enzyme-linked immtmosorbent assay (ELISA). Results Compared with the control serum group, the expressions of RANTES, Eotaxin, TGF-β1 mRNA of HASMCs and those protein in HASMCs culture supematants were significantly increased in the asthmatic serum group and the control serum + FN group and the control serum + Col Ⅰ group ( P < 0.05 ). The expressions of RANTES, Eotaxin, TGF-β1 mRNA of HASMCs and protein in HASMCs culture superna-tants were significantly increased in the asthmatic serum + FN group and the asthmatic serum + Col Ⅰ group. 50 μmoL/L LY294002 could significantly inhibit the expressions of RANTES, Eotaxin, TGF-β1 mRNA of HASMCs and protein in HASMCs culture supematants(P < 0.05). Conclusion These results suggest ex-tracellular matrixe may regulate immunomodulatory function of HASMCs passively sensitized with asthmatic serum and PI3K signaling pathway may play an important role in the process.
9.The application of GeXP based multiplex RT-PCR assay for respiratory viral infection in children

Objective To detect twenty types/subtypes of respiratory viruses with the GeXP (genomelab genetic analysis system) based mRT-PCR (multiplex reverse transcription-polymerase chain reaction) assay,and discuss its value in pediatric respiratory viral infection.Methods A retrospective study was conducted on a total of 1 305 inpatient children (0-6 years old) admitted to the Department of Respiratory in Children's Hospital of Hebei Province from May 2014 to November 2014.The sputum samples were collected and the viral DNA or RNA was extracted.After examining the specificity and sensitivity,the GeXP-based mRT-PCR assay was performed to test 20 types/subtypes of respiratory viruses.The obtained results were evaluated in comparison with those of fluorescence real-time PCR and direct sequencing.Chisquare test was conducted by SPSS16.0.Results Among 1 305 hospitalized children,the positive rate of respiratory virus was 58.31% (761/1 305).The positive rate of respiratory syncytial virus (RSV,17.32%,226/1 305) was the highest followed by the parainfluenza virus 3 (PIV-3,16.02%,209/ 1 305),human rhinovirus (HRV,11.95%,156/1 305),adenovirus (ADV,5.06%,66/1 305),human bocavirus (HBoV,3.14%,41/1 305) and the parainfluenza virus 1 (PIV-1,2.07%,27/1 305).Multiple virus infection was found in 92 children (7.05%).The highest positive rate of respiratory virus was observed in 1-2 years old children (83.28%,249/299);the lowest rate was found in 5-6 years old children (37.68%,26/69).Conclusions The proposed GeXP-based mRT-PCR assay possessed the advantage of high throughput,sensitivity,specificity and rapidity in the detection of twenty types/subtypes of respiratory viruses.It provided a thorough investigation of viral pathogens involved in acute respiratory infection thus could satisfy the clinic requirement and provide epidemic data on the disease prevention.
10.Observation on the Efficacy of Acupuncture at Xinming Points plus Strong Reinforcing Manipulation in Treating Optic Atrophy

Objective To observe the clinical efficacy of acupuncture at Xinming points (Extra) plus strong rein-forcing mani- pulation in treating optic atrophy.Methods Forty (56 eyes) optic atrophy patients were randomly allocated to a treatment group of 20 cases (29 eyes) and a control group of 20 cases (27 eyes). The control group received conventional medications, and the treatment group received acupuncture at Xinming points (Extra) plus strong reinforcing manipulation in addition. The visual acuity, the mean visual sensitivity (MS) of visual field and the P100 wave latency of P-VEP were recorded in the two groups before and after the treatment. The clinical therapeutic effects were compared between the two groups.Results The visual acuity, the MS of visual field and the P100 wave latency of P-VEP were significantly changed after treatment in both two groups (P<0.05). After the treatment, there were statistically significant differences in the visual acuity, the MS of visual field and the P100 wave latency of P-VEP between the two groups (P<0.05). The total efficacy rate was 75.0% in the treatment group versus 35.0% in the control group, and the difference was statistically significant (P<0.05).Conclusions Acupuncture at Xinming points (Extra) plus strong reinforcing manipulation is an effective method for optic atrophy.
