1.The correlation between diabetes and pancreatic cancer and the imaging findings

Diabetes mellitus ( DM) is widely considered to be associated with pancreatic cancer ( PC) .Some studies have shown that DM has been recognized as a risk factor for PC and the early clinical manifestation of PC , however the exact relationship re-mains unclear .The conventional imaging techniques and molecular imaging techniques can evaluate the abnormal condition during the occurrence and development of PC and diabetes .This article will review the correlation between PC and DM and the potential molecular mechanism, such as hyperglycemia, insulin-like growth factors, oxidative stress, islet amyloid polypeptide and Kruppel sample factorⅡgene mutations, etc.In addition, the value of imaging diagnosis for PC and DM is also reviewed .
2.Glucocorticosteroid membrane implanted into perichoroidal space protects retina of rabbit endophthalmitis

Objective To evaluate the effect of implantation of chitosan membrane with triamcinolone(TA) into perichoroidal space combined with intraocular vancomycin injection on the protection of retina in rabbit endophthalmitis.Methods The drug membrane was made of gelatin-chitosan.The right eyes of 30 normal pigment rabbits were inoculated intravitreally with 0.1 ml mixture of 105 CFU standard strain Staphylococcus aureus(ATCC 25923).Twenty-four hours later,rabbits were equally randomized to intravitreal vancomycin group(Group A),intravitreal vancomycin and TA eye dropping(Group B),and intravitreal vancomycin combined with TA membrane implantation(Group C).Clinical observation of ocular fundus was done every day with indirect ophthalmofundoscope.B ultrasound examination was carried out in 24 h after inoculation and on the 14th day after therapy.Electroretinogram(ERG) was done before therapy and the 14th day after therapy.Histopathologic grading and electron microscopy were carried out at the 14th day.Results After implanting the drug membrane into perichoroidal space,the inflammation in group C was markedly less than the other 2 groups,and its inflammation score was significantly lowest(P
3.Observation on ultrastructure of blood-brain barrier around hematoma in patients with hypertensive cerebral hemorrhage

Objective To observe the ultrastructure of blood-brain barrier (BBB) around hematoma in patients with hypertensive intracerebral hemorrhage (ICH).Methods Brain tissue around hematoma in 10 patients with hypertensive ICH was obtained using stereotaxic apparatus targeted by CT scan. Ultrastructure of BBB was observed under electron microscope. Results Astrocytic end feet swelled and pinocytotic vesicles increased 12 h after ICH. Mitochondrion of endothelial cells swelled and ridges disrupted 24 h after ICH. Astrocytic end feet and endothelial cells showed apparent edema, cell nuclei tumefied 48 h after ICH. Cytoplasm loosened and fundus membranes ruptured 72 h after ICH. Endothelial cell membranes and nuclei dissolved and capillary vessels dented 5 d after ICH. Conclusion Prevention of BBB from damage is important in treating ICH at early stage.

Objective To analyze and summarize the clinical diagnosis and therapy features of chronic liver disease patients with limbs infection.Methods A retrospective analysis was performed on 29 chronic liver disease patients with serious limbs infection in our hospital.Results Chronic liver disease patients with limbs infection specially vibrio infection,had a high ratio of MODS,so early diagnosis,early therapy with antibiotics and early operation to expose and cut lesion has a good effect.Conclusions The chronic liver disease patients with limbs infection should early diagnosed,early treated by antibiotics,and early exposed the swelling limbs or cut lesion by surgery.
6.Significance of EGFR, Her-2 and VEGF expressions in cervical intraepithelial neoplasia and squamous cell carcinoma

Objective To investigate the expressions of EGFR,Her-2 and VEGF in cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcinoma,as well as its clinic value of carcinogenesis,progression and metastasis in ccrvical squamous cell carcinoma.Methods EGFR,Her-2 and VEGF were evaluated by immunohistochemistry in tissues from normal cervix,CIN and squamous cell carcinoma.Results The expressions of EGFR,Her-2 and VEGF were negative in normal cervix.The positive rate of EGFR,Her-2 and VEGF in CIN tissues were 35.0 % (14/40),37.5 % (15/40),42.5 % (17/40),while in cervical carcinoma tissues were 70.0 % (35/50),78.0 % (39/50),76.0 % (38/50) respectively,which had statistical significance.There was relationship between the expressions of EGFR,Her-2,VEGF proteins with pathological grade andlymph node metastasis of squamous cell carcinoma.The expressions of Her-2 and VEGF correlated with the clinical stage of quamous cell carcinoma.Conclusion The expressions of EGFR,Her-2 and VEGF may be correlated with the occurrence,development and metastasis of cervical carcinoma.It may be used as an important marker to evaluate the malignant degree of cervical carcinoma and it can be helpful in the treatment of cervical carcinoma.
7.In vitro growth characteristics of human Tenon capsule fibroblasts from patients with bleb scaring after antiglaucoma filtration surgery

Background Human Tenon fibroblasts (HTFs) are the main cell component in scar tissue after antiglaucomatous filtrating surgery.To study the in vitro growth and proliferation features will offer an approach to the preventing and treating of scarring following antiglaucomatous filtrating surgery.Objective The goal of present study was to investigate the growth characteristics of HTFs in vitro from patients with bleb scarring after antiglaucomatous filtrating surgery.Methods This study process was approved by Ethic Committee of Beijing Chaoyang Hospital,and informed consent was obtained from each subject prior to the initaion of the study.The specimens of scaring tissue were obtained during the secondary trabeculectomy from 8 eyes with bleb scaring after initial antiglaucomatous filtrating surgery with the tissue size of 4 mm×5 mm,and the normal Tenon capsule specimens were acquired during the strabismus surgery from 8 eyes in the same way.HTFs were primarily cultured and passaged by tissue explants adherent method and identified using immunoinfluorescence technique with vemintin antibody.Nest PCR was used to exclude the mycoplasma infection during the culturing process.The morphology,growth curve and population doubling time (PDT) of the fifth generation of cells were examined and calculated by luminescence method cell vitality method 0,4,7,11,14 days and compared between the patients and normal groups.The stability of the cell growth was assessed by comparing the morphology,growth curve and PDT between the fifth generation and fifteenth generation of HTFs.Results Primarily cultured cells reached confluence 1-2 weeks with the similar shape to HTFs.The growth properties of the cells were same between the scarring group and normal group and showed positive response for vemintin antibody.No react band for mycoplasma was detected.The PDT was (20.54±3.51) hours and (18.86±2.91) hours in the scarring group and normal group,respectively,showing insignificant difference between them (t=0.634,P=0.561).No significant differences were found in the number of cells in 4,7,11 and 14 days after passage (t =2.663,P =0.081; t =0.194,P =0.863 ; t =3.338,P =0.053 ; t =0.627,P =0.565),with a consistent growth curve with the lapse of time between the two groups.The HTFs from scarred Tenon capsule fibrosis were cultured and passaged until the fifteenth generation.The PDT was gained to be (20.54 ±3.51) hours in the fifth generation of cells and (22.84±4.15) hours in the fifteenth generation of cells,without significant difference between them (t =0.733,P =0.505).The number of cellsin 4,7,11 and 14 days was not significantly different between the fifteenth generation and the fifth generation of cells (t=1.528,P=0.235;t=0.269,P=0.786;t=1.954,P=0.139;t =0.730,P =0.506).In addition,a good and stable growth curve also was exhibited in the fifteenth generation of cells compared with the fifth generation of cells.Conclusions Bleb scar-derived HTFs present good and stable growth in vitro.This result demonstrates HTFs were target cells in antifibrosis study after antiglaucoma filtrating surgery.
8.Application Research of Lung Care Software of Dual-source CT in Solitary Pulmonary Nodules

Objective To evaluate the auxiliary diagnostic ability and applicability of the Lung Care software of dualsource CT in the study of solitary pulmonary nodule(SPN). Methods 40 cases with SPN proved by pathologically were examined by dual-source CT. R-MPR and VOI of Lung Care software three-dimension rebuilding image were obtained to analyze imaging features of SPN and compared with routine chest CT. 20 cases had regular visits during 12 months which were divided into three stages to measure the growing speed or doubling time. Results There were 28 cases of malignant nodules and 12 cases of benign nodules in 40 cases of SPN. The accuracy for malignancy of routine chest CT and LungCare software three-dimension rebuilding are 53.57% ,82.14% respectively. The accuracy for benign nodules is 41.67%,83.33% respectively. 9.Utility of ~(18)F-FDG PET/CT in the Evaluation of Solitary Pulmonary Nodule: A Comparative Study with LungCare Software of Dual Source CT

Objective To evaluate the auxiliary diagnostic ability and applicability of ~(18)F-FDG PET/CT and the LungCare soft-ware of dual source CT in solitary pulmonary nodule(SPN). Methods 28 patients with SPN proved by pathologically were examined by ~(18)F-FDG PET/CT and LungCare software of dual source CT. ~(18)F-FDG PET/CT and LungCare software of dual source CT data were analysed comparatively in terms of the diagnostic accuracy in distinguishing malignant and benign. All data were analyzed for significance by using the χ~2-test,the value of P<0.05 was accepted as significance. Results There were 15 malignant and 9 benign nodules. In evaluating SPN,the diagnostic sensitivity, specificity, accuracy, positive predictive value and negative predictive value were 80.0% ,88.8% ,83.3%,92.3% and 72.7% with ~(18)F-FDG PET/aT respectively,and 93.3% ,77.7% ,87.5%,87.5% and 87.5% with LungCare software, respectively. To evaluate SPN,LungCare software and ~(18)F-FDO PET/CT was not significant difference in accuracy(P>0.05) ,while there were significant differences in sensitivity , specificity , positive and negative predictive value (P< 0.05). The sensitivity and negative predictive values with~(18)F-FDG PET/aT were much better than that with LungCare software, but the specificity and positive predictive values were less than that with LungCare software. Conclusion ~(18)F-FDG PET/CT has certain auxiliary function in distinguishing benign SPN from malignant SPN,it combining with LungCare software , the accuracy of diagnosis can be improved.
10.Gene expression of matrix metalloprotease and their inhibitors in liver tissue of rats with Vibrio vulnificus sepsis

Objective To investigate gene expressions of matrix metalloprotease (MMP-2, MMP-9) and tissue inhibitor of metalloproteinase-2 (TIMP-2) and RECK ( reversion inducing cysteine-rich protein with Kazal motifs) in the liver tissue of rats with Vibrio vulnificus sepsis. Methods Vibrio vulnificus sepsis model was induced by injection of 1 mL/100 g Vibrio vulnificus (9 × 106 cfu/mL) at hind limbs in 50 Sprague-Dawley male rats of clean grade, each time 10 animals were sacrificed at 2, 6, 9, 12 and 16 h after injection and liver samples were taken. Ten rats served as control group. The ethological changes were observed. The total RNA was extracted from liver tissue and the gene expressions of MMP-2, MMP-9, TIMP-2 and RECK were evaluated by semi-quantitative RT-PCR. Results The rats manifested shortness of breath, fever and swelling in hind limbs at 4 h after injection of Vibrio vulnificus. These symptoms gradually deteriorated, and the rats presented cyanosis and convulsion at 12 h. The gene expressions of MMP-2 and MMP-9 were markedly up-regulated, while those of TIMP-2 and RECK were down-regulated. Conclusion The study suggests that the elevation of MMP-2 and MMP-9 and down-regulation of TIMP-2 and RECK is associated with the development of Vibrio vulnificus sepsis.
