1.The combination experimental therapy with antimicrobial agents in Vibrio vulnif icus infection in mouse

Objective We conducted an in vivo study with the mouse model of Vib rio vulnificus infection to evaluate the efficacies of combination therapy with antimicrobia l agents. Methods Vibrio vulnificus (6.0?10 8 cfu/ml)was injected intraperitoneally into the right abdominal cavity. One hour, 2 hour and 3 hour after inoculation, 7 antimicrobial agents were given alone or in combination intraperitoneally at human therapeutic dose level. The numbers of survial mouse and the supermicrostr ucture change of organs w ere observed. 7 antimicrobial agents were Imipenem, Chloramphenicol, Doxycycline Hydrochloride, Netilmicin, Cefoperazone, Piperacillin, Levofloxacin. Res ults Two hours after infection, the mouse survival rates of groups trea ted with Chloramphenicol, Levofloxacin, Netilmicin, Cefoperazone was 100%. Howev er, the survival rates in the mouse treated by Piperacillin was 60% and the surv ival rate of in the mouse treated with Imipenem or Doxycycline Hydrochloride was 20%. The survival rate in the mouse treated with Cefoperazone combined with Lev ofloxacin, Cefoperazone combined with Netilmicin, or Netilmicin combined with Do xycycline Hydrochloride, was 100%. The supermicrostructure injure of the organs in the mouse recovered. Conclusions These results indicate that Chloramphenicol, Netilmicin, Cefoperazone and Levofl oxacin alone had satisfactory efficacy in the treatment of experimental Vibrio v ulnificus infection in mouse. The combination therapies of Cefoperazone with Lev ofloxacin, Cefoperazone with Netilmicin, and Netilmicin with Doxycycline Hydroch loride are more advantageous than using antimicrobial agent alone.
2.Influences of Stress on Attentional Bias of Elite Athletes

Objective To demonstrate the influence of stress on attentional bias in athletes,in order to make guidance for athletes' attention training and control.Method Using dot-probe paradigm,we analyzed the attention behavior of 25 athletes either under stress or not when viewing different pictures of emotional face.The reaction time and the score of the attention bias were recorded and compared.Result The reaction time to negative face pictures under stress was significantly shorter than that to the positive and neutral ones.The value of the attention bias to negative pictures was significantly bigger than O.Conclusion Stress tends to induce attention bias of athletes to negative information.
3.In vitro growth characteristics of human Tenon capsule fibroblasts from patients with bleb scaring after antiglaucoma filtration surgery

Background Human Tenon fibroblasts (HTFs) are the main cell component in scar tissue after antiglaucomatous filtrating surgery.To study the in vitro growth and proliferation features will offer an approach to the preventing and treating of scarring following antiglaucomatous filtrating surgery.Objective The goal of present study was to investigate the growth characteristics of HTFs in vitro from patients with bleb scarring after antiglaucomatous filtrating surgery.Methods This study process was approved by Ethic Committee of Beijing Chaoyang Hospital,and informed consent was obtained from each subject prior to the initaion of the study.The specimens of scaring tissue were obtained during the secondary trabeculectomy from 8 eyes with bleb scaring after initial antiglaucomatous filtrating surgery with the tissue size of 4 mm×5 mm,and the normal Tenon capsule specimens were acquired during the strabismus surgery from 8 eyes in the same way.HTFs were primarily cultured and passaged by tissue explants adherent method and identified using immunoinfluorescence technique with vemintin antibody.Nest PCR was used to exclude the mycoplasma infection during the culturing process.The morphology,growth curve and population doubling time (PDT) of the fifth generation of cells were examined and calculated by luminescence method cell vitality method 0,4,7,11,14 days and compared between the patients and normal groups.The stability of the cell growth was assessed by comparing the morphology,growth curve and PDT between the fifth generation and fifteenth generation of HTFs.Results Primarily cultured cells reached confluence 1-2 weeks with the similar shape to HTFs.The growth properties of the cells were same between the scarring group and normal group and showed positive response for vemintin antibody.No react band for mycoplasma was detected.The PDT was (20.54±3.51) hours and (18.86±2.91) hours in the scarring group and normal group,respectively,showing insignificant difference between them (t=0.634,P=0.561).No significant differences were found in the number of cells in 4,7,11 and 14 days after passage (t =2.663,P =0.081; t =0.194,P =0.863 ; t =3.338,P =0.053 ; t =0.627,P =0.565),with a consistent growth curve with the lapse of time between the two groups.The HTFs from scarred Tenon capsule fibrosis were cultured and passaged until the fifteenth generation.The PDT was gained to be (20.54 ±3.51) hours in the fifth generation of cells and (22.84±4.15) hours in the fifteenth generation of cells,without significant difference between them (t =0.733,P =0.505).The number of cellsin 4,7,11 and 14 days was not significantly different between the fifteenth generation and the fifth generation of cells (t=1.528,P=0.235;t=0.269,P=0.786;t=1.954,P=0.139;t =0.730,P =0.506).In addition,a good and stable growth curve also was exhibited in the fifteenth generation of cells compared with the fifth generation of cells.Conclusions Bleb scar-derived HTFs present good and stable growth in vitro.This result demonstrates HTFs were target cells in antifibrosis study after antiglaucoma filtrating surgery.
4.Changes and clinical significane of plasma coagulation factor Ⅶ and antithrombin III in patients with acute myocardial infarction

Objective To observe changes of plasma coagulation factor Ⅶ(FVII) and antithrombin III( AT-Ⅲ) in patients with acute myacardial infarction(AMI) . Methods The blood samples of 40 patients with AMI and 20 healthy volunteers were obtained through vein at the moment of admission. The plasma activated factors( FVIIag,FVI- la,FVII: C and AT-Ⅲ) were detected with different methods. Results Compared with control group,the plasina levels of FVIIag, FVIIa and the FVII: C in patients with AMI were significantly higher( P < 0.05, P < 0. 01) and especially in FVIIa(P <0.01) ,the AT-Ⅲ activity decreased significantly. Conclusion FVIIa and AT-III play important role in thrombosis of AMI,and the changes of their activities could predict the formation of acute cardiovascular event.
5.Primary clear cell carcinoma of the cervix:report of five cases and review of the literature

Objective To review the survival outcomes in patients with endometrial stromal sarcoma(ESS)in Cancer Hospital,Peking Union Medical College,Chinese Academy of Medical Sciences,and to discuss prognostic factors and the role of post-operative adjuvant radiotherapy and chemotherapy.Methods Hospital records and pathology reports for 97 patients with ESS were reviewed.Among 97 patients,69 had low-grade ESS(LGESS),16 had high-grade ESS(HGESS)and 12 had unclear grade.The median age at diagnosis was 44.0 years.The median follow-up time was 62 months(5-277 months).Atypical vaginal bleeding(43%)and prolonged and increased menses(36%)were the main symptoms.Results Totally 2-year and 5-year cumulative survival rates were 93%and 84%,respectively.Cumulative survival curves were significantly different between LGESS and HGESS,and so did cumulative survival curves between stage Ⅰ-Ⅱ and stage Ⅲ-Ⅳ(P<0.05).Totally,34 patients(37%)had local or distant recurrence.The median time-to-recurrence(TTR)was 27 months.The recurrence rates of the patients with or without preserve of ovary were 89%and 24%,respectively(P=0.000).The local-control-rates of the patients who received or did not receive post-operative radiotherapy were 81%and 43%,respectively(P=0.011).Conclusions The prognosis of HGESS is obviously worse than that of LGESS.The risk of recurrence of patients with preserve of ovary was remarkably higher than that of patients without preserve of ovary.Postoperative radiotherapy could increase the local-control-rates.
6.Regulation Mechanism and Significance of TCMBu-Shen Gu-Jin Prescription on IL-1 and TNF-αof Knee Osteoarthritis

This study was aimed to observe the effect of traditional Chinese medicine (TCM)Bu-Shen Gu-Jin Prescription (BSGJP) on the expression and action mechanism of interleukin-1 (IL-1) and tumour necrosis factor-alpha (TNF-α) in knee osteoarthritis (KOA) rabbit model. A total of 76 8-month healthy male New Zealand white rabbits were randomly divided into the control group, model group, TCM BSGJP group and western medicine group, with 19 rabbits in each group. Except the control group, improved Hulth modeling method was used in the animal model establishment. After 8-week consecutive medication, the serum, part of the synovial membrane tissues on the anterior side of the knee joint, articular cartilage and subchondral bone of the medial condyle of the femur were collected. Enzyme-linked immunosorbent assay (ELISA) was used in the content determination of IL-1 and TNF-α. The results showed that the narrowing of the joint space of the western medicine group were in between the model group and the TCM BSGJP group with obvious osteophyte formation. The TCM BSGJP group had slight narrowing of the joint space, mild rough of the articular cartilage surface, and slight osteophyte formation. The contents of IL-1 and TNF-α in the model group were higher than than of the control group (P < 0.01). Both medication groups were lower than the model group (P < 0.01). There was no significant difference between the two medication groups. It was concluded that BSGJP effectively reduced the secretion of IL-1 and TNF-α, delayed the deterioration of cartilage, which provided scientific experimental evidences for the clinical applications.
7.Risk factors and outcomes of patients with acute renal injury after intra-coronarystent implantation

Objective To investigate the risk factors and outcomes of acute kidney injury (AKI) in patients after intra--coronary stent implantation.Methods A retrospective and case control study was done with data analysis in 325 patients who underwent intra-coronary stent implantation from January 2010 to March 2011.The patients were divided into two groups as per the criteria of AKI identified on the 7th day after implantation of stent.The variables to be studied included:(1) age,gender,hypertension,diabetes,cerebrovascular disease,left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate,hyperuricemia,proteinuria,emergency operation,hydration,and medication (ACEI/ARB,statins) before operation; (2) dose of contrast media,operation time,hypotension during intra-operative period; and (3) postoperative:hypotension.The variables were analyzed with the process of One-way ANOVA and multivariate Logistical regression analysis.Consequently,the independent risk factors of AKI in patients after intra-coronary stent implantation could be found.Further,the prognosis of AKI patients was analyzed.Results Of the 325 patients,51 (15.7%) developed AKI.Compared the normal group,hospital stay (P < 0.01 ) and in-hospital mortality (P < 0.05) increased significantly in the AKI group.Monofactorial analysis showed that age,pre-operative laboratory and clinical data including left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate, hyperuricemia, proteinuria, hydration and emergency operation, and intraoperative information such as operation time and hypotension,and postoperative hypotension in AKI patients group were significantly different in comparison with control group ( P < 0.05 ). Multivariate logistic regression analysis revealed that elderly age (OR =0.253),pre-operative proteinuria (OR =5.351 ),preoperative left ventricular insufficiency ( OR =8.704),eGFR ≤ 60 ml/ ( min · 1.73 m2 ) ( OR =6.677 ),prolonged operation time ( OR =1.017),intra-operative hypotension ( OR =25.245 ) were independent risk factors of AKI ( P < 0.05 ).Conclusions AKI is a common complication and associated with increase in mortality after intra-coronary stent implantation.Increase in age,pre-operative proteinuria,pre-operative left ventricular insufficiency,pre-operative low estimated glomerular filtration rate,prolonged operation time,intra-operative hypotension are the independently risk factors associated with AKI.
8.Relationship between orthostatic hypotension and antihypertensive therapy in the elderly hypertensive population

Objective To investigate the relationship between orthostatic hypotension (OH) and antihypertensive therapy in the elderly hypertensive population.Methods A total of 1174 elderly retired people (849 cases with hypertension and 325 cases without hypertension) in Guangzhou Military Region underwent health physical examination.All people were surveyed by questionnaires.The orthostatic blood pressure and heart rate were measured in supine position after resting for more than 5 minutes and then at 0 and 2 minutes after standing.Comparative analysis was made on the prevalence of OH and the relationship between OH and antihypertensive therapy.Results A significant difference in the prevalence of OH was observed between hypertensive patients and nonhypertensive patients [29.4% (250/849) vs.15.7% (51/325),x2 =23.32,P<0.01].There was no significant difference in the prevalence of orthostatic hypotension in patients with versus without antihypertensive therapy [30.5% (220/721) vs.23.4% (30/128),x2 =2.62,P>0.05].Conclusions Orthostatic hypotension is common observed in elderly hypertensive patients.Antihypertensive therapy may not increase the prevalence of OH.
9.Correlation between expression of resistin in adipose tissues and glycometabolism in rats with traumatic brain injury

Objective To investigate the expression pattern of resistin in subcutaneous adipose tissue and its effect on glycometabolism in rats with traumatic brain injury (TBI).Methods Fifty-six SD rats were randomly divided into TBI group (n =48) and control group (n =8) according to the random number table.mRNA and protein expressions of resistin in subcutaneous adipose tissue were detected with real-time PCR and Western-blot.Concentrations of serum insulin and serum fasting blood glucose were evaluated using the ELISA method and quantitative estimation of insulin sensitivity was performed.Indices measured and their correlations were statistically analyzed.Results Levels of the resistin,serum insulin and FBG were significantly higher in TBI group than in control group (P < 0.05).Quantitative estimation of insulin sensitivity lowered in TBI group compared to control group (P < 0.05).Single factor linear correlation analysis showed negative correlation between resistin expression and quantitative insulin sensitivity check index in TBI group (P < 0.05).Conclusions Resistin is shown to have significant expression in subcutaneous adipose tissue and involve in glycometabolism.Obviously,resistin may play a significant role in insulin resistance after TBI in rats.
10.Biological activity assays and cellular imaging of anti-human sperm protein 17 immunomagnetic nanoparticles

AIM: To prepare anti-Sperm protein 17 (Sp17) immunomagnetic nanoparticles (IMNPs), and make foundation for target diagnosis of ovarian cancer by magnetic resonance imaging. METHODS: The anti-human Sp17 IMNPs were prepared by grafting anti-Sp17 antibodies on the surface of chitosan-coated magnetic nanoparticles (MNPs) using the linker of EDC/NHS (1-ethyl-3-[3-dimethylaminopropyl]carbodiimide/N-hydroxysuccinimide). The morphology and properties of the nanoparticles were characterized by transmission electronic microscopy (TEM), the conjugation of the antibodies was evaluated by native-polyacrylamide gel electrophoresis, the immunologic activity of IMNPs was evaluated by enzyme linked immunosorbent assay (ELISA). A set of in vitro magnetic resonance imaging (MRI) experiments were performed after incubated the IMNPs with human Sp17 gene transfected ovarian cancer HO-8910 cells. RESULTS: We had successfully grafted the MNPs with anti-Sp17 antibody and the IMNPs kept good bioactivity. The MRI showed that the IMNPs were targeted successfully to the positive cells, and no obviously non-specific adsorption was observed. CONCLUSION: The anti-Sp17 IMNPs with good specificity can used for further study of ovarian cancer target therapy.
